Verfahren zur Herstellung neuer Hydrazine
Gegenstand des Patents ist ein Verfahren zur Herstellung von Hydrazinen der Formel Ar-O-R-NH-NH2 I, worin R einen gegebenenfalls substituierten Rest der Formel -(CH2)11-, worin n für eine ganze Zahl von 2-8 steht, und Ar einen gegebenenfalls substituierten aromatischen Kohlenwasserstoffrest bedeutet und worin mindestens einer der Reste Ar oder R einen Substituenten enthält.
Die Reste Ar sind vorzugsweise höchstens zweikernige aromatische Reste, wie Phenyl- oder Naphthylreste. Als Substituenten in diesen Resten kommen vor allem freie oder substituierte Oxygruppen', wie durch niedere aliphatische oder cycloaliphatische Kohlenwasserstoffreste, besonders niedere Alkylreste, wie Methyl-, Sithyl-, gerade oder verzweigte Propyl-, Butyl- oder Amylreste, substituierte Oxygruppen, niedere Alkylreste, wie die oben genannten, oder Halo genatome, besonders Chlor oder Brom, in Betracht.
Als Substituenten des Restes der Formel (CH)n kommen insbesondere freie oder substituierte, wie acylierte Oxygruppen oder niedere Alkylreste, z. B. die genannten, in Frage. Vorteilhaft besitzt dieser Rest zusammen mit allfälligen Alkylsubstituenten 2 bis 8, insbesondere 2-6 Kohlenstoffatome.
Acylreste sind vor allem diejenigen von Carbonsäuren, besonders aliphatischen Carbonsäuren, wie niederen Fettsäuren, oder solche von aromatischen oder heterocyclischen Carbonsäuren.
Die neuen Hydrazine besitzen eine wertvolle zentralerregende Wirkung mit aussergewöhnlicher Verteilung der zentralen Angriffspunkte. Sie können daher zur Steigerung zentraler Funktionen, wie z. B. bei zentralen Depressionszuständen Verwendung finden.
Besonders wertvoll sind die der Formel I entsprechenden Verbindungen der Formel Ph-O-Alk-X-Alk-NH-NH2 und ihre Salze, worin Ph einen Phenylrest bedeutet, der wie eingangs gezeigt, vor allem aber durch Halogenatome oder niedere Alkoxygruppen, besonders Methoxy- oder Butoxygruppen substituiert sein kann, Alk einen Alkylenrest mit 1-4 Kohlenstoffatomen, vorzugsweise Methylen, Äthylen, Äthyliden oder Propyliden, und X eine direkte Bindung oder eine Methylen- oder Oxymethylengruppe darstellt. Die Gruppe Alk-X-Alk enthält dabei besonders 2-4 Kohlenstoffatome und stellt in erster Linie eine l2ithy- len-, Propylen-(1,2)-, Propylen-(1, 3)-oder 2-Oxy propylen-(1,3)-gruppe dar.
Besonders hervorzuheben ist das 2-Phenoxy-1 methyl-äthyl-(1)-hydrazin der Formel

das 3 - (o-Methoxy-phenoxy)-2-oxy-propyl-(1 )-hydra- zin und das 2-(p-Chlorphenoxy)-äthyl-hydrazin und ihre Salze.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, dass man ein Amin der Formel
Ar-O-R-NH2 mit einem reaktionsfähigen Ester von Hydroxylamin, z. B. einem N-Halogenamin, wie Chloramin, oder Hydroxylamin-O-sulfonsäure, umsetzt.
Erhaltene acylierbare Verbindungen, worunter auch Verbindungen mit freien Oxygruppen zu verstehen sind, können in üblicher Weise acyliert werden,
Die verfahrensgemässe Reaktion kann in üblicher Weise in An- oder Abwesenheit von Verdünnungsmit teln, Kondensationsmitteln und/oder Katalysatoren, bei gewöhnlicher oder erhöhter Temperatur, gegebe nenfalls unter Druck, durchgeführt werden.
Die Ausgangsstoffe sind bekannt oder lassen sich nach an sich bekannten Methoden herstellen. Sie können unter den Reaktionsbedingungen hergestellt werden.
