Verfahren zur Herstellung neuer polycyclischer Guanidine
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Imino-alkyl-guanidinen der Formel

worin R einen an mindestens einen isocyclischen oder heterocyclischen Rest aromatischer Natur anellierten, gegebenenfalls durch Kohlenwasserstoffreste substituierten Alkylenrest bedeutet, wobei der durch R und N gebildete Ring 4-9 Ringkohlenstoffatome enthält, und in denen A eine Alkylengruppe mit 2-5 Kohlenstoffatomen darstellt.
In den neuen Verbindungen ist die Guanidogruppe vornehmlich unsubstituiert; sie kann jedoch auch substituiert sein, z. B. durch aliphatische Kohlenwasserstoffreste, vorzugsweise Niederalkylgruppen, wie Methyl, Äthyl, oder i-Propyl.
Als Reste aromatischer Natur kommen vornehmlich monocyclische Reste in Frage, z. B. Benzol- oder Pyridinkerne, aber auch bicyclische Reste, wie Naphthalin- oder Chinolinkerne.
Als Beispiele für die oben beschriebenen, R enthaltenden Iminoreste können angeführt werden: Indolinyl-(l), Isoindolinyl-(2), Tetrahydrochinolyl-(1), Tetrahydroisochinolyl - (2), 2,3-Benzo- 2,3-dehydro- hexylenimino, 3,4 - Benzo -3 ,4-dehydro-hexylenimino, 4,5-Benzo-4,5-dehydro-hexylenimino, 2,3-Benzo-2,3- dehydro-heptylenimino, 3,4 -Benzo-3,4-dehydro-hep- tylenimino, 4,5-Benzo-4,5-dehydro-heptylenimino, 2,3;6,7-Dibenzo-2,3;6,7-bis-dehydro-hexylenimino oder 3,4-Naphtho-(2',3')-3,4-dehydro-heptylenimino.
Die genannten Ringsysteme sind im nichtaromatischen Teil vornehmlich unsubstituiert, sie können aber auch durch Kohlenwasserstoffreste, z. B. Niederalkylgruppen, wie Methyl- oder Äthylgruppen, substitiert sein. Der aromatische Teil kann beispielsweise durch folgende Gruppen substituiert sein: Niederalkyl, wie Methyl oder Äthyl, freies, veräthertes oder verestertes Hydroxy, z. B. Niederalkoxy, wie Methoxy, Äthoxy, oder i-Propoxy, Niederalkylendioxy, wie Methylendioxy, Aralkoxy, wie Benzyloxy, oder Niederalkoxy-carbonyloxy, wie Methoxycarbonyloxy, Niederalkanoyloxy, wie Acetoxy oder Propionyloxy, Halogen, wie Fluor, Chlor oder Brom, oder Niederalkylmercapto, wie Methyl- oder Äthylmercapto, Niederalkanoyl, wie Acetyl oder Propionyl, Nitro, Amino, mono-substituiertes Amino, z. B.
Niederalkylamino, wie Methyl- oder Äthylamino, vornehmlich disubstituiertes Amino, z. B. Di-niederalkylamino, wie Dimethylamino, Methyl-äthylamino oder Diäthylamino, oder Alkylenimino, worin die Alkylenkette, die auch durch Sauerstoff- oder Stickstoffatome unterbrochen sein kann, 6 Ringkohlenstoffatome enthält, z. B.
Pyrrolidino, Piperidino, 2-Methyl-piperidino, Hexamethylenimino, Morpholino, 4-Methyl-piperazino oder 4-ss-Hydroxy-äthyl-piperazino, oder Halogen-niederalkyl, wie Trifluormethyl.
In den neuen Verbindungen wird die Imino- von der Guanidogruppe durch 2-5, vornehmlich aber durch 2-3 Kohlenstoffatome des Alkylenrestes getrennt. Dieser bedeutet somit speziell 1, 2-Äthylen, 1,2- 2,3- oder 1,3-Propylen, aber auch 1,3- oder 1, 4-Butylen oder 1,5-Pentylen.
Als Salze der neuen Verbindungen kommen vornehmlich therapeutisch anwendbare Säureadditionssalze, z. B. solche von anorganischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefel- oder Phosphorsäuren, oder von organischen Säuren, z. B. Essig-, Propion-, Glykol-, Milch-, Brenztrauben-, Oxal-, Malon-, Bernstein-, Malein-, Fumar-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Citracon-, Hydroxymalein- oder Dihydroxymaleinsäure, oder Benzoe-, Phenylessig-, 4-Amino-benzoe-, 4-Hydroxybenzoe-, Anthranil-, Zimt-, Mandel-, Salicyl-, 4-Aminosalicyl-, 2-Phenoxybenzoe- oder 2-Acetoxy-benzoesäure, oder Methansulfon-, Äthansulfon-, 2-Hydroxy-äthansulfon- oder p-Toluolsulfonsäure in Frage. Hiervon können Monooder Polysalze gebildet werden.
Die neuen Guanidinverbindungen und ihre Salze verhindern die hypertensive, d. h. blutdruckerhöhende Wirkung von l-Phenyl-2-amino-propan (Amphetamin) oder ähnlichen Sympathikomimetika, ohne deren stimulierende Wirksamkeit zu beeinträchtigen.
Sie können deshalb beispielsweise in Kombination mit Sympathikomimetika des Amphetamin-Typs verwendet werden, um die hypertensive Nebenwirkung der letztgenannten Drogen auszuschalten und ihre sehr erwünschte stimulierende Wirkung voll zur Geltung zu bringen.
Weiter sind die neuen Guanidinverbindungen und ihre Salze durch antihypertensive Eigenschaften cha rakterisiert; auch verursachen sie eine Steigerung der peripheren Blutzirkulation. Sie können deshalb sowohl als Mittel gegen hohen Blutdruck, speziell gegen neurogene, renale oder essentielle Hypertonie, als auch zur Behandlung peripherer Gefässerkrankungen, z. B. der Reynaudschen Krankheit, verwendet werden. Die von den neuen Verbindungen hervorgerufenen Wirkungen dauern relativ lang an, weshalb sie sich besonders gut zur Behandlung von chronischen Hypertonieformen eignen.
Eine bevorzugte Gruppe der neuen Guanidine bilden solche der Formeln

