die in 1,2-Stellung eine weitere Doppelbindung auf weisen können, der aeroben Einwirkung von Enzymen unterwirft, die die Hydroxylgruppe in die 11-Stellung einzuführen vermögen. In erhaltenen Verbindungen können, falls erwünscht, freie Hydroxygruppen ver- estert oder zu Oxogruppen dehydriert werden.
Die verwendeten Enzyme können mikrobiologi scher Herkunft sein, wobei man die Ausgangsstoffe z. B. mit lebenden Mikroorganismen aerob irrkubiert, die Sauerstoff in 11-Stellung einzuführen vermögen. Die verwendeten Mikroorganismen sind z.
B. solche der Gruppen Mucorales, Penicillium, Actinomyceten, wie Streptomyceten, Aspergillus, Cunninghamella, Curvularia, insbesondere Rhizopus nigricans, Cur- vularia lunata, Cunninghamella blakesleana, Cunning- hamella bainieri,
Cunninghamella elegans, Cunning- hamella echinulata.
Zur Durchführung des mikrobiologischen Verfah rens kann man die Ausgangsstoffe mit Kulturen der genannten Mikroorganismen unter an sich bekannten aeroben Bedingungen inkubieren. Das Wachstum er folgt in Oberflächenkultur oder, technisch vorteilhaft, submers, wobei man schüttelt oder rührt. Die Kul turen enthalten assimilierbaren Kohlenstoff, insbeson dere Kohlenhydrate, sowie gegebenenfalls Wuchs stoffe, beispielsweise Maisquellwasser oder Bierwürze, und anorganische Salze. Es sind somit natürliche, synthetische oder halbsynthetische Nährlösungen brauchbar.
Das praktisch einfachste Verfahren ist im folgenden geschildert, ohne dass die Erfindung durch diese Angaben beschränkt sein soll: Man züchtet die Organismen in Apparaturen und unter- ähnlichen Be dingungen, wie sie bei der Antibiotika-Fabrikation als sog. Tieftankverfahren bekannt sind. Nach Entwick lung der Kulturen gibt man die genannten Ausgangs stoffe in feiner Dispersion oder Lösung, beispiels weise in Methanol, Aceton oder Äthylenglykol zu und irrkubiert weiter.
Schliesslich trennt man vom Mycel ab, extrahiert das Filtrat und/oder die Mycelmasse und isoliert aus dem Extrakt die 11-Hydroxyverbin- dungen in an sich bekannter Weise, z. B. durch Ent- mischungsverfahren, Adsorption, Chromatographie, Kristallisation, Überführung in funktionelle Derivate, wie Ester und dergleichen.
Dieselben Umsetzungen lassen sich auch durchführen, indem man die wirk samen Enzyme aus entsprechenden aeroben Kulturen der genannten Organismen zuerst abtrennt und unter Ausschluss der wachsenden Kulturen verwendet. So kann man z. B. das von entsprechenden aeroben Kul turen der genannten Organismen gebildete Mycel ab trennen, in Wasser oder Pufferlösungen suspendieren, die genannten Ausgangsstoffe diesen Aufschlämmun- gen zugeben und inkubieren.
Anstelle von Enzymen mikrobiologischer Her kunft können auch z. B. solche aus tierischen Or ganen, insbesondere Nebennieren von Rindern oder Schweinen, Verwendung finden. Das hiezu einzu schlagende Verfahren, in erster Linie die Anwendung von Homogenaten, ist ebenfalls an sich bekannt und wird in den Beispielen näher beschrieben.
Die erhaltenen Verbindungen lassen sich ins besondere in 21-Stellung verestern. Für die Gewin nung der Ester dienen Anhydride, Halogenide oder Thiolderivate von gesättigten oder ungesättigten ali- phatischen oder cycloaliphatischen, von aromatischen oder heterocyclischen Carbonsäuren, z. B. solchen mit 2 bis 12 Kohlenstoffatomen.
Für die Überführung von 11a- und 11/3-Hydroxy- verbindungen in die entsprechenden I1-Ketone ver wendet man die üblichen Dehydrierungsmittel, z. B. Chromtrioxyd in Eisessig oder den Chromtrioxyd- Pyridin-Komplex.
