Verfahren zur Herstellung von neuen heterocyclischen Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 5-Amino-l ,5-diazacyclo- octan-Derivaten der Formel I,

Das Symbol R in den obigen Formeln kann sowohl für einen Alkylrest, wie beispielsweise den Me- thyl-, Athyl-, Propyl-, Isopropyl oder Butylrest, als auch für einen Aralkylrest, wie z. B. den Benzylrest, stehen.
Die Ausführung des Verfahrens gestaltet sich beispielsweise wie folgt: Die wässerige saure Lösung (pH 3-4) einer Verbindung der Formel II, z.
1 -Methyl- 1 5 diazacyclooctan, wird bei Raumtemperatur mit einer wässerigen Natriumnitritlösung versetzt und das Gemisch bei dieser Temperatur gehalten. Anschliessend wird das Reaktionsgemisch unter Eiskühlung mit überschüssigem Kaliumhydroxyd alkalisch gestellt und erschöpfend mit Äther extrahiert. Die Nitrosoverbindung wird nach Verdampfen des Ethers durch Destillation bei vermindertem Druck gereinigt.
Die Reduktion der Nitrosoverbindung kann nach bekannten Methoden, z. B. mit Zink und Essigsäure oder mit Lithiumaluminiumhydrid, ausgeführt werden.
Die Reduktion mit Zink und Essigsäure erfolgt beispielsweise so, dass man die wässerige Lösung der Nitrosoverbindung zuerst mit Zinkstaub, dann im Verlauf von 30-60 Minuten in der Kälte mit verdünnter Essigsäure versetzt. Zur Vervollständigung der Reaktion wird das Reaktionsgemisch noch 2-3 Stunden bei Raumtemperatur gehalten und anschlie ssend noch 2-3 Stunden auf höhere Temperatur (etwa 800) gebracht. Nach Filtration und Alkalischmachen in der Kälte wird das erhaltene Amin nach bekannten Methoden, z. B. durch Extraktion mit einem indifferenten Lösungsmitrel, vorzugsweise Diäthyläther, isoliert, und durch Destillation und/oder Kristallisation gereinigt.
Die Reduktion mit Lithiumaluminiumhydrid erfolgt in der Weise, dass man die Lösung der Nitrosoverbindung in einem inerten organischen Lösungsmittel, wie z. B. Äther, Tetrahydrofuran oder Dioxan, in der Siedehitze mit dem im gleichen Lösungsmittel gelösten Reduktionsmittel behandelt. Nach 2- bis 4-stündigem Erhitzen versetzt man mit Wasser und macht das Reaktionsgemisch mit einem Alkalihydroxyd, z. B. Kaliumhydroxyd, alkalisch. Das Endprodukt wird wie oben beschrieben isoliert.
Die verfahrensgemäss hergestellten neuen 5-Amino 1,5-diazacyclooctan-Derivate bilden mit geeigneten anorganischen oder organischen Säuren beständige bei Raumtemperatur kristallisierte Säureadditionssalze. Solche Salze sind beispielsweise die Dihydrochloride, Dihydrobromide, Dimaleinate, Difumarate, Dioxalate usw.
Die neuen Verfahrensprodukte sowie ihre Salze besitzen mannigfache pharmakologische Wirkungen, die sie zu wertvollen Medikamenten machen. Sie senken beispielsweise den Blutdruck und hemmen die Wirkung der Monoaminoxydase. Ferner besitzen sie eine spezifische toxische Wirkung auf Enteroparasiten.
Ausserdem können die nach dem vorliegenden Verfahren hergestellten neuen Verbindungen auch als Zwischenprodukte zur Herstellung von Medikamenten verwendet werden.
Die als Ausgangssubstanzen verwendeten 1 ,5-Di- azacyclooctan-Derivate der Formel II können beispielsweise hergestellt werden, indem man 1 ,2-Tri- methylen-pyrazolidin in einem geeigneten wasserfreien Lösungsmittel, wie beispielsweise abs. Aceton, mit einer organischen Halogen-Verbindung der Formel R-Hal, worin Hal Chlor, Brom oder Jod bedeutet, in die quartäre Verbindung der Formel IV

überführt und diese anschliessend reduziert. Die Reduktion wird vorzugsweise in einem offenkettigen oder cyclischen Äther, wie z. B. Diäthyläther oder Tetrahydrofuran, mit amalgamierten Aluminiumspänen ausgeführt.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelzund Siedepunkte sind nicht korrigiert.