Endstoffe des Verfahrens sind z. B. die folgenden: 2-(2,3-Benzo-2,3-dehydro-hexylenimino)-äthyl- guanidin, 2-(2,3 ;6,7-Dibenzo-2,3 ;6,7-didehydro-hexylenimino)- äthyl-guanidin, 2-[1 ,2,3,4-Tetrahydro-chinolyl-(l )j-äthyl-guarndin, 2-[7-Chlor- 1,2,3 ,4-tetrahydrcchinolyl-( 1 )]-äthyl- guanidin, 3-[ 1 ,2,3,4-Tetrahydro-chinolyl-(l)j-propyl-guanidin, 3-[Isoindolinyl-(2)]-propyl-guanidin, 2-[Isoindolinyl-(2)]-äthyl-guanidin, 2-[4-Methoxy-isoindolinyl-(2)]-äthyl-guanidin, 2-(4,5-Benzo-4,5-dehydro-hexylenimino)-äthyl- guanidin, 2-(2, 3 -Benzo-2, 3 -dehydro-heptylenimino)-äthyl- guanidin, 3-[1,2,3,4-Tetrahydro-isochinolyl-(2)]-propyl guanidin,

Die neuen Guanidine und ihre Salze können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man entsprechende Imino-alkylamine oder deren Salze mit Verbindungen der Formel I

