Verfahren zur Herstellung von Benzoesäure-Derivaten Die Erfindung betrifft ein Verfahren zur Her stellung von neuen Benzoesäure-Derivaten, die eine tranquillisierende Wirkung besitzen.
Es wurde gefunden, dass Benzoesäureamide, in welchen das Stickstoffatom des Amids ein Glied eines Ringes ist, und der Benzolring zwei oder drei Meth- oxygruppen enthält, tranquillisierende oder atarak- tische Wirkung besitzen. Entsprechende Benzoesäure- derivate sind z.
B. die 3,4-Dimethoxy-benzoesäure, 3,4,5-Trimethoxy-benzoesäure, 2,3,4-, 2,4,5- und 2,4,6-Trimethoxy-benzoesäure. In der zur Bildung der Säueramide verwendeten zyklischen sekundären Aminen ist die NH-Gruppe zweckmässig ein Glied eines 5- oder 6gliedrigen Ringes. Ausser dem Amin- Stickstoffatom können auch weitere Heteroatome, wie Sauerstoff, Schwefel oder Stickstoff in dem Ring zu gegen sein. Bei dem Verfahren gemäss der Erfindung können z.
B. folgende sekundäre zyklische Amine verwendet werden: Piperidin, Pyrrolidin, Morpholin, Thiomorpholin und die hydrierten Derivate von Chinolin und Isochinolin. Diese zyklischen Amine können an den Kohlenstoffatomen auch Substituen- ten, wie Alkyl, Alkoxy, Aryl, Aralkyl und Halogen enthalten.
Die besagten zyklischen Amide der Ben- zoesäure werden erfindungsg-,mäss hergestellt, indem man mit 2 oder 3 Methoxygruppen substituierte Ben- zoesäure oder deren zur Bildung von Säureamid ge eignete Derivate mit einem sekundären Amin zur Reaktion bringt, in welcher die NH-Gruppe ein Glied eines Ringes ist.
So können die sekundären zyklischen Aminc mit Benzoesäurehalogeniden, Estern oder Aziden zu den gewünschten Säureamiden umgesetzt werden. Die Reaktion wird vorteilhaft in inertem Lösungsmittel, wie Benzol Chloroform, durchgeführt, man kann aber auch ohne Lösungsmittel oder in wässrig alkalischem Medium nach Schotten-Baumann arbeiten. Falls bei der Reaktion Säure freigesetzt wird, verwendet man als säurebindendes Mittel vorteilhaft das bei der Reaktion benützte sekundäre Amin oder ein tertiäres Amin. Das erhaltene Reaktionsprodukt kann aus dem Reaktionsgemisch in an sich bekann ter Weise, z.
B. durch Filtrieren oder Eindampfen oder durch Auskristallisieren gewonnen werden. Die Salzsäuresalze gewisser neuer Verbindungen, wie z. B. die der Pyrrolidin- und Piperidinderivate sind in Was ser gut löslich.
Die gemäss der Erfindung erzeugten neuen zyk lischen Amide der zwei oder drei Methoxygruppen tragenden Benzoesäure sind sehr wertvolle tranquilli- sierende Mittel niedriger Toxizität und mit sehr vor teilhafter therapeutischer Wirkung. So ist z. B. das 3,4,5-Trimethoxy-benzoyl-morpholin per os halb so toxisch, aber doppelt so aktiv als das bekannte 2 Methyl-2-N-propyl-1,3-propandiol-dicarbamat, wel ches in der Therapie als Tranquillizer oder Atarakti- kum benützt wird.
Ein weiterer, sehr wertvoller Vor teil des oben angeführten Morpholids, besteht darin, dass es die Haltungsreflexe und Koordinationsbewe gungen kaum beeinflusst. Durch diese Eigenschaften ist es ein vorzügliches, tagsüber zu verabreichendes Sedativmittel. Ein weiterer Vorteil besteht darin, dass es eine sedative Wirkung hat, ohne die Reflex-Irri- tabilität zu beeinflussen.