Die Verfahrensprodukte zeichnen sich durch eine hohe biologische Wirkung im Leberglykogen- und Granulomtest aus. Im Gegensatz zu manchen in 6a- Stellung nicht halogenierten Corticosteroiden zeigen die neuen Verfahrensprodukte die Nebenwirkung der Retention von Natrium nicht oder nur in untergeord netem Masse.
Die als Ausgangsstoffe dienenden d4-Pregnen- und dl.-I-Pregnadienverbindungen lassen sich nach dem in den Patenten Nrn. 372663, 374660, 377343 und 372661 beschriebenen Verfahren gewinnen.
In den nachfolgenden Beispielen sind die Tem peraturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 1 Liter einer sterilisierten Nährlösung, enthaltend 20 g Pepton und 5 cm3 Corn steep liquor, wird mit 30 cm3 einer wachsenden Kultur von Rhizopus nigri- cans, die durch Impfen mit Sporen des genannten Pil zes (auf Agar gezüchtet) erhalten wird, vermischt. Die Mischung wird 24 Std. bei 28 unter Belüftung ge rührt.
Am Ende der Inkubationszeit beträgt das pH der Kultur 3 bis 4, und das Wachstum des Pilzes ist gut entwickelt. 33 cms einer äthanolischen Lösung von 14-6a-Chlorpregnen-17a,21-diol-3,20-dion, ent haltend 330 mg des Steroids, wird dann zur Kultur gegeben und die Mischung 24 Std. bei 28 unter Be lüftung gerührt. Die Inkubationsmischung wird mit Methylenchlorid extrahiert, der Extrakt mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und im Vakuum vorsichtig konzentriert.
Die ein geengte Lösung wird an Silicagel chromatographiert. Elution mit Chloroform-Äther liefert das reine :-F-6a- Chlor-pregnen-11 a,17a,21-triol-3,20-dion; .?",;a, 238 m, ,<I>log e</I> = 4,11, [a],, = +74 . Dasselbe Umsetzungsprodukt erhält man, wenn von einem 21-Ester, z. B. einem solchen mit 2 bis 12 Kohlenstoffatomen, von 4-1-6a-Chlor-pregnen-17a,21- diol-3,20-dion ausgegangen wird.
1 g des erhaltenen Triols löst man in 5 cm:' Pyri- din, kühlt auf 0 ab, vermischt bei etwa 0 - mit 0,28 cm-' Acetanhydrid (etwa 1,1 Äquivalente) und lässt die Mischung 1 Std. bei 0 und 4 Std. bei Raum temperatur stehen. Dann wird in Wasser gegossen und mit Methylenchlorid extrahiert. Der Extrakt wird anschliessend mit verdünnter Salzsäure, 5 /oiger Sodalösung und Wasser gewaschen, über Natrium sulfat getrocknet, filtriert und zur Trockne ein gedampft.
Kristallisation des Rückstandes liefert das reine 21 -Acetat von d-1-6a-Chlor-pregnen-11rt,17a,21- triol-3,20-dion; d""@, 238 m,u, log f = 4,14, [a]D = + 69 .
Wird für die Veresterung anstelle von Acetan- hydrid Propionsäureanhydrid verwendet, so erhält man als Endstoff das 21-Propionat von J-1-6a-Chlor- pregnen -11 a,17a,21- triol - 3,20-dion; 7"" @, 237 mp, log E = 4,09, lab = +66 .
Eine Lösung von 1 g des erhaltenen 21-Acetates von 4-1-6a <I>-</I> Chlor-pregnen-11 a,17a,21-triol-3,20-dion in 30<B>CM 3</B> Eisessig wird unter Rühren tropfenweise mit 5 cm3 einer 80 /eigen Lösung von Chromsäure in Eisessig vermischt, 2 Std. bei Raumtemperatur ste hengelassen und in Wasser gegossen. Das Oxydations produkt wird mit Essigester extrahiert, der Extrakt mit Wasser, 511/aiger Sodalösung und Wasser ge waschen, über Natriumsulfat getrocknet, filtriert und zur Trockne verdampft.