oder ihren Salzen, umsetzt, worin X einen Alkylmercapto-, Alkoxy- oder einen l-Pyrazolrest bedeutet, Z eine Aminogruppe darstellt, N-Y für eine Iminogruppe steht oder X und Y zusammen mit der CM- Doppelbindung eine CN-Dreifachbindung bilden.
Wenn erwünscht, können erhaltene Salze in die freien Verbindungen oder erhaltene freie Verbindungen in ihre Salze umgewandelt werden.
Verbindungen der Formel I sind beispielsweise S-Niederalkyl-isothioharnstoffe oder O-Niederalkylisoharnstoffe oder Cyanamide oder 1-Guanyl-pyrazole, die gegebenenfalls auch N-substituiert sein können.
Als N-Substituenten kommen vornehmlich Niederalkylgruppen in Frage, z. B. Methyl, Äthyl, oder i-Propyl. 1-Guanyl-pyrazole sind vornehmlich auch im Kern, z. B. durch Niederalkylgruppen, substituiert.
Die genannten Verbindungen können auch in Form von Säureadditionssalzen, vornehmlich Mineralsäure- additionssalzen, vorliegen.
Eine besonders bevorzugte Verbindung der Formel I ist S-Methyl-isothioharnstoff und seine Mineralsäure-additionssalze, speziell das S-Methyl-isothioharnstoff-sulfat. Weiter können als bevorzugte Verbindungen nach O-Methyl-isorharnstoff-sulfat, Cyanamid oder 1-Guanyl-3, 5-dimethyl-pyrazol-nitrat genannt werden.
In dem als Ausgangsprodukt verwendeten Iminoalkylamin steht die Aminogruppe vornehmlich für eine primäre Aminogruppe, aber auch für eine sekundäre Aminogruppe, z. B. für eine Niederalkyl-aminogruppe, wie eine Methyl- oder Äthylaminogruppe.
Diese Verbindungen werden zumeist in Form der freien Basen umgesetzt.
Die Reaktion mit S-Niederalkyl-isothioharnstoffen wird vorzugsweise in Gegenwart von Verdünnungsmitteln durchgeführt, deren Auswahl im Hinblick auf die Löslichkeiten der Reaktionskomponenten erfolgt.
Als Verdünnungs- oder Lösungsmittel kommen beispielsweise Wasser oder mit Wasser mischbare organische Lösungsmittel in Frage, z. B. Alkanole, wie Methanol, Äthanol, Propanol, i-Propanol oder tert.
Butanol, Äther, wie Dioxan oder Tetrahydrofuran, Ketone, wie Aceton oder Methyl-äthylketon, Niederalkancarbonsäuren, wie Essigsäure, oder Formamide, wie Formamid oder Dimethyl-formamid, oder deren wässrige Gemische. Die Reaktion kann bei Raumtemperatur oder, falls erforderlich, bei erhöhter Temperatur, z. B. bei der Siedetemperatur des Lösungsoder Verdünnungsmittels, bei normalem oder erhöhtem Druck oder in Gegenwart eines Inertgases, z. B.
Stickstoff, durchgeführt werden.
In analoger Weise kann auch die Umsetzung mit O-Niederalkyl-isoharnstoffen durchgeführt werden.
Die Reaktion mit Cyanamiden wird beispielsweise so vorgenommen, dass man eine Mischung des Alkenyleniminoniederalkylamins der oben genannten Art, speziell eines Salzes davon, z. B. eines Additionssalzes mit einer Mineralsäure, wie Salzsäure, Bromwasserstoffsäure oder Schwefelsäure, und des Cyanamids erhitzt, die erhaltene Schmelze in einem Lösungsmittel, z. B. einer Niederalkancarbonsäure, wie Essigsäure, löst und das Reaktionsprodukt in an sich bekannter Weise isoliert. Obige Umsetzung kann aber auch in Gegenwart eines Lösungs- oder Verdünnungsmittels, z. B. eines niederen Alkanols, wie Äthanol, oder einer konzentrierten wässrigen Säure, wie Salzsäure, durchgeführt werden, wobei sich das Säureadditionssalz des Ausgangsproduktes auch intermediär bilden kann.
Desgleichen kann sich auch das Cyanamid intermediär bilden, beispielsweise bei Verwendung von l-Nitroso-3-methyl-guanidin das Methyl-cyanamid, womit man die entsprechenden l-(Imino-alkyl)-3- methyl-guanidine herstellen kann. Die Reaktion kann exotherm verlaufen, falls notwendig, erhitzt man auf Temperaturen zwischen ungefähr 80 und 2000, eventuell in Gegenwart eines Inertgases, z. B. Stickstoff.
Die Umsetzung der oben beschriebenen Iminoalkylamine mit einem l-Guanyl-pyrazol kann in Anoder Abwesenheit eines Verdünnungsmittels erfolgen, beispielsweise durch Erhitzen der Mischung auf deren Schmelzpunkt oder, bei Anwesenheit eines Lösungsmittels, auf deren Siedepunkt. Vorteilhaft schliesst man die Gegenwart von Kohlendioxyd durch Anwendung eines Inertgases aus.
Die oben angeführten Ausgangsstoffe sind bekannt oder lassen sich, falls neu, nach an sich bekannten Methoden herstellen. S-Niederalkyl-isothioharnstoffe oder O-Niederalkyl-isoharnstoffe können beispielsweise durch Alkylierung von mindestens 1 Wasserstoffatom enthaltenden Thioharnstoffen oder Harnstoffen mit Niederalkyl-halogeniden, wie Methyloder Äthylchlorid, -bromid - oder -jodid, oder Diniederalkylsulfaten, wie Dimethyl- oder Diäthylsulfat, erhalten werden.
Die genannten Imino-alkylamine erhält man beispielsweise durch Reaktion eines entsprechenden Imins mit einem reaktionsfähig veresterten Cyan-niederalkanol, z. B. einem Cyan-niederalkyl-halogenid, worin Halogen z. B. für Chlor oder Brom steht, oder mit einem Niederalkenyl-cyanid, worin die Doppelbindung durch die Cyangruppe aktiviert ist. In den erhaltenen substituierten Niederalkylcyaniden kann sodann die Cyangruppe durch Reduktion in die Methylenaminogruppe übergeführt werden, beispielsweise durch katalytische Hydrierung, wie Behandlung mit Wasserstoff, in Gegenwart eines ein Metall der 8. Gruppe des periodischen Systems enthaltenden Katalysators, z. B.
Palladiummohr oder Raney-Nickel, oder vorzugsweise durch Behandlung mit einem Dileichtmetallhydrid, wie Lithium-, Natrium- oder Magnesium-aluminiumhydrid, oder mit Aluminium hydrid oder Aluminiumborhydrid, falls notwendig, in Gegenwart eines Aktivators, wie Aluminiumchlorid.
Die neuen Guanidinverbindungen können entweder als freie Verbindungen oder in Form ihrer Salze erhalten werden. Ein Salz kann in an sich bekannter Weise, beispielsweise durch Behandlung mit einem stark alkalischen Mittel, wie wässrigem Alkalimetallhydroxyd, z. B. Lithium-, Natrium- oder Kaliumhydroxyd, oder mit starken Anion-Austauscherharzen, wie quaternären Ammonium-Austauscherharzen, in die freie Verbindung übergeführt werden. Von den freien Basen können mit geeigneten, beispielsweise den eingangs erwähnten, anorganischen oder organischen Säuren therapeutisch anwendbare Additionssalze hergestellt werden. Die Umsetzung mit Säuren erfolgt vorzugsweise in geeigneten Verdünnungsmitteln, z. B. Niederalkanolen, wie Methanol, Äthanol, n-Propanol oder i-Propanol, Äthern, wie Diäthyläther oder Dioxan, Estern, wie Essigsäure äthylester, oder Mischungen dieser.
Hierbei können basische, neutrale, saure oder gemischte Salze erhalten werden.
Vornehmlich werden solche Ausgangsmaterialien verwendet, die die eingangs erwähnten bevorzugten Verbindungen ergeben.
Die Temperaturen sind in den folgenden Beispielen in Celsiusgraden angegeben.
Beispiel 1
Eine homogene Mischung von 9,1 g 2-[1,2,3,4- Tetrahydro-isochinolyl-(2)]-äthylamin, 7,2 g S-Me- thyl-isothioharnstoff-sulfat und 9 cm3 Wasser lässt man bis zur Beendigung der Methylmercaptan-Entwicklung (etwa 6 Stunden) am Rückflusskühler sieden. Beim Stehen über Nacht fällt ein Niederschlag aus, den man abfiltriert und aus wasserfreiem Äthanol umkristallisiert. Man erhält so das 2-[1,2,3,4-Tetra- hydro-isochinolyl-(2)]-äthyl-guanidin der Formel