<I>Beispiel 1</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-piperidin-chlorhydrat werden 11,5 g 3,4,5-Tri- methoxy-benzoylchlorid in 100 ml wasserfreiem Ben zol gelöst und 10 ml Triäthylamin und unter Kühlen allmählich 4,7 g Piperidin hinzugefügt. Das Reak- tionsgemisch wird 2 Stunden lang unter Rückfluss gekocht. Das ausgeschiedene Triäthylamin-chlorhydrat wird abfiltriert, die Lösung mit Wasser gewaschen und eingedampft.
Der ölige Rückstand wird in 50 ml Äther gelöst und unter Eiskühlung mit Salzsäure- gas gesättigt. Die ausgeschiedene weisse Kristallmasse wird abfiltriert und aus wasserfreiem Äthanol umkri stallisiert. Ausbeute 14 g (88ö). Schmp. 133-134 C. Die Verbindung ist in Wasser leicht löslich.
<I>Beispiel 2</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-pyrrolidin-chlorhydrat werden 23 g 3,4,5-Tri- methoxy-benzoylchlorid in 100 ml Benzol gelöst und 20 ml Triäthylamin und unter Kühlen 8 g Pyrrolidin hinzugefügt. Das Gemisch wird 2 Stunden lang un ter Rückfluss gekocht. Das ausgeschiedene Triäthyl- amin-chlorhydrat wird abfiltriert, die Benzollösung mit Wasser gewaschen und eingedampft.
Das zu rückbleibende Öl wird in 100 ml Äther gelöst und die Lösung mit Salzsäuregas gesättigt. Das ausgeschie dene Chlorhydrat wird abfiltriert und aus Alkohol umkristallisiert. Die erhaltenen farblosen Lamellen sind in Wasser gut löslich. Ausbeute: 28 g (90ö). Schmp. 159-160 C.
<I>Beispiel 3</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-morpholin werden 46 g 3,4,5-Trimethoxy-ben- zoyl-chlorid in 300 ml Benzol gelöst und danach 25 g Triäthylamin und unter Eiskühlung allmählich 19 g Morpholin hinzugefügt. Die Lösung wird 2 Stunden lang unter Rückfluss gekocht. Die Lösung wird fil triert und danach mit verdünnter Schwefelsäure, dann mit Natriumhydrocarbonatlösung und zuletzt mit Wasser gewaschen und eingedampft. Die zurückblei bende gelbe ölige Substanz wird bald kristallin.
Die kristalline Masse wird mit Äther aufgenommen, fil triert und aus 90 ö igem Äthanol umkristallisiert. Die prismenartigen Kristalle sind in Wasser schwach lös lich. Ausbeute: 90,'rj, Schmp. 120-122 C.
Beispeil <I>4</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-thiomorpholid werden 5,12g Thiomorpholin in 40 ml Benzol gelöst und 10,3 g 3,4,5-Trimethoxy- benzoyl-chlorid, dann unter Eiskühlung und Rühren 7 ml Triäthylamin hinzugefügt. Das Reaktionsgemisch wird 2 Stunden lang unter Rückfluss gekocht.
Nach Abkühlen wird das ausgeschiedene Triäthylamin- chlorhydrat abfiltriert, das Filtrat mit verdünnter Salzsäure und dann mit Natriumhydrocarbonatlösung gewaschen, über Natriumsulfat getrocknet und ein gedampft. Die zurückbleibende ölige Substanz wird zweimal aus 50 öigem Alkohol umkristallisiert. Aus beute: 11 g, Schmp. 102-103' C.
<I>Beispiel 5</I> Zur Herstellung von N-(3,4-Dimethoxy-benzoyl)- morpholin werden 17,5 g 3,4-Dimethoxy-benzoyl- chlorid in 150 ml Benzol gelöst und unter Eiskühlung und Rühren ein Gemisch von 8,4 ml Morpholin und 12,2 ml Triäthylamin tropfenweise hinzugefügt. Das Reaktionsgemisch wird 1 Stunde lang gekocht, ab gekühlt, 100 ml Wasser hinzugefügt und die Benzol schicht abgetrennt. Die Benzollösung wird mit ver dünnter Salzsäure dann mit Natriumcarbonatlösung gewaschen und danach über Kaliumcarbonat getrock net.
