Verfahren zur Herstellung von substituierten 2-Phenyl-7-aminoalkoxy-chromonen
In der deutschen Patentschrift Nur.1018874 wird die Herstellung von N-substituierten 2-Methyl 7-aminoalkoxy-chromonen beschrieben. Man erhält diese Produkte durch Umsetzung der Alkalisalze von 2-Methyl-7-oxychromon mit basisch substituierten Alkylhalogeniden. Pharmakologisch zeichnen sich diese Verbindungen durch khellinähnliche Eigenschaften aus, wobei als Vorteile gegenüber Khellin die grössere therapeutische Breite und die gute Wasserlöslichkeit anzusehen sind.
Es wurde nun gefunden, dass die als Salze eben falls gut t wasserlöslichen N- und gegebenenfalls im Phenylrest substituierten 2- Phenyl - 7-aminoalkoxy- chromone auf analogem Wege aus 2-Phenyl-7-oxy chromon und basisch substituierten Alkylhalogeniden herstellbar sind, jedoch in ihren pharmakologischen Eigenschaften den Produkten der oben erwähnten Patentschrift überlegen sind. Dies kommt besonders zum Ausdruck, wenn man die muskulotrop-spasmolytische Wirkung am Meers chweinchend arm prüft.
Die Phenylchromone wirken in allen Fällen erheblich stärker und sind noch ungiftiger als die analogen Methylderivate. Zur Erläuterung diene folgende Tabelle: Pharmakologischer Vergleich von Verbindungen der Formel

Am isolierten Meerschweinchenherz wirken alle untersuchten Substanzen ähnlich gut coronarerweiternd wie die Methylchromonen, doch zeigen sich in einigen Fällen auch hier Vorteile.
So zeigt das 4' Methoxy - 2-phenyl-7-(N-dimethylaminoäthoxy)-cllro- mon-hydrochlorid eine besonders starke Coronarerweiterung, die etwa das 7fache der Theophyllin Wirkung erreicht, ohne dass dabei ein negativ inotroper Effekt auftritt, der bei den Methylchromonen immer zu beobachten ist.
Zur Prüfung der peripheren Gefässwirkung wurden Versuche am isolierten durchströmten Kaninchenohr durchgeführt. In dieser Versuchsanordnung wirkten 100 7-Piperidino-äthoxy-2-phenyl-chromonhydrochlorid etwa 4mal so stark wie 100 7-Piperidino-äthoxy-2-methyl-chromon-hydrochlorid.
Der 2-Phenylrest der erfindungsgemässen Verbindungen kann gegebenenfalls auch substituiert sein. Besonders bevorzugt sind als Substituenten Alkoxygruppen, insbesondere die Methoxygruppe. Die Alkoxygruppe steht vorzugsweise in der p-Stellung des 2-Phenylrestes.
Beispiel 1
1,15 g Natrium werden in 65 cm3 absolutem Methylalkohol gelöst und 12 g 7-Oxy-2-phenylchromon zugegeben. Man destilliert das Lösungsmittel auf dem Wasserbad im Vakuum ab und nimmt den Rückstand in 95 cm3 absolutem Dioxan auf. Anschliessend gibt man eine Lösung von 6,7 g N-y-Dimethylaminopropylchlorid in 20 cm3 absolutem Dioxan dazu, rührt 4 Stunden bei 90-100 Innentemperatur und destilliert das Dioxan im Vakuum ab. Der Rückstand wird mit Wasser verrührt, nach kurzem Stehen abgesaugt, mit Wasser gewaschen und im Exsiccator getrocknet. Die so erhaltene Base wird in absolutem Äthylalkohol gelöst und mit alkoholischer Salzsäure schwach angesäuert.
Nach dem Abkühlen wird das reine 2- Phenyl-7-(N-yimethylaminopropoxy)chro- mon-hydrochlorid abgesaugt und in der Trockenpistole bei 1300 getrocknet. Man erhält 7,5 g vom Schmelzpunkt 219-220 .
Beispiel 2
Es wird analog wie im Beispiel 1 gearbeitet, jedoch wird an Stelle von N-,-Dimethylamino-propyl- chlorid N-,R-Ohloräthyl-piperidin verwendet. Ausgehend von 20 g 7-Oxy-2-phenylchromon erhält man 22 g 2- Phenyl - 7-y-piperidinoäthoxy)-chromon- hydrochlorid, das nach dem Umkristallisieren aus Äthylalkohol bei 248-2490 schmilzt.
Beispiel 3
Es wird analog wie im Beispiel 1 gearbeitet, jedoch wird an Stelle von N-;}-Dimethylamino-propyl- chlorid N-, B-Dimethylamino-äthylchlorid verwendet.
Der Schmelzpunkt des erhaltenen 2-Phenyl-7-(N-fi- dimethylaminoäthoxy)-chromon-hydrochlorid beträgt 216-220 .
