Verfahren zur Herstellung von 6-Chlor-7-sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd Die Verbindung 6-Chlor-7-sulfamyl-1,2,4-benzo- thiadiazin-1,1-dioxyd hat sich als ein wertvolles Diuretikum erwiesen. Diese Verbindung ist durch Behandeln von 5-Chlor-2,4-disulfamyl-anilin mit Ameisensäure in grossem überschuss erzeugt wor den, der grössere Teil der letzteren Verbindung konnte nur mit Schwierigkeiten wiedergewonnen werden.
überraschenderweise ist nun gefunden worden, dass statt Ameisensäure die viel billigere Oxalsäure vorteilhafterweise benützt werden kann, von welcher überdies ein beträchtlich kleiner überschuss erforder lich ist. Auf diese Weise, u. a., ist es möglich, in einer Apparatur mit gewissen Dimensionen viel grössere Partien als bei Gebrauch von Ameisensäure zu konvertieren. Es ist leicht einzusehen, was dies, zusammen mit einem kleineren Verbrauch an Chemi kalien, für den Prozess vom ökonomischen Stand punkt aus bedeutet.
Das erfindungsgemässe Verfahren zur Herstel lung von 6-Chlor-7-sulfamyl-1,2,4-benzothiadiazin- 1,1-dioxyd ist dadurch gekennzeichnet, dass man 5-Chlor-2,4-disulfamylanilin mit Oxalsäure umsetzt. Die Reaktion kann durch Erhitzen auf eine Tempe ratur im Bereich von 125-250 C durchgeführt wer den, aber man gelangt zu den besten Ausbeuten, wenn die Umsetzung bei einer Temperatur von 170 bis 200 C erfolgt.
<I>Beispiel 1</I> 1 Gewichtsteil 5-Chlor-2,4-disulfamylanilin wird sorgfältig mit 2 Gewichtsteilen wasserfreier Oxalsäure vermischt. Das so erhaltene Gemisch wird in ein mit einem Rückflusskühler versehenes Reaktionsgefäss gebracht und hernach während 4 Stunden in einem Ölbad, dessen Temperatur zwischen 190 und 200 C gehalten wird, erhitzt. Nach dem Abkühlen wird Wasser hinzugefügt, und das erhaltene 6-Chlor-7- sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd wird ab filtriert.
Falls ein reines Produkt gewünscht wird, kann es leicht durch Rekristallisation aus verdünn tem Alkohol erhalten werden. <I>Beispiel 2</I> 11,43 g 5-Chlor-2,4-disulfamylanilin (0,04 Mole) werden mit 50,5 g fein pulverisiertem Oxalsäure- dihydrat (0,4 Mole) vermischt. Das Gemisch wird dann unter Rühren mit einer Aufheizungszeit von ungefähr 10 Minuten auf 130 C gebracht und die Schmelze während 1,5 Minuten bei 130 C gehalten.
Bei einer Temperatur von 90 C werden dann 200 ml Wasser hinzugefügt, das Gemisch auf Zimmertempe ratur abgekühlt, der ausgefallene Niederschlag fil triert und aus 300 ml kochendem Wasser umkristal- lisiert; es wurde ein Rohprodukt mit einer Ausbeute von 8,5 g erhalten.
4,25 g des so erhaltenen Rohproduktes werden in einer Lösung von 4 g Natriumbicarbonat in 250 ml Wasser suspendiert und während 2 Stunden gerührt. Der erhaltene Niederschlag wird abfiltriert, das klare Filtrat im Vakuum auf 50 ml konzentriert und mit 2n Chlorwasserstoffsäure angesäuert..
Der sich bildende Niederschlag wird abfiltriert, mit Wasser gewaschen, und nach Umkristallisieren aus Wasser erhält man 800 mg 3-Carboxy-6-chlor-7- sulfamyl -1,2,4 -- benzothiadiazin-1,1-dioxyd-mono- hydrat mit einem Schmelzpunkt von 285-287 C.