Nach Abtrennen des Kaliumcarbonats wird das Lösungsmittel in Vakuum abgetrieben. Der Rückstand wird fraktioniert. 13,22 g N-(3,4-Dimethoxy-ben- zoyl)-morpholin wird erhalten, das unter 0,4 mm Druck bei 182-185 C siedet. Man erhält eine Mass gelbe, sehr viskose Substanz, welche in Wasser schwach löslich ist.
<I>Beispiel 6</I> Zur Herstellung von N-(2,3,4-Trimethoxy-ben- zoyl)-morpholin werden 10,23 g 2,3,4-Trimethoxy- benzoyl-chlorid in 65 ml Benzol gelöst und unter Kühlen und Rühren ein Gemisch von 4,5 ml Morpholin und 7 ml Triäthylamin tropfenweise hinzugefügt. Das Reaktionsgemisch wird 2 Stunden lang gekocht, gekühlt, filtriert und das Lösungsmittel in Vakuum abgetrieben. Der Rückstand wird zweimal aus Wasser umkristallisiert und mit Aktivkohle geklärt. Ausbeute: 7,22 g, Schmp. 84-86 C.
<I>Beispiel 7</I> Zur Herstellung von N-(2,4,6-Trimethoxy-ben- zoyl)-morpholin werden zu einem Gemisch von 6,79 g 2,4,6-Trimethoxybenzoesäure 50 ml Benzol und 2,6 ml Pyridin, 2,3 ml Thionylchlorid in 20 ml Ben zol tropfenweise hinzugefügt. Das Reaktionsgemisch wird 2 Stunden lang stehengelassen und danach unter Eiskühlung ein Gemisch von 8,4 ml Morpholin und 5 ml Benzol hinzugefügt. Das Reaktionsgemisch wird 5 Stunden lang gekocht, abgekühlt und 50 ml Wasser hinzugefügt.
Danach wird die Benzolschicht abge trennt, mit verdünnter Salzsäure und nachher mit einer Kaliumhyrdroxydlösung gewaschen. Die Benzol lösung wird über Kaliumcarbonat getrocknet und nach Abtrennen des Kaliumcarbonats der Benzol in Vakuum abgetrieben. Der Rückstand wird aus einem Gemisch von 7 ml Wasser und 1 ml Äthanol um kristallisiert. Ausbeute: 5,9 g, Schmp. 130-131'J C.
<I>Beispiel 8</I> Zur Herstellung von N-(2,4,5-Trimethoxy-ben- zoyl)-morpholin werden 10 g 2,4,5-Trimethoxy-ben- zoesäure-chlorid in 100 ml Benzol gelöst und 10 g Morpholin gelöst in 100 ml Benzol unter Kühlung hinzugefügt. Das Reaktionsgemisch wird über Nacht stehengelassen, das ausgeschiedene Morpholin-chlor- hydrat abfiltriert, der Benzol abgetrieben und der Rückstand in Vakuum destilliert. Unter 0,07 mm Druck siedet es bei 160-162' C und man erhält 9,5 g (8510) einen viskosen gelben Sirup.
Der Sirup wird in wenig warmen Benzol gelöst, bei Abkühlen scheidet das Morpholid kristallin aus. Die Abschei- dung wird durch Hinzufügung von Äther vervollstän digt, die ausgeschiedene Substanz wird filtriert und mit Äther gewaschen. Man erhält 8,5 g gelblichgrüne Kristalle, welche in Wasser wenig löslich sind. Schmp. 91-92' C, Ausbeute: 70%.
