Die Einführung des Restes R' in die Verbindung der Formel VI oder deren beispielsweise durch Ko chen mit alkoholischer Kalilauge erhaltene Hydro- lysenprodukte kann z.
B. durch Umsetzung mit einem reaktionsfähigen Ester eines niedermolekularen Alka- nols, wie Dimethylsulfat, Diäthylsulfat, Methyljodid, p-Toluolsulfonsäuremethylester, 2,4-Dinitrobenzolsul- fonsäuremethylester, Äthylbromid, Äthyljodid, n-Pro- pylbromid, n-Butylbromid oder Isobutylbromid in Gegenwart von Kaliumhydroxyd, oder z.
B. auch durch Umsetzung mit Diazomethan als niedermole kularem Alkylierungsmittel erfolgen. An die Stelle der vorgenannten alipliatischen Halogenide können beispielsweise auch Benzylhalogenide und Benzhy- drylhalogenide treten. Erfolgt die Hydrolyse nach der Verätherung, ist gegebenenfalls die Aufarbeitung des Äthers nicht nur unnötig, sondern wegen allfälliger, bereits bei der Verätherung eingetretener teilweiser Hydrolyse sogar unzweckmässig.
Werden anstelle von eigentlichen Äthern Verbindungen von Acetalcharak- ter, z. B. durch Umsetzung von Verbindungen der Formel VI mit Dihydropyran oder mit Chlordime- thyläther hergestellt,
so ist bei der anschliessenden Hydrolyse die Verwendung alkalischer Mittel wie der bereits genannten alkoholischen Kalilauge naturgemäss besonders an2ezeiat. Als alkalische Kondensationsmittel für die Um setzung von Verbindungen der Formel 11 mit reak tionsfähigen Estern von basischen Alkoholen der For mel<B>111</B> eignen sich insbesondere Natriumamid, Lithiumamid, Kaliumamid, Natrium, Kalium, Butyl- lithium, Phenyllithium,
Natriumhydrid oder Lithium- hydrid. Die Umsetzung kann in An- oder Abwesen heit eines inerten organischen Lösungsmittels, wovon als Beispiel Benzol, Toluol, Xylole genannt seien, durchgeführt weiden.
Als reaktionsfähige Ester von basischen Alko holen der Formel<B>111</B> kommen insbesondere die Halogenide in Frage; im einzelnen seien genannt: Dimethylaminoäthylchlorid, Diäthylaminoäthylchlorid, Methyläthylaminoäthylchlorid, /3-Dimethylamino-propylchlorid, /3-Dimethylamino-isopropy lehlorid, y-Dimethylamino-propylchlorid, a-Dimethylamino-butylchlorid, /3-(Di-n-propylamino)-äthylchlorid, n-(N-Methyl-isopropylamino)-äthylchlorid,
/3-(Di-n-butyl-amino)-äthylchlorid, a-(Di-isobutyl-amino)-äthylchlorid, Pyrrolidinoäthylchlorid, Piperidinoäthylchlorid, y-Pyrrolidino-propylchlorid, y-Piperidino-propylchlorid, Morpholinoäthy lchlorid, ss-(4-Methylpiperazino)-ätliylchlorid, ss-(4-Acetoxyäthyl-piperazino)-äthylchlorid, ;
,-(4-Acetoxyäthyl-piperazino)-propylchlorid und 1-Methyl-piperidyl-(3)-methylchlorid sowie die entsprechenden Bromide und Jodide.
Die gegebenenfalls durchzuführende Hydrogeno- lyse von Verbindungen der Formel IV kann z. B. in neutraler oder schwachsaurer methanolischer oder äthanolischer Lösung in Gegenwart von Palladium kohle bereits bei Normaldruck und Raumtemperatur erfolgen.
Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phos phorsäure, Methansulfonsäure, Äthandisulfonsäure., Essigsäure, Bernsteinsäure, Fumarsäure, Maleinsäure. Äpfelsäure, Weinsäure, Zitronensäure, Benzoesäure und Phthalsäure bilden die erfindungsgemäss erhal tenen Verbindungen der Formel 1 Salze, welche zum Teil wasserlöslich sind.
