Verfahren zur Herstellung von 9a- xysteroiden Diese Erfindung bezieht sich auf ein Verfahren zur mikrobiologischen Oxydation von Steroiden und den so erzeugten neuartigen Verbindungen.
Insbeson dere bezieht sie sich auf ein Verfahren zur Umwand lung eines 9a-Wasserstoff-1 1-oxysteraids der Pregnan- reihe oder der Allopregnanreihe zu einem 9a Hy- droxy-steroid durch die oxydierende Aktivität einer Art von Pilzen der Gattungen Cunninghamella und Helicostylum.
Vorzugsweise werden 11ss,21- Dioxy - 4,17(20)- pregnadien - 3 - an oder ein 21-Kohlenwasserstoff- carbonsäureester hiervon (USA-Patent Nr. 2 844 604) mit den oxydierende Eigenschaften besitzenden Pilz- arten der Gattungen Cunninghamella oder Helico- stylum behandelt, wobei 9a,21 Dioxy-4,17(20)- pregnadien-3,11-dion gebildet wird.
Diese Verbin dung kann in das bekannte physiologisch aktive 9a-Oxycortisonacetat umgewandelt werden (Fried et a1., J. Am. Chem. Soc. 79, 1130 [1957]), durch oxy- dative Oxylierung, nach Umwandlung in sein 21-Acetat, mit Osmiumtetroxyd und einem Oxyda tionsmittel gemäss dem Verfahren der USA-Patente Nrn. 2 769 823 und 2 769 825 und den nach stehend angeführten Beispielen.
Die oxydierenden Pilze, die in dem Verfahren dieser Erfindung verwendet werden, sind Arten der Gattung Cunninghamella aus der Familie Choane- phoraceae und der Ordnung Mucorales, und Arten der Gattung Helicostylum aus der Familie Thanni- diaceae, ebenfalls aus der Ordnung Mucorales. Diese Gattungen sind gut definiert, und typische Arten können von Kultursammlungen, wie z B.
der American Type Culture Collection, bezogen werden. Pilzarten, die indem Verfahren dieser Erfindung ver wendet werden können, umfassen C. blakesleeana, C. elegans, C. verticullata, C. echinulata, C. bertholle- tia und H. piriforme.
Die Verwendung dieser Pilze bei der mikrobiolo- gischen Umwandlung gewisser Steroide ist bereits be kannt. Im USA-Patent Nr. 2 602 769 wird ein Bei spiel einer Hydroxylierung eines 11 Methylensteroids mit Cunninghamella blakesleeana angeführt. USA- Patent Nr.
2 812 286 beschreibt die 11a Hydroxy- lierung eines 11-Methylensteroids mit Cunning- hamella echinulata. Ein Beispiel einer 8- und einer 14 - Hydroxylierung eines 11- Methylensteroids mit Helicostylum piriforme wird in den USA-Patenten Nm. 2 703 806, 2 602 769 und 2 670 358 bekannt gemacht.
Die 9 Hydroxylierung eines 11-Oxysteroids mit einem Pilz, wie er im Verfahren dieser Erfindung verwendet wird, war bisher unbekannt. Diese Pilze sind innerhalb der Ordnung der Pilze, die im Ver fahren vom USA-Patent Nr 2 602 769 verwendet werden, aber ihr Gebrauch zur Bioumwandlung eines 11-Oxysteroids um 9a-Oxy-ll-ketosteroid herzustel len, ist darin nicht beschrieben.
Das vorliegende Ver fahren liefert eine Methode, um das physiologisch aktive 9a-Oxycortison und seine 21 Ester oder Zwi schenprodukte für die Herstellung derselben darzu stellen, ohne das verhältnismässig teure Hydrocortison- acetat als Zwischenprodukt zu verwenden. Statt dessen kann J 1ss,21 Dioxy-4,17(20)-pregnadien-3-on und seine Ester, die nun einfach aus Progesteron dar gestellt werden können (USA-Patent Nr. 2 844 604), verwendet werden.
Ausgangsmaterialien für das Verfahren dieser Erfindung sind: 9a-Wasserstoff-11-oxysteroide der Pregnan-oder Allopregnanreihe, vorzugsweise Steroide der llss-Oxypregnanreihe. Besonders bevorzugt sind d4-3 Ketö-9-Wasserstoff-1lss-oxysteroide der Pregnan- reihe. Die Ausgangssteroide können z. B. die in den Ecken sitzenden Methylgruppen, die für die meisten Steroide charakteristisch sind, enthalten, eine Me- thylgruppe in den 2-, 6-, 7-, 16-, 17- und 21-Stel- lungen, Halogen, z.
