Verfahren zur Herstellung von neuen 3-Acylamino-2,4,6-trijod-benzoesäuren und Verwendung derselben als Röntgenkontrastmittel
Gegenstand des vorliegenden Patents ist ein Verfahren zur Herstellung von neuen 3-Acylamino-2,4,6trijod-benzoesäuren der Formel

in welcher R1 Methylen oder Ethyliden, R2 Äthylen oder Isopropylen, R5 Methyl oder Äthyl, n die Zahlen 1 bis und mit 7 darstellen.
Die neuen Säuren der Formel I und ihre Salze mit organischen oder anorganischen Basen sind als Röntgenkontrastmittel bzw. als schattengebende Substanzen für Röntgenkontrastmittel geeignet. Sie können für bronchographische Zwecke, für die Ausscheidungsurographie, für Lymphographie, Salpingographie, Darstellung der Gallenwege usw. verwendet werden.
Das Verfahren zur Herstellung der neuen 3 Acylamino-2,4,6-trijod-benzoesäuren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

mit einem Säurehalogenid der Formel HalOC-Rl-(o-R2) ll-R3 III umsetzt.
Die Umsetzung erfolgt vorzugsweise in einem inerten Lösungsmittel, wie z. B. Benzol, Toluol, Xylol, Chlorbenzol, Dichlorbenzol usf.
Die Säurehalogenide der Formel III sind z. T. bekannt, insoweit sie unbekannt sind, wird ihre Herstellung im experimentellen Teil eingehend beschrieben.
Das vorliegende Patent bezieht sich auch auf die Verwendung von Säuren der Formel I und ihren Salzen mit organischen oder anorganischen Basen als Röntgenkontrastmittel.
Solche Röntgenkontrastmittel können in Form von Emulsionen, Suspensionen, Lösungen usf. vorliegen. Sie können auch viskositätserhöhende Substanzen, wie z. B. Carboxymethylcellulose, Carboxymethylstärke usf., enthalten.
Der Einfachheit und des besseren Verständnisses halber werden die Polyalkoxyfettsäuren wie folgt benannt (Beispiel):

