Verfahren zur Herstellung eines neuen Antibiotikums Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung des neuen Antibiotikums <B>A20388,</B> das dadurch gekennzeichnet ist, dass man den S.treptomyces-Stamm A 20388 züchtet und hier auf das Antibiotikum <B>A20388</B> isoliert.
Das Antibiotikum A 20388 entsteht bei der Kul tur eines neuen Stammes der Gattung Streptomyces, der aus einer in Ceylon gesammelten Bodenprobe isoliert wurde, und in unseren Laboratorien und in der Eidg. Technischen Hochschule, Institut für spe zielle Botanik, Zürich, unter der Bezeichnung <B>A20388</B> aufbewahrt wird.
Der Stamm A 20388 bildet ein zimtbraunes Luft- mycel. Die Sporenketten ,sind monopodial verzweigt mit engen, geschlossenen -Spiralen. Die Oberfläche der einzelnen Sponen ist glatt. Beim Wachstum auf peptonhaltigen Nährböden wird keine melanoide Verfärbung beobachtet.
Das Wachstum ist relativ wenig abhängig von der Temperatur, sowohl bei 18 als auch bei 40aentwickelt sich der Pilz gut.
Zur weiteren Charakterisierung wird im folgen den das Wachstum des Stammes<B>A20388</B> auf ver schiedenen Nährmedien beschrieben. Die Nähr medien 1-7 und 10 wurden nach W. Lindenbein, Arch. M'dcrobiol. <I>17,</I> 361 (1952) hergestellt.
1. Synthetischer Agar: Wachstum dünn, schleierartig, weissgelb 2. Synthetische Lösung: feine, weissgelbe Trübung. 3. Glukose-Agar: Wachstum dünn, schleierartig, weissgelb bis hellgelb.
4. Glukose-Asparagin-Agar: Wachsture dünn, schleierartig, hellgelb, Luftmycel sammetig, kreideweiss 'bis zimtbraun. 5. Calciummalat-Agar: Wachstum dünn, .schleierartig, weissgelb, Luft- mycel spärlich, meist milchweiss, an einigen Stellen zimtbraun.
6. Gelatinstich (18 ): Wachstum spärlich, dünn, .schleierartig, weissgelb bis bräunlichgelb; Verflüssigung spärlich, nach 60 Tagen 2 min. 7. Stärkeplatte: Wachstum dünn, schleierartig weissgelb, Luftmycel fehlt, Hydrolyse spärlich, auch nach 10 Tagen nur Spur.
B. Kartoffeln: Wachstum pustelig bis dünn, schleierartig, bräunlichgelb; Luftmycel sammetig, anfangs schneeweiss bis weissgrau, später Masskarmin und zimtbraun.
9. Karotten: Wachstum spärlich, pustelig; Luftmycel spärlich, weissgrau.
10. Lackmusmilch: Ringwachstum und pusteliges Oberflächen- wachstum, weissgelb, Lackmus rötlich, sehr starke Peptonisierung und starke Koagulation.
In seinen wesentlichen Merkmalen, wie Luft mycelfarbe, Morphologie der einzelnen Sporen sowie der Sporenketten und melanoide Verfärbung von pep- tonhaltigen Nährböden, stimmt der Stamm A 20388 mit Streptomyces violaceo-niger (Waksman et Curtis)
überein und wird deshalb vorläufig dieser Art zuge zählt. Es ist bekannt, dass Idas Antibiotikum Nigericin von einem Streptomyces-Stamm produziert wird, der ebenfalls der Gattung S. violaceo-niger angehört [vgl.
Harned et a1., Äntibiotics and Chemotherapy 1, 594 (1951)]. Das Nigericin ist eine organische Säure, die leicht mit Alkali titriert werden kaum (Äquiva- 1,nt-Gewicht 728).
Wie weiter unten gezeigt wird, handelt es sich beim neuen Antibiotikum<B>A20388</B> und bei dessen Komponenten N 1 und N 2 um neu trale oder phenolische Substanzen, so dass eine Iden tität mit Nibericin ausgeschlossen werden kann.
Zur Herstellung des Antibiotikums A 20388 kön nen auch Varianten, wie sie z. B. durch Selektionie- rung oder Mutation, insbesondere unter der Einwir kung von Ultraviolett- oder Röntgenstrahlen oder von Stickstoff-Senfölen gewonnen werden, Verwen dung finden.