B. ein Fluoratom in den 2-, 4-, 6-, 16- und 21-Stellungen, eine Hydroxy- oder Acyl oxygruppe in den 1-, 3-, 5-, 6-, 7-, 8-, 12-, 14-, 15-, 16-, 17- und 21-Stellungen, eine Ketogruppe in den 3-, 6-, 7-, 12-, 16- und 20-Stellungen, eine Doppel bindung in den 1-, 4-, 5-, 6-, 7-, 14-, 16-, 17(20)
- und 20-Stellungen und andere Gruppen in andern Stellungen des Moleküls, wie z. B. Cyan, Alkoxy-, Maleinsäureanhydrid-Addukt, Epoxy-, Carboalkoxy-, Carboxy-, Vinyl-, Äthinyl-, aldehydische und Lakton- gruppen.
Die als Ausgangsmaterialien verwendeten Steroide der Pregnanreihe, die 9a Wasserstoff-11-oxygruppe besitzen, umfassen die Progesterone, Pregnenolone, Pregnane, Pregnene, Pregnadiene und Pregnatriene. Bevorzugt sind die 44-3-Ketopregnene. Die l lss-Oxy- steroide sind die bevorzugten Ausgangsmaterialien, die in den meisten Fällen im Verfahren dieser Er findung zu 11-Ketosteroiden oxydiert werden.
Die 9ss-Oxy-11-oxydierten Produkte des Verfahrens die ser Erfindung umfassen eine grosse Anzahl physiolo gisch aktiver Vertreter (siehe brit. Patent Nr. 777 996, USA-Patente Nrn. 2 840 572, 2 840 578, 2 840 599 und 2 840 580), z. B. 9a-Hydroxycortison, die ent zündungshemmende Wirksamkeit besitzen. Andere Verbindungen, wie sie in diesem Verfahren herge stellt werden, z.
B. solche ohne die 44-3-Ketogruppe oder 20-Ketogruppe in der Pregnanreihe, können in Verbindungen der Progesteronreihe, z. B. d4-3,20- Diketo-progesteron, durch die bekannten Reaktionen umgewandelt werden.
Bei der Ausführung des Ver fahrens dieser Erfindung wird das ausgewählte Aus- gangssteroidsubstrat, welches 9a Wasserstoff-11-oxy- steroid wie oben beschrieben umfasst und im wesent lichen frei von 11-Methylensteroid ist, der oxydieren den Aktivität einer Kultur von Pilzen der Gattung Cunninghamella oder Helicostylum, vorzugsweise unter Bedingungen einer aeroben, bewegten, unter getauchten Gärung in einem Nährmedium,
welches assimilierbaren Stickstoff, Kohlenwasserstoffe und vorzugsweise auch Phosphor enthält, ausgesetzt.
Steroidsubstrat bedeutet das zugefügte Steroid und umfasst nicht diejenigen Steroide, die von dem Pilz auf natürliche Weise erzeugt werden. Man lässt den Pilz vorzugsweise einige Zeit, z. B. 24-48 Stun den, wachsen, bevor man das Steroidsubstrat hinzu- fügt. Nach der Zugabe des Steroids werden die Gä rungsbedingungen, vorzugsweise während einiger Stunden, z. B. 24 Stunden und mehr, beibehalten, bevor die Steroidumwandlungsprodukte abgetrennt werden.
Eine vollständige Diskussion verschiedener Gärungsbedingungen findet sich im USA-Patent Nr. 2 602 769.
Die Steroidumwandlungsprodukte werden mit Vorteil durch Extraktion des Gärungspilzkörpers und des Mediums mit einem Lösungsmittel isoliert. Zum Beispiel wird das Mycel filtriert und mit Aceton ge waschen, und das Medium wird mit Methylenchlorid oder einem andern mit Wasser nicht mischbaren Lösungsmittel extrahiert. Gewöhnlich findet man den grössten Teil des Steroidumwandlungsproduktes im Pilzkörper.
Die extrahierten Steroide können dann durch die bekannten Verfahren, z. B. fraktionierte Kristallisa tion oder Chromatographie, gereinigt werden. <I>Beispiel 1</I> 9a,21-Dioxy-4,17(20)-(cis)-pregnadien-3,11-dion Ein Gärapparat, der 100 Liter eines sterilen Me diums bei einem pH von 4,9, das aus handelsüblicher Dextrose (10 g pro Liter) und Corn steep-Feststoffen (20 g pro Liter) hergestellt wurde, enthält, wurde mit 5 Liter einer vegetativen Kultur von Helico- stylum piriforme Bain (A. T. C. C. 8992) geimpft.
