Es ist bekannt, dass sich Indole mit Zink und Mineralsäure zu Indolinen reduzieren lassen. Es wurde nun erstmals gezeigt, dass sich auch Lyserg- säure- und Dihydrolysergsäure-Derivate mit nascie- rendem Wasserstoff in 2,3-Stellung reduzieren lassen.
Diese Tatsache ist deshalb überraschend, weil Lyserg- säure- und Dihydrolysergsäüre-Derivate gegenüber Mineralsäuren äusserst empfindlich sind und damit leicht zu Naphthalin Derivaten umgelagert werden können [A. Stoll und Th. Petrzilka, Helv. <B><I>36,</I></B> 1125 (1953)].
Es ist überdies. ein überraschender Befund, dass bei der Anwendung der Reduktionsmethode entspre chend dem vorliegenden Verfahren die Doppelbin dung in 9,10-Stellung nicht angegriffen wird.
Die erfindungsgemässe Reduktion der Lyserg- säure- und Dihydrolysergsäure-Derivate in 2,3-Stel- lung erfolgt, wie schon gesagt, mit nascierendem Wasserstoff, vorzugsweise mit Zink und Mineral säure.
Das erfindungsgemässe Verfahren wird beispiels weise ausgeführt, indem man einer Lösung eines Lysergsäure- oder Dihydrolysergsäure-Derivates der Formel 1I oder dessen Salz 10 bis 200 Gewichtsteile Zinkstaub suspendiert und hierauf unter Rühren Mi neralsäure in gasförmiger oder flüssiger Phase mit einer solchen Geschwindigkeit zugibt, dass der pH- Wert zwischen 1 und 2 liegt.
Als Lösungsmittel eignen sich dabei Wasser, wässrige Säuren, Alkohole, alkoholische Säuren und Gemische von Alkoholen mit Wasser. Für das Ge lingen der Reaktion ist es von Vorteil, bei einer Re aktionsdauer von 3 bis 8 Stunden und einer Reak tionstemperatur zwischen 10 und 50 , vorzugsweise zwischen 15 und 25 , zu arbeiten. Das Ende der Umsetzung ist mit dem Kellerschen Farbreagenz [Eisen-(111)-chlorid enthaltender Eisessig und konz. Schwefelsäure] durch Verschwinden der Farbreaktion erkennbar.
Man filtriert anschliessend vom nicht um gesetzten Zink ab, macht das Filtrat unter Kühlung alkalisch, z. B. mit Ammoniak, und schüttelt mit einem mit Wasser nicht mischbaren Lösungsmittel, wie z. B. Chloroform, aus. Die Reinigung der End produkte erfolgt durch Kristallisation oder chromato, graphische Adsorption.
Die gegebenenfalls durchzuführende katalytische Hydrierung der Lysergsäure Derivate in 9,10-Stel- lung erfolgt nach bekannten Verfahren mit Kataly satoren der B. Gruppe des periodischen Systems, wie z. B. Palladium, Rhodium, Platin oder Nickel. Als Lösungsmittel für die katalytische Hydrierung kön nen Alkohole, Eisessig, Essigsäureäthylester oder Gemische derselben verwendet werden.
Die erfindungsgemäss hergestellten neuen 2,3-Di- hydroverbindungen der Lysergsäure- und Dihydro- lysergsäure-Reihe sind teils ölige, teils kristalline Substanzen. Mit anorganischen oder organischen Säuren bilden sie beständige, kristallisierte zwei basische Salze. Die Kellersche und Van Urksche Farb- reaktionen sind negativ.
Mit dem Dragendorffschen Reagenz wird eine orange Färbung erhalten. Die er findungsgemäss hergestellten Verbindungen sind in Wasser wenig, in polaren organischen Lösungsmitteln im allgemeinen gut löslich, schwer löslich hingegen in Benzol und Petroläther.
Die in den nachfolgenden Beispielen beschrie benen Verbindungen zeichnen sich durch therapeu tisch verwertbare pharmakodynamische Eigenschaf ten aus, die sie zu wertvollen Medikamenten machen. So wirken sie beispielsweise sedativ auf motorische und psychische Funktionen und beeinflussen den Tonus der Gefässe. Ausserdem können die Ver fahrensprodukte als Zwischenprodukte für die Her stellung von Medikamenten verwendet werden.