Beispiel I 1-Methyl-5-amino-1, ,5-diazacyclooctan a) l-Methyl-5-nitroso- 1,5-diazacyclooctan
Man stellt die Lösung von 9,2 g 1-Methyl-1,5- diazacyclooctan in 10 cm3 dest Wasser unter Eiskühlung mit 15,5 g 500,'oiger Schwefelsäure auf ein pH von 3-4. Die saure Lösung wird darauf bei Raumtemperatur mit der Lösung von 16,5 g Natriumnitrit in 21,5 cm Wasser versetzt.
Nach 16stündigem Rühren bei Raumtemperatur versetzt man unter guter Eiskühlung mit festem Kaliumhydroxyd im Überschuss, extrahiert das Reaktionsgemisch mit Ather und destilliert den nach dem Verdampfen des Athers erhaltenen Rückstand bei einem Druck von 11 mm Hg, wobei das 1-Methyl-5-nitroso-l ,5-diazacyclooctan bei 132-134 übergeht.
Das als Ausgangsmaterial verwendete l-Methyl- 1,5-diazacyclooctan wird wie folgt hergestellt: Man versetzt die im Eisbad abgekühlte Lösung von 11,2 g 1,2- Trimethylenpyrazolidin (Sdp. 169-171 ) in 200 cm3 abs. Aceton mit 17,0 g Methyljodid. Nach einstündigem Stehen filtriert man den farblosen Niederschlag ab und kristallisiert das entstandene l-Me- thyl-trimethylenpyrazolidiniumjodid aus Isopropanol um. Smp. 250-251".
27 g amalgamierte Aluminiumspäne werden mit 400 cm3 Äther überschichtet, im Eisbad gekühlt und nach Zusatz von 30,5 g l-Methyl-trimethylenpyrazolidiniumjodid unter zeitweiligem Umschwenken im Verlauf einer Stunde mit 55 cm3 Wasser versetzt.
Man lässt langsam aufwärmen und hält das Reaktionsgemisch über Nacht am Rückfluss zum Sieden. Sodann wird unter Eiskühlung mit überschüssigem Kaliumhydroxyd versetzt, die ätherische Phase abgetrennt und der Rückstand erschöpfend mit Ather extrahiert. Die vereinigten ätherischen Lösungen liefern nach dem Eindampfen im Vakuum das reine l-Methyl-l ,5-diazacyclooctan vom Sdp. 175-176 < 5. b) 1 -Methyl-5-amino- 1 ,5-diazacyclooctan
Die Lösung von 6,9 g 1-Methyl-5-nitroso-l,5- diazacyclooctan in 35 cm3 Wasser wird mit 25 g Zinkstaub und darauf im Verlauf von 45 Min. unter Eiskühlung und Rühren mit 25 g 750/0 der Essigsäure versetzt.
Anschliessend rührt man 3 Std. bei Raumtemperatur und erhitzt noch 21H2 Std. im Ölbad auf 80t ;. Darauf wird heiss filtriert, das Filtrat nach Abkühlen im Eisbad mit festem Kaliumhydroxyd im Überschuss versetzt und dreimal mit je 100 cm3 Äther extrahiert. Die vereinigten Ätherlösungen liefern nach dem Eindampfen ein farbloses Öl, das in Äthanol gelöst und mit 480/obiger Bromwasserstoffsäure in das Hydrobromid übergeführt wird. Das erhaltene l-Methyl-5- amino-l ,5-diazacyclooctan-dihydrobromid wird wie folgt gereinigt: Man löst das Dihydrobromid in möglichst wenig heisser, 480/0 iger Bromwasserstoffsäure und versetzt mit Athanol, wobei das reine Dihydrobromid kristallin ausfällt.

Beispiel 2 l-Methyl-5-amino-1, 5-diazacyclooctan
Das nach Beispiel 1 es) erhaltene t-Methyl-5- nitroso-1,5-diazacyclooctan kann auch mit Lithiumaluminiumhydrid zum gewünschten Amino-Derivat reduziert werden. Man verfährt wie folgt: Die Lösung von 9,3 g 1 -Methyl-5-nitroso- 1 ,5-diazacyclooctan in 70 cm abs. Tetrahydrofuran lässt man vorsichtig in die siedende Lösung von 4,5 g Lithiumaluminiumhydrid in 70 cm3 abs. Tetrahydrofuran zutropfen und erhitzt das Reaktionsgemisch anschliessend 31/2 Std. am Rückfluss. Nach Abkühlen im Eisbad versetzt man das Reaktionsgemisch vorsichtig mit 18 cm9 dest.
