Die Reduktion führt man vorzugsweise durch Behandeln mit Lithiumaluminiumhydrid in einem inerten Lösungsmittel wie Dioxan, Tetrahydrofuran u. a. und bei erhöhter Temperatur, z. B. 50-100 , durch.
Die neuen Verbindungen obiger Formel I sind farblose, meist schön kristallisierende Substanzen.
In organischen Lösungsmitteln sind sie mässig bis gut, in Wasser dagegen schwer löslich. Mit organischen und anorganischen Säuren bilden sie beständige, kristallisierte Salze, welche gut bis sehr gut wasserlöslich sind. Sie gegen mit dem Kellerschen Reagens (Eisen III-chlorid enthaltender Eisessig und konz. Schwefelsäure) und dem Van Urkschen Reagens (p-Dimethylaminobenzaldehyd und verd. Mineralsäure) charakteristische Farbreaktionen.
Die Verbindungen gehören zur pharmakologischen Gruppe der sogenannten Reserpin-Antagonisten, das heisst sie vermögen gewisse pharmakodynamische Effekte des Reserpins, insbesondere seine krampffördernde Wirkung, zu unterdrücken. Sie entfalten weiterhin gewisse antikonvulsive Eigenschaften, die sich u. a. an einer Hemmung der durch Cardiazol, Tryptamin, Nicotin oder Elektroschock erzeugten Streckkrämpfe zeigen. Ferner weisen sie eine Hemm wirkung gegenüber der Monoaminoxydase auf. Die Substanzen können zur Behandlung verschiedener psychischer Erkrankungen, insbesondere bei depressi ven Störungen, verwendet werden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden, Die Schmelzpunkte sind korrigiert.
Beispiel 1 3-(2'-Amino-butyl)-4-methyl-indol
Unter Feuchtigkeitsausschluss mischt man 36, 5 cm3 Dimethylformamid und 10 cm3 Phosphoroxychlorid bei 10-20 . Anschliessend gibt man während 30 Min. bei 20-300 eine Lösung von 13,1 g 4-Methyl-indol in 25 cms Dimethylformamid hinzu, erwärmt 45 Min. auf 35-37 und giesst dann das Reaktionsgemisch unter Rühren auf 100 g Eis und 100 cm3 Wasser.
Hierauf versetzt man innerhalb 30 Min. mit einer Lösung von 19,3 g Natriumhydroxyd in 200 cm3 Wasser, erhitzt 3 Min. zum Sieden, filtriert nach Erkalten und wäscht fünfmal mit je 50 cm3 Wasser.
Der 4-Methyl-indol -3- aldehyd kristallisiert aus Methanol/Chloroform in schrägen Würfeln vom
Kellersche Farbreaktion: negativ.
Smp. 192-1940.
Van Urksche Farbreaktion: orangerot.
9,84 g des so erhaltenen 4-Methyl-indol-3-aldehyds, 3,74 g Ammoniumacetat und 80 cm3 l-Nitropropan werden 6 Std. unter Rühren auf 105 erhitzt, dann abgekühlt und nach Zusatz von 100 cm3 Wasser kristallisieren gelassen. Das 3-(2'-Athyl-2'-nitrovinyl)- 4-methyl-indol kristallisiert in tief dunkelroten, 1 bis 2 cm langen Prismen. Umkristallisiert wird aus Methanol/Chloroform; Smp. 1721730.
Kellersche Farbreaktion: orange, nach 1 Sek. gelb.
Van Urksche Farbreaktion: orangegelb (Eigenfarbe).
Zu einer auf 50 erwärmten Lösung von 27 g Lithiumaluminiumhydrid in 270 cm3 abs. Tetrahydrofuran tropft man eine Lösung von 12,4 g des gemäss obigen Ausführungen erhaltenen 3 -(2'-Äthyl-2'-nitro- vinyl)-4-methyl-indols in 270 cm3 abs. Tetrahydrofuran und rührt 5 Std. bei 50". Anschliessend zersetzt man den Komplex und überschüssiges Lithiumaluminiumhydrid mit 80 cm3 Methanol und 130 cm3 gesättigter Natriumsulfatlösung, filtriert, wäscht das Filtrat mit Chloroform nach und verdampft es zur Trockne. Man schüttelt den Rückstand zwischen Äther und Weinsäure aus, stellt die saure Lösung unter Eiskühlung mit verd.
Natronlauge alkalisch, schüttelt mehrmals mit Äther rasch aus, trocknet die vereinigten Ätherextrakte über Natriumsulfat und dampft den Äther ab. 3-(2'-Aminobutyl)-4-methyl-indol bildet aus Chloroform Kristalle vom Smp. 1641660.
