Verfahren zur Herstellung von 16-Methylen-17a-hydroxy-ster oiden Es wurde gefunden, dass die therapeutisch wert vollen 16-Methylen-17a-hydroxy-steroide durch Be handlung der entsprechenden 16a,17a-Oxido-16ss- methyl-steroide mit mindestens katalytischen Mengen einer starken Säure oder einer Lewis-Säure erhalten werden können.
Diese Reaktion ist von grundlegender Bedeutung. Insbesondere ist es überraschend, dass bei der Be handlung von 16a,17a-Oxido-16p'-methyl-steroiden mit Säuren nicht die entsprechenden 16,17-Di- hydroxy-16-methyl-steroide gebildet werden, sondern direkt Steroide mit einer exocyclischen Methylen- gruppe am C-Atom 16 und einer alphaständigen Hydroxylgruppe am C-Atom 17 erhalten werden.
Gegenstand der Erfindung ist dementsprechend ein Verfahren zur Herstellung von 16-Methylen-17a- hydroxy-steroiden, das darin besteht, dass man ein 16r(,17(/-Oxido -16ss - methyl-steroid durch Behand lung mit mindestens katalytisch wirkenden Mengen einer starken Säure oder einer Lewis-Säure in Ge genwart eines inerten Lösungsmittels in das entspre chende 16-Methylen-17a-hydroxy-steroid überführt.
Als Säuren kann man für die Aufspaltungsreak- tion nach der Erfindung vorzugsweise Mineralsäuren, wie z. B. Schwefelsäure, Salzsäure, Bromwasserstoff- säure usw., verwenden. Sehr gut geeignet sind auch Alkyl- oder Arylsulfonsäuren, insbesondere p-Toluol- sulfonsäure. Die Säuren können sowohl in sehr ge ringen, z. B. katalytisch wirkenden Mengen, als auch in molaren oder überschüssigen Mengen angewendet werden.
Die Umsetzung wird in Gegenwart eines inerten Lösungsmittels durchgeführt. Als Lösungsmittel sind z. B. verwendbar: Alkohole wie Methanol, Äthanol, Propanol, Isopropanol, Butanol, tert.-Butanol oder Amylalkohol, Kohlenwasserstoffe wie Benzol oder Toluol, chlorierte Kohlenwasserstoffe wie Tetrachlor- kohlenstoff, Chloroform,
Methylenchlorid oder Di- chloräthan, ferner Äther wie Dimethyläther oder Di- äthyläther, oder Ester wie Essigsäuremethyl- oder äthylester oder Keton wie Aceton oder Methyl-äthyl- keton, aber auch Eisessig und Acetanhydrid. Grund sätzlich sind alle diejenigen Lösungsmittel verwend bar, die mit dem als Ausgangsmaterial eingesetzten 16a,
17a-Oxido-16ss-methyl-steroid sowie dem ent stehenden 16-Methylen-17a-hydroxy-steroid nicht reagieren und das Ausgangssteroid wenigstens in ge ringem Masse lösen.
Das Verfahren nach der Erfindung ist allgemein anwendbar. Die als Ausgangsmaterial verwendeten 16a,17a - Oxido -16ss - methyl - steroide können der Testan-, Östran-, Pregnan- oder Androstan-Reihe angehören und in den verschiedensten Stellen funk tionell substituiert sein sowie eine Doppelbindung oder mehrere Doppelbindungen enthalten. Zum Bei spiel können sich Sauerstoffunktionen an den C-Atomen 3, 11, 20 und/Oder 21 befinden.
Halogen atome können an den C-Atomen 2, 4, 6, 9 und 21 stehen, Doppelbindungen können sich befinden zwi schen den C-Atomen 1 und 2, 3 und 4, 4 und 5, 5 und 6, 6 und 7 und 9 und 11, Alkylgruppen an den C-Atomen 2 und 6.
Nach der Erfindung kann man aus den entspre chenden 16a,1.7a-Oxido-16ss-methyl-steroiden z. B. die folgenden 16-Methylen-steroide herstellen: 16-Methylen-5-pregnen-3,Q,17a-diol-20-on- 3-acetat, 16-Methylen-4-pregnen-17a-ol-3,20-dion, 6(/-Fluor-1 6-methylen-4-pregnen-17a-ol- 3,20-dion, 6pi-.Fluor-1 6-methylen-4-pregnen 17a-ol- 3,20-dion, 16-Methylen-5-pregnen-3f3,
17a-diol-20-on- acetat usw.