Kristallisation des Rückstan des aus Aceton-Hexan liefert das 21-Acetat von Jl- 6a-Chlor-pregnen-17rt,21-diol-3,11,20-trion; F. 197 , [a]D = + 176 .
<I>Beispiel 2</I> 1 Liter einer sterilisierten Nährlösung, enthaltend 20 g Pepton und 50 ein- Corn steep liquor, wird mit 30 cm3 einer wachsenden Kultur von Curvularia lunata, die durch Impfen mit Sporen des genannten Pilzes (auf Agar gezüchtet) erhalten wird, vermischt. Die Mischung wird 48 Std. bei 28 unter Belüftung gerührt. Am Ende der Inkubationszeit beträgt der pH der Kultur 3 bis 4, und das Wachstum des Pilzes ist gut entwickelt.
Das Kulturmedium wird dann mit 33 cm3 einer äthanolischen Lösung von d-1-6a-Chlor- pregnen-17a,21-diol-3,20-dion, enthaltend 330 mg des Steroides, vermischt und 48 Std. unter Belüftung gerührt.
Das Umsetzungsprodukt wird mit Methylen- chlorid extrahiert und der Extrakt gewaschen, ge trocknet, konzentriert und an Silicagel wie in Beispiel 1 beschrieben chromatographiert, wobei das<I>44-6a-</I> Chlor - pregnen - l lss,17a,21- triol-3,20-dion erhalten wird, das mit einem authentischen Muster identisch ist.
Eine Lösung von 1 g des erhaltenen Triols in 5 cm 3 Pyridin wird mit 1 cm3 Acetanhydrid ver mischt, 4 Std. bei Raumtemperatur stehengelassen und in Wasser gegossen. Anschliessend wird mit Methylen- chlorid extrahiert und der Extrakt mit verdünnter Salzsäure, 5 /aiger Sodalösung und Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und im Va kuum zur Trockne verdampft.
Kristallisation des Rückstandes aus Aceton-Hexan liefert das 21-Acetat von A4-6a-Chlor-pregnen-11/3,17a,21-triol-3,20-dion, das mit einem authentischen Muster identisch ist; F. 174-176 , [a]D = +97 .
Wird anstelle der freien Verbindung das 21 - Propionat von 44- 6a - Chlorpregnen-17a,21- diol-3,20-dion eingesetzt, so erhält man eben falls das 44 - 6a - Chlor - pregnen - l lss,17a, 21 - triol-3,20-dion, das bei der Veresterung mit Pro- pionsäureanhydrid das entsprechende 21-Propionat liefert; A",a, 238 my, log E = 4,12.
<I>Beispiel 3</I> 1 g 44 - 6a - Chlor-pregnen-17a,21-diol-3,20-dion oder ein 21-Ester davon wird nach den Angaben in Beispiel 2 inkubiert und aufgearbeitet. Das erhaltene Rohprodukt wird mit 5 cm3 Pyridin und 1 cm3 Acetanhydrid in 21-Stellung verestert.
Zur Reinigung wird das erhaltene rohe 21-Acetat an 20 g Alu miniumoxyd chromatographiert. Elution mit Benzol- Äther (60:40) liefert das 21-Acetat von 44-6a Chlor - pregnen - l lss,17a,21-triol-3,20-dion, das mit dem in Beispiel 2 beschriebenen Präparat identisch ist.
In analoger Weise erhält man das 21-Propionat von 44-6a-Chlor-pregnen-11/3,17a,21-triol-3,20-dion. Das 21-Acetat von 44-6a-Chlor-pregnen-llss,17a, 21-triol-3,20-dion wird nach den Angaben in Beispiel 1 mit Chromsäure oxydiert. Das erhaltene 21-Acetat von 44-6a -Chlor - pregnen-17a,21-diol-3,11,20-trion ist mit dem in Beispiel 1 beschriebenen Präparat iden tisch.
Ausgehend vom entsprechenden 21-Propionat er hält man das 21-Propionat von 44-6a-Chlor-pregnen- 17a,21-diol-3,11,20-trion.