Nach 24 Stunden kräftigen Rührens und Belüftung mit einem Volumen von 2,0 Liter pro Minute wur den 20,0 g von llss,21-Dioxy-4,17(20)-(cis)-pregna- dien-3-on (USA-Patent Nr. 2 844 604) als Lösung in 500 ml Aceton hinzugefügt und die Gärung 48 Stunden lang unter den gleichen Bedingungen fort gesetzt. Die Umwandlungsprodukte wurden nach der Methode von D. H. Peterson et a1., J.
Am. Chem. Soc. 74, 5933 (1952) extrahiert und ergaben etwa 42 g halbkristallinen Niederschlag, der in etwa 1 Liter trockenem Methylenchlorid aufgelöst wurde und über 2 kg Magnesiumsilikat (Florisil) chromato- graphiert wurde.
Die Kolonne wurde mit sechs 2-Liter-Portionen von jedem der folgenden Lösungs- mittelkompositionen entwickelt: Hexane (Skellysolve B) plus 180/9 Aceton, Hexane plus 20% Aceton, Hexane plus 2511/o Aceton und Hexane plus 500/a Aceton.
Die Rückstände der Fraktion 9-21, die Methy- lenfraktion mitgezählt, wurden vereinigt, in heissem Aceton aufgelöst, mit 2 g Holzkohle (Darco G-60) entfärbt, filtriert und das Filtrat konzentriert, bis reichliche Kristallisation einsetzte. Die Mischung wurde in einem Kühlschrank 2-3 Stunden lang ge kühlt und dann filtriert, wobei 10,33 g von 9a,21- Dioxy-4,17(20)-(cis)-pregnadien-3,11-dion mit einem Schmelzpunkt von 217-219 erhalten wurden.
Zu sätzliche Mengen an Kristallen wurden gesammelt, bis 12,31 g erhalten wurden, die aus Aceton um kristallisiert wurden und 9a,21-Dioxy-4,17(20)-(cis)- pregnadien-3,11-dion ergab, das bei 219,5-221 C schmolz und einen WD von + 173 C in Dioxan, einen .? mäh von 238,5 mu (a,%, 15,725), ---- vNuioi 3440, 3330, 1700, 1640 und 1605 cm-' max und die folgende Analyse aufwies:

Nach 24 Stunden war das pH 4,4. Dieser Mischung wurden 3,0 g l lfl,21-Dioxy-4,17(20)- (cis)-pregnadien-3-on in 30 ml heissen Propylengly- kols zugegeben und die Belüftung und Rührung während 24 Stunden fortgesetzt. Dann wurde der Inhalt des Gärbottichs mehrmals mit Methylen- chlorid extrahiert.
Das 9a,21-Dioxy-4,17(20)-(cis)-pregnadien-3,11- dion wurde aus dem Bierextrakt auf die gleiche Weise wie im Beispiel 1 beschrieben isoliert und seine Identität mit dem 9a,21-Dioxy-4,17(20)-(cis)- pregnadien-3,11-dion, das gemäss der Methode des Beispiels 1 erhalten worden war, bewiesen.
Gemäss dem Verfahren der Beispiele 1 und 2 wird 11/3,21-Dioxy-4,17(20)-(trans)-pregnadien-3-on in 9a,21 Dioxy-4,17(20)-(trans)-pregnadien- 3,11-dion, 11 a-Oxyprogesteron in 9a,lla-Dihydroxyprogesteron, 11f-Hydroxyprogesteron in 9a-Oxy-11-ketoprogesteron, 11,B,21 Dihydroxyprogesteron-21-acetat in 9a,21-Dioxy-11-ketoprogesteron umgewandelt.

Das erhaltene 9a,21-Dioxy-4,17(20)-(cis)-pregna- dien-3,11-dion kann folgendermassen verestert werden: Zu einer Lösung von 5,0 g 9a,21-Dioxy-4,17(20)- (eis)-pregnadien-3,11-dion in 15 ml trockenem Pyri- din von 20 C wurden 15 ml Essigsäureanhydrid zu gegeben. Nach 17 Stunden bei Zimmertemperatur wurde die Lösung in 450 ml einer Eiswasser mischung gegossen. Das kristalline Produkt, das dabei ausfiel, wurde gut mit Wasser gewaschen und getrocknet.
Man erhielt 5,53 g 9a,21-Dioxy-4,17(20)- (cis)-pregnadien-3,11-dion-21-acetat, das bei 172,5 bis 177 C schmolz. Die Umkristallisierung dieser Kristalle aus heissem Aceton, das filtriert und dann bis zum Einsetzen der Kristallisation auf einem Dampfbad konzentriert wurde, erhöhte den Schmelz punkt auf 188,5-190 C.