Wasser und trägt nach einer weiteren Stunde bei O11 festes Kaliumhydroxyd im Überschuss ein. Das Lösungsmittel wird abgetrennt und der Rückstand erschöpfend mit Äther extrahiert. Die vereinigten organischen Phasen werden bei vermindertem Druck eingeengt und der Rückstand, das l-Methyl-5-amino- 1 ,5-diazacyclooctan, bei einem Druck von 11 mm Hg destilliert. Sdp. 94 /11 mm Hg.
Dihydrobromid: Die erhaltene Base wird in Athanol gelöst und mit 48 /Oiger Bromwasserstoffsäure versetzt. Smp. 197-199o (Zers.).
Beispiel 3 l-Isopropyl-5-amino- 1,5-diazacyclooctan a) I-Isopropyl-5-nitroso-1, ,5-diazacyclooctan
Die Lösung von 11,3 g l-Isopropyl-1,5-diaza- cyclooctan in 10 cm3 Wasser wird unter Eiskühlung mit 19,0 g 500/oiger Schwefelsäure und anschliessend bei Raumtemperatur mit der Lösung von 16,5 g Natriumnitrit in 22 cm3 Wasser versetzt. Nach 24stündigem Rühren bei Raumtemperatur versetzt man das Reakionsgemisch mit festem Kaliumhydroxyd im Überschuss und extrahiert erschöpfend mit Äther.
Das nach dem Abdampfen des ethers erhaltene Öl wird bei 12 mm/Hg im Kugelrohr destilliert, wobei das l-Isopropyl-5 -nitroso-1,5 -diazacyclooctan zwischen 150 und 1600 übergeht. b) l-Isopropyl-5-amino- 1,5-diazacyclooctan
Die Lösung von 7,4 g 1-Isopropyl-5-nitroso-1 5- diazacyclooctan in 35 cm3 Wasser wird mit 25 g Zinkstaub und darauf unter Eiskühlung innert 30 Min. mit 25 g 75 0/obiger Essigsäure versetzt. Anschliessend rührt man 3 Std. bei Raumtemperatur und erhitzt noch 2 Std. auf 800. Darauf wird heiss filtriert, das klare Filtrat nach Abkühlung im Eisbad mit festem Kaliumhydroxyd im Überschuss versetzt und erschöpfend mit Äther extrahiert.
Der nach dem Eindampfen der vereinigten Ätherlösungen erhaltene Rückstand wird bei 12 mm/Hg im Kugelrohr destilliert, wobei das 1-Isopropyl-5-amino-1, 5-diazacyclooctan zwischen 110 und 1150 übergeht.
Das als Ausgangsmaterial verwendete l-Isopropyl-l ,5-diazacyclooctan wird wie folgt hergestellt:
Man versetzt die Lösung von 139 cm3 Trimethylendibromid in 1000 cm3 Athanol mit 75,1 g Isopropylhydrazin-hydrochlorid und 360 g wasserfreiem Natriumcarbonat und erhitzt anschliessend 8 Std. zum Sieden. Nach dem Abkühlen auf etwa 00 filtriert man von unlöslichem Salz ab und dampft das Filtrat im Vakuum ein. Der erhaltene Rückstand wird zur Entfernung neutraler, ätherlöslicher Verunreinigungen erschöpfend mit Äther extrahiert. Anschliessend werden 60 g amalgamierte, mit 750 cm3 Äther überschichtete Aluminiumspäne mit dem erhaltenen Rückstand, und darauf unter Eiskühlung portionenweise mit insgesamt 130 cm3 dest. Wasser versetzt.
Man lässt das Reaktionsgemisch etwa 12 Std., zuerst bei etwa Oa dann bei Raumtemperatur, stehen. Das Reaktionsgemisch wird sodann bei etwa 0a mit festem Kaliumhydroxyd im Überschuss versetzt, die organische Phase abgetrennt und der Rückstand erschöpfend mit Äther extrahiert. Die vereinigten ätherischen Lösungen werden eingedampft und der Rückstand unter einem Druck von 9 mm/Hg destilliert, wobei das l-Isopropyl-1,5-diazacyclooctan zwischen 85 und 860 übergeht.