Analyse (als Monohydrat): Berechnet: C 26,86 H 2,25 H20 5,04 Gefunden: C 26,67 H 2,28 H20 4,8 500 mg 3 - Carboxy= 6 - chlor-7-sulfamyl-1,2,4- benzothiadiazin-1,1-dioxyd-monohydrat werden in 8 ml Eisessig während 4 Stunden unter Rückfluss erhitzt.
Das Lösungsmittel wird dann im Vakuum entfernt und der Rückstand aus Wasser umkristalli- siert. Es werden 375 mg, entsprechend einer Aus beute von 86()/o, 6-Chlor-7-sulfamyl-1,2,4-benzothia- diazin-1,1-dioxyd mit dem Schmelzpunkt von 355 C (Zers.) erhalten.
<I>Beispiel 3</I> 100 mg 5-Chlor-2,4-disulfamylanilin werden mit 400 mg fein pulverisiertem Oxalsäuredihydrat ver mischt und das Gemisch in einem offenen Probe röhrchen bei einer ölbadtemperatur von 190-195 C während 15 Minuten erhitzt. Nach dem Abkühlen wird das pulverisierte Produkt zweimal mit je 10 ml Wasser extrahiert und dann in verdünntem Ammo- niumhydroxyd gelöst.
Die erhaltene Lösung wird mit 5 mg Norit entfärbt, hernach filtriert und mit 2,5n Chlorwasserstoffsäure angesäuert. Das aus fallende 6-Chlor - 7 - sulfamyl-1,2,4-benzothiadiazin- 1,1-dioxyd wird abfiltriert, mit Wasser gewaschen, und nach dem Trocknen an der Luft erhält man 58 mg Endprodukt mit einem Schmelzpunkt von 350 bis 352 C (Zers.).
<I>Beispiel 4</I> 11,43 g 5-Chlor-2,4-disulfamylanilin (0,04 Mol) und 50,5 g fein pulverisiertes Oxalsäuredihydrat (0,4 Mol) werden vermischt und auf einem Ölbad bei einer Temperatur von 190-195 C unter Rühren in einer 3-Halsflasche, die mit einem Rückflusskühler versehen ist, erhitzt. -Das Reaktionsgemisch wird dann wie in Beispiel 2 beschrieben aufgearbeitet, wobei man. 13,8 g eines Materials erhält, welches grösstenteils aus 3-Carboxy-6-chlor-7-sulfamyl-1,2,4- benzothiadiazin-1,1-dioxyd besteht.
2 g dieses Materials werden dann in Ameisen säure während 4 Stunden unter Rückflussbedingungen behandelt, wobei man reines 6-Chlor-7-sulfamyl- 1,2,4-benzothiadiazin-1,1-dioxyd erhält.
Eine 2-g-Probe des ursprünglichen Reaktions gemisches wird in Eisessig während 4 Stunden unter Rückflussbedingungen erhitzt, und man erhält rohes 6-Chlor-7-sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd. <I>Beispiel 5</I> Ein gut vermahlenes Gemisch von 11,43 g 5-Chlor-2,4-disulfamylanilin und 50,44 g Oxalsäure- dihydrat wird bei 126 C während 1i/2 Minuten zum Schmelzen erhitzt. Das Gemisch wird dann auf 110 C abgekühlt, mit 200 ml Wasser behandelt und hernach auf Zimmertemperatur gekühlt.
Der gebil dete Niederschlag wird abfiltriert, mit Wasser ge waschen, und man erhält 10,5g Rohprodukt, das auf Grund von physikalischen Messungen aus unrei nem 3 - Carboxy-6-chlor-7-sulfamyl-1,2,4-benzothia- diazin-1,1-dioxyd besteht. 1,35 g des so erhaltenen Produktes wurden mit 7 ml Diphenyläther bei 220 bis 230 C während 3/4 Stunden unter ständiger Ent wicklung von Kohlendioxyd erhitzt. Das Gemisch wird hernach auf Zimmertemperatur abgekühlt und mit 10 ml Äther versetzt. Es werden 1,15g Nieder schlag erhalten, der abfiltriert und mit Äther gewa schen wird.