Durch die erfindungsgemässe Reduktion in 2,3-Stellung und nachfolgende Dehydrierung gelingt es, die empfind liche Indol-Gruppierung in Lysergsäure- bzw. Di- hydrolysergsäure-Verbindungen vorübergehend auf- zuheben, wodurch Lysergsäure- bzw. Dihydrolyserg- säure-Verbindungen völlig neuen Umsetzungen zu gänglich werden, wie z. B. Oxydationen, säurekataly- sierten Reaktionen, Substitutionen im Benzol-Kern, u. a.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelz punkte sind urkorrigiert. <I>Beispiel 1</I> 1-Methyl-2, 3-dihydro-d-lysergsäure- D-propanolamid-(2') Eine Lösung von 5,0 g 1-Methyl-d-lysergsäure- D-propanolamid-(2') in 500 cm3 Alkohol wird mit 500g Zinkstaub versetzt. Bei einer Temperatur von 10 werden im Dunkeln und unter starkem Rühren total 1,1 1 konz. Salzsäure tropfenweise während 3 Stunden zugegeben.
Hierauf rührt man noch 11/" Stunden bei der gleichen Temperatur, filtriert vom überschüssigen Zink ab und stellt das Filtrat unter Kühlung mit Ammoniak alkalisch. Man schüttelt mit Methylenchlorid aus und kristallisiert das durch Verdampfen des Lösungsmittels gewonnene Rohpro dukt aus Alkohol/Essigsäureäthylester um. Farblose Nadeln vom Smp. 190-191'. Keller-Reaktion: nega tiv. Dihydrochlorid: farblose Nädelchen vom Smp. 213-216 .
<I>Beispiel 2</I> 1-Methyl-2,3,9,10-tetrahydro-d-lysergsäure- D-propanolamid-(2') Eine Lösung von 0,96 g 1-Methyl-2,3-dihydro-d- lysergsäure-D-propanolamid-(2') in 20 cm3 Eisessig wird mit 0,1 g vorhydriertem Platinoxyd und Wasser stoff geschüttelt. Nach Aufnahme von 70 cm Wasserstoff bricht man die Hydrierung ab, filtriert vom Katalysator ab, engt das Filtrat bis zur Sirup konsistenz ein und schüttelt den Rückstand zwischen Natriumbicarbonatlösung und Methylenchlorid aus.
Das durch Verdampfen des Lösungsmittels erhalten Rohprodukt chromatographiert man an 150 g Alu miniumoxyd, wobei das 1-Methyl-2,3,9,10-tetra- hydro-d-lysergsäure-D-propanolamid-(2') mit Chloro form und 1 % Methanol ins Filtrat gewaschen wird. Die Verbindung kristallisiert schlecht.
Bimaleinat: farblose, rechteckige Prismen aus Methanol/Aceton, Smp. 165-170 .
<I>Beispiel 3</I> 2,3-Dihydro-d-lysergsäure-D-propanolamid-(2') In einer Lösung von 5 g Ergobasin-bimaleinat in 200 cm3 Wasser suspendiert man 500 g Zinkstaub und lässt unter intensivem Rühren während 6 Stun den im Dunkeln und bei einer Temperatur von 15 bis 20 total 1,1 1 konz. Salzsäure zutropfen. Nach dieser Zeit ist die Kellersche Farbreaktion negativ.
Man filtriert vom überschüssigen Zinkstaub ab, neu tralisiert unter Kühlung durch Einleiten von gas förmigem Ammoniak, fügt noch einen überschuss an 25ö igem wässrigem Ammoniak hinzu und schüttelt mit Methylenchlorid aus. Das Rohprodukt wird aus Essigsäureäthylester/Alkohol umkristallisiert. Farb lose Nadeln vom Smp. 193-194 . Kellersche Farb- reaktion: negativ.
<I>Beispiel 4</I> 1-Methyl-2,3-dihydro-d-lysergsäure-(+)- butanolamid-(2') Man suspendiert 440 g Zinkstaub in einer Lö sung von 5 g 1-MethyT-d-lysergsäure-(+)-butanol- amid-(2') in 200 cm3 Wasser und tropft während 41/2 Stunden unter kräftigem Rühren und bei einer Temperatur von 15-20 und gleichzeitigem Durch leiten von Stickstoff 0,9 1 konz. Salzsäure hinzu. Nach dieser Zeit fällt die Kellersche Farbreaktion negativ aus. Die Aufarbeitung erfolgt analog Bei spiel 1.
1-Methyl-2,3 -dihydro-d-lysergsäure-(+)-butanol- amid-(2') wird aus Alkohol/Essigsäureäthylester um kristallisiert. Farblose Kristalle vom Smp. 200-202 . Kellersche Farbreaktion: negativ.