Die Reaktionszeiten für die Umsetzungen nach der Erfindung liegen zwischen wenigen Minuten und etwa 6 Stunden. In der Regel benötigt man eine Re aktionszeit von 1-4 Stunden. Die Umsetzung führt man zweckmässigerweise in der Wärme, z. B. durch kochen des Reaktionsgemisches am Rückfluss, durch. Man kann aber die Aufspaltung auch durchführen, indem man das Reaktionsgemisch bei Zimmertempe ratur einige Tage stehenlässt.
Das Verfahren nach der Erfindung stellt eine völlig neue Methode zur Einführung einer exocycli- schen Methylengruppe in Steroide dar und bedeutet gegenüber den bisher bekannten Verfahren zur Her stellung von 16-Methylen-steroiden einen wesent lichen technischen Fortschritt, da die Anzahl der be nötigten Reaktionsstufen wesentlich geringer ist als die bei den bekannten Verfahren (vgl. z.
B. USA- Patente Nrn. 2 865 808 und 2 902 483) und die er haltenen Ausbeuten um ein Vielfaches grösser sind als die Ausbeuten, nach den in den genannten USA- Patentschriften beschriebenen Verfahren.
Die als Ausgangsmaterial benötigten 16a,17a- Oxido-16/3-methyl-steroide sind nach bekannten Methoden durch Epoxydierung der zu Grunde lie genden 16-Methyl-16-dehydro-steroide zugänglich. <I>Beispiele</I> 1. .16-Methylen-5-pregnen-3/3,17a-diol-20-on- 3-acetat.
Eine Lösung von 9,3 g 16a,17a-Oxido-16/3-me- thyl-5-pregnen-3j3-ol-20-on-3-acetat (1) in 50 ml wasserfreiem Benzol wird nach Zugabe von 650 mg p-Toluolsulfonsäure 2 Stunden unter kontinuierli cher Wasserabscheidung am Rückfluss erhitzt. Die benzolische Lösung wird anschliessend zuerst mit 5a/aiger Natriumhydrogencarbonat-Lösung, dann mit Wasser gewaschen und eingedampft. Der bräunliche kristalline Rückstand schmilzt bei 187-193 .
Durch Umkristallisieren aus Methanol erhält man reines 16-Methylen - 5 - pregnen-3/3,17 a-diol-20-on-3-acetat vom Schmp. 196-198"; WD -140 (Chloroform).
2. 16-Methylen-4-pregnen-17a-ol-3,20-dion.
10 g 16a,17a-Oxido-16/3-methyl-4.-pregnen-3,20- dion und 680 mg p-Toluolsulfonsäure werden in 300 ml Benzol unter kontinuierlicher Wasserabschei- dung 4 Stunden am Rückfluss erhitzt. Dann wird wie in Beispiel 1 beschrieben aufgearbeitet.
Aus dem erhaltenen Rückstand wird durch Umkristallisieren aus viel Methanol reines 16-Methylen-4-pregnen- 17a-ol-3,20-dion vom Schmelzpunkt 221-223 erhal ten. (a)D-10,7 (Chloroform). 240-241 m,a, e = 17 300. 3. 6ci-Fluor-16-methylen-4-pregnen-17r@-ol- 3,20-dion.
13 g 6a-Fluor-16a,17a - oxido -16i> - methyl-4- pregnen-3,20-1ion und 430 mg p-Toluolsulfonsäure werden in 1300m1 Benzol 4 Stunden unter konti nuierlicher Wasserabscheidung am Rückfluss erhitzt. Die Lösung wird wie unter 1 aufgearbeitet und der erhaltene Rückstand aus Äthanol umkristallisiert.
Das reine 6p,-Fluor-16-methylen-4-pregnen-17a-ol- 3,20 - dion schmilzt bei 206-208". (a)D -16,0 (Chloroform). 2.",;" 235 my, e <I>=</I> 15 950.
4. 613-Fluor-16-methylen-4-pregnen-17a-ol- 3,20-dion.