In den nachstehenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu ems. Die Temperaturen werden in Celsiusgrader angegeben.
<I>Beispiel 1</I> a) 25,2 Teile 3-Amino-5-acetyl-iminodibenzy werden in 150 Volumteilen Wasser und 30 Volum. teilen konz. Schwefelsäure gelöst und bei 0 mit ; Teilen Natriumnitrit in 20 Volumteilen Wasser diazo tiert. Nach viertelstündigem Stehenlassen wird diE Lösung auf 80 erhitzt. Nach beendeter Stickstoff entwicklung wird die Lösung abgekühlt und das har zige Reaktionsprodukt abgetrennt.
Es wird in heissem Äther gelöst, die Ätherlösung wird zur Entfernung von unlöslichem rötlichem Nebenprodukt filtriert, und das Filtrat wird zur Trockne eingedampft. Der Rück stand geht beim Versetzen mit wenig Benzol vorüber gehend in Lösung und kristallisiert sofort wieder aus. Aus Benzol umkristallisiert, erhält man ein Benzol enthaltendes Produkt (etwa -'/;, Mol Benzol), das scharf bei 104-104,5 schmilzt. Durch kurzes Er hitzen über den Schmelzpunkt erhält man das benzol- freie 3-Hydroxy-5-acetyl-imidodibenzyl, das bei 78 bis 821, schmilzt.
b) 25,3 Teile 3-Hydroxy-5-acetyl-iminodibenzyl werden in Äther gelöst und zu dieser Lösung 5 Teile Diazomethan in Äther gegeben, wobei sofort Stick stoff entwickelt wird. Nach längerem Stehenlassen bei Zimmertemperatur wird überschüssiges Diazome- than durch Zugabe von einigen Tropfen Essigsäure zerstört. Nicht umgesetztes 3-Hydroxy-5-acetyl-imino- dibenzyl wird durch Schütteln mit verdünnter Natron lauge aus der Ätherlösung entfernt. Nach dem Wa schen mit Wasser und Trocknen der Ätherlösung wird diese zur Trockne eingedampft.
Der grünliche ölige Rückstand wird in 250 Volumteilen Alkohol, enthaltend 10 Teile Kaliumhydroxyd, 4 Stunden zum Sieden erhitzt. Beim Abkühlen kristallisiert das Reak tionsprodukt aus. Nach dem Verdünnen mit Wasser werden die Kristalle abgenutscht, mit Wasser gründ lich gewaschen und getrocknet. Durch Umkristalli sation aus Hexan erhält man das 3-Methoxy-imino- dibenzyl in Form von gelblichen Kristallen, die bei 94 schmelzen.
(-) 22,5 Teile 3-Methoxy-iminodibenzyl werden in 250 Volumteilen abs. Xylol gelöst und mit 4,3 Teilen in Toluol pulverisiertem Natriumamid 3 Stunden un ter Rühren auf 90-100 erhitzt. Hierauf wird y-Di- methylamino-propylchlorid (freigesetzt aus 16 Teilen des Hydrochlorides und aufgenommen in Xylol) hin zugefügt und während 20 Stunden unter Rückfluss erhitzt.
Die Base wird aus dem Reaktionsgemisch mit 2n Salzsäure extrahiert und hierauf mit 5n Na tronlauge freigesetzt. Die freie Base wird in Äther aufgenommen. Nach dem Trocknen mit Natrium sulfat und Abdampfen des Äthers verbleibt ein Öl, das im Hochvakuum bei 140-142 0,001 mm destil liert. Wird die gereinigte Base in 2n Salzsäure ge geben, so kristallisiert nach mehrstündigem Stehen das Hydrochlorid des 3-Methoxy-5-(y-dimethylamino- propyl)-iminodibenzyls vom Smp. 177 .