Beispiel 4 1-Benzyl-5-amino-1, 5-diazacyclooctan a) l-Benzyl-5-nitroso-l ,5-diazacyclooctan
Die Lösung von 9,5 g 1-Benzyl-1,5-diazacyclo- octan in 10 cmS Wasser wird bei 0-5O mit 10,1 g 500/obiger Schwefelsäure und anschliessend mit der Lösung von 3,5 g Natriumnitrit in 5,0 cms Wasser versetzt. Nach 3stündigem Rühren des Reaktionsgemisches bei Raumtemperatur und nach Zusatz von 50 cms Äther versetzt man das Reaktionsgemisch unter guter Eiskühlung mit festem Kaliumhydroxyd im Überschuss und filtriert anschliessend vom ausgefallenen Niederschlag ab.
Die ätherische Phase des Filtrates wird abgetrennt und die wässerige Phase noch zweimal mit je 50 cm3 Äther extrahiert. Die vereinigten ätherischen Lösungen werden über Magnesiumsulfat getrocknet und eingeengt. Der erhaltene Rückstand wird bei 0,02 mm/Hg destilliert, wobei das 1 -Benzyl-5-nitroso-1 ,5-diazacyclooctan zwischen 142 und 1470 übergeht. b) 1-Benzyl-5-amino-1 ,5-diazacyclooctan
Zu der siedenden Lösung von 3,2 g Lithium aluminiumhydrid in 70 cm3 abs. Tetrahydrofuran lässt man unter Rühren im Verlaufe von 21/2 Std. die Lösung von 9,7 g 1-Benzyl-5-nitroso-1,5-diazacyclooctan in 70 cm3 abs. Tetrahydrofuran zutropfen und erhitzt das Reaktionsgemisch weitere 41/2 Std. am Rückfluss.
Nach Abkühlen im Eisbad lässt man vorsichtig 15 cm3 Wasser zutropfen und versetzt nach einer weiteren halben Stunde mit festem Kaliumhydroxyd im Über- schuss. Das Lösungsmittel wird abgetrennt und der Rückstand erschöpfend mit Äther extrahiert. Der nach dem Abdampfen des äther erhaltene Rückstand wird bei 13 mm/Hg destilliert, wobei das l-Ben zyl-5-amino- 1 ,5-diazacyclooctan zwischen 180 und 1820 überdestilliert.
Das als Ausgangssubstanz verwendete 1-Benzyl 1,5-diazacyclooctan wird analog wie das l-Isopropyl 1,5-diazacyclooctan (vergleiche Beispiel 3) hergestellt, indem man Trimethylendibromid mit Benzylhydrazin kondensiert und das entstandene 1-Benzyl- 1, 2-trime- thylen-pyrazolidiniumbromid mit amalgamierten Aluminiumspänen reduziert. Sdp. 155-1589/10 mm Hg.

The process is carried out, for example, as follows: The aqueous acidic solution (pH 3-4) of a compound of the formula II, e.g.
1 -Methyl- 1 5 diazacyclooctane, an aqueous sodium nitrite solution is added at room temperature and the mixture is kept at this temperature. The reaction mixture is then made alkaline with excess potassium hydroxide while cooling with ice and extracted exhaustively with ether. After evaporation of the ether, the nitroso compound is purified by distillation under reduced pressure.
The reduction of the nitroso compound can be carried out by known methods, e.g. B. with zinc and acetic acid or with lithium aluminum hydride.
The reduction with zinc and acetic acid is carried out, for example, by first adding zinc dust to the aqueous solution of the nitroso compound, then adding dilute acetic acid over the course of 30-60 minutes in the cold. To complete the reaction, the reaction mixture is kept at room temperature for another 2-3 hours and then brought to a higher temperature (about 800) for another 2-3 hours. After filtration and making alkaline in the cold, the amine obtained is by known methods, for. B. isolated by extraction with an inert solvent, preferably diethyl ether, and purified by distillation and / or crystallization.
The reduction with lithium aluminum hydride is carried out in such a way that the solution of the nitroso compound in an inert organic solvent, such as. B. ether, tetrahydrofuran or dioxane, treated at the boiling point with the reducing agent dissolved in the same solvent. The novel 5-amino 1,5-diazacyclooctane derivatives prepared according to the process form acid addition salts which are stable at room temperature and which are stable with suitable inorganic or organic acids. Such salts are, for example, the dihydrochlorides, dihydrobromides, dimaleinates, difumarates, dioxalates, etc.
The new process products and their salts have manifold pharmacological effects that make them valuable medicines. For example, they lower blood pressure and inhibit the effect of monoamine oxidase. They also have a specific toxic effect on enteroparasites.
In addition, the new compounds produced by the present process can also be used as intermediates for the production of medicaments.