1 g des so erhaltenen Materials wird in 15 ml Wasser gelöst und 2 ml 2,5n Natriumhydroxyd hinzu gefügt. Die Lösung wird mit Essigsäure auf einen pH von 7,8 gebracht, der gebildete Niederschlag ab filtriert und das Filtrat mit 2,5n Chlorwasserstoff stark sauer gemacht. Nach 2 Stunden wird weiterer Niederschlag abfiltriert und mit Wasser gewaschen, wobei man 150 mg 6-Chlor-7-sulfamyl-1,2,4-benzo- thiadiazin-1,1-dioxyd mit einem Schmelzpunkt von 347-353 C (Zers.) erhält.
Process for the preparation of 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide The compound 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide has proven to be a valuable diuretic. This compound was produced by treating 5-chloro-2,4-disulfamyl-aniline with formic acid in large excess; the greater part of the latter compound could only be recovered with difficulty.
Surprisingly, it has now been found that instead of formic acid, the much cheaper oxalic acid can advantageously be used, of which, moreover, a considerably small excess is required. In this way, u. a., it is possible to convert much larger portions in an apparatus of certain dimensions than when using formic acid. It is easy to see what this, together with a smaller consumption of chemicals, means for the process from an economic point of view.
The process according to the invention for the production of 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide is characterized in that 5-chloro-2,4-disulfamylaniline is reacted with oxalic acid. The reaction can be carried out by heating to a temperature in the range of 125-250 ° C, but the best yields are obtained when the reaction is carried out at a temperature of 170-200 ° C.
<I> Example 1 </I> 1 part by weight of 5-chloro-2,4-disulfamylaniline is carefully mixed with 2 parts by weight of anhydrous oxalic acid. The mixture obtained in this way is placed in a reaction vessel provided with a reflux condenser and then heated in an oil bath, the temperature of which is kept between 190 and 200 ° C., for 4 hours. If a pure product is desired, it can easily be obtained by recrystallization from dilute alcohol. <I> Example 2 </I> 11.43 g of 5-chloro-2,4-disulfamylaniline (0.04 moles) are mixed with 50.5 g of finely powdered oxalic acid dihydrate (0.4 moles). The mixture is then brought to 130 ° C. with stirring with a heating time of approximately 10 minutes and the melt is held at 130 ° C. for 1.5 minutes.
200 ml of water are then added at a temperature of 90 ° C., the mixture is cooled to room temperature, the precipitate which has separated out is filtered and recrystallized from 300 ml of boiling water; a crude product was obtained in a yield of 8.5 g.
4.25 g of the crude product thus obtained are suspended in a solution of 4 g of sodium bicarbonate in 250 ml of water and stirred for 2 hours. The precipitate obtained is filtered off, the clear filtrate concentrated in vacuo to 50 ml and acidified with 2N hydrochloric acid.
The precipitate that forms is filtered off, washed with water, and after recrystallization from water, 800 mg of 3-carboxy-6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide monohydrate are obtained with a melting point of 285-287 C.

<I> Example 3 </I> 100 mg of 5-chloro-2,4-disulfamylaniline are mixed with 400 mg of finely pulverized oxalic acid dihydrate and the mixture is heated in an open sample tube at an oil bath temperature of 190-195 ° C. for 15 minutes. 2 g of this material are then treated in formic acid for 4 hours under reflux conditions, pure 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide being obtained.
A 2 g sample of the original reaction mixture is heated under reflux conditions in glacial acetic acid for 4 hours, and crude 6-chloro-7-sulfamyl-1,2,4-benzothiadiazine-1,1-dioxide is obtained. <I> Example 5 </I> A well-ground mixture of 11.43 g of 5-chloro-2,4-disulfamylaniline and 50.44 g of oxalic acid dihydrate is heated to melt at 126 ° C. for 1½ minutes. The mixture is then cooled to 110 ° C., treated with 200 ml of water and then cooled to room temperature.
The precipitate formed is filtered off, washed with water, and 10.5 g of crude product is obtained which, on the basis of physical measurements, consists of impure 3-carboxy-6-chloro-7-sulfamyl-1,2,4-benzothiazine -1,1-dioxide. 1.35 g of the product thus obtained were heated with 7 ml of diphenyl ether at 220 to 230 C for 3/4 hours with constant development of carbon dioxide.