<I>Beispiel 2</I> n) 18,6 Teile 2-Hydroxy-iminodibenzyl, das z. B. durch Umwandlung des 2-Amino-5-acetyl-iminodi- benzyls in 2-Hydroxy-5-acetyl-iminodibenzyl analog Beispiel 1 und Abspaltung des Acetylrestes durch Kochen der letzteren Verbindung mit alkoholischer Kalilauge erhältlich ist, werden in einer Lösung von 6 Teilen Kaliumhydroxyd in 180 Volumteilen abs. Alkohol gelöst, mit 11,5 Teilen Benzylchlorid ver setzt und 18 Stunden unter Rückfluss gekocht.
Hier auf destilliert man den Alkohol ab und nimmt den Rückstand in Äther auf. Die ätherische Lösung wird mit verdünnter Natronlauge und anschliessend mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird aus Äther-Pentan umkristallisiert. Das so erhaltene 2-Benzyloxy-iminodibenzyl schmilzt bei 92-94 .
b) Die aus 9 Teilen y-Dimethylaminopropylchlo- rid freigesetzte Base und 14,3 Teile 2-Benzyloxy- iminodibenzyl werden in 200 Volumteilen Benzol gelöst und bei 50 mit einer Suspension von 2,5 Tei len Natriumamid in Toluol versetzt. Anschliessend kocht man das Reaktionsgemisch 18 Stunden unter Rückfluss. Hierauf kühlt man es ab und zersetzt es mit Wasser.
Die benzolische Phase wird viermal mit je 2,5 Volumteilen 1n Salzsäure ausgeschüttelt und die sauren Auszüge werden mit konz. Natronlauge alkalisch gestellt. Die ausgeschiedene Base wird in Äther aufgenommen, die Ätherlösung gründlich mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird im Hochvakuum destilliert, das 2- Benzyloxy-5-(y-dimethylamino-propyl)- iminodibenzyl geht unter 0,002 mm Druck bei 212-215a über.
c) 1,23 Teile der vorstehenden Verbindung wer den in 30 Volumteilen Methanol und 5 Volumteilen 1n Salzsäure gelöst und in Gegenwart von 0,36 Teilen Palladium-Kohle bei Raumtemperatur bis zur Auf nahme der theoretischen Wasserstoffmenge geschüt telt. Hierauf saugt man vom Katalysator ab und engt das Filtrat im Vakuum ein. Der zurückbleibende Rückstand wird mit konz. Ammoniak alkalisch ge stellt, wobei das 2-Hydroxy-5-(y-dimethylamino-pro- pyl)-iminodibenzyl kristallin ausfällt.
Es kann aus Aceton-Äther umkristallisiert werden, Smp. 132 bis 133 .
Analog dem ersten Abschnitt dieses Beispiels kann das im Beispiel 1 und a) beschriebene 2-Hy- droxy-5-acetyl-iminodibenzyl in das 3-Benzyloxy-5- acetyl-iminodibenzyl umgewandelt werden, welches man noch als Rohprodukt durch mehrstündiges Ko chen mit alkoholischer Kalilauge analog dem zweiten Teil von Beispiel 1 b) zum 3-Benzyloxy-iminodibenzyl hydrolysieren kann, aus welchem man analog dem zweiten und dritten Abschnitt des vorliegenden Bei spiels das 3-Benzyloxy-5-(y-dimethylamino-propyl)
- iminodibenzyl und das 3-Hydroxy-5-(y-dimethyl- amino-propyl)-iminodibenzyl erhält. <I>Beispiel 3</I> n) 25,3 Teile 3-Hydroxy-5-acetyl-iminodibenzyl werden in einer aus 2,3 Teilen Natrium und 200 Volumteilen abs. Alkohol bereiteten Natriumalkoho- latlösung gelöst. Zur roten Lösung werden 12,7 Teile Benzylchlorid hinzugefügt, worauf sich sofort Na- triumchlorid auszuscheiden beginnt.
Nach 15stündi- gem Kochen unter Rückfluss wird das Natriumchlorid abfiltriert und die Lösung im Vakuum eingeengt. Der Rückstand wird in Äther aufgenommen und allfällige nicht umgesetzte Hydroxyverbindungen mit 2n Na tronlauge ausgeschüttelt. Die Ätherlösung wird hier auf mit Wasser gewaschen, mit Natriumsulfat ge trocknet und zur Trockne eingedampft.
