worin Hal für Chlor oder Brom steht, umsetzt. Die praktische Ausführung des Verfahrens gestal tet sich z. B. wie folgt: Die Lösung von 4-Hydroxy- piperidin und 2'-Bromäthyl-p-chlorbenzhydryläther in abs. Äthanol wird in Gegenwart eines halogenwasser- stoffbindenden Mittels, wie beispielsweise wasserfreies Natriumcarbonat, mehrere Stunden zum Sieden am Rückfluss erhitzt.
Nach beendeter Reaktion werden aus dem Reaktionsgemisch die anorganischen Salze abgetrennt. Das Endprodukt der Formel I wird durch fraktionierte Destillation im Hochvakuum isoliert und gereinigt und gewünschtenfalls mit anorganischen oder organischen Säuren in Salze übergeführt.
Die erfindungsgemäss hergestellten neuen Piperi- dinderivate sind bei Zimmertemperatur ölig oder fest. Es sind basische Verbindungen, die mit anorganischen oder organischen Säuren beständige kristallisierte Salze bilden.
Ist in Verbindungen der allgemeinen Formel I R1 verschieden von Wasserstoff, dann fällt das Reak tionsprodukt als Racemat an, das gewünschtenfalls nach bekannten Methoden getrennt werden kann.
Die nach dem vorliegenden Verfahren hergestell ten, bisher unbekannten Piperidinderivate sind auf Grund ihrer ausgezeichneten pharmakodynamischen Eigenschaften zur Verwendung als Heilmittel in hohem Masse geeignet. Sie zeichnen sich bei geringer Toxizität sowohl durch acetylcholinhemmende Wir kung (atropinähnliche Wirkung) als auch durch eine bemerkenswerte histaminhemmende Wirkung aus.
Wichtig ist dabei vor allem die lange Dauer des Anti- histamineffektes. Die neuen Verbindungen sollen des halb in der Therapie als Antihistaminica Verwendung finden. Sie dienen aber auch als Zwischenprodukte für die Herstellung von Medikamenten.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelz- und Siedepunkte sind nicht korrigiert.
<I>Beispiel I</I> 1-(2'-p-Chlorbenzhydryloxy-äthyl)-4-hydroxy- piperidin Die Lösung von 4 g 4-Hydroxypiperidin, 13 g 2'-Bromäthyl-p-chlorbenzhydryläther in 50 cm3 abs. Äthanol und 2,5g wasserfreies Natriumcarbonat wer den 8 Stunden am Rückfluss zum Sieden erhitzt, wo bei nach je 4 Stunden nochmals total 7,5g wasser freies Natriumcarbonat in zwei Portionen zugegeben werden. Zur Vervollständigung der Reaktion wird noch 12-l4 Stunden erhitzt.
Nach dem Erkalten wird von anorganischen Salzen abfiltriert und das Filtrat im Vakuum eingedampft. Der Rückstand wird in Benzol gelöst, von ungelösten Bestandteilen filtriert und das Lösungsmittel wieder im Vakuum verdampft. Der Rückstand wird im Vakuum destilliert, wobei bei 0,02 mm Hg und einer Badtemperatur von 190 bis 210 das 1- (2' - p - Chlorbenzhydryloxy-äthyl)-4-hy- droxy-piperidin als schwach gelb gefärbtes, viskoses Öl übergeht, das in der Kälte harzartig erstarrt.
Zur Herstellung des neutralen Naphthalin-1,5- disulfonates wird die Lösung von 8,12 g 1-(2'-p- Chlorbenzhydryloxy - äthyl) - 4 - hydroxy-piperidin in 20 cm3 Äthanol mit 4,62 g 76,3%iger Naphthalin- 1,5-disulfonsäure versetzt. Nach Zusatz von Äther kristallisiert das neutrale Naphthalin-1,5-disulfonat aus. Nach Umkristallisieren aus Methanol schmilzt das Salz bei 189-190 .
Der als Ausgangsmaterial zur Verwendung ge langende 2' - Bromäthyl - p-chlorbenzhydryläther wird wie folgt hergestellt: 11,85 g p-Chlor-diphenyl-chlor- methan, 8,98g Äthylenbromhydrin und 5,12 g was serfreies Natriumcarbonat werden 8 Stunden unter Rühren auf 120 erhitzt.
Nach Zusatz von 50 cm- Benzol filtriert man die ausgefallenen anorganischen Salze ab, dampft die Benzollösung im Vakuum ein und destilliert den Rückstand. Sdp. 152-153o/ 0,05 mm Hg. <I>Beispiel 2</I> 1-(2'-Benzhydryloxy-äthyl)-4-hydroxy-piperidin Die Lösung von 5 g 4-Hydroxy-piperidin, 15 g 2'-Bromäthyl-benzhydryläther in 50 cm-- absolutem Athanol und 5,
5g wasserfreies Natriumcarbonat wer den 8 Stunden am Rückfluss zum Sieden erhitzt. Man gibt anschliessend weitere 5,5 g wasserfreies Na- triumcarbonat zu und erhitzt, zur Vervollständigung der Reaktion, nochmals 15 Stunden. Nach dem Er kalten wird von anorganischen Salzen abfiltriert und das Filtrat im Vakuum eingedampft. Der Rückstand wird in Benzol aufgenommen, von ungelösten Be standteilen filtriert und das Lösungsmittel wieder im Vakuum verdampft.
Der Rückstand wird im Kugel rohr destilliert, wobei unter einem Druck von 0,02 mm Hg das 1-(2'-Benzhydryloxy-äthyl)-4-hy- droxypiperidin bei 200-210 als viskoses Öl übergeht.
Das Hydrochlorid kristallisiert aus Wasser mit einem Mol Kristallwasser. Smp. 110-112 (Zers.). Analog wie oben beschrieben werden aus 4- Hydroxy-piperidin und einem 2'- Bromäthyl - benz- hydryläther der Formel 1I die folgenden Verbindun gen erhalten:

If R1 is different from hydrogen in compounds of the general formula I, the reaction product is obtained as a racemate which, if desired, can be separated by known methods.
The previously unknown piperidine derivatives produced by the present process are highly suitable for use as medicinal products due to their excellent pharmacodynamic properties. With low toxicity, they are distinguished by both an acetylcholine-inhibiting effect (atropine-like effect) and a remarkable histamine-inhibiting effect.
The long duration of the antihistamine effect is particularly important. The new compounds should therefore be used in therapy as antihistamines. But they also serve as intermediate products for the manufacture of medicines.
In the following examples, all temperatures are given in degrees Celsius. The melting and boiling points are not corrected.
<I> Example I </I> 1- (2'-p-chlorobenzhydryloxy-ethyl) -4-hydroxypiperidine The solution of 4 g of 4-hydroxypiperidine, 13 g of 2'-bromoethyl-p-chlorobenzhydryl ether in 50 cm3 of abs .