<I>Beispiel 4</I> 1 Liter einer sterilisierten Nährlösung, enthaltend 20 g Pepton und 50 cm3 Corn steep liquor, wird mit 30 cm3 einer wachenden Kultur von Curvularia lunata, die durch Impfen mit Sporen des genannten Pilzes (auf Agar gezüchtet) erhalten wird, vermischt. Die Mischung wird dann 48 Std. bei 28 unter Be lüftung gerührt. Am Ende der Inkubationszeit be trägt das pH der Kultur 3 bis 4, und das Wachstum des Pilzes ist gut entwickelt.
Pro 1/9 Liter der erhal- tenen Kultur gibt man 33 cms einer 1 % igen äthanoli- schen Lösung von 41,4-2-Methyl-6a-chlor-pregnadien- 17a,21-diol-3,20-dion. Die Mischung wird unter Be lüftung 48 Std. bei 28 gerührt, mit Methylenchlorid extrahiert und der Extrakt mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und im Va kuum ohne überhitzen auf ein kleines Volumen ein gedampft.
Die konzentrierte Lösung aus verschie denen einzelnen Inkubationen wird vereinigt und an Silicagel chromatographiert. Elution der Säule mit Mischungen von Chloroform-Äther liefert kristalline Fraktionen, die vereinigt und aus Aceton-Hexan um kristallisiert werden, wobei das 41,4-2-Methyl-6a- chlor-pregnadien-11ss,17a,21-triol-3,20-dion erhalten wird; A."., 248 my, log a = 4,19, [a]D = +70 .
Diese Verbindung lässt sich wie in den vorher gehenden Beispielen beschrieben in ihre 21-Ester überführen.
In analoger Weise wird das d4-2a-Methyl-6a- chlor - pregnen -17a,21-diol-3,20-dion in das 44-2a Methyl - 6a -chlor-pregnen-11 ss,17a,21-triol-3,20-dion übergeführt;
A ",a238 my, log a = 4,09, [a]D = +l0311. <I>Beispiel 5</I> Es wird eine Kultur von Rhizopus nigricans durch Impfen eines wässrigen Mediums, enthaltend 21% Pepton und 5 m/a Corn steep liquor, mit dem genann ten Pilz und Inkubation während 24 Std.
unter Be lüftung und Rühren hergestellt. Zu jedem Liter der Kultur gibt man 30 cm3 einer äthanolischen Lösung von<I>44-6a</I> - Fluor - pregnen-17a,21-diol-3,20-dion, die 300 mg des Steroides enthält. Die Mischung wird während 24 Std. bei 20 unter Belüftung gerührt. Das Inkubationsprodukt wird dann mit mehreren Portio nen Methylenchlorid extrahiert, der Extrakt mit Was ser gewaschen, über Natriumsulfat getrocknet, filtriert und im Vakuum auf ein kleines Volumen eingeengt.
Chromatographie des Konzentrates an Silicagel liefert das 44-6a-Fluor-pregnen-lla,17a,21-triol-3,20-dion; Ama, 237 my, log E = 4,22, lab = +160 .
Eine Mischung von 1 g der obigen Verbindung, 10 cm3 Pyridin und 1 cm3 Acetanhydrid lässt man über Nacht bei Raumtemperatur stehen, giesst dann in Wasser und erhitzt 1/2 Std. auf dem Dampfbad.
Nach dem Abkühlen wird abgenutscht, der Filterkuchen ge waschen, getrocknet und durch Kristallisation aus Aceton-Hexan gereinigt, wobei das 21-Acetat von <I>d4 - 6a</I> - Fluor-pregnen-11 a,17a,21-triol-3,20-dion er halten wird; Am., 237 my, log a = 4,23, [a]D = +152 .
1 g des obigen 21-Acetates wird in 50 cm3 Aceton gelöst, auf 0 gekühlt und mit einer 8n Chromsäure lösung, die durch Auflösen von 1,6 g Chromsäure in verdünnter Schwefelsäure erhalten wird, versetzt. Die Mischung rührt man 5-10 Minuten bei 0 , giesst in Wasser, extrahiert mit Äther, wäscht mit Wasser, trocknet den Äther über Natriumsulfat und verdampft ihn im Vakuum. Aus dem Rückstand erhält man durch Kristallisation aus Aceton-Hexan das 44-6a Fluor-pregnen-17a,21-diol-3,11,20-trion.