Das zurück bleibende kristalline 3-Benzyloxy-5-acetyl-iminodi- benzyl kann aus wenig Alkohol umkristallisiert wer den und schmilzt bei 91 .
b) 34,3 Teile 3-Benzyloxy-5-acetyl-iminodibenzyl werden durch 10stündiges Kochen mit alkoholischer Kalilauge (12 Teile KOH in 250 Volumteilen Alko hol) hydrolysiert. Hierauf wird die Lösung im Va kuum eingeengt. Der ölige Rückstand wird mit Was ser versetzt und das 3-Benzyloxy-iminodibenzyl mit Äther extrahiert. Die getrocknete Ätherlösung wird zur Trockne eingedampft. Das erhaltene Öl kristalli siert nach etwa 24stündigem Stehenlassen.
Nach Um kristallisieren aus Benzol-petroläther schmilzt das 3- Benzyloxy-iminodibenzyl bei 111 .
c) 4,3 Teile rohes 3-Benzyloxy-iminodibenzyl wer den in 60 Volumteilen abs. Toluol gelöst und die unter Rückfluss kochende Lösung tropfenweise mit 1,6 Teilen einer 33o/oigen Suspension von Natrium- amid in abs. Toluol versetzt.
Nach 2 Stunden wird eine durch Freisetzung der Base aus dem Hydro- chlorid frisch bereitete Lösung von 2 Teilen ss-Piperi- dino-äthylchlorid in 40 Volumteilen abs. Toluol zu gefügt und anschliessend noch 0,8 Volumteile der Natriumamid-Suspension. Nach 14 Stunden Kochen unter Rückfluss wird das Reaktionsgemisch abgekühlt, mit Wasser versetzt und die Phase getrennt. Die organische Schicht wird dreimal mit 2n Salzsäure extrahiert,
die vereinigten salzsauren Extrakte werden mit konz. Ammoniaklösung basisch gestellt und die erhaltene Suspension mit Äther extrahiert. Der Äther extrakt wird über Natriumsulfat getrocknet und ein gedampft, wobei das rohe 3-Benzyloxy-5-(ss-piperi- dino-äthyl)-iminodibenzyl zurückbleibt.
Dieses wird mittels äthanolischer Chlorwasserstofflösung in das Hydrochlorid übergeführt und letzteres aus Methanol/ Aceton umkristallisiert. Smp.208-209 nach Um wandlung in feine Nadeln oberhalb 185 .
In analoger Weise erhält man das 3-Benzyloxy- 5-(y-di-methylamino-propyl)-iminodibenzyl vom Kpo,o25 202 ausgehend von 3-Benzyloxy-iminodiben- zyl und y-Dimethylamino-propylchlorid; und das 3- Benzyloxy- 5 - [y- (4'- methyl-piperazinyl-1')-ss-methyl- propyl]-iminodibenzyl vom Kpo,oo8 260 unter Ver wendung von 3-Benzyloxy-iminodibenzyl und y-(4 Methyl-piperazinyl-1)-ss-methyl-propyl-chlorid.
b) Durch Hydrogenolyse analog Beispiel 1 c) er hält man aus den obigen 5-substituierten 3-Benzyl- oxy-iminodibenzylen die entsprechenden 5-substituier- ten 3-Hydroxy-iminodibenzyle, d. h. 3-Hydroxy-5-(ss- piperidino-äthyl)-iminodibenzyi, 3-Hydroxy-5-(y-di- methylamino-propyl)-iminodibenzyl und 3-Hydroxy 5-[y-(4'-methyl-piperazinyl-1')-ss-methyl-propyl]-imino- dibenzyl.

<I> Example 1 </I> a) 25.2 parts of 3-amino-5-acetyl-iminodibenzy are in 150 parts by volume of water and 30 parts by volume. share conc. Sulfuric acid dissolved and at 0 with; Parts of sodium nitrite in 20 parts by volume of water diazo benefits. After standing for a quarter of an hour, the solution is heated to 80. After the evolution of nitrogen has ended, the solution is cooled and the resinous reaction product is separated off.