Der Streptomyceten-Stamm A 20388 kann z. B. in wässriger, anorganische Salze, eine Kohlenstoff- und Stickstoffquelle. und gegebenenfalls wachstums fördernde Stoffe enthaltender Nährlösung aerob,
also beispielsweise in ruhender Oberflächenkultur oder vorzugsweise submers unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den bekannten Fe,rmentern gezüchtet werden. Als Temperatur eignet sich eine solche zwischen 18 und 40 . Eine wesentliche antibakterielle Wirkung zeigt die Nährlösung dabei im allgemeinen nach 11/2 bis 5 Tagen.
Die Nährlösung enthält als anorganische Salze beispielsweise Chloride, Nitrate, Carbonate, Sulfate von Alkalien, Erdalkalien, Magnesium, Eisen, Zink, Mangan. Als stickstoffhaltige Verbindungen und gegebenenfalls zuzusetzende Kohlehydrate und wachstumsfördernde Stoffe seien z.
B. genannt: Aminosäuren und ihre Gemische, Peptid@e und Pro teine sowie ihre Hydrolysate, wie Pepton oder Tryp- ton, Fleischextrakte, wasserlösliche Anteile von Ge treidekörnern, wie Mais und Weizen, von Destilla-. tionsrückständen bei der Alkoholherstellung, von Hefe, Bohnen, insbesondere der Sojapflanze, von Samen, beispielsweise der Baumwollpflanze, ferner Glukose,
Saccharose, Lakto.s:e, Stärke, Mannit, Glyce rin usw.
Bei dieser Fermentation wird neben dem Anti- biotikum A 20388, das speziell antibakteriell wirk sam ist, auch eine pilzaktive Substanz gebildet. Zur Isolierung des Antibiotikums dienen z. B. folgende Verfahren: Man trennt das Mycel vom Kulturfiltrat ab, wonach die Hauptmenge .des Antibiotikums A 20388 im Kulturfiltrat gefunden wird. Die pilz aktive Substanz bleibt am Mycel adsorbie:rt und kann daraus extrahiert werden.
Dazu eignen sich besonders organische, mindestens teilweise wasserlösliche Lö sungsmittel, wie Alkohole, z. B. Methanol, Äthanol und Butanole, oder Ketone, z. B. Aceton und Methyl- äthylke:ton. Aus :diesen Mycelextrakten kann man beispielsweise durch Behandlung mit Essigester die pilzaktive Substanz in fester Form isolieren.
Im Ultra- violett-Absorptionsspektrum sind Maxima bei fol genden Wellenlängen sichtbar: 360 m (log E l@"' = 2,71), <I>342</I> mp. (log E l% il - 2,73), 325 my (log E 1 il = 2,57) und 309 mu. <B>(log</B> Ei eil = 2,32). Dies deuttet daraufhin, dass es sich bei dieser Substanz um ein Pentaen handelt.
Zur Isolierung des Antibiotikums A 20388 extra hiert man dass Kulturfiltrat mit einem mit Wasser nicht mischbaren, organischen Lösungsmittel, wie Ester n=iederer Fettsäuren, beispielsweise Äthylacetat oder Amyl'@acetat, Kohlenwasserstoffei, z. B. Benzol, chlorievten Kohlenwasserstoffen, z. B. Äthylenchlorid, Methyl@enchlorid oder Chloroform, Ketonen, z. B.
Methylpropylketon, Methylamylketo.n oder Diiso- butylketon, Alkoholen, wie Butylalkoholen oder Amylalkoholen, Ä.them, z.
B. Äthyläther, Diisopro- pyläth.ex, Di:butyläthern oder Glykoläthern und der gleichen. Anstelle einer Lösungsmnittel-Extraktion der Kulturen oder in Kombination mit einer solchen als weitere Reinigungsoperation kann man das Anti biotikum auch durch Adsorption gewinnen, beispiels weise an Aktivkohle oder an aktivierten Erden, wie Fullererd@e oder Floridin,
und anschliessende Extrak tion des Adsoirbate.s z. B. mit einem in Wasser wenig- stens teilweise löslichen organischen Lösungsmittel, wie Aceton, Butanol oder Me,thyläthylketon.