Die neuen Verbindungen bilden Salze mit anorganischen oder organischen Säuren. Als salzbildende Säuren kommen beispielsweise in Frage:
Halogenwasserstoffsäuren, Schwefelsäure,
Phosphorsäure, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie
Ameisen-, Essig-, Propion-, Oxal-, Bernstein-,
Glykol-, Milch-, ipfel-, Wein-, Zitronen-,
Ascorbin-, Oxymalein-, Dioxymalein- oder
Brenztraubensäure;
Phenylessig-, Benzoe-, p-Aminobenzoe-,
Anthranil-, p-Oxybenzoe-, Salicyl- oder p-Aminosalicylsäure;
Methansulfon-, Sithansulfon-, Oxyäthansulfon-, Äthylensulfonsäure;
Toluolsulfon-, Naphthalinsulfonsäuren oder
Sulfanilsäure.
Erhaltene Salze können in an sich bekannter Weise in die freien Basen verwandelt werden.
Die neuen Verbindungen, ihre Salze oder entsprechende Gemische können z. B. in Form pharmazeutischer Präparate Verwendung finden, welche die genannten Verbindungen in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial enthalten.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
88,3 g (0,53 Mol) fl-(o-Methoxyphenoxy)-äthyl- amin werden in einem Gemisch von 100 cm3 Wasser und 150 cm3 Äthanol zusammen mit 9,2 g Kaliumhydroxyd am Rückfluss erhitzt und innerhalb von 30 Minuten mit 9,5 g Hydroxylamin-O-sulfonsäure in 50 cm3 Wasser versetzt. Das Reaktionsgemisch wird auf die Hälfte eingeengt, mit 100 cm3 5n Natronlauge versetzt und mehrmals mit Chloroform extrahiert. Das nach dem Trocknen und Eindampfen des Lösungsmittels verbleibende Öl wird destilliert.
Nach einem Vorlauf des überschüssigen Ausgangsamins vom Kp.11: 138-1420 erhält man im Hochvakuum das 2 (o-Methoxyphenoxy)-äthyl-hydrazin der Formel

als gelbliches Öl. Kp. oo : 108-1120.
Das Bisoxalat bildet, aus Äthanol-Äther umkristallisiert, farblose Kristalle vom F. 139-1400.
Das als Ausgangsstoff verwendete Amin wird z. B. wie folgt erhalten:
Zu einer Lösung von 11,5 g (0,5 Mol) Natrium in 250 cm3 absolutem Äthanol lässt man 62 g (0,5 Mol) in 100 cm3 absolutem Äthanol gelöstes Gujakol zufliessen. Nachdem der Kolbeninhalt auf 0 abgekühlt worden ist, werden 282 g (1,5 Mol) Xithylen- bromid zugegeben. Unter kräftigem Rühren kocht man das Reaktionsgemisch unter Rückfluss, bis es neutral reagiert (etwa 4 Stunden). Nach dem Abkühlen filtriert man das entstandene Natriumbromid ab und engt im Wasserstrahlvakuum ein.
Der Rückstand wird in 200 cm3 Chloroform aufgenommen, zweimal mit je 50 cm3 2n Natronlauge ausgezogen, dann mit Wasser neutralgewaschen und schliesslich über wasserfreiem Natriumsulfat getrocknet. Nach dem Abdestillieren des Lösungsmittels kühlt man den Rückstand in Eis und filtriert das als Nebenprodukt entstandene 1,2-Di-(o-Methoxyphenoxy)-äthan (F. 148-150 ) ab.
Das Filtrat wird am Vakuum destilliert. Das o-Methoxyphenoxy-äthylbromid ist eine farblose Flüssigkeit vom Kp.,,: 141-1420, welche nach einiger Zeit kristallinisch erstarrt und dann einen F. 40-420 aufweist.
Zu 40 g (0,173 Mol) o-Methoxyphenoxy-äthyl- bromid in 200 cm3 Methanol gibt man 40 g flüssiges Ammoniak und erhitzt im Schüttelautoklaven 10 Stunden lang auf 100. Nach dem Abkühlen wird das überschüssige Ammoniak abgeblasen und das Methanol verdampft. Den Rückstand nimmt man in Chloroform auf, wäscht zweimal mit Wasser aus und trocknet über wasserfreiem Natriumsulfat. Nach dem Verdampfen des Lösungsmittels wird der Rückstand im Wasserstrahlvakuum destilliert. Das ss-(o-Methoxy- phenoxy)-äthylamin der Formel

ist eine farblose Flüssigkeit vom Kp. 11 : 139-1410.
Neutralisiert man die Base mit einer Lösung von Chlorwasserstoff in Essigester, so erhält man ein farbloses Hydrochlorid, welches nach Umkristallisieren aus Athanol-Ather bei 116-117 schmilzt.