Zu einer eiskalten Lösung von 387 mg (1 mM.) 9a,21-Dioxy-4,17(20)-(eis) pregnadien-3,11-dion-21- acetat in 18,5 ml tertiären Butylalkohols, der 0,93 ml Pyridin und 0,185 ml Wasser enthielt, wurden 370 mg N - Methylmorpholinoxyd, 800 mg Phenyljodoso- acetat und 4,0 mg Osmiumtetroxyd zugegeben.
Die Aufschlämmung wurde 2 Tage lang bei 0-5 C ge rührt, 600 mg Filtersubstanz (Magnesoi) wurde zu gefügt, gefolgt von einer Lösung von 150 mg Na triumsulfit in 10 ml Wasser.
Die sich ergebende Mi schung wurde 15 Minuten lang bei Zimmertempera tur gerührt. Die Mischung wurde filtriert und das Filtrat unter vermindertem Druck konzentriert, bis die erste Spur einer Fällung einsetzte. Die Lösung wurde mit einem Minimum an tertiärem Butylalkohol geklärt und bei Zimmertemperatur 1 Stunde gerührt. Der sich ausscheidende Niederschlag wurde abfil- triert,
mit tert. Butylalkohol und Wasser gewaschen und dann getrocknet. Man erhielt 245 mg 9a,17a,21- Trihydroxy-4-pregnen-3,11,20-trion 21-acetat, das bei 233-236 C schmolz. Eine polymorphe Modifika tion wurde erhalten, welche bei 212-214 C schmolz. Umkristallisation aus einer Mischung von Aceton und Hexanen erhöhte den Schmelzpunkt auf 237 bis 239 C.

Process for the Preparation of 9Axysteroids This invention relates to a process for the microbiological oxidation of steroids and the novel compounds so produced.
In particular, it relates to a process for converting a 9a-hydrogen-11-oxysteraids of the Pregnan series or the Allopregnan series to a 9a hydroxy-steroid by the oxidizing activity of a type of fungus of the genera Cunninghamella and Helicostylum.
Preference is given to 11ss, 21-dioxy - 4,17 (20) - pregnadiene - 3 - an or a 21-hydrocarbon carboxylic acid ester thereof (US Pat. No. 2,844,604) with the oxidizing properties of the fungus species of the genera Cunninghamella or Helico-stylum treated, whereby 9a, 21 dioxy-4,17 (20) - pregnadiene-3,11-dione is formed.
This compound can be converted into the known physiologically active 9a-oxycortisone acetate (Fried et al., J. Am. Chem. Soc. 79, 1130 [1957]), by oxidative oxylation, after conversion into its 21-acetate, with osmium tetroxide and an oxidizing agent according to the method of US Pat. Nos. 2,769,823 and 2,769,825 and the examples below.
The oxidizing fungi used in the method of this invention are species of the genus Cunninghamella from the family Choanephoraceae and the order Mucorales, and species of the genus Helicostylum from the family Thannidiaceae, also from the order Mucorales. These genera are well defined, and typical species can be found in cultural collections such as
the American Type Culture Collection. Species of fungi that can be used in the method of this invention include C. blakesleeana, C. elegans, C. verticullata, C. echinulata, C. bertholle- tia, and H. piriforme.
The use of these mushrooms in the microbiological conversion of certain steroids is already known. U.S. Patent No. 2,602,769 gives an example of hydroxylation of an 11 methylene steroid with Cunninghamella blakesleeana. USA patent no.
2,812,286 describes the 11a hydroxylation of an 11-methylene steroid with Cunninghamella echinulata. An example of 8- and 14- hydroxylation of an 11-methylene steroid with Helicostylum piriforme is given in U.S. Patents Nm. 2 703 806, 2 602 769 and 2 670 358.
The 9 hydroxylation of an 11-oxysteroid with a fungus as used in the method of this invention was heretofore unknown. These mushrooms are within the order of the mushrooms used in the United States Patent No. 2,602,769, but their use to bioconvert an 11-oxysteroid to produce 9a-oxy-II-ketosteroid is not described therein.
The present process provides a method to represent the physiologically active 9a-oxycortisone and its 21 esters or intermediates for the production of the same without using the relatively expensive hydrocortisone acetate as an intermediate. Instead, J 1ss, 21 dioxy-4,17 (20) -pregnadien-3-one and its esters, which can now be easily made from progesterone (US Pat. No. 2,844,604), can be used.