The 1,5-diazacyclooctane derivatives of the formula II used as starting substances can be prepared, for example, by adding 1,2-trimethylene pyrazolidine in a suitable anhydrous solvent, such as abs. Example I 1-methyl-5-amino-1,, 5-diazacyclooctane a) 1-methyl-5-nitroso-1,5-diazacyclooctane
The solution of 9.2 g of 1-methyl-1,5-diazacyclooctane in 10 cm3 of distilled water is adjusted to a pH of 3-4 with 15.5 g of 500% sulfuric acid while cooling with ice. The acidic solution is then mixed with a solution of 16.5 g of sodium nitrite in 21.5 cm of water at room temperature.
After stirring for 16 hours at room temperature, excess solid potassium hydroxide is added with good ice cooling, the reaction mixture is extracted with ether and the residue obtained after evaporation of the ether is distilled at a pressure of 11 mm Hg, the 1-methyl-5-nitroso-l , 5-diazacyclooctane at 132-134.
The 1-methyl-1,5-diazacyclooctane used as starting material is prepared as follows: The solution, cooled in an ice bath, of 11.2 g of 1,2-trimethylene pyrazolidine (bp. 169-171) in 200 cm3 of abs. Acetone with 17.0 g methyl iodide. After standing for one hour, the colorless precipitate is filtered off and the l-methyltrimethylene pyrazolidinium iodide formed is recrystallized from isopropanol. 27 g of amalgamated aluminum shavings are covered with a layer of 400 cm3 of ether, cooled in an ice bath and, after adding 30.5 g of 1-methyl-trimethylene pyrazolidinium iodide, 55 cm3 of water are added over the course of an hour while swirling from time to time.
The mixture is allowed to warm up slowly and the reaction mixture is refluxed overnight. Excess potassium hydroxide is then added while cooling with ice, the ethereal phase is separated off and the residue is extracted exhaustively with ether. The solution of 6.9 g of 1-methyl-5-nitroso-1,5-diazacyclooctane in 35 cm3 of water is mixed with 25 g of zinc dust and then in the course of 45 minutes with ice-cooling and stirring with 25 g of 750/0 acetic acid .
The mixture is then stirred for 3 hours at room temperature and heated to 80t in an oil bath for a further 21H2 hours. It is then filtered hot, the filtrate, after cooling in an ice bath, is treated with excess solid potassium hydroxide and extracted three times with 100 cm3 of ether each time. After evaporation, the combined ether solutions give a colorless oil which is dissolved in ethanol and converted into the hydrobromide with 480% hydrobromic acid. The l-methyl-5-amino-l, 5-diazacyclooctane dihydrobromide obtained is purified as follows: The dihydrobromide is dissolved in as little hot 480/0 hydrobromic acid as possible and ethanol is added, the pure dihydrobromide precipitating in crystalline form.

Example 2 1-methyl-5-amino-1,5-diazacyclooctane
The t-methyl-5-nitroso-1,5-diazacyclooctane obtained according to Example 1 es) can also be reduced to the desired amino derivative with lithium aluminum hydride. The procedure is as follows: The solution of 9.3 g of 1-methyl-5-nitroso-1,5-diazacyclooctane in 70 cm abs. Water and, after a further hour, enters solid potassium hydroxide in excess at O11. The solvent is separated off and the residue is extracted exhaustively with ether. The combined organic phases are concentrated under reduced pressure and the residue, the 1-methyl-5-amino-1, 5-diazacyclooctane, is distilled at a pressure of 11 mm Hg. Bp 94/11 mm Hg.
Dihydrobromide: The base obtained is dissolved in ethanol and treated with 48% hydrobromic acid. 197-199o (dec.).
Example 3 l-Isopropyl-5-amino-1,5-diazacyclooctane a) I-Isopropyl-5-nitroso-1,5-diazacyclooctane
The solution of 11.3 g of 1-isopropyl-1,5-diaza-cyclooctane in 10 cm3 of water is mixed with 19.0 g of 500% sulfuric acid while cooling with ice and then at room temperature with the solution of 16.5 g of sodium nitrite in 22 cm3 Water added. After stirring for 24 hours at room temperature, the reaction mixture is treated with excess solid potassium hydroxide and extracted exhaustively with ether.

The mixture is then stirred for 3 hours at room temperature and heated for a further 2 hours to 800. It is then filtered hot, the clear filtrate, after cooling in an ice bath, mixed with excess solid potassium hydroxide and extracted exhaustively with ether.
The residue obtained after evaporation of the combined ether solutions is distilled at 12 mm / Hg in a bulb tube, the 1-isopropyl-5-amino-1,5-diazacyclooctane passing over between 110 and 1150.
