Verfahren zur Herstellung von 10-Hydroxydecen-2-carbonsäure-1
Bekanntlich ist 1 0-Hydroxydecen-2-carbonsäure- 1 ein Bestandteil des Gelee Royal der Bienen, und es besitzt bestimmte therapeutische Eigenschaften.
Für die aus Gelee Royal isolierte 10-Hydroxydecen- 2-carbonsäure-1 wurde ein Schmelzpunkt von 54 bis 560 C angegeben. Die vorliegende Erfindung betrifft nun Verfahren zur Herstellung des cis-Isomeren vom Fp. 73,5 bis 74,5 und des trans-Isomeren dieser Carbonsäure vom Fp. 64 bis 650.
Erfindungsgemäss wird 10-Hydroxydecen-2-car- bonsäure-l hergestellt, indem man die Dreifachbindung der entsprechenden acetylenisch ungesättigten Säure, d. h. der 1 O-Hydroxydecin-2-carbonsäure- 1 katalytisch oder unkatalytisch partiell hydriert. Die partielle Hydrierung der Dreifachbindung durch katalytische Hydrierung mit einem Katalysator nach Lindlar, einem reduzierten Palladium-Blei-Katalysator, liefert das cis-Isomer, das heisst die cis-10 Hydroxydecen-2-carbonsäure-1, eine bislang in der Literatur nicht beschriebene Verbindung, während man bei der unkatalytischen Hydrierung, z. B. mit einem Alkalimetall in flüssigem Ammoniak, das trans-Isomer der 1 O-Hydroxydecen-2-carbonsäure- 1 erhält.
Zur Herstellung des Ausgangsmaterials wird zweckmässig ein Äther oder ein Acylderivat des 7 Chlor- oder 7-Bromheptanols mit einem Alkalijodid, vorzugsweise Natriumjodid, umgesetzt, worauf man das erhaltene Jodid oder gegebenenfalls das entsprechende Bromid mit Natriumacetylid behandelt, das gebildete Acetylenderivat nacheinander mit Magnesiumdiäthyl und fester Kohlensäure zur Reaktion bringt und den erhaltenen Äther oder das Acylderivat der 10-Hydroxydecin-2-carbonsäure-1 zur Abspaltung der Äther- oder Acylgruppe verseift. Anschliessend wird dann erfindungsgemäss katalytisch oder unkatalytisch partiell hydriert.
Die ebenfalls in der Literatur nicht beschriebene lO-Hydroxydecin-2-carbonsäure-1 kann also durch hydrolytische Spaltung eines Äther- oder Acylderivates, z. B. des Tetrahydropyranyläthers, hergestellt werden.
Äther oder Acylderivate der 10-Hydroxydecin-2carbonsäure-l können durch Behandlung der entsprechenden Acetylenderivate zunächst mit Magnesiumdiäthyl und anschliessend mit fester Kohlensäure hergestellt werden. Auf diese Weise kann das 9-2' Tetrahydropyranyloxynonin-1, ebenfalls eine bisher unbekannte Verbindung, in den Tetrahydropyranyl äther der 10-Hydroxydecin-2-carbonsäure-1 umgewandelt werden.
Ein Acetylenderivat, wie es weiter oben beschrieben wurde, kann durch Behandlung des entsprechenden Jodids oder Bromids mit Natriumacetylid, vorzugsweise in Dimethylformamid als Verdünnungsmittel, hergestellt werden. Beispielsweise kann der Tetrahydropyranyläther des 7-Jodheptanols, eine bisher unbekannte Verbindung, in das 9-2'-Tetrahydropyranyloxynonin-l überführt werden.
Ein Äther oder Acylderivat des 7-Jodheptanols kann aus der entsprechenden Chlor- oder Bromverbindung durch Umsetzung mit einem Alkalijodid, vorzugsweise Natriumjodid, gewonnen werden. Beispielsweise lässt sich der Tetrahydropyranyläther des 7 Chlorheptanols, eine bisher unbekannte Verbindung, in den Tetrahydropyranyläther des 7-Jodheptanols umwandeln.
7-Chlorheptanol ist z. B. von Coleman und Bywater in der Zeitschrift Journal of the American Chemical Society, 66, 1944, Seite 1821, beschrieben.