In analoger Weise können aus entsprechenden Ausgangsstoffen noch folgende Verbindungen erhalten werden: 1. 3-(o-Methoxyphenoxy)-2-hydroxy-propyl-( 1)- hydrazin-oxalat vom F. 15y155C (aus Methanol-Essigester).
2. 2-(p-Chlorphenoxy)-äthyl-hydrazin- hydrochlorid vom F. 108-1090 (aus Sithanol-Essigester).
3. 2-(o,p-Dichlorphenoxy)-äthyl-hydrazin-oxalat vom F. 1700 (Zers.) (aus 2ithanol-Wasser).
4. 2-(o,o'-Dimethoxyphenoxy)-äthyl-hydrazin hydrochlorid vom F. 161-162 (aus Methanol-Athanol).
5. 2-[m-(n)-Butoxyphenoxy]-äthyl-hydrazin- hydrochlorid vom F. 94-95 (aus Methanol-Xthanol).
6. 1 -Phenoxy-propyl-(2)-hydrazin, Kp.,,:145-1470;
Dihydrochlorid F. 124-1250 (Zers.),
Bis-oxalat F. 1480 (Zers.) (aus Äthanol-Essigester).
Ferner durch übliche Acetylierung 7. N-[ 1 -Phenoxy-propyl-(2)j-N'-acetylhydrazin vom Kp.0,08: 140-1430.
Process for the production of new hydrazines
The subject of the patent is a process for the preparation of hydrazines of the formula Ar-OR-NH-NH2 I, in which R is an optionally substituted radical of the formula - (CH2) 11-, in which n is an integer from 2-8, and Ar denotes an optionally substituted aromatic hydrocarbon radical and in which at least one of the radicals Ar or R contains a substituent.
The Ar radicals are preferably at most binuclear aromatic radicals, such as phenyl or naphthyl radicals. Substituents in these radicals are primarily free or substituted oxy groups, such as lower aliphatic or cycloaliphatic hydrocarbon radicals, particularly lower alkyl radicals, such as methyl, sithyl, straight or branched propyl, butyl or amyl radicals, substituted oxy groups, lower alkyl radicals, such as those mentioned above, or halogen atoms, especially chlorine or bromine, into consideration.
Substituents of the radical of the formula (CH) n are in particular free or substituted, such as acylated oxy groups or lower alkyl radicals, e.g. B. those mentioned in question. This radical advantageously has 2 to 8, in particular 2-6, carbon atoms together with any alkyl substituents.
Acyl radicals are above all those of carboxylic acids, especially aliphatic carboxylic acids such as lower fatty acids, or those of aromatic or heterocyclic carboxylic acids.
The new hydrazines have a valuable central stimulating effect with an exceptional distribution of the central points of attack. You can therefore increase key functions, such as B. find use in central states of depression.

The bisoxalate, recrystallized from ethanol-ether, forms colorless crystals with a mp of 139-1400.
The amine used as starting material is z. B. obtained as follows:
62 g (0.5 mol) of gujakol dissolved in 100 cm3 of absolute ethanol are allowed to flow into a solution of 11.5 g (0.5 mol) of sodium in 250 cm3 of absolute ethanol. After the contents of the flask have cooled to 0, 282 g (1.5 mol) of xithylene bromide are added. The reaction mixture is refluxed with vigorous stirring until it reacts neutrally (about 4 hours). After cooling, the sodium bromide formed is filtered off and concentrated in a water jet vacuum.
The residue is taken up in 200 cm3 of chloroform, extracted twice with 50 cm3 of 2N sodium hydroxide solution each time, then washed neutral with water and finally dried over anhydrous sodium sulfate. After the solvent has been distilled off, the residue is cooled in ice and the 1,2-di (o-methoxyphenoxy) ethane (mp 148-150) formed as a by-product is filtered off.
The filtrate is distilled in vacuo. The o-methoxyphenoxy-ethyl bromide is a colorless liquid of bp.: 141-1420, which solidifies crystalline after some time and then has a m.p. 40-420.
40 g of liquid ammonia are added to 40 g (0.173 mol) of o-methoxyphenoxy-ethyl bromide in 200 cm3 of methanol and heated to 100 for 10 hours in a shaking autoclave. After cooling, the excess ammonia is blown off and the methanol is evaporated. The residue is taken up in chloroform, washed twice with water and dried over anhydrous sodium sulfate.