Eine weitere Anreicherung des Antibiotikums A 20388 lässt sich dadurch erzielen, dass man die antibiotikumhaltigen organischen Extrakte zuerst mit einer sauren wässrigen Lösung mit einem pH unter 5 und dann :mit einer alkalischen wässrigen Lösung mit einem pH über 8 wiederholt auszieht, wobei die Hauptmenge der antibakteriellen Aktivität in der organischen Phase bleibt, aus der das Antibiotikum A 20388 isoliert wird.
Es wurde gefunden, dass das Antibiotikum A 20388 aus zwei Komponenten besteht, die mit N 1 oder Lankavamycin und N 2 oder Lankavacidin be zeichnet werden. Diese lassen sich leicht mittels Gegenstromverteilung in einheitlicher Form isolieren. Für diese Auftrennung eignen sich speziell zwei- phasige Lösungsmittelsysteme, die aus einem Ge misch von chlorierten Kohlenwasserstoffei, Alko holen und Wasser bestehen.
Beispielsweise hat sich für diesen Zweck ein Gemisch von Tetrachlorkohlen- stoff, Chloroform, Methanol und Wasser gut bewährt.
Ein gutes Reinigungsverfahren sowohl für das neue Antibiotikum als auch für seine Komponenten stellt wiederum die Verteilung zwischen einer alko holischen wässrigen Lösung und einem mit Wasser nicht mischbaren Lösungsmittel dar. Zweckmässig erfolgt die Verteilung nach dem Gegenstroanverfahren in entsprechenden Apparaten. Auch Chromatogra- phie, beispielsweise am Aluminiumoxyd, ist zur Reinigung, sehr geeignet.
Diese Reinigungsverfahren können einzeln oder in Kombination miteinander angewandt werden.
Die Gewinnung der reinen Komponenten Lanka vamycin und Lankavacidin in kristalliner Form nimmt man z: B. aus organischen Lösungsmitteln, wie aus Aceton, Methanol, Äthanol, Essigester, Chloroform, Aceton-Methanol-Gemischen, Aceton- Äther-Gemischen, Aceton-Petroläther-Gemischen oder Essigester-Petroläther-Gemischen vor. Zum Umkristallisieren dienen z.
B. dieselben Lösungsmittel oder auch wässrig-organische Lösungen, wie ver dünnte Alkohole, verdünntes Aceton usw.
Das Antibiotikum A 20388 erhält man als gelb liches amorphes Pulver. Die Komponente Lanka- vamycin bildet farblose Kristalle, die .einen doppelten Schmelzpunkt bei 147-150 und bei 181-182 zeigen, [a] D = - 94 (in a@bs. Äthanol).
Die Elementar analyse liefert folgende Werte: C = 60,18 %, H = 8,75 %, O = 31,06 %, CH3C0 = 11,49 %, OCH3 = 8,67 0/0, (C)
CH3 = 20,54 %.
Das Ultraviolett-Absorptio@nsspektrum zeigt ein schwaches Maximum bei 289 mit (log E = 1,50). Im Infrarot-Spektrum in Kaliumbromid (vgl. Fig.l) sind Banden u. a. bei folgenden Wellenlängen sicht bar:

löslich in Alkohol, Aceton, Chloroform, Essigester, Äther und Benzol und nur schwer löslich in Wasser und Petroläther. In Aceton gelöst, ist sie bei Zimmertemperatur be ständig gegen Kaliumpermanganat. Bei der Hydro lyse mit 0,5n Schwefelsäure bei 100 wird ein redu zierender Zucker abgespalten und bei der Behan d lung mit Alkali wird Essigsäure freigesetzt.
Die Komponente Lankavacidin bildet Massgelbe Kristalle, die unscharf bei 165-168 schmelzen und mit Ferrichloridlösung eine grüne Farbreaktion geben. Sie sind u. a. löslich in. Alkohol, Aceton, Chloroform und Essigester und nur mässig oder schwer löslich in Petroläther und Wasser.
Die Elernenbaranalyse liefert die folgenden Werte: C = 62,71 /o, H = 7,11%, N = 2,870/0, (C)CH3 = 12,60 0/<B>0</B>.
[a]D = - 161 (in aibs. Äthanol).
Das Ultraviolettabsorptionsspektrum zeigt ein Maximum bei 227 m/s (log E i%m = 2,92) und eine m Inflexion bei 285 mu (log EM= 2,95).