Die Stufen in der Synthese der cis- und trans-10 Hydroxydecen-2-carbonsäure- 1 sind in der beiliegenden Zeichnung angegeben.
Es ist klar, dass die verschiedenen Stufen dieser Synthese Abwandlungen gestatten. Beispielsweise kann die Tetrahydropyranylgruppe auf irgendeine Stufe nach der Umsetzung mit Natriumacetylid abgespalten und die endständige Hydroxylgruppe wieder gebildet werden.
Weiterhin können die oben bezeichneten Carbonsäuren, das heisst die Verbindungen V, VI, VII und VIII, in Form von Estern, Salzen oder Glycosiden vorliegen, aus denen die gewünschte 10-Hydroxy- decen-2-carbonsäure-1 auf chemischem oder enzymatischem Wege erhalten werden kann.
Therapeutische Zubereitungen können cis- 10- Hydroxydecen-2-carbonsäure- 1 und ein ungiftiges Verdünnungsmittel oder einen Nährstoff enthalten.
Herstellung von 7-Chlor-1-2'-tetrahydropyranyl oxyheptan (II)
Ein Gemisch von 38 g 7-Chlorheptanol (I) und 25 g 2,3-Dihydropyran wird auf 0 abgekühlt, mit 2 Tropfen konzentrierter Salzsäure versetzt und über Nacht stehengelassen. Das Produkt wird über Kaliumhydroxydplätzchen destilliert. Ausbeute an 7 Chlor-l -2'-tetrahydropyranyloxyheptan (II) 53 g, 91 % derTheorie, Kp. 92 bis 950/0,2 mm Hgn 2D5 =1,4605.
Herstellung von 7-Jod-1-2'-tetrahydropyranyl- oxyheptan (III)
13,5 g Chlortetrahydropyranyloxyheptan (II) (57,5 mMol), in 150 ml über Magnesiumsulfat getrocknetem Aceton gelöst, werden mit 18,0 g (120 mMol) Natriumjodid 5 Stunden unter Rückfluss gekocht und anschliessend vom ausgefällten Natriumchlorid (2,2 g) abfiltriert. Es werden nochmals 2 g Natriumjodid zur Lösung gegeben und das Gemisch weitere 7 Stunden unter Rückfluss gekocht. Hierauf wird erneut vom Natriumchlorid (1 g) abfiltriert. Ins gesamt werden 3,2 g Natriumchlorid, 95 so % der Theo- rie, isolierte. Das Filtrat wird mit einem grossen Volumen an Äther verdünnt, mit verdünnter Natriumthiosulfatlösung und mit Wasser gewaschen und über Magnesiumsulfat getrocknet.
Nach dem Abdestillieren des Lösungsmittels hinterbleiben 18 g des rohen Jodids, das unmittelbar in die nächste Stufe eingesetzt wird.
Herstellung von 9-2'-Tetrakydropyranyl- oxynonin-l (IV)
5,3 g Natrium (230 Milligramm-Atome) werden in 500 ml flüssigem Ammoniak in einem l-Liter Kolben unter gleichzeitiger Zugabe von trockenem Acetylen gelöst, bis die blaue Farbe verschwunden ist. Hierauf werden 18 g des vorstehend beschriebenen Jodids in 150 ml Dimethylformamid, das über Magnesiumsulfat getrocknet und destilliert wurde, langsam zugegeben. Man lässt das Ammoniak langsam verdampfen (ein zu rasches Verdampfen verursacht starkes Schäumen). Das Reaktionsgemisch wird anschliessend unter trockenem Stickstoff 4 Stunden auf 700 erwärmt und gerührt. Hierauf wird das Reaktionsgemisch mit Eis abgekühlt und dreimal mit Äther extrahiert.
Der Ätherextrakt wird dreimal mit eiskalter verdünnter Salzsäure, dann zweimal mit 2-n Natronlauge und schliesslich mit Wasser gewaschen, getrocknet und der Äther abdestilliert. Der Rückstand wird an einer kurzen Kolonne destilliert. Nach einem bei 68 bis 720/0,3 mm Hg überdestillierenden geringen Vorlauf geht das 9-2'-Tetrahydropyranyloxynonin-l als farbloses Öl bei 90 bis 910/0,30 mm Hg über. Ausbeute 8,25 g, 64 % der Theorie, bezogen auf das eingesetzte Chlorid; n D23 = 1,4618.