It is dissolved in hot ether, the ethereal solution is filtered to remove insoluble reddish by-product, and the filtrate is evaporated to dryness. The residue goes into solution when a little benzene is added and immediately crystallizes out again. Recrystallized from benzene, a benzene-containing product is obtained (about - '/; mol benzene) which melts sharply at 104-104.5. Briefly heating above the melting point gives benzene-free 3-hydroxy-5-acetyl-imidodibenzyl, which melts at 78 to 821.
b) 25.3 parts of 3-hydroxy-5-acetyl-iminodibenzyl are dissolved in ether and 5 parts of diazomethane in ether are added to this solution, with nitrogen being developed immediately. After standing for a long time at room temperature, excess diazomethane is destroyed by adding a few drops of acetic acid. Unreacted 3-hydroxy-5-acetyl-imino-dibenzyl is removed from the ethereal solution by shaking with dilute sodium hydroxide solution. After washing with water and drying the ethereal solution, it is evaporated to dryness.
The greenish oily residue is heated to boiling for 4 hours in 250 parts by volume of alcohol containing 10 parts of potassium hydroxide. The reaction product crystallizes out on cooling. After diluting with water, the crystals are suction filtered, washed thoroughly with water and dried. Recrystallization from hexane gives 3-methoxy-imino-dibenzyl in the form of yellowish crystals which melt at 94.
(-) 22.5 parts of 3-methoxy-iminodibenzyl are abs in 250 parts by volume. Dissolved xylene and heated with 4.3 parts of sodium amide powdered in toluene for 3 hours under stirring to 90-100. Then y-dimethylamino-propyl chloride (released from 16 parts of the hydrochloride and taken up in xylene) is added and refluxed for 20 hours.
The base is extracted from the reaction mixture with 2N hydrochloric acid and then released with 5N sodium hydroxide solution. The free base is taken up in ether. After drying with sodium sulfate and evaporation of the ether, an oil remains, which distils 0.001 mm in a high vacuum at 140-142. If the purified base is given in 2N hydrochloric acid, the hydrochloride of 3-methoxy-5- (γ-dimethylaminopropyl) -iminodibenzyl of melting point 177 crystallizes after standing for several hours.
<I> Example 2 </I> n) 18.6 parts of 2-hydroxy-iminodibenzyl, the z. B. by converting the 2-amino-5-acetyl-iminodibenzyl into 2-hydroxy-5-acetyl-iminodibenzyl analogously to Example 1 and splitting off the acetyl radical by boiling the latter compound with alcoholic potassium hydroxide solution are available in a solution of 6 Parts of potassium hydroxide in 180 parts by volume of abs. Dissolved alcohol, ver with 11.5 parts of benzyl chloride and refluxed for 18 hours.
Here the alcohol is distilled off and the residue is taken up in ether. The ethereal solution is washed with dilute sodium hydroxide solution and then with water, dried and evaporated. The residue is recrystallized from ether pentane. The 2-benzyloxy-iminodibenzyl thus obtained melts at 92-94.
b) The base released from 9 parts of γ-dimethylaminopropyl chloride and 14.3 parts of 2-benzyloxy-iminodibenzyl are dissolved in 200 parts by volume of benzene and, at 50, a suspension of 2.5 parts of sodium amide in toluene is added. The reaction mixture is then refluxed for 18 hours. It is then cooled and decomposed with water.
The benzene phase is extracted four times with 2.5 parts by volume of 1N hydrochloric acid and the acidic extracts are mixed with conc. Sodium hydroxide solution made alkaline. The precipitated base is taken up in ether, the ether solution is washed thoroughly with water, dried and evaporated. The residue is distilled in a high vacuum, the 2-benzyloxy-5- (γ-dimethylamino-propyl) - iminodibenzyl passes under 0.002 mm pressure at 212-215a.
c) 1.23 parts of the above compound who dissolved in 30 parts by volume of methanol and 5 parts by volume of 1N hydrochloric acid and shaken in the presence of 0.36 parts of palladium-carbon at room temperature until the theoretical amount of hydrogen is absorbed. The catalyst is then filtered off with suction and the filtrate is concentrated in vacuo. The remaining residue is treated with conc. Ammonia is made alkaline, the 2-hydroxy-5- (γ-dimethylamino-propyl) -iminodibenzyl precipitating in crystalline form.