<I>Beispiel 5</I> d,1-1-Methyl-6-acetyl-2,3,9,10-tetrahydro- nor lysergsäure-methylester 3,26 g d,1-1-Methyl-6-acetyl-9,10-dihydro-nor- lysergsäure-(I)-methylester werden in 250 cm3 mit HCl-Gas gesättigtem Methanol abs. gelöst und auf 10 abgekühlt. Zu dieser Lösung gibt man portionen- weise unter gutem Rühren und Kühlen insgesamt 25 g Zinkpulver, wobei die Reaktionstemperatur innerhalb 10-15 gehalten wird.
Nach 2 Stunden Reaktionszeit werden nochmals 25g Zinkpulver zu gegeben und weitere 50 cm3 mit HCl-Gas gesättigtes Methanol zugetropft. Nach etwa 5 Stunden zeigt eine Probe der Reaktionslösung keine positive Keller- Reaktion mehr. Man filtriert die Lösung vom über schüssigen Zink ab und entfernt das Lösungsmittel im Vakuum, bis ein dickflüssiger farbloser Sirup zurückbleibt.
Dieser wird mit Eiswasser verdiinnt, mit konz. Ammoniaklösung 25 % unter Kühlen alka- lisiert und mit Chloroform mehrmals extrahiert. Die vereinigten Extrakte werden einmal mit gesättigter Natriumchloridlösung gewaschen und mit Natrium sulfat getrocknet. Als roher Eindampfrückstand ver bleiben 3,05 g krist. Rohprodukt.
Eine einmalige Umkristallisation aus Essigsäureäthylester ergibt 2,210 g farblose Kristalle vom Smp. 172-174 . <I>Beispiel 6</I> d,1-6-Acetyl-2,3,9,10-tetrahydro-nor- lysergsäure-methylester 1,5 g d,l-6-Acetyl-9,10-dihydro-nor-lysergsäure- methylester werden in 150 cm3 abs. Methanol gelöst und bei 0 bis 5 mit Salzsäure-Gas gesättigt.
Hierauf werden insgesamt 100 g Zinkpulver in vier Portionen zugegeben und das Reduktionsgemisch stark gerührt während insgesamt 4 Stunden. Nach dieser Zeit zeigt eine Probe der Lösung keine positive Keller-Reak- tion mehr. Man filtriert vom überschüssigen Zink ab und engt die Lösung im Vakuum ein. Das dick flüssige farblose Öl wird in Chloroform aufgenom men und bei 0 bis 10 mit Ammoniakgas alkalisiert. D.ie Chloroformlösung wird noch mit Wasser gewa schen und über Natriumsulfat getrocknet.
Nach dem Eindampfen des Lösungsmittels im Vakuum erhält man einen Schaum, aus dem sich in F-ssigsäureäthyl- ester/Ather-Gemisch Kristalle vom Smp. 145-146 abscheiden. Diese erweisen sich laut Analyse, IR- Spektrum und UV-Spektrum als d,l - 6 - Acetyl- 2,3,9,10-tetrahydro-nor-lysergsäure-methylester. <I>Beispiel 7</I> 2,3,9,10-Tetrahydro-d-lysergsäure- D-propanolamid-(2') Man suspendiert 500 g Zinkstaub und 5 g 9,
10- Dihydro - d - lysergsäure - D - propanolamid - (2') in 200 cm3 Wasser und lässt im Dunkeln während 7 Stunden unter intensivem Rühren und bei einer Temperatur von 15-20 und gleichzeitigem Durch leiten von Stickstoff 1,1 1 konz. Salzsäure zutropfen. Nach dieser Zeit fällt die Kellersche Farbreaktion negativ aus. Zur Aufarbeitung filtriert man vom überschüssigen Zinkstaub ab.
In das klare Filtrat leitet man unter kräftigem Rühren und intensivem Kühlen so lange gasförmigen Ammoniak ein, bis sich ein intermediär gebildeter Niederschlag von Zinksalzen wieder gelöst hat. Man schüttelt mit Me- thylenchlorid aus und kristallisiert den nach dem Verdampfen des Lösungsmittels erhaltenen Rück stand aus Äthanol um. Farblose Nädelchen vom Smp. 243-245 . Keller-Reaktion negativ. [a]20 = -169 (c = 1 in Pyridin).