als Sulfat vom F. 20P2070. Das Salz enthält 2 Mol Base pro Mol Schwefelsäure und ein halbes Mol Kristallwasser.
Das Ausgangsprodukt kann folgendermassen gewonnen werden:
Eine Mischung von 30,6 g 1 ,2,3,4-Tetrahydro- isochinolin, 17,3 g Chloracetonitril, 95,2 g wasserfreiem Kaliumcarbonat und 200 cmd Toluol hält man 41/o Stunden unter Rühren im Sieden. Nach dem Erkalten filtriert man, dampft das Lösungsmittel unter vermindertem Druck ab und destilliert das verbleibunde 1,2,3 ,4-Tetrahydro - isochinolyl - (2) - acetonitril Kp.24 170-190 .
Zu einer Lösung von 23,3 g des erhaltenen Nitrils in 100 cm Diäthyläther fügt man bei Raumtemperatur unter Rühren innerhalb einer halben Stunde eine Mischung von 6,1 g Lithium-Aluminiumhydrid in 150 cm Äther und rührt 2 Stunden bei Raumtemperatur weiter. Hierauf versetzt man das Reaktionsgemisch nacheinander mit 18,5 cm Essigsäure äthylester, 6,3 cm3 Wasser, 12,3 cm5 15Siger wässriger Natronlauge und 18,5 cm3 Wasser und trennt die organische Phase ab. Nach dem Abdampfen des Äthers erhält man durch Destillation das 2-[1,2,3,4- Tetrahydro-isochinolyl-(2)]-äthylamin vom Kp.0,1 82 bis 880.
Analog kann man folgende Verbindungen erhalten: 3-[1,2,3,4- Tetrahydro- isochinolyl- (2)]- propyl- guanidin, 2-[6-Methoxy-1,2,3,4-tetrahydro-isochinolyl (2)]-äthyl-guanidin, 2-[6,7-Dimethoxy-1,2,3,4-tetrahydro-isochinolyl- (2)] - äthyl - guanidin, 2 - [7 - Chlor- 1,2,3 ,4-tetrahydro-isochinolyl- (2)]-äthyl-guanidin, 1 {2-[1,2,3,4-Tetrahydro-isochinolyl-(2)]-äthyl}-3-methyl-guanidin, vornehmlich als Säureadditionssalze, z. B. als Sulfat.
Beispiel 2
Eine Mischung von 2 g 2-(2,3;6,7-Dibenzo-2,3; 6,7-didehydro-hexylenimino)-äthylamin, 1,2 g S-Me- thyl-isothioharnstoff-sulfat und 5 cm3 Wasser lässt man 4 Stunden sieden und erhält nach dem Einengen des Reaktionsgemisches das 2-(2,3;6,7-Dibenzo- 2,3;6,7-didehydro-hexylenimino)-äthyl-guanidin der Formel