Heerstellung von 10-Hydroxydecin-2-carbon säure-1 (V)
Dioxan wird getrocknet, dann mit Äthylmagnesiumbromid in Äther versetzt, bis die starke Reaktion nachlässt. Hierauf wird die Suspension über eine Kolonne destilliert und die zwischen 99 und 1010 übergehende Fraktion aufgefangen. n 2r3 des gereinigten Dioxans 1,4206.
Äthylmagnesiumbromid wird in üblicher Weise aus 0,8 g Magnesium (33 mMol) und 5,25 g (48 mMol) Äthylbromid in trockenem Äther unter Stickstoff hergestellt und anschliessend durch Kochen unter Rückfluss für 5 Stunden mit besonders gereinigtem Dioxan (2,9 ml, 33 mMol) in Magnesiumdiäthyl überführt.
6,7 g (30 mMol) Tetrahydropyranyloxynonin (III), in 20 ml trockenem Äther gelöst, werden hierauf zum Magnesiumdiäthyl zugegeben und unter Rühren 1 Stunde unter Rückfluss gekocht. Hierauf wird das Gemisch auf feste Kohlensäure in Äther gegossen.
Das Reaktionsgemisch wird für 1 Stunde zeitweilig gerührt, dann mit gesättigter Ammoniumchloridlösung und einigen Tropfen verdünnter Salzsäure versetzt, das Gemisch geschüttelt und abtrennen gelassen. Die Ätherschicht wird zweimal mit 2n Natriumcarbonatlösung gewaschen und die vereinigten wässrigen Anteile angesäuert. Aus dem Ätherextrakt werden 2,6 g (39 %) reines unverändertes 9-2'-Tetrahydropyranyloxynonin-1 wiedergewonnen. Durch Extraktion der angesäuerten wässrigen Anteile mit Äther, Waschen des äther mit Wasser und anschliessendes Trocknen und Verdampfen des Äthers werden 4,6 g (56% der Theorie) rohes 10-2'-Tetrahydropyranyloxydecin-2-carbonsäure- 1 (V) als farbloses Öl erhalten.
Die Carbonsäure wird in einem Gemisch von 30 mI Methanol und 30 ml Wasser gelöst, das einige Kristalle p-Toluolsulfonsäure enthielt, und zur Abspaltung der Tetrahydropyranylgruppe 1 Stunde unter Rückfluss gekocht. Das Lösungsmittel wird anschlie ssend abdestilliert und der Rückstand aus Petroläther, Kp. 60 bis 800, bis zur Schmelzpunktkonstanz umkristallisiert. Ausbeute an 1 0-Hydroxydecin-2-carbon- säure-l (VI) 2,1 g, 38% der Theorie, Fp. 72,0 bis 73,00.
Herstellung von cis-10-Hydroxydecen-2- carbonsäure-1 (Vll)
1,52 g 10-Hydroxydecin-2-carbonsäure-1 (8,26 mMol) in 50 ml Äthylacetat werden mit 400 mg Lindlars Katalysator und einem Tropfen Chinolin so lange hydriert, bis 210 cms Wasserstoff bei 200 C und 741 ml Hg aufgenommen sind. Diese Menge entspricht 8,4 mMol Wasserstoff. Die Hydrierung wird abgebrochen, der Katalysator abfiltriert und das Lösungsmittel abdestilliert. Nach Umkristallisation aus wenig Petroläther, Kp. 60 bis 800, enthaltendem Ather erhält man 1,35 g (90% der Theorie) cis-10 Hydroxydecen-2-carbonsäure-1 vom Fp. 73,5 bis 74,50.

Production of 7-chloro-1-2'-tetrahydropyranyl oxyheptane (II)

Preparation of 9-2'-tetrakydropyranyl-oxynonine-1 (IV)
5.3 g of sodium (230 milligram atoms) are dissolved in 500 ml of liquid ammonia in a 1 liter flask with the simultaneous addition of dry acetylene until the blue color has disappeared. 18 g of the above-described iodide in 150 ml of dimethylformamide, which has been dried over magnesium sulfate and distilled, are then slowly added. Preparation of 10-hydroxydecyne-2-carboxylic acid-1 (V)