It can be recrystallized from acetone-ether, m.p. 132 to 133.
Analogously to the first section of this example, the 2-hydroxy-5-acetyl-iminodibenzyl described in Example 1 and a) can be converted into 3-benzyloxy-5-acetyl-iminodibenzyl, which can be converted into the crude product by cooking for several hours Alcoholic potassium hydroxide solution can be hydrolyzed to 3-benzyloxy-iminodibenzyl analogously to the second part of Example 1 b), from which 3-benzyloxy-5- (y-dimethylamino-propyl) can be obtained analogously to the second and third sections of the present example

Alcohol prepared sodium alcohol solution dissolved. 12.7 parts of benzyl chloride are added to the red solution, whereupon sodium chloride begins to separate out immediately.
After refluxing for 15 hours, the sodium chloride is filtered off and the solution is concentrated in vacuo. The residue is taken up in ether and any unreacted hydroxy compounds are extracted with 2N sodium hydroxide solution. The ether solution is washed here with water, dried with sodium sulfate and evaporated to dryness.
The remaining crystalline 3-benzyloxy-5-acetyl-iminodibenzyl can be recrystallized from a little alcohol and melts at 91.
b) 34.3 parts of 3-benzyloxy-5-acetyl-iminodibenzyl are hydrolyzed by boiling for 10 hours with alcoholic potassium hydroxide solution (12 parts of KOH in 250 parts by volume of alcohol). The solution is then concentrated in vacuo. What water is added to the oily residue and the 3-benzyloxy-iminodibenzyl is extracted with ether. The dried ether solution is evaporated to dryness. The oil obtained crystallizes after standing for about 24 hours.
After crystallizing from benzene petroleum ether, the 3-benzyloxy-iminodibenzyl melts at 111.
c) 4.3 parts of crude 3-benzyloxy-iminodibenzyl who abs in 60 parts by volume. Dissolved toluene and the refluxing solution with 1.6 parts of a 33% suspension of sodium amide in abs. Toluene added.
After 2 hours, a solution, freshly prepared by liberating the base from the hydrochloride, of 2 parts of β-piperidinoethyl chloride in 40 parts by volume of abs. Toluene added and then 0.8 parts by volume of the sodium amide suspension. After boiling under reflux for 14 hours, the reaction mixture is cooled, water is added and the phase is separated. The organic layer is extracted three times with 2N hydrochloric acid,
the combined hydrochloric acid extracts are treated with conc. Ammonia solution made basic and the resulting suspension extracted with ether. The ether extract is dried over sodium sulfate and evaporated, whereby the crude 3-benzyloxy-5- (ss-piperidino-ethyl) -iminodibenzyl remains.
This is converted into the hydrochloride by means of an ethanolic hydrogen chloride solution and the latter is recrystallized from methanol / acetone. Mp 208-209 after conversion into fine needles above 185.
In an analogous manner, 3-benzyloxy-5- (y-dimethylamino-propyl) -iminodibenzyl is obtained from Kpo, o25 202 starting from 3-benzyloxy-iminodibenzyl and y-dimethylamino-propyl chloride; and the 3-benzyloxy-5 - [y- (4'-methyl-piperazinyl-1 ') - ss-methyl-propyl] -iminodibenzyl from Kpo, oo8 260 using 3-benzyloxy-iminodibenzyl and y- (4 Methyl-piperazinyl-1) -ss-methyl-propyl chloride.
b) By hydrogenolysis analogous to Example 1 c) he obtains the corresponding 5-substituted 3-hydroxy-iminodibenzyls from the above 5-substituted 3-benzyloxy-iminodibenzyls, ie. H. 3-hydroxy-5- (ss- piperidino-ethyl) -iminodibenzyl, 3-hydroxy-5- (y-dimethylamino-propyl) -iminodibenzyl and 3-hydroxy 5- [y- (4'-methyl-piperazinyl- 1 ') - ss-methyl-propyl] -imino-dibenzyl.