<I>Beispiel 3</I> 2,3-Dihydro-d-lysergsäure-(+)-butanolamid-(2') Eine Lösung von 5 g d-Lysergsäure-(+)-butanol- amid-(2')-bimaleinat in 200 cm3 Wasser wird analog Beispiel 7 mit 500 g Zinkstaub und 0,9 1 konz. Salz säure innert 61/2 Stunden reduziert. Nach dieser Zeit ist die Kellersche Farbreaktion negativ. Die Auf arbeitung erfolgt wieder analog Beispiel 7.
Das Rohprodukt wird in Methanol gelöst, durch Aktiv kohle filtriert und aus Methanol/Aceton kristallisiert. Farblose Nadeln vom Smp. 182,5-184 . Kellersche Farbreaktion negativ. [a] D = -19,5 (c - 1 in Chloroform). Bimaleinat: Smp. 143 bis 144 aus Äthanol.
Beispiel <I>9</I> 2,3-Dihydro-d-isolysergsäure-(+)-butanolamid-(2') 4 g d-Isolysergsäure-(+)-butanolamid-(2') wer den mit 400 g Zinkstaub und 0,95 1 konz. Salzsäure analog Beispiel 7 innert 6 Stunden reduziert. Nach dieser Zeit fällt die Farbreaktion nach Keller negativ aus.
Die Aufarbeitung wird durchgeführt wie im Beispiel 7 beschrieben und liefert 2,8 g öliges Roh- produkt. Durch Verteilungschromatographie an einer Säule von 100 g Silicagel mit einem Wassergehalt von 100 lässt sich das 2,3-Dihydro-d-isolysergsäure-(-;-)- butanolamid-(2') kristallin von den übrigen Reak tionsprodukten abscheiden.
Kristallisation aus Benzol liefert ein Produkt vom Smp. 152-153 . Kellersche Farbreaktion negativ. [a] D = + 263 (c - 0,7 in Chloroform). <I>Beispiel 10</I> 1-Methyl-2,3,9,10-tetrahydro-d-lysergsäure- (+)-butanolamid-(2') 5 g 9,10-Dihydro-l-methyl-dlysergsäure-(+)- butanolamid-(29 werden mit 500g Zinkstaub und 1 1 konz. Salzsäure analog Beispiel 7 innert 51/2 Stun den reduziert. Nach dieser Zeit ist die Kellersche Farbreaktion negativ.
Das Reaktionsgemisch wird analog Beispiel 7 aufgearbeitet. Man erhält ein teil weise kristallines Rohprodukt, das zur weiteren Rei nigung zunächst an 150 g Aluminiumoxyd chromato- graphiert wird. Mit Benzol/Aceton im Volumen verhältnis (1 : 1) wird das 1-Methyl-2,3,9,10 tetra- hydro - d - lysergsäure-(+)-butanolamid-(2') von der Säule eluiert und anschliessend aus Äthanol/Benzol kristallisiert.
Farblose Nadeln vom Smp. 215-218''. Kellersche Farbreaktion negativ. [a] = D -101,6 (c = 1 in Dioxan). <I>Beispiel 11</I> 2,3-Dihydro-d-lysergsäure-diäthylamid Man reduziert 5 g d-Lysergsäure-diäthyl'amid mit 500g Zinkstaub und 1,2 1 konz. Salzsäure analog Beispiel 7. Das Ende der Reaktion wird durch den negativen Ausfall der Kellerschen Farbreaktion nach 7 Stunden angezeigt. Die Aufarbeitung erfolgt wie derum analog Beispiel 7.
Das 2,3-Dihydro-d-lyserg- säure-diäthylamid fällt dabei als amorpher Schaum an. Die Kellersche Farbreaktion ist negativ, die Re aktion von Van Urk schwach gelb.
<I>Beispiel 12</I> 1-Methyl-2,3-dihydro-d-lysergsäure-diäthylamid 4 g 1-Methyl-d lysergsäure-diäthylamid-bitartrat werden mit 500 g Zinkstaub und 0,8 1 konz. Salz säure analog Beispiel 7 innert 4 Stunden reduziert.
Nach dieser Zeit ist die Kellersche Farbreaktion negativ geworden. -Die Aufarbeitung erfolgt analog Beispiel 7 und liefert ein amorphes Produkt, das durch Chromatographie an 150g Aluminiumoxyd weiter gereinigt wird. Gelber, amorpher Schaum. Kellersche Farbreaktion negativ. <I>Beispiel 13</I> 2,3-Dihydro-d-lysergsäureamid 2 g d-Lysergsäureamid werden mit 200 g Zink staub und 0,45 1 konz. Salzsäure analog Beispiel 1 innert 6 Stunden reduziert.