<I>Beispiel 6</I> Wird in Beispie17 die Kultur von Rhizopus nigricans durch eine solche von Curvularia lunata ersetzt, die un ter ähnlichen Bedingungen durch 48stündige Inkuba tion erhalten wird, so erhält man ausgehend von d4- 6a-Fluor-pregnen-17a,21-diol-3,20-dion nach 48stün- diger Inkubation bei 29 und Aufarbeitung gemäss den Angaben in Beispiel 5 das 44-6a-Fluor-pregnen-llss, 17a,21-triol-3,20-dion (F. l92-201 ,
[a]D = +127 ), das sich nach den Angaben in den vorhergehenden Beispielen in das 21-Acetat bzw. einen andern 21- Ester und durch Chromsäureoxydation in das 21- Acetat bzw. einen andern 21-Ester von d4-6a-Fluor- pregnen -17a,21- diol-3,11,20-trion überführen lässt; das 21-Acetat davon schmilzt bei 215-2l6 , [a]D = <B>+1900.</B>
Eine gerührte Lösung von 500 mg des 21-Acetates von 44-6a-Fluor-pregnen-11ss,17a,21-triol-3,20-dion in 300 cm3 Eisessig wird vorsichtig mit einer Lösung von 130 mg Chromsäure in 1 cm3 Wasser und 8 cm Eisessig versetzt. Die Mischung lässt man 2 Std. bei Raumtemperatur stehen, giesst in Wasser, nutscht ab, wäscht und trocknet den Rückstand und kristallisiert ihn aus Aceton-Hexan, wobei das 21-Acetat von 44-6a-Fluor-pregnen-17a,21-diol-3,11,20-trion erhal ten wird; F. 215-216 , [a]D = +190 .
<I>Beispiel 7</I> Es werden die Lösungen A, B und C in destillier tem Wasser wie folgt bereitet: Lösung A durch Mischen von 425 cm- einer 1,742o/oigen Dikalium- hydrogenphosphat-Lösung mit 75 cm3 einer 1,38 o/a.- igen Mononatriumdihydrogenphosphat-Lösung;
Lö sung B wird erhalten durch Verdünnen von 1 Liter einer 4,5 11/aigen Natriumchlorid-Lösung, 40 cm3 einer 5,75 % igen Kaliumchlorid-Lösung und 10 cm3 einer 19,1 o/o.igen Magnesiumsulfat-Lösung auf 5 Liter;
Lösung C wird erhalten durch Auflösen von 20,9 g Fumarsäure und 14,4 g Natriumhydroxyd in 1 Liter Wasser und Verdünnen auf 1,2 Liter. Dann werden 475 cm3 Lösung A, 4,32 Liter Lösung B und 1,2 Liter Lösung C vermischt.
Nebennieren von frisch geschlachteten Rindern werden vom Fett befreit und dann in einer Fleisch- hackmaschine behandelt, bis eine homogene Masse erhalten wird. 3 kg dieser Masse werden in 6 Liter des obigen Gemisches der Lösungen A, B und C gegeben und stark gerührt.
Dann gibt man eine Lösung von 3 g des 21- Acetates von 44-6a-Chlor-pregnen-17a,21-diol-3,20- dion in 16 cm3 Propylenglykol zu und rührt die Mischung bei 28-37 während 3 Std. Darauf gibt man 40 Liter Aceton zu und rührt noch 1 Std. bei Raum temperatur.
Die feste Masse wird durch Filtration entfernt und zweimal mit je 10 Liter Aceton gewaschen. Die ver einigten Filtrate werden im Vakuum unter 30 auf etwa 5 Liter eingeengt. Der wässrige Rückstand wird nun dreimal mit 4 Liter Hexan gewaschen. Die so be handelte wässrige Lösung wird zweimal mit je 3 Liter Methylenchlorid extrahiert, der Extrakt mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum unterhalb Raumtemperatur auf etwa 300 cm-' eingeengt. Die eingeengte Lösung wird auf eine Säule aus einer Mischung von 90 g Silicagel und 90 g Celite gegeben.