Kellersche Farbreaktion: braunviolett.
Van Urksche Farbreaktion: hellbraun.
Nach der gleichen Methode wie in Beispiel 1 ausführlich beschrieben, aber bei Anwendung von Nitroäthan anstelle von l-Nitropropan bzw. bei Einsatz von 4-Brom- anstelle von 4-Methyl-indol gelangt man über die entsprechenden Zwischenprodukte der Formeln III und W zu folgenden weiteren Verbindungen: Beispiel Endprodukt Smp.

Beispiel 5
4-Chlor-3-(2'-amino-prokyl)-indol
Man erhitzt 10,8 g 4-Chlor-3-(2'-methyl-2'-nitrovinyl)-indol in 100 cm3 abs.
Tetrahydrofuran mit 17 g Lithiumaluminiumhydrid 4 Std. unter Rückfluss, zersetzt den Komplex und überschüssiges Reduktionsmittel durch Eintropfen von Methanol, versetzt mit gesättigter Natriumsulfatlösung, filtriert und schüttelt das Filtrat zwischen Weinsäure und Äther aus. Man trennt die wässrige Phase ab, macht sie mit Natronlauge alkalisch, schüttelt mit Chloroform aus und kristallisiert den durch Verdampfen des Chloroforms erhaltenen Rückstand aus Essigsäure-äthylester. Prismen vom Smp. 123-1250.
Kellersche Farbreaktion: dunkellila.
Van Urksche Farbreaktion: hellrosa.
Das 4-Chlor-3-(2'-methyl - 2' - nitro - vinyl) - indol wird wie folgt hergestellt:
4-Chlor-indol wird mittels Dimethylformamid und Phosphoroxychlorid in bekannter Weise in 4-Chlor-indol-3-aldehyd übergeführt; Smp. 160-163 nach Umkristallisieren aus Wasser. Dann erhitzt man den Aldehyd mit Nitroäthan und Ammoniumacetat auf 105 . Nach Verdünnen mit Wasser kristallisiert die orange-rote Nitroverbindung in Prismen vom Smp. 195-196" C.
Beispiel 6
4-Chlor-3 -(2'-amino-butyl)-indol
Man erhitzt 16,3 g 4-Chlor-3 -(2'-äthyl-2'-nitrovinyl)-indol und 26 g Lithiumaluminiumhydrid in 100 cm3 abs. Tetrahydrofuran 4 Std. unter Rückfluss und arbeitet dann wie im vorhergehenden Beispiel auf.
Das rohe 4-Chlor-3-(2'-amino-butyl)-indol wird direkt in sein Methansulfonat übergeführt; Prismen vom Smp. 255-256" aus Essigsäureäthylester.
Kellersche Farbreaktion: blauviolett.
Van Urksche Farbreaktion: praktisch negativ.
Das Ausgangsprodukt, 4-Chlor-3-(2'-äthyl-2'nitrovinyl)-indol, erhält man durch Erhitzen von 4-Chlor-indol-3-aldehyd, der seinerseits wie in Beispiel 5 beschrieben erhalten wird, mit Nitropropan und Ammoniumacetat auf 1200 und Kristallisation des Rohproduktes aus Wasser. Orange-rote Prismen vom Smp. 196-197".

The reduction is preferably carried out by treating with lithium aluminum hydride in an inert solvent such as dioxane, tetrahydrofuran and the like. a. and at elevated temperature, e.g. B. 50-100 by.
The new compounds of the above formula I are colorless, mostly nicely crystallizing substances.
They are moderately to well soluble in organic solvents, but sparingly soluble in water. With organic and inorganic acids they form stable, crystallized salts, which are easily to very easily soluble in water. They counteract characteristic color reactions with Keller's reagent (glacial acetic acid and conc. Sulfuric acid containing iron III chloride) and Van Urk's reagent (p-dimethylaminobenzaldehyde and dilute mineral acid).
The compounds belong to the pharmacological group of so-called reserpine antagonists, that is to say they are able to suppress certain pharmacodynamic effects of reserpine, in particular its anti-convulsive effect. They continue to develop certain anticonvulsant properties, which may result. a. show an inhibition of the stretching convulsions caused by cardiazole, tryptamine, nicotine or electric shock. They also have an inhibitory effect on monoamine oxidase. The substances can be used for the treatment of various mental illnesses, in particular in the case of depressive disorders.
In the following examples, all temperatures are given in degrees Celsius. The melting points have been corrected.