Das eingesetzte Amin lässt sich analog der in Beispiel 1 angegebenen Methode durch Umsetzung von 19,5 g 2,3;6,7-Dibenzo-2,3;6,7-bis-dehydrohexylenimin mit 7,6 g Chloracetonitril in 75 cm3 Benzol in Gegenwart von 5,3 g wasserfreiem Natriumcarbonat und darauffolgender Reduktion des erhaltenen Nitrils mit Lithium-Aluminiumhydrid erhalten.
Beispiel 3
Eine Mischung von 1,5 g 2-[1,2,3,4-Tetrahydro chinolyl-(1)J-äthylamin, 1,2 g S-Methyl-isothioharnstoff-sulfat und Wasser ergibt nach 4stündigem Rückflusskochen das 2-[1,2,3 4-Tetrahydro-chinolyl-(l)]- äthyl-guanidin der Formel

Das Ausgangsprodukt erhält man z. B. durch Erhitzen eines Gemisches von 13,3 g 1,2,3,4-Tetra- hydrochinolin, 7,6 g Chloracetonitril, 5,3 g wasserfreiem Natriumcarbonat und 75 cm3 Benzol und darauffolgender Reduktion des erhaltenen 1,2,3,4- Tetrahydro-chinolyl-acetonitrils mit Lithium-Aluminiumhydrid in ätherischer Lösung.
Analog der angegebenen Methode können noch folgende Guanidine hergestellt werden: 2-[6-Methoxy- 1,2,3,4 - tetrahydro-chinolyl-( 1)]- äthyl-guanidin, 2-[7 Chlor 1,2,3,4 - tetrahydro-chinolyl-(1)]-äthyl-guanidin oder 3-[1 2, 3, 4-Tetrahydro-chinolyl (l)]-propyl-guani- din, vornehmlich in Form der Säureadditionssalze, z. B. der Sulfate.
Beispiel 4
Eine Mischung von 2 g 3-[Isoindolinyl-(2)]-propylamin, 1,8 g S-Methyl-isothioharnstoff-sulfat und 7 cm3 Wasser hält man 4 Stunden im Sieden, worauf nach teilweisem Abdampfen des Wassers das 3-[Isoindolinyl-(2)]-propyl-guanidin der Formel

in Form des Sulfats ausfällt.
Das Ausgangsprodukt wird z. B. wie folgt erhalten:
Zu einer Mischung von 11,9 g Isoindolin und 21,2 g Acrylnitril fügt man einige Tropfen Benzyltrimethylammoniumhydroxyd und erhitzt das Gemisch nach dem Abklingen der Reaktion einige Stunden zum Sieden. Das durch fraktionierte Destillation erhaltene/3-fIsoindolinyl-(2)]-propionitril reduziert man analog der in Beispiel 1 angegebenen Methode zum erwünschten Amin.
In ähnlicher Weise erhält man 2-[Isoindolinyl (2)] -äthyl-guanidin, 2- [4-Methoxy-isoindolinyl-(2)]- äthyl-guanidin oder 2-(2, 3-Benzo-2, 3-dehydro-hepty- lenimino)-äthyl-guanidin bzw. deren Sulfate.
Beispiel 5
Eine Mischung von 2 g 2-(2,3-Benzo-2,3-dehydrohexylenimino)-äthylamin, 1,5 g S-Methyl-isothioharnstoff-sulfat und 5 cm3 Wasser lässt man 4 Stunden am Rückflusskühler sieden, kühlt ab und erhält nach dem Einengen das 2-(2,3-Benzo-2,3-dehydro-hexylen- imino)-äthyl-guanidin der Formel

14,7 g 2,3-Benzo-2,3 -dehydro-hexylenimin und 7,6 g Chloracetonitril fügt man unter Rühren zu einer Suspension von 5,3 g wasserfreiem Natriumcarbonat und 75 cm3 Benzol, hält das Reaktionsgemisch 4 Stunden im Sieden und filtriert nach dem Erkalten.
Nach dem Abdampfen des Benzols erhält man das 2,3 -Benzo-2,3-dehydro-hexylenimino-acetonitril. Dieses löst man in trockenem Diäthyläther, fügt Lithium Aluminiumhydrid zu und lässt das Gemisch einige Zeit sieden. Nach dem üblichen Aufarbeiten erhält man das 2-(2,3 -Benzo- 2,3 -dehydro-hexylenimino)- äthylamin.