Eine Lösung von 3,2 g 61i-Fluor-16a,17a-oxido- 16/3 - methyl - pregnan - 5c < - ol-3,20-dion und 25 mg p-Toluolsulfonsäure in 650 ml Benzol wird unter kontinuierlicher Wasserabscheidung 6 Stunden am Rückfluss erhitzt und dann wie üblich aufgearbeitet. Der kristalline Rückstand wird aus Methanol umkri stallisiert.
Das reine 6(3-Fluor-16-methylen-4-pregnen- 17cc-ol-3,20-dion schmilzt bei 253-255 . (a)D -90,6 (Chloroform). Z""" 232 my, ± = 13 700.
5. 16-Methylen-5-pregnen-3/3,17a-diol-20-on- 3-acetat.
5 g 16f)-Methyl-16a,17a-oxido-5-pregnen-3i3-o1- 20-on-acetat werden in 300 ml Methanol gelöst. Die Lösung wird nach Zugabe von 0,25 g p-Toluolsulfon- säure 5 Stunden am Rückfluss erhitzt und dann in 2 1 Wasser eingegossen. Der Niederschlag wird abge saugt, getrocknet und mit 50 ml Essigsäureanhydrid und 50 ml Pyridin 1 Stunde auf dem Dampfbad er hitzt. Die Lösung wird dann in 1 1 Wasser eingegos sen, das rohe 16-Methylen-5-pregnen-3@,17a-diol- 20-on-3-acetat abgesaugt, getrocknet und aus Me thanol umkristallisiert.
6. 16-Methylen-5-pregnen-313,17cc-diol-20-on- 3-acetat.
Die Lösung von 5 g 16ss-Methyl-16a,17a-oxido- 5-pregnen-3/3-ol-20-on-acetat in 50 ml Dioxan wird mit 0,1 ml konzentrierter Schwefelsäure versetzt, 1 Stunde auf 100 erhitzt und dann in 1 1 Wasser eingegossen. Der Niederschlag wird abgesaugt und aus Methanol umkristallisiert.
7. 16-Methylen-5-pregnen-3[3,17rc-diol-20-on- 3-acetat.
Die Lösung von 2 g 16j3-Methyl-16cc,17a-oxido- 5-pregnen-313-ol-20-on-acetat in 20 ml Dioxan wird nach Zugabe von 0,1 g Benzolsulfonsäure 5 Stunden zum Sieden erhitzt und dann in Wasser eingegossen. Der Niederschlag wird nach dem Absaugen aus Me thanol umkristallisiert.
B. 16-Methylen-5-pregnen-3i3,17a-diol-20-on- 3-acetat.
2 g 16/3-Methyl-16ci,17a-oxido-5-pregnen-3j3-ol- 20-on-acetat werden in 80 ml Eisessig unter Erwär men gelöst. Die Lösung wird nach Zugabe von 0,3 ml einer 5,6n/oigen (18,2 mg) Lösung von Salzsäure in Eisessig 48 Stunden bei Raumtemperatur stehen gelassen und dann in Wasser eingegossen. Der aus geschiedene Niederschlag wird aus Methanol umkri stallisiert.
9. 16-Methylen-5-pregnen-3ss,17a-diol-20-on- 3-acetat.
Die Lösung von 2 g 16ss-Methyl-16a,17a-oxido- 5-pregnen-3ss-ol-20-on-acetat in 20 ml wasserfreiem Chloroform, die 18,2 mg Salzsäure enthalten, wird 1 Stunde am Rückfluss erhitzt und danach zur Trockne eingedampft. Der Rückstand besteht aus reinem 16-Methylen-5-pregnen-3j3,17a-diol-20-on-3- acetat.
10. 16-Methylen-4-pregnen-17a-ol-3,20-dion. Die Lösung von 2 g 16ss-Methyl-16a,17a-oxido- 4-pregnen-3,20-dion in 20 ml wasserfreiem Benzol, die 21,3 mg Salzsäure enthalten, wird 6 Stunden am Rückfluss erhitzt und dann zur Trockne eingedampft, Der erhaltene Rückstand wird aus Methanol um kristallisiert.
11. 16-Methylen-5-pregnen-3/3,17a-diol-20-on- acetat.