Die Säule wird nun mit einer Mischung von 900 cm- Methylenchlorid und 100 cm3 Aceton und schliesslich mit einer solchen von 1600 cm.' Methylenchlorid und 400 cm3 Aceton gewaschen. Mit dem letzteren Lösungsmittelgemisch wird das 44-6a-Chlor-pregnen-11/3,17(,i,21-triol-3,20-dion elu- iert, das durch Kristallisation aus Äther gereinigt wird; A",1\ 236-238 my, log a = 4,14.
In analoger Weise erhält man, ausgehend vom i4-6a-Fluor-pregnen-17a, 21-diol-3,20-dion, das J4-6a-Fluor-pregnen-11/3,17a, 21-triol-3,20-dion.
1 g des Triols wird in 10 cm3 Pyridin gelöst und mit Acetanhydrid vermischt. Die Mischung lässt man über Nacht bei Raumtemperatur stehen, giesst in Eis wasser, rührt 1/2 Std. und filtriert vom Niederschlag ab, wäscht und trocknet ihn und kristallisiert aus Aceton-Hexan, wobei das 21-Acetat von 44-6a-Chlor- pregnen -1 l/3, 17a,21 - triol - 3,20-dion erhalten wird; F. 174-176 , [a]j) <I>= +97 .</I>
In analoger Weise lassen sich höhere 21-Ester von J4-6a-Chlor-pregnen-11/3,17a,21-triol-3,20-dion her stellen.
<I>Beispiel 8</I> Analog den Angaben in Beispiel 7 werden 3 g 44-6a-Fluor-pregnen-17a,21-diol-3,20-dion in 16 em3 Propylenglykol mit 3 kg Nebennierenbrei und 6 Liter der Mischung der Lösungen A, B und C umgesetzt und aufgearbeitet. Der Methylenehlorid-Extrakt wird im Vakuum auf etwa 300 cm3 eingeengt und auf eine Säule aus 90 g Silicagel und 90 g Celite gegeben.
Die Säule wird dann mit 3 Liter Methylenchlorid und einer Mischung von 900 cm3 Methylenehlorid und 100 cm3 Aceton eluiert. Das Umsetzungsprodukt wird mit Methylenchlorid-Aceton (80:20 und 70:30)- Mischungen eluiert. Durch Eindampfen dieser Eluate und Kristallisation des Rückstandes aus Essigester er hält man das 44-6a-Fluor-pregnen-llss,17a,21-triol- 3,20-dion; F. 197-201 , [a]D = +127 .
Wird in analoger Weise das 41.4-6a-Fluor-pregna- dien-17a,21-diol-3,20-dion inkubiert, so erhält man das d1,4 - 6a-Fluor-pregnadien-11/3,17a,21-triol-3,20- dion; F. 208-213 , [a]D = +92 .
Die erhaltenen llss-Hydroxyverbindungen lassen sich nach den Angaben in Beispiel 7 in 21-Stellung verestern, z. B. mit Säuren, die 2 bis 12 Kohlenstoff- atome enthalten.
Das 21-Acetat von 41,4-6a-Fluor-pregnadien-l 1ss, 17a,21-triol-3,20-dion kann wie in Beispiel 6 be schrieben zum 21-Acetat von 41A-6a-Fluor pregna- dien-17a,21-diol-3,11,20-trion oxydiert werden; F. 228-230 , [a]D = +142 .
<I>Beispiel 9</I> Analog der Angaben in Beispiel 7 werden 2 g d4-2a-Methyl-6a-fluor-pregnen-17a,21-diol-3,20-dion in 16 cm3 Propylenglykol mit 3 kg Nebennierenbrei urid 6 Liter der Mischung der Lösungen A, B und C inkubiert. Der Methylenchlorid-Extrakt wird im Va kuum auf etwa 300 cm3 eingeengt und auf eine Säule aus 90 g Silicagel und 90 g Celite gegeben.

Das Inkubationsprodukt wird mit Methylenchlorid extrahiert und der Extrakt mit Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum auf ein kleines Volumen eingeengt. Auf diese Weise werden total 3 g des Ausgangsstoffes um gesetzt.
