Verfahren zur Herstellung von Toxiferinen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Nordihydrotoxiferin oder von am N3 und/oder am NJ-Atom quaternisierten Derivaten davon, das dadurch gekennzeichnet ist, dass man Caracurin V der Formel

oder entsprechende quartäre oder bisquartäre Salze von Caracurin V bzw.
Va mit Halogenwasserstoff be handelt und anschliessend einer Hydrogenolyse unter wirft. Erhaltene nichtquaternisierte Produkte können erwünschtenfalls quaternisiert oder in ihre gewöhn lichen Salze übergeführt werden. Es ist zweckmässig, als Halogenwasserstoff Brom wasserstoff zu verwenden und die Reaktion in einem Lösungsmittel, z. B. in Eisessig, durchzuführen. Vor teilhaft arbeitet man unter Ausschluss von Licht und Feuchtigkeit sowie bei Raumtemperatur. Die Hydro- genolyse wird vorteilhaft mit Zinkstaub und Eisessig bewirkt.
Als Quaternisierungsmittel sind z. B. orga nische Halogenide oder Sulfate geeignet, z. B. Alkyl- oder Alkylenhalogenide, wie z. B. Methyljodid, Me- thylbromid, Allylbromid, oder Dimethylsulfat. Die allfällige Quaternisierung von erfindungsgemäss erhal tenem Nordihydrotoxiferin erfolgt zweckmässig in einem inerten Lösungsmittel, z. B. Benzol.
Zur Bil dung gewöhnlicher Salze kann man anorganische oder organische Säuren verwenden, z. B. Schwefelsäure, Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Phosphorsäure, Oxalsäure, Tohiolsulfosäure, Methan- sulfosäure, Pikrinsäure.
Die erfindungsgemäss erhaltenen Verfahrenspro dukte können z. B. durch Chromatographie oder Kri stallisation, z. B. über ihre schwerlöslichen Dijodide oder Dipikrate, gereinigt werden.
Die genannten Verfahrensendstoffe sind kristalline Verbindungen, die sich durch ihre ausgeprägten Wir kungen auf das Nervensystem auszeichnen; ein Teil derselben, insbesondere die bisquartären Salze haben curarisierende Wirksamkeit. Sie sollen als Heilmittel verwendet werden.
<I>Beispiel 1</I> Man tropft, unter Rühren, eine Lösung von 96 mg Caracurin V in 1 ml Eisessig innerhalb von 10 Minu ten in 30 ml Bromwasserstoff/Eisessig (bei 0 gesät tigte Lösung, 3: 100 verdünnt). Die gelbe Lösung wird 62 Stunden unter Licht- und Feuchtigkeitsaus schluss bei Raumtemperatur aufbewahrt. Anschliessend dampft man die Reaktionsmischung, aus welcher sich ein geringfügiger Niederschlag ausgeschieden hat, im Vakuum bei 40 ein. Der Rückstand wird mit 50 ml Eisessig und 5 g Zinkstaub 90 Minuten und nach Zugabe von 10 ml absolutem Methanol noch weitere 30 Minuten geschüttelt. Hierauf wird filtriert und der Rückstand mit viel Methanol gewaschen.
Das Filtrat wird im Vakuum eingedampft, in Wasser gelöst, mit wässrigem Ammoniak versetzt und hierauf mit Chloro form erschöpfend ausgeschüttelt. Der Chloroform- extrakt wird in benzolischer Lösung an 15g Alu miniumoxyd (Brockmann, 121/o Wasser) chromato- graphiert, wobei die Eluate papierchromatographisch verfolgt werden und aus den Nordihydrotoxiferin ent haltenden Fraktionen dieses isoliert wird.
Das erhal tene Nordihydrotoxiferin kann man danach durch Be handeln mit Methyljodid in Benzol in Dihydrotoxi- ferin-dijodid überführen, letzteres aus Aceton/Wasser umkristallisieren und an Amberlite IRA 400 (Chlo- ridform, eingetragene Marke)
in das Dihydrotoxiferin- dichlorid umwandeln und dieses zweimal aus Metha- nol/Äther umkristallisieren [a p = -605 5 (c = 0,377; 50 proz. Äthanol)]. Das so erhaltene Dihydro- toxiferin-dichlorid kann auf übliche Weise in Dihydro- toxiferin-dipikrat übergeführt werden, welches man einmal aus Aceton/Wasser umkristallisiert. Schmelz punkt l80-185 .
<I>Beispiel 2</I> 100 mg Caracurin Va werden in 1 ml Eisessig gelöst und mit 30 ml Bromwasserstoff/Eisessig (bei 0 gesättigte Lösung, 3 : 100 verdünnt) versetzt. Nach 60 Stunden dampft man im Vakuum ein und arbeitet nach den Angaben des Beispiels 1 weiter. Das erhal tene Nordihydrotoxiferin kann danach quaternisiert und als Dihydrotoxiferin-dichlorid wie im Beispiel 1 identifiziert werden.
<I>Beispiel 3</I> 100 mg C-Toxiferin-dichlorid löst man in 1 ml Eisessig und fügt 50 ml Bromwasserstoff/Eisessig (bei 0 gesättigte Lösung, 3 : 100 verdünnt) hinzu. Nach 60 Stunden dampft man im Vakuum ein. Den Rück stand schüttelt man mit<B>100</B> ml Eisessig und über schüssigem Zinkstaub während 90 Minuten, worauf man 20 ml absolutes Methanol zusetzt und noch 30 Minuten weiterschüttelt. Anschliessend filtriert man und wäscht mit absolutem Methanol nach. Die Flüs sigkeit wird im Vakuum eingedampft und der Rück stand mit Wasser aufgenommen.
Aus der wässerigen Lösung fällt man das Alkaloidgemisch mit wässeriger Pikrinsäurelösung, worauf die Pikrate durch Ionen austausch in die entsprechenden Chloride übergeführt werden. Das Dihydrotoxiferin-dichlorid wird durch Kristallisation abgetrennt und durch Umlösen aus Metanol/Äther gereinigt.
In ähnlicher Arbeitsweise lässt sich auch Caracurin V-dimethochlorid in Dihydrotoxiferin-dichlorid über führen.

The alkaloid mixture is precipitated from the aqueous solution with aqueous picric acid solution, whereupon the picrates are converted into the corresponding chlorides by ion exchange. The dihydrotoxiferin dichloride is separated off by crystallization and purified by dissolving from methanol / ether.
Caracurin V dimethochloride can also be converted into dihydrotoxiferin dichloride in a similar way.