Die Lösung von 5 g 16l-Methyl-16a,17a-oxido- 5-pregnen-3/3-ol-20-on-acetat wird in einem Gemisch aus 300 ml Äther und 100 ml Benzol mit 15,5 ml einer 14,450/aigen Lösung von p-Toluolsulfonsäure in Äther versetzt und 60 Stunden bei Raumtempera tur stehengelassen. Dann wird unter vermindertem Druck bei einer Badtemperatur von 20 zur Trockne eingedampft. Der Rückstand wird mit 25 ml Me thanol aufgeschlemmt und das reine 16-Methylen- 5-pregnen-3ss,17a-diol-20-on-acetat abgesaugt.
12. 16-Methylen-5-pregnen-3J3,17a-diol-20-on- acetat.
1 g 16ss-Methyl-16a,17a-oxido-5-pregnen-3ss-ol- 20-on-acetat wird in 10 ml Benzol gelöst und mit 0,036 ml Bortrifluorid-Ätherat versetzt. Nach 16- stündigem Stehen bei Raumtemperatur wird die Lö sung mit einer wässrigen Natriumhydrogencarbonat- lösung und Wasser gewaschen, getrocknet und ein gedampft. Der Rückstand wird aus Methanol um kristallisiert.
13. 16-Methylen-5-pregnen-3j3,17a-diol-20-on- acetat.
2 g 16ss-Methyl-16a,17a-oxido-5-pregnen-3ss-ol- 20-on-acetat werden in 20 ml Dioxan gelöst und mit 0,05 ml einer 70%igen Perchlorsäure 1 Stunde am Rückfluss gekocht. Die Lösung wird in Wasser ein gegossen, der Niederschlag abgesaugt und aus Me thanol umkristallisiert.
14. 16-Methylen-5-pregnen-3j3,17a-diol-20-on- acetat.
2 g 16j3-Methyl-16a,17a-oxido-5-pregnen-3ss-ol- 20-on-acetat werden mit 0,1g Methansulfonsäure in 60 ml Benzol 3 Stunden unter kontinuierlicher Wasserabscheidung am Rückfluss gekocht. Die Lö sung wird mit wässriger Natriumhydrogencarbonat- lösung und Wasser gewaschen, getrocknet und einge dampft. Der Rückstand wird aus Methanol umkri stallisiert.
15. 16-Methylen-5 pregnen-3ss,17a-diol-20-on- acetat.
2 g 16ss-Methyl-16a,17a-oxido-5-pregnen-3/3-ol- 20-on-acetat werden in 20 ml Benzol, die 21 mg Bromwasserstoff enthalten, 2 Stunden am Rückfluss gekocht. Die Lösung wird eingedampft und der Rückstand aus Methanol umkristallisiert.
16. 16-Methylen-5 pregnen-3ss,17a-diol-20-on- 3-acetat.
Die Lösung von 2 g 16ss-Methyl-16a,17a-oxido- 5 pregnen-3ss-ol-20-acetat in 45 ml wasserfreiem Essigester, die 18 mg Salzsäure enthalten, wird 2 Stunden am Rückfluss erhitzt und dann zur Trockne eingedampft. Der Rückstand von rohem 16-Methy- len-5-pregnen-3ss,17a-diol-20-on-3-acetat wird aus Methanol umkristallisiert.
17. 16-Methylen-5-pregnen-3/3,17a-diol-20-on- 3-acetat.
2 g 16ss Methyl-16a,17a-oxido-5-pregnen-3ss-ol- 20-on-acetat werden in 40 ml Aceton gelöst. Die Lö- sung wird nach Zugabe von 0,22 ml einer 8,2 % igen Salzsäurelösung in Aceton 2 Stunden am Rückfluss erhitzt und dann zur Trockne eingedampft.
18. 16-Methylen-4-pregnen-17a-ol-3,20-dion. Die Lösung von 2 g 16/3-Methyl-16a,17a-oxido- 4 - pregnen - 3,20 - dion in 20 ml Chloroform, die 21,4 mg Salzsäure enthalten, wird 16 Stunden am Rückfluss erhitzt und dann zur Trockne eingedampft. Der Rückstand wird aus Methanol umkristallisiert und ergibt reines 16-Methylen-4-pregnen-17a-ol- 3,20-dion.
Bei Verwendung von Aceton oder Essigester als Lösungsmittel beträgt die Reaktionszeit nur 2 Stun den.