Beispiel 4
Es wird analog wie im Beispiel 1 gearbeitet, jedoch wird an Stelle von N-;-Dimethylamino-propyl- chlorid N-fl-Diäthylamino-äthylchlorid verwendet.
Der Schmelzpunkt des erhaltenen 2-Phenyl-7-(N-p- diäthylaminoät, hoxy)-chromon-hydrochlorid beträgt 187-188 .
Beispiel 5
Es wird analog wie im Beispiel 1 gearbeitet, jedoch wird an Stelle von N-;-Dimethylamino-propyl- chlorid N-4-Chloräthyl-morpholin verwendet. Man erhält 2-Phenyl-7-{N-fi-morpholinoäthoxy)hromon hydrochlorid vom Schmelzpunkt 253-2540.
Beispiel 6
Zu einer Lösung von 0,66 g Natrium in 100 cm3 absolutem Methanol werden 7,7 g 7-Oxy-4'-methoxy2-phenyl-chromon zugesetzt. Man rührt, bis alles gelöst ist und dampft im Vakuum zur Trockne ein. Der Rückstand wird in der Reibschale zerrieben, in 75 cm3 reinem Dioxan suspendiert und dazu 3,4 g N-fl-Dimethylamino-äthylchlond gegeben. Nun kocht man 3 Stunden unter Rühren am Rückfluss, destilliert das Dioxan im Vakuum ab und extrahiert den Rückstand mit Äther. Die nach dem Abdestillieren des ethers erhaltene Base vom Schmelzpunkt 118 bis 119 wird durch Lösen in Äthylalkohol, Ansäuern mit alkoholischer Salzsäure und Fällen mit absolutem Äther in das Hydrochlorid übergeführt.
Man erhält so 8 g 4'-Methoxy-2-phenyl-7-(N-B-dimethylamino- äthoxy)-chromon-hydrochlorid, das durch Umkristallisieren aus Isopropylalkohol gereinigt wird. Der Schmelzpunkt beträgt 220-2210.
Beispiel 7
Das aus 10 g 7-Oxy-4'-methoxy-flavon hergestellte Natriumsalz wird in 100 cms Dioxan suspendiert, und bei 800 werden 5,45 g Dimethylamino- propylchlorid - in wenig Dioxan gelöst - zugetropft.
Dann kocht man zwei Stunden am Rückfluss, filtriert, säuert mit alkoholischer Salzsäure an und saugt das Rohprodukt nach einigem Stehen ab. Nach Umkristallisieren aus Äthylalkohol erhält man 10,3 g 2- (p-Methoxyphenyl)-7-dimethylaminopropoxy-ohro- mon-hydrochlorid vom Schmelzpunkt 231-2330.
Analog wurden folgende Verbindungen gewonnen: 1. 2-(p-Methoxyphenyl)-7-piperidinoäthoxy- chromon-hydrochlorid vom Schmelzpunkt
237-2380.
2. 2-(p-Methoxyphenyl)-7-diäthylaminoäthoxy- chromon-hydrochlorid vom Schmelzpunkt 231-233 .
3. 2-(p-MethoxyphenyD-7-pyrolidinoäthoxy- chromon-hydrochlorid vom Schmelzpunkt 236-2380.
4. 2- (p-Methoxyphenyl) -7-pyrolidinopropoxy- chromon-hydrochlorid vom Schmelzpunkt 230-2320.
Process for the preparation of substituted 2-phenyl-7-aminoalkoxy-chromones
The German patent Nur.1018874 describes the preparation of N-substituted 2-methyl 7-aminoalkoxy-chromones. These products are obtained by reacting the alkali metal salts of 2-methyl-7-oxychromone with basic substituted alkyl halides. Pharmacologically, these compounds are characterized by properties similar to khellin, the advantages over khellin being the greater therapeutic range and good water solubility.
It has now been found that the salts which are also readily water-soluble N- and 2-phenyl-7-aminoalkoxy-chromones which are optionally substituted in the phenyl radical can be prepared in an analogous way from 2-phenyl-7-oxy-chromone and basic substituted alkyl halides, however are superior in their pharmacological properties to the products of the above-mentioned patent. This is particularly evident when the musculotropic-spasmolytic effect is tested on poor guinea pigs.
In all cases, the phenylchromones have a considerably stronger effect and are even less toxic than the analogous methyl derivatives. The following table serves as an illustration: Pharmacological comparison of compounds of the formula

In the isolated guinea pig heart, all the substances investigated have a similarly good coronary-expanding effect as methylchromones, but in some cases they also have advantages.
Thus, the 4 'methoxy-2-phenyl-7- (N-dimethylaminoethoxy) -clro- mono hydrochloride shows a particularly strong coronary expansion, which reaches about 7 times the theophylline effect, without a negative inotropic effect occurring in the Methyl chromones is always observed.