Eine Mischung von 6,5 g 2-(4,5-Benzo-4,5- dehydro-hexylenimino)-äthylamin, 4,8 g S-Methylisothioharnstoff-sulfat und 8 cm3 Wasser lässt man 5 Stunden am Rückflusskühler sieden und erhält nach dem Abkühlen des Reaktionsgemisches das 2-(4,5 Benzo-4,5-dehydro-hexylenimino)-äthyl-guanidin der Formel

Das Gemisch von 75 g 1,2-Di-cyanomethyl-ben- zol, 10 g Raney-Nickel und 500 cm3 mit Ammoniak gesättigtem Äthanol wird 5 Stunden bei 75 at und 1000 hydriert, das erhaltene Reaktionsgemisch filtriert und das Filtrat zur Trockne eingeengt. Der Rückstand wird sodann unter vermindertem Druck destilliert und man erhält 4,5-Benzo-4,5-dehydrohexylenimin vom Kp. s 123-124 und nD = 1,5600 neben einer wesentlich geringeren Menge 1 ,2-(ss- Amino-äthyl)-benzol vom Kp.0,15 90-98 und n2,5 = 1,5571.
Eine Mischung von 20,4 g 4,5-Benzo-4,5-de- hydro-hexylenimin, 11,6 g Chloracetonitril, 59,5 g wasserfreiem Natriumcarbonat, 0,5 cm3 Wasser und 70 cm3 Toluol erhitzt man unter Rühren 12 Stunden zum Sieden. Das heisse Reaktionsgemisch wird hierauf filtriert, das Lösungsmittel unter vermindertem Druck abgedampft und der Rückstand aus Äthanol kristallisiert. Man erhält so dass (4,5-Benzo-4,5- dehydro-hexylenimino)-acetonitril vom F. 113-116 .
Eine Lösung von 19,2 g des erhaltenen Nitrils in 100 cm3 Diäthyläther und 35 cm3 Tetrahydrofuran fügt man unter Rühren während einer Stunde einer Suspension von 8,0 g Lithiumaluminiumhydrid und 300 cm3 Diäthyläther zu, rührt noch 2 Stunden weiter und lässt das Reaktionsgemisch über Nacht bei Raumtemperatur stehen. Hierauf versetzt man nacheinander mit 24 cm3 Essigsäureäthylester, 8 cm5 Wasser, 16 cm 15 % iger wässriger Natronlauge und nochmals mit 24 cins Wasser, filtriert das Gemisch und engt das Filtrat ein. Der Rückstand wird unter vermindertem Druck destilliert und man erhält so das erwünschte 2 - (4,5- Benzo - 4,5- dehydro-hexylenimino)-äthylamin vom Kp. 005 117-1400.
Beispiel 7
Die Mischung von 4,5 g 2-[Indolinyl-(1)]-äthyl- amin, 3,9 g S-Methyl-isothioharnstoff-sulfat und 2 cm3 Wasser lässt man 5 Stunden am Rückflusskühler sieden. Nach dem Abkühlen verreibt man das keah- tionsgemisch mehrmals mit Aceton, filtriert den wei ssen Rückstand ab und kristallisiert ihn aus wässrigem Äthanol-Aceton. Den Rückstand löst man in Äthanol und säuert die Lösung mit äthanolischer Salzsäure an. Beim Versetzen der sauren Lösung mit Diäthyläther kristallisiert das gebildete 2-[Indolinyl- (1)] - äthyl- guanidin-dihydrochlorid aus; es schmilzt nach dem Kristallisieren aus Äthanol Diäthyläther bei 1740 unter Zersetzung.
Das als Ausgangsprodukt verwendete 2-[lndo- linyl (l) l-äthylamin kann nach folgender Methode hergestellt werden:
Eine Mischung von 23,8 g Indolin, 16,7 g Chloracetonitril, 42,4 g wasserfreiem Natriumcarbonat, 0,25 cm3 Wasser und 250 cm3 Toluol lässt man unter Rühren 11 Stunden am Rückflusskühler sieden. Das Reaktionsgemisch wird hierauf filtriert, der Rückstand mit Toluol gewaschen, das Filtrat zur Trockne eingedampft und das verbleibende Indolinyl-(l)-acetonitril unter vermindertem Druck destilliert; Kr.15 152-1720, nr5 = 1,5710.
Eine Lösung von 8,5 g des erhaltenen Nitrils in 50 cm3 Diäthyläther lässt man während 11/2 Stunden unter Rühren zu einer unter Stickstoff befindlichen Suspension von 4,3 g Lithium-aluminiumhydrid in 150 cm3 Diäthyläther zutropfen. Nach dem Stehen über Nacht bei Raumtemperatur versetzt man das Reaktionsgemisch nacheinander mit 12 cm3 Essigsäureäthylester, 4 cm3 Wasser, 8 cm3 15/0iger Natronlauge und noch 2 cm3 Wasser und filtriert. In the new compounds, the guanido group is mainly unsubstituted; however, it can also be substituted, e.g. B. by aliphatic hydrocarbon radicals, preferably lower alkyl groups such as methyl, ethyl, or i-propyl.
Monocyclic radicals are primarily suitable as radicals of an aromatic nature, e.g. B. benzene or pyridine nuclei, but also bicyclic radicals such as naphthalene or quinoline nuclei.
As examples of the above-described, R-containing imino radicals, there may be mentioned: indolinyl- (1), isoindolinyl- (2), tetrahydroquinolyl- (1), tetrahydroisoquinolyl- (2), 2,3-benzo- 2,3-dehydro- hexylenimino, 3,4 - benzo -3, 4-dehydrohexylenimino, 4,5-benzo-4,5-dehydrohexylenimino, 2,3-benzo-2,3-dehydro-heptylenimino, 3,4 -benzo 3,4-dehydro-heptylenimino, 4,5-benzo-4,5-dehydro-heptylenimino, 2,3; 6,7-dibenzo-2,3; 6,7-bis-dehydrohexylenimino or 3, 4-naphtho- (2 ', 3') -3,4-dehydro-heptylenimino.