3, 6, 9-Trioxa-decansäure-(l).
Beispiel I
132 g 2,4,6-Trijod-3-amino-benzoesäure werden in 2,5 1 abs. Toluol erhitzt und etwa 150 cm Toluol abdestilliert. Zu der kochenden Lösung gibt man 58,6 g 3, 6-Dioxa-heptansäure-(l)-chlorid, gelöst in 50 cms abs. Toluol. Nach Beendigung des Eintropfens wird noch eine Stunde unter Rühren zum Rückfluss erhitzt. Nach ungefähr einer Stunde ist die Salzsäureentwicklung beendet, und das Endprodukt be ginnt auszukristallisieren. Nach dem Stehen über Nacht wird das Ausgefallene abgesaugt, mit Ather gewaschen und bei 400 C im Vakuum getrocknet.
Man erhält 109,3 g rohe 2,4,6-Trijod-3-(3',6'-dixoa- heptanoyl-l'-amino)-benzoesäure, die bei 193-196 C schmilzt. Die neue Säure wird durch Umfällen aus verdünnter Natronlauge gereinigt. Das Reinprodukt schmilzt bei 196-198 C; es stellt eine farblose kristalline Substanz dar. Die Auswage beträgt 91,3 g, das sind 56% d. Th.
Löslichkeiten: Die neue Säure ist in kaltem Wasser unlöslich, in heissem sehr wenig löslich; in kaltem Äthanol und Aceton wenig, in der Hitze aber gut löslich; in Äther, Petroläther und Chloroform praktisch unlöslich. 100 cm3 Wasser von 200 C lösen etwa 0,2 g der Verbindung. 100 cm3 einer wässerigen 10% eigen Triäthylenglycollösung Iösen ungefähr 0,6 g der Verbindung.
Das als Ausgangsprodukt verwendete 3,6-Dioxaheptansäure-(l)-chlorid wird z. B. wie folgt hergestellt:
55 g 3,6-Dioxa-heptansäure-(l) und 170 cm3 dest. Thionylchlorid werden in 170 cm3 abs. Benzol 11/2 Stunden bei 500 C gerührt. Anschliessend engt man die Lösung im Vakuum bei ungefähr 500 C ein, gibt zu dem Rückstand zweimal je 170 cm3 abs.
Toluol und destilliert im Vakuum wieder ab. Der Rückstand stellt das 3, 6-Dioxa-heptansäure-(1 )-chio- rid dar. Dieses wird nicht weiter gereinigt, sondern zum Umsatz sofort weiter verwendet.
Beispiel 2
Erhitzt man 154 g 2,4,6-Trijod-3 -amino-benzoe- säure mit 90 g 3,6,9-Trioxa-decansäure-(1)-chlorid in 2,9 1 abs. Toluol zum Sieden und arbeitet dann wie in Beispiel 1 angegeben auf, so erhält man 172 g der bei 1441470 C schmelzenden rohen 2,4,6-Trijod- 3-(3',6',9'-trioxa- decanoyl- l'-amino)-benzoesäure.
Nach dem Umfällen aus verdünnter Natronlauge- Salzsäure erhält man 150 g eines reineren Produktes.
Durch Umknstallisation aus Methanol erhält man 130 g der bei 148-150 C schmelzenden Reinsubstanz. Die neue Säure löst sich in kaltem Wasser sehr wenig, in heissem wenig. In kaltem Äthanol und Aceton ist sie kalt mässig, heiss gut löslich. 100 cm3 Wasser von 200 C lösen ungefähr 1 g der Säure. 100 cm3 einer 10 % igen Triäthylenglycollösung lösen ungefähr 1,7 g der Verbindung.
Beispiel 3
154 g 2,4,6-Trijod-3-amino-benzoesäure werden in 2,9 1 abs. Toluol unter Rühren zum Sieden erhitzt.
Zwecks Entfernung von Feuchtigkeit werden etwa 250 cm3 Benzol abdestilliert. Dann gibt man zu der kochenden Lösung 110 g 3,6,9,12-Tetraoxa-tridecan- säure-(l)-chlorid in einer Lösung von 70 cm3 abs.
Toluol. Die Zugabe erfolgt innerhalb von 15-30 Minuten in kleinen Portionen. Das Ganze wird bis zum Abklingen der HCl-Entwicklung erhitzt und dann die erhaltene Lösung im Vakuum eingeengt.
Nach dem Umfällen aus verdünnter Natronlauge/HCl wird noch aus Aceton umkristallisiert. Man erhält 94 g der bei 102-1040 C schmelzenden 2,4,6-Trijod- 3-(3',6',9',12'-tetraoxa-tridecanolyl-1'-amino)-benzoesäure. Die neue Säure löst sich in kaltem Wasser wenig, in heissem Äthanol und Aceton sehr gut und in kaltem Chloroform leicht, in Äther und Petroläther ist sie unlöslich. 100 cm3 Wasser von 800 C lösen ungefähr 1,5 g der Säure. 100 cm3 einer 10% eigen Triäthylenglycollösung lösen etwa 2,4 g der Säure.
Beispiel 4
I. 2,5 g 2,4,6-Trijodid-3-(3',6',9'-trixoa-decanoyl- l'-amino)-benzoesäure werden in 300 cm3 abs. Toluol heiss gelöst, die Lösung filtriert und zum Filtrat 5 cm3 Diäthylamin gegeben. Nach dem Stehen über Nacht wird die ausgefallene Kristallmasse abgesaugt, mit Toluol und Äther gewaschen und im Exsiccator getrocknet. Man erhält 2,6 g des farblosen Salzes, das bei 1631660 C schmilzt. Das neue Salz ist in Wasser sehr leicht löslich, in kaltem Alkohol mässig und in heissem Alkohol gut löslich.
II. 3,4 g 2,4,6-Trijod-3-(3',6',9'-trioxa-decanoyl- l'-amino) benzoesäure werden in eine Lösung von 0,11 g Natrium in 10 cm3 abs. Äthanol gegeben. Man lässt einen Tag im Eisschrank stehen und saugt die abgeschiedenen Kristalle ab. Nach dem Waschen mit wenig eiskaltem Äthanol und Äther wird im Exsiccator getrocknet. Man erhält 2,4 g des bei 145 bis 1470 C schmelzenden Natriumsalzes. Das Salz ist in Wasser und Äthanol kalt gut löslich und in heissem Dioxan ziemlich gut löslich.
In gleicher Weise, wie in den Beispielen beschrieben, erhält man weiter die folgenden Säuren:
2, 4, 6-Trijod-3-(3',6',9', 11', 1 4'-pentaoxa pentadecanoyl- 1 )-amino-benzoesäure;
2,4,l6-Trijod-3-(3',6',9',11',14',17'-hexaoxa octadecanoyl-1')-amino-benzoesäure.
Die erfindungsgemässen Säuren werden vorteilhaft in Form ihrer Salze mit anorganischen und organischen Basen appliziert.
Als anorganische Basen kann man zur Salzbildung verwenden:
Natriumhydroxyd, Calciumhydroxyd,
Ammoniumhydroxyd usf.
Als organische Basen kann man zur Salzbildung verwenden:
Methylamin, Äthylamin, Propylamin, Butylamin,
Dimethylamin, Diäthylamin, Dipropylamin,
Dibutylamin, Äthanolamin, Diäthanolamin,
Triäthanolamin, Diäthylaminoäthanol,
Dimethylaminoäthanol, Dipropylaminoäthanol,
Morpholinoäthanol, Diäthylaminoacetamid,
Dimethylaminoacetamid, Piperidinoacetamid,
Pyrrolidinoacetamid, Morpholinoacetamid,
Piperazin, N-Methylpiperazin,
N,N'-Dimethylpiperazin, Piperazinoäthanol, Piperazin-N,N'-diäthanol, Glucamin,
N-Methylglucamin, N-Athylglucamin usf.
Ob die erfindungsgemässen Säuren in Form ihrer Salze oder in freier Form angewendet werden sollen, hängt vom Verwendungszweck ab.

Example I.
132 g of 2,4,6-triiodo-3-aminobenzoic acid are in 2.5 1 abs. Heated toluene and distilled off about 150 cm of toluene. 58.6 g of 3,6-dioxa-heptanoic acid (l) chloride, dissolved in 50 cms of abs, are added to the boiling solution. Toluene. 154 g of 2,4,6-triiodo-3-aminobenzoic acid in 2.9 l abs. Toluene heated to boiling with stirring.
To remove moisture, about 250 cm3 of benzene are distilled off. 110 g of 3,6,9,12-tetraoxa-tridecanoic acid (l) chloride in a solution of 70 cm3 abs are then added to the boiling solution.
Toluene. It is added in small portions within 15-30 minutes. The whole is heated until the evolution of HCl has subsided and the resulting solution is then concentrated in vacuo.
After reprecipitation from dilute sodium hydroxide solution / HCl, it is recrystallized from acetone. 94 g of 2,4,6-triiodo-3- (3 ', 6', 9 ', 12'-tetraoxa-tridecanolyl-1'-amino) -benzoic acid which melt at 102-1040 ° C. are obtained.