<I>Beispiel 9</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-morpholin wird ein Gemisch von 10,6 g 3,4,5- Trimethoxybenzoesäure, 50 ml Toluol, 4,4 ml Mor- pholin und 4,4 ml Phosphortrichlorid unter Rück fluss 4 Stunden lang gekocht. Das Lösungsmittel wird dann in Vakuum abgetrieben und der Rückstand mit 10 iger Natriumcarbonatlösung digeriert. Das ausgeschiedene gelbe Pulver wird abfiltriert und aus Wasser umkristallisiert.
Ausbeute: 9,55 g, Schmp. 120 bis 122' C. Die Mutterlaugen werden mit Benzol extrahiert, wonach man noch 0,22 g Substanz erhält. <I>Beispiel 10</I> Zur Herstellung von N-(3,4,5-Trimethoxy-ben- zoyl)-2-methyl-morpholin werden 23,1 g 3,4,5-Tri- m2thoxy-benzoylchlorid in 120 ml Benzol gelöst und 11 ml 2-Methylmorpholin und danach 17 ml Tri- äthylamin bei 0 C tropfenweise hinzugefügt und das Reaktionsgemisch 3 Stunden lang unter Rückfluss gekocht.
Nach Abkühlen wird die ausgeschiedene Substanz abfiltriert und die Lösung eingedampft. Dir Rückstand wird dreimal aus verdünntem Alkohol umkristallisiert. Ausbeute: 12,5 g, Schmp. 115 bis 117' C.
<I>Beispiel 11</I> Zur Herstellung von N-(2,3,4-Trimethoxy-ben- zoyl)-piperidin werden zu 10,97 g 2,3,4-Trimethoxy- K,nzoylchlorid in 60 ml Benzol gelöst 11 ml Piperidin unter Eiskühlung tropfenweise hinzugefügt und das Reaktionsgemisch wird danach 2 Stunden lang unter Rückfluss gekocht.
Nach Abkühlung wird die aus geschiedene Substanz abfiltriert. Die Benzollösung wird mit verdünnter Salzsäurelösung und dann mit Natriumhydrocarbonatlösung gewaschen und über wasserfreiem Kaliumcarbonat getrocknet. Nach Ent fernen des Kaliumcarbonats wird die Lösung ein gedampft. Der Rückstand wird aus acetonhaltigem Petroläther zweimal umkristallisiert. Ausbeute 7,94 g, Schmp. 77-57 C.

<I> Example 2 </I> To produce N- (3,4,5-trimethoxy-benzoyl) -pyrrolidine chlorohydrate, 23 g of 3,4,5-trimethoxy-benzoyl chloride are dissolved in 100 ml of benzene and 20 ml of triethylamine and, with cooling, 8 g of pyrrolidine added. The mixture is refluxed for 2 hours. The precipitated triethylamine chlorohydrate is filtered off, the benzene solution washed with water and evaporated.
The oil that remains is dissolved in 100 ml of ether and the solution is saturated with hydrochloric acid gas. The precipitated chlorohydrate is filtered off and recrystallized from alcohol. The colorless lamellae obtained are readily soluble in water. Yield: 28 g (90 °). M.p. 159-160 C.
<I> Example 3 </I> To prepare N- (3,4,5-trimethoxy-benzoyl) -morpholine, 46 g of 3,4,5-trimethoxy-benzoyl chloride are dissolved in 300 ml of benzene and then 25 g of triethylamine and 19 g of morpholine gradually added while cooling with ice. Example <I> 4 </I> To prepare N- (3,4,5-trimethoxy-benzoyl) -thiomorpholide, 5.12 g of thiomorpholine are dissolved in 40 ml of benzene and 10.3 g of 3,4,5- Trimethoxybenzoyl chloride, then 7 ml of triethylamine were added while cooling with ice and stirring. <I> Example 8 </I> To prepare N- (2,4,5-trimethoxy-benzoyl) -morpholine, 10 g of 2,4,5-trimethoxy-benzoic acid chloride are dissolved in 100 ml of benzene and 10 g of morpholine dissolved in 100 ml of benzene are added with cooling. The reaction mixture is left to stand overnight, the precipitated morpholine chlorohydrate is filtered off, the benzene is driven off and the residue is distilled in vacuo.