In the new compounds the imino group is separated from the guanido group by 2-5, but primarily by 2-3 carbon atoms of the alkylene radical. This means specifically 1,2-ethylene, 1,2-2,3- or 1,3-propylene, but also 1,3- or 1,4-butylene or 1,5-pentylene.
The salts of the new compounds are mainly therapeutically applicable acid addition salts, eg. B. those of inorganic acids such as hydrochloric acid, hydrobromic acid, sulfuric or phosphoric acids, or of organic acids, e.g. B. vinegar, propion, glycol, milk, pyranium, oxal, malon, amber, maleic, fumaric, apple, wine, lemon, ascorbic, citracon, hydroxymalein or Dihydroxymaleic acid, or benzoic, phenylacetic, 4-amino-benzoic, 4-hydroxybenzoic, anthranil, cinnamon, almond, salicylic, 4-aminosalicylic, 2-phenoxybenzoic or 2-acetoxy-benzoic acid, or Methanesulfonic, ethanesulfonic, 2-hydroxy-ethanesulfonic or p-toluenesulfonic acid in question. The new guanidine compounds and their salts prevent hypertensive, i.e. H. Hypertensive effects of l-phenyl-2-aminopropane (amphetamine) or similar sympathomimetics without impairing their stimulating effectiveness.
They can therefore be used, for example, in combination with sympathomimetics of the amphetamine type in order to eliminate the hypertensive side effects of the latter drugs and to bring their very desirable stimulating effect to full advantage.
Furthermore, the new guanidine compounds and their salts are characterized by antihypertensive properties; they also cause an increase in peripheral blood circulation. They can therefore be used both as a means against high blood pressure, especially against neurogenic, renal or essential hypertension, and for the treatment of peripheral vascular diseases, e.g. B. Reynaud's disease can be used. The effects produced by the new compounds last for a relatively long time, which is why they are particularly suitable for the treatment of chronic forms of hypertension.

Butanol, ethers such as dioxane or tetrahydrofuran, ketones such as acetone or methyl ethyl ketone, lower alkanecarboxylic acids such as acetic acid, or formamides such as formamide or dimethylformamide, or their aqueous mixtures. The reaction can be carried out at room temperature or, if necessary, at an elevated temperature, e.g. B. at the boiling point of the solvent or diluent, at normal or elevated pressure or in the presence of an inert gas, e.g. B.
Nitrogen.
The reaction with O-lower alkyl isoureas can also be carried out in an analogous manner.
The reaction with cyanamides is carried out, for example, in such a way that a mixture of the alkenylenimino-lower alkylamine of the type mentioned above, especially a salt thereof, e.g. B. an addition salt with a mineral acid, such as hydrochloric acid, hydrobromic acid or sulfuric acid, and the cyanamide heated, the resulting melt in a solvent, for. The above-described iminoalkylamines can be reacted with an l-guanyl-pyrazole in the presence or absence of a diluent, for example by heating the mixture to its melting point or, in the presence of a solvent, to its boiling point. The presence of carbon dioxide is advantageously eliminated by using an inert gas.
The starting materials listed above are known or, if new, can be prepared by methods known per se. S-lower alkyl isothioureas or O-lower alkyl isoureas can be obtained, for example, by alkylating thioureas or ureas containing at least 1 hydrogen atom with lower alkyl halides, such as methyl or ethyl chloride, bromide or iodide, or di-lower alkyl sulfates, such as dimethyl or diethyl sulfate .

The new guanidine compounds can be obtained either as free compounds or in the form of their salts. A salt can in a manner known per se, for example by treatment with a strongly alkaline agent, such as aqueous alkali metal hydroxide, e.g. B. lithium, sodium or potassium hydroxide, or with strong anion exchange resins, such as quaternary ammonium exchange resins, are converted into the free compound. Therapeutically applicable addition salts can be prepared from the free bases with suitable inorganic or organic acids, for example those mentioned at the beginning. The reaction with acids is preferably carried out in suitable diluents, e.g. B. lower alkanols such as methanol, ethanol, n-propanol or i-propanol, ethers such as diethyl ether or dioxane, esters such as ethyl acetate, or mixtures of these.
Basic, neutral, acidic or mixed salts can be obtained here.
Primarily those starting materials are used which give the preferred compounds mentioned at the outset.