Nach dieser Zeit fällt die Kellersche Farbreaktion negativ aus. Die Aufarbei tung wird analog Beispiel 7 durchgeführt. Das Roh produkt wird zur Abtrennung einer kleinen Menge von gebildetem 2,3-Dihydro-d-isolysergsäureamid an 60 g Aluminiumoxyd chromatographiert. Mit M;- thylenchlorid, das 1 ä Methanol enthält, wird das 2,3-Dihydro-d-lysergsäureamid in das Filtrat ge waschen.
Kristallisation aus Äthanol/Essigsäureäthyl- ester unter Zusatz von wenig Heptan liefert farblose, strahlige Drusen vom Smp. 182-184 (Zers.). Kellersche Farbreaktion negativ. [(@] D = + 11 (c = 0,5 in Chloroform). <I>Beispiel 14</I> 2,3,9,10-Tetrahydro-lysergsäureamid 3 g 9,10-Dihydro-lysergsäureamid werden mit 300 g Zinkstaub und 0,9 1 konz. Salzsäure analog Beispiel 7 innert 6 Stunden reduziert.
Nach dieser Zeit ist die Kellersche Farbreaktion negativ gewor den. Die Aufarbeitung wird analog Beispiel 7 durch geführt. Kristallisation des Rohproduktes aus Äthanol/Essigsäureäthylester ergibt farblose Nädel- chen vom Smp. 239-2411 (unter partieller Sublima tion in Nadeln und Zersetzung).
Kellersche Farb- reaktion negativ. [a] D = -90 (in Chloroform). <I>Beispiel 15</I> 1-Methyl-2,3-dihydro-d-lysergsäureamid und 1-Methyl-2,3-dihydro-d-isolysergsäureamid 2,6 g 1-Methyl-lysergsäureamid werden mit 260 g Zinkstaub und 0,35 1 konz. Salzsäure analog Bei spiel 7 innert 6 Stunden reduziert.
Nachdem die Kellersche Farbreaktion negativ geworden ist, wird analog Beispiel 7 aufgearbeitet und das Rohprodukt an 130 g Aluminiumoxyd chromatographiert. Das 1-Methyl-2,3-dihydro-D-isolysergsäureamid wird be reits mit Methylenchlorid und 0,5110 Methanol in das Filtrat gewaschen. Die Kristallisation aus Äthanol. liefert farblose, rechteckige Platten vom Smp. 205 bis 206 . Kellersche Farbreaktion negativ. [c < ] D = + 257 (in Chloroform).
Mit Methylenchlorid und 1 ö Methanol wird anschliessend das 1-Methyl-2,3- dihydro-D-lysergsäureamid von der Säule eluiert, welches aus Athanol umkristallisiert einen Smp. von l91-192 zeigt. Kellersche Farbreaktion negativ. [a] - -76 (in Chloroform).

It is known that indoles can be reduced to indolines with zinc and mineral acid. It has now been shown for the first time that lysergic acid and dihydrolysergic acid derivatives can also be reduced with nascent hydrogen in the 2,3-position.
This fact is surprising because lysergic acid and dihydrolysergic acid derivatives are extremely sensitive to mineral acids and can therefore easily be rearranged to naphthalene derivatives [A. Stoll and Th. Petrzilka, Helv. <B><I>36,</I> </B> 1125 (1953)].
It is moreover. a surprising finding that when using the reduction method in accordance with the present process, the double bond in the 9,10-position is not attacked.
The reduction according to the invention of the lysergic acid and dihydrolysergic acid derivatives in the 2,3-position takes place, as already mentioned, with nascent hydrogen, preferably with zinc and mineral acid.
The inventive method is carried out, for example, by suspending a solution of a lysergic acid or dihydrolysergic acid derivative of the formula 1I or its salt 10 to 200 parts by weight of zinc dust and then adding mineral acid in the gaseous or liquid phase with stirring at such a rate that the pH value is between 1 and 2.
Suitable solvents are water, aqueous acids, alcohols, alcoholic acids and mixtures of alcohols with water. For the success of the reaction, it is advantageous to work with a reaction time of 3 to 8 hours and a reaction temperature between 10 and 50, preferably between 15 and 25. The end of the reaction is with Keller's color reagent [iron (111) chloride containing glacial acetic acid and conc. Sulfuric acid] recognizable by the disappearance of the color reaction.