To test the peripheral vascular effect, experiments were carried out on isolated rabbit ears with blood flow. In this experimental set-up, 100 7-piperidino-ethoxy-2-phenylchromone hydrochloride was about 4 times as effective as 100 7-piperidino-ethoxy-2-methylchromone hydrochloride.
The 2-phenyl radical of the compounds according to the invention can optionally also be substituted. Particularly preferred substituents are alkoxy groups, especially the methoxy group. The alkoxy group is preferably in the p-position of the 2-phenyl radical.
example 1
1.15 g of sodium are dissolved in 65 cm3 of absolute methyl alcohol and 12 g of 7-oxy-2-phenylchromone are added. The solvent is distilled off on a water bath in vacuo and the residue is taken up in 95 cm3 of absolute dioxane. A solution of 6.7 g of N-γ-dimethylaminopropyl chloride in 20 cm3 of absolute dioxane is then added, the mixture is stirred for 4 hours at an internal temperature of 90-100 and the dioxane is distilled off in vacuo. The residue is stirred with water, filtered off with suction after standing for a short time, washed with water and dried in a desiccator. The base obtained in this way is dissolved in absolute ethyl alcohol and weakly acidified with alcoholic hydrochloric acid.
After cooling, the pure 2-phenyl-7- (N-yimethylaminopropoxy) chromium hydrochloride is filtered off with suction and dried at 1300 in a drying gun. 7.5 g with a melting point of 219-220 are obtained.
Example 2
The procedure is analogous to that in Example 1, but instead of N -, - dimethylamino-propyl chloride, N-, R-chloroethyl piperidine is used. Starting from 20 g of 7-oxy-2-phenylchromone, 22 g of 2-phenyl-7-y-piperidinoethoxy) -chromone hydrochloride are obtained, which, after recrystallization from ethyl alcohol, melts at 248-2490.
Example 3
The procedure is analogous to that in Example 1, but instead of N -;} - dimethylamino-propyl chloride, N-, B-dimethylamino-ethyl chloride is used.
The melting point of the 2-phenyl-7- (N-fi- dimethylaminoethoxy) chromone hydrochloride obtained is 216-220.
Example 4
The procedure is analogous to that in Example 1, except that N-fl-diethylamino-ethyl chloride is used instead of N -; - dimethylamino-propyl chloride.
The melting point of the 2-phenyl-7- (N-p-diethylaminoät, hoxy) -chromon hydrochloride obtained is 187-188.
Example 5
The procedure is analogous to that in Example 1, except that N-4-chloroethyl morpholine is used instead of N -; - dimethylamino-propyl chloride. 2-Phenyl-7- {N-fi-morpholinoethoxy) hromone hydrochloride with a melting point of 253-2540 is obtained.
Example 6
7.7 g of 7-oxy-4'-methoxy2-phenylchromone are added to a solution of 0.66 g of sodium in 100 cm3 of absolute methanol. The mixture is stirred until everything has dissolved and evaporated to dryness in a vacuum. The residue is ground up in the mortar, suspended in 75 cm3 of pure dioxane and 3.4 g of N-fl-dimethylamino-ethyl chloride are added. The mixture is then refluxed for 3 hours with stirring, the dioxane is distilled off in vacuo and the residue is extracted with ether. The base obtained after the ether has been distilled off and has a melting point of 118 to 119 is converted into the hydrochloride by dissolving it in ethyl alcohol, acidifying it with alcoholic hydrochloric acid and precipitating it with absolute ether.
This gives 8 g of 4'-methoxy-2-phenyl-7- (N-B-dimethylaminoethoxy) chromone hydrochloride, which is purified by recrystallization from isopropyl alcohol. The melting point is 220-2210.
Example 7
The sodium salt prepared from 10 g of 7-oxy-4'-methoxy-flavone is suspended in 100 cms of dioxane, and at 800, 5.45 g of dimethylaminopropyl chloride - dissolved in a little dioxane - are added dropwise.
Then it is refluxed for two hours, filtered, acidified with alcoholic hydrochloric acid and the crude product is filtered off with suction after standing for a while. After recrystallization from ethyl alcohol, 10.3 g of 2- (p-methoxyphenyl) -7-dimethylaminopropoxy-ohromone hydrochloride with a melting point of 231-2330 are obtained.

2. 2- (p-Methoxyphenyl) -7-diethylaminoethoxychromone hydrochloride, melting point 231-233.
3. 2- (p-MethoxyphenyD-7-pyrolidinoethoxychromone hydrochloride, melting point 236-2380.
4. 2- (p-Methoxyphenyl) -7-pyrolidinopropoxychromone hydrochloride, melting point 230-2320.