A mixture of 2 g of 2- (2,3-benzo-2,3-dehydrohexylenimino) ethylamine, 1.5 g of S-methyl isothiourea sulfate and 5 cm3 of water is allowed to boil for 4 hours on a reflux condenser, then cooled and obtained after concentrating the 2- (2,3-benzo-2,3-dehydro-hexylenemino) -ethyl-guanidine of the formula

The starting material can be obtained as follows:
14.7 g of 2,3-benzo-2,3-dehydrohexylenimine and 7.6 g of chloroacetonitrile are added with stirring to a suspension of 5.3 g of anhydrous sodium carbonate and 75 cm 3 of benzene, the reaction mixture is kept at the boil for 4 hours filtered after cooling.
After the benzene has been evaporated off, 2,3-benzo-2,3-dehydrohexylenimino-acetonitrile is obtained. This is dissolved in dry diethyl ether, lithium aluminum hydride is added and the mixture is allowed to boil for some time. After the usual work-up, 2- (2,3-benzo- 2,3 -dehydrohexylenimino) ethylamine is obtained.
Example 6
A mixture of 6.5 g of 2- (4,5-benzo-4,5-dehydrohexylenimino) ethylamine, 4.8 g of S-methylisothiourea sulfate and 8 cm3 of water is allowed to boil for 5 hours on the reflux condenser and is obtained cooling the reaction mixture, the 2- (4,5 benzo-4,5-dehydro-hexylenimino) -ethyl-guanidine of the formula

as sulfate. This melts after recrystallization from aqueous ethanol-acetone at 239-242.
The starting product can be obtained as follows:
The mixture of 75 g of 1,2-dicyanomethylbenzene, 10 g of Raney nickel and 500 cm3 of ethanol saturated with ammonia is hydrogenated for 5 hours at 75 atmospheres and 1000, the resulting reaction mixture is filtered and the filtrate is concentrated to dryness. The residue is then distilled under reduced pressure and 4,5-benzo-4,5-dehydrohexylenimine of bp. 123-124 and nD = 1.5600 is obtained, along with a much smaller amount of 1,2- (s-amino-ethyl ) -benzene with a boiling point of 0.15 90-98 and n2.5 = 1.5571.
A mixture of 20.4 g of 4,5-benzo-4,5-de-hydrohexylenimine, 11.6 g of chloroacetonitrile, 59.5 g of anhydrous sodium carbonate, 0.5 cm3 of water and 70 cm3 of toluene is heated with stirring 12 Hours to simmer. The hot reaction mixture is then filtered, the solvent is evaporated off under reduced pressure and the residue is crystallized from ethanol. This gives (4,5-benzo-4,5-dehydrohexylenimino) acetonitrile with a melting point of 113-116.
A solution of 19.2 g of the nitrile obtained in 100 cm3 of diethyl ether and 35 cm3 of tetrahydrofuran is added with stirring over the course of one hour to a suspension of 8.0 g of lithium aluminum hydride and 300 cm3 of diethyl ether, stirring is continued for a further 2 hours and the reaction mixture is left overnight stand at room temperature. Thereafter, 24 cm3 of ethyl acetate, 8 cm5 of water, 16 cm of 15% aqueous sodium hydroxide solution and another 24 cm of water are added, the mixture is filtered and the filtrate is concentrated. The residue is distilled under reduced pressure and the desired 2- (4,5-benzo-4,5-dehydrohexylenimino) ethylamine of bp 005 117-1400 is obtained.
Example 7
The mixture of 4.5 g of 2- [indolinyl (1)] ethylamine, 3.9 g of S-methyl isothiourea sulfate and 2 cm3 of water is allowed to boil for 5 hours on a reflux condenser. After cooling, the reaction mixture is rubbed several times with acetone, the white residue is filtered off and it is crystallized from aqueous ethanol-acetone. The 2- [indolinyl- (1)] -ethyl-guanidine of the formula is obtained

An aqueous solution of 2.5 g of 2- [indolinyl (l)] ethyl guanidine sulfate is made alkaline with concentrated sodium hydroxide solution while cooling, the precipitated oil is extracted with chloroform, the extract is dried and evaporated to dryness. The residue is dissolved in ethanol and the solution is acidified with ethanolic hydrochloric acid. A solution of 8.5 g of the nitrile obtained in 50 cm 3 of diethyl ether is added dropwise over 11/2 hours with stirring to a nitrogen suspension of 4.3 g of lithium aluminum hydride in 150 cm 3 of diethyl ether. After standing overnight at room temperature, the reaction mixture is successively mixed with 12 cm3 of ethyl acetate, 4 cm3 of water, 8 cm3 of 15% sodium hydroxide solution and another 2 cm3 of water and filtered. The filter residue is washed with diethyl ether, the filtrate is evaporated to dryness and the remaining 2- [indolinyl- (1)] -ethylamine is distilled under reduced pressure; Kp.17 = 1541600.