Starting materials for the process of this invention are: 9a-hydrogen-11-oxysteroids of the pregnane or allopregnan series, preferably steroids of the llss-oxypregnan series. Particularly preferred are d4-3 keto-9-hydrogen-lss-oxysteroids of the pregnan series. The starting steroids can e.g. B. contain the methyl groups in the corners, which are characteristic of most steroids, a methyl group in the 2-, 6-, 7-, 16-, 17- and 21-positions, halogen, e.g.
B. a fluorine atom in the 2-, 4-, 6-, 16- and 21-positions, a hydroxy or acyl oxy group in the 1-, 3-, 5-, 6-, 7-, 8-, 12- , 14-, 15-, 16-, 17- and 21-positions, a keto group in the 3-, 6-, 7-, 12-, 16- and 20-positions, a double bond in the 1-, 4- , 5-, 6-, 7-, 14-, 16-, 17 (20)
- and 20 positions and other groups in other positions of the molecule, e.g. B. cyano, alkoxy, maleic anhydride adduct, epoxy, carboalkoxy, carboxy, vinyl, ethynyl, aldehydic and lactone groups.
The steroids of the pregnan series used as starting materials, which have 9a hydrogen-11-oxy group, include the progesterones, pregnenolones, pregnans, pregnenes, pregnadienes and pregnatrienes. The 44-3 ketopregnenes are preferred. The lss-oxy steroids are the preferred starting materials which in most cases are oxidized to 11-keto steroids in the process of this invention.
The 9ss-oxy-11-oxidized products of the process of this invention comprise a large number of physiologically active agents (see British Patent No. 777,996, U.S. Patent Nos. 2,840,572, 2,840,578, 2,840,599 and 2) 840 580), e.g. B. 9a-hydroxycortisone, which have anti-inflammatory effectiveness. Other compounds, as they are Herge in this process, z.
B. those without the 44-3-keto group or 20-keto group in the pregnane series can be used in compounds of the progesterone series, e.g. B. d4-3,20-diketo-progesterone, can be converted by the known reactions.
In practicing the method of this invention, the selected starting steroid substrate, which comprises 9a hydrogen-11-oxysteroid as described above and is essentially free of 11-methylene steroid, will oxidize the activity of a culture of fungi of the genus Cunninghamella or Helicostylum, preferably under conditions of aerobic, agitated, submerged fermentation in a nutrient medium,
which contains assimilable nitrogen, hydrocarbons and preferably also phosphorus.
Steroid substrate means the steroid added and does not include those steroids that are naturally produced by the fungus. The mushroom is preferably left for some time, e.g. B. 24-48 hours before adding the steroid substrate. After the addition of the steroid, the fermentation conditions, preferably for a few hours, e.g. B. 24 hours and more, before the steroid conversion products are separated.
A full discussion of various fermentation conditions can be found in U.S. Patent No. 2,602,769.
The steroid conversion products are advantageously isolated by extracting the fermentation fungus body and the medium with a solvent. For example, the mycelium is filtered and washed with acetone, and the medium is extracted with methylene chloride or some other water immiscible solvent. Most of the steroid conversion product is usually found in the mushroom body.
The extracted steroids can then be prepared by known methods, e.g. B. fractional crystallization or chromatography, be purified. <I> Example 1 </I> 9a, 21-Dioxy-4,17 (20) - (cis) -pregnadiene-3,11-dione A fermentation apparatus that can produce 100 liters of a sterile medium at a pH of 4.9 , which was produced from commercial dextrose (10 g per liter) and corn steep solids (20 g per liter) was inoculated with 5 liters of a vegetative culture of Helico-stylum piriforme Bain (ATCC 8992).
After 24 hours of vigorous stirring and aeration with a volume of 2.0 liters per minute, 20.0 g of llss, 21-dioxy-4,17 (20) - (cis) -pregnadien-3-one (USA -Patent No. 2 844 604) was added as a solution in 500 ml of acetone and fermentation continued for 48 hours under the same conditions. The conversion products were determined by the method of D. H. Peterson et al., J.
At the. Chem. Soc. 74, 5933 (1952) and gave about 42 g of semicrystalline precipitate, which was dissolved in about 1 liter of dry methylene chloride and chromatographed over 2 kg of magnesium silicate (Florisil).
The column was developed with six 2 liter portions of each of the following solvent compositions: hexanes (Skellysolve B) plus 180/9 acetone, hexanes plus 20% acetone, hexanes plus 2511 / o acetone and hexanes plus 500 / a acetone.
The residues of fraction 9-21, including the methylene fraction, were combined, dissolved in hot acetone, decolorized with 2 g of charcoal (Darco G-60), filtered and the filtrate concentrated until abundant crystallization began.