Example 1 3- (2'-Amino-butyl) -4-methyl-indole
With the exclusion of moisture, mix 36.5 cm3 of dimethylformamide and 10 cm3 of phosphorus oxychloride at 10-20. A solution of 13.1 g of 4-methylindole in 25 cms of dimethylformamide is then added over 30 minutes at 20-300, the mixture is heated to 35-37 for 45 minutes and the reaction mixture is then poured onto 100 g of ice and 100 g of ice with stirring cm3 of water.
A solution of 19.3 g of sodium hydroxide in 200 cm3 of water is then added within 30 minutes, the mixture is heated to boiling for 3 minutes, filtered after cooling and washed five times with 50 cm3 of water each time.

9.84 g of the 4-methyl-indole-3-aldehyde thus obtained, 3.74 g of ammonium acetate and 80 cm3 of l-nitropropane are heated to 105 for 6 hours with stirring, then cooled and, after adding 100 cm3 of water, allowed to crystallize. The 3- (2'-ethyl-2'-nitrovinyl) -4-methyl-indole crystallizes in deep, dark red, 1 to 2 cm long prisms. It is recrystallized from methanol / chloroform; 1721730.
Keller's color reaction: orange, yellow after 1 sec.
Van Urk's color reaction: orange-yellow (inherent color).
To a heated to 50 solution of 27 g of lithium aluminum hydride in 270 cm3 of abs. Tetrahydrofuran is added dropwise to a solution of 12.4 g of the 3 - (2'-ethyl-2'-nitro-vinyl) -4-methyl-indole in 270 cm3 of abs. Tetrahydrofuran and stirred for 5 hours at 50 ". The complex and excess lithium aluminum hydride are then decomposed with 80 cm3 of methanol and 130 cm3 of saturated sodium sulfate solution, filtered, the filtrate is washed with chloroform and evaporated to dryness. The residue is shaken between ether and tartaric acid off, the acidic solution is ice-cooled with dil.
Sodium hydroxide solution is alkaline, shakes out quickly several times with ether, dries the combined ether extracts over sodium sulfate and evaporates the ether. 3- (2'-Aminobutyl) -4-methyl-indole forms crystals from chloroform with a melting point of 1641660.
Keller's color reaction: brown-violet.
Van Urk's color reaction: light brown.
Using the same method as described in detail in Example 1, but when using nitroethane instead of l-nitropropane or when using 4-bromine instead of 4-methyl-indole, the corresponding intermediates of the formulas III and W lead to the following further compounds: example end product m.p.

10.8 g of 4-chloro-3- (2'-methyl-2'-nitrovinyl) indole are heated in 100 cm3 of abs.
Tetrahydrofuran with 17 g of lithium aluminum hydride for 4 hours under reflux, decomposes the complex and excess reducing agent by dropping in methanol, mixed with saturated sodium sulfate solution, filtered and shakes the filtrate between tartaric acid and ether. The aqueous phase is separated off, made alkaline with sodium hydroxide solution, extracted with chloroform and the residue obtained by evaporation of the chloroform is crystallized from ethyl acetate. Prisms of m.p. 123-1250.
Keller's color reaction: dark purple.
Van Urk's color reaction: light pink.
The 4-chloro-3- (2'-methyl - 2 '- nitro - vinyl) - indole is produced as follows:
4-chloro-indole is converted into 4-chloro-indole-3-aldehyde in a known manner using dimethylformamide and phosphorus oxychloride; 160-163 after recrystallization from water. The aldehyde is then heated to 105 with nitroethane and ammonium acetate. After dilution with water, the orange-red nitro compound crystallizes in prisms with a melting point of 195-196 "C.
Example 6
4-chloro-3 - (2'-aminobutyl) indole
16.3 g of 4-chloro-3 - (2'-ethyl-2'-nitrovinyl) indole and 26 g of lithium aluminum hydride are heated in 100 cm3 of abs. Tetrahydrofuran under reflux for 4 hours and then works up as in the previous example.
The crude 4-chloro-3- (2'-aminobutyl) indole is converted directly into its methanesulfonate; Prisms with a melting point of 255-256 "made from ethyl acetate.
Keller's color reaction: blue-violet.
Van Urk's color reaction: practically negative.
The starting product, 4-chloro-3- (2'-ethyl-2'nitrovinyl) indole, is obtained by heating 4-chloro-indole-3-aldehyde, which in turn is obtained as described in Example 5, with nitropropane and Ammonium acetate to 1200 and crystallization of the crude product from water. Orange-red prisms of m.p. 196-197 ".