Im Infrarotspektrum in Kaliumbromid @(vgl. Fig. 2) sind Banden u. a. bei folgenden Wellenlängen sicht bar:

Brei der Hydrolyse mit 6n Salzsäure bei 110 werden keine Spaltstücke gebildet, die mit N'n ydrin eine Farbreaktion geben.
Das Antibiotikum A20388 und seine Kompo nenten Lankavamycin und Lankavacidin besitzen eine sehr hohe antibiotische Wirksamkeit :
gegenüber verschiedenen Testorganismen. Verwendet man ass Testmethode in vitro Verdünnungsreihen (Zehner potenzen) in Glukosebouillon, die während 24 Stun- den bei 37 bebrütet werden, so ergeben sich folgende noch hemmende Konzentrationen:

Escherichla coli, Sahnonella typhosa, Salmonella schottmuelleri, Shigella -sonnei, Pseudomonas aerugi- nosa, Klebsiella pneumoniae (Typ A), Pasteurella pestis, Vibrio comma (El Tor), Streptococcus faecalis,
ferner Pilze, wie Candida albicans und Tricho- phyton interdigitale, werden durch Konzentrationen von 100 /tg/cm3 der beiden, Verbindungen nicht ge hemmt.
Wie aus der Tabelle hervorgeht, sind Lanka- vamycin und Lankavacidin hauptsächlich gegenüber grampositiven Mikroorganismen in vitro wirksam. Lankavacidin zeigt in vivo eine ausgeprägte chemo therapeutische Aktivität.
Mit Streptococcus pyogenes bzw. Staphylococcus aureus infizierte Mäuse über- leben den 10. Versuchstag zu 100 %, wenn sie innert 30 Stunden nach der Infektion 5 X 33 mg/kg Lanka- vacidin subcu:
tan erhalten. Die unter gleichen Bedin- gungen wirksame Dosis per os beiträgt 5 X 100 mg/kg. Lankavamycin ist in 10facher Dosis nur sehr schwach wirksam bzw. unwirksam.
Gleichzeitige Anwendung von Lankavacidin und Lankavarnycin ergibt in. vitro wie in vivo bessere antibakterielle Effekte, als der Wirkung der Komponenten allein zukommt. Der synergistische Effekt zeigt sich z.
B. auf Staphylo- coccen beimpften Agarplätten, auf die mit Lösungen vorn Lankavacidin und Lankavamycin getränkte Filterpapierstreifen gelegt wurden.
Fig. 3 zeigt einen Kreuzstreifente-st gegen Staphylococcus aureus mit Lankavamycin (L') und Lankavacidin (L") in ver schiedenen Konzentrationen (mg/cm3), die am rechten bzw.
unteren Streifenende angegeben sind. Die Konzentrationen von 0,1 mg/cm3 Lankavaanycin allein bzw. 0,01 mg/cm3 Lankavacidin allein sind un wirksam..
In vivo ist die Dosis, die 100 fl/o der mit Staphylo- coccen infizierten Mäuse schützt, 10 mg/kg Lanka- vacidin und 100 mg/kg Lankavamycin, das heisst eine Dosis, bei der die einzelnen Komponenten allein nicht wirksam. sind.
Das Antibiotikum A 20388, seine Komponenten Lankavamycin und Lankavacidin oder entsprechende Gemische können als Heilmittel, z. B. in Form phar mazeutischer Präparate, Verwendung finden.
Diese enthalten die genannten Verbindungen in Mischung mit ,einem für die enterale, parenterale oder lokale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.
B. Gelatine, Milchzucker, Stärke, Magnesiumstea at, Talk, pflanz- liche öle, Benzylalkohole, Gummi, Polyalkylen- glykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.
B. als Tabletten, Dragees, Pulver, Salben, Cremen, Suppositorien oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabili- sierungs-, Netz- oder Emulgiermüttel. Sie können auch noch andere therapeutisch werftvolle Stoffe enthalten.
In den nachfolgenden Beispielen sind die Tem peraturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Man bereitet eine Nährlösung der Zusammen- setzung: 20 g Distillers solubles, 20 g Laktose, 1 g Natriumnitrat, 5 .g Natriumchlorid und 1 Liter Leitungswasser und stellt sie auf pH 7,5 ein. Diese, bzw.
ein Vielfaches derselben, wird .in, 500-cm3-Erlen- meyern (mit je 100 cm3 Nährlösung) oder in 500- Liter-Fermentern (mit je 300 Liter Nährlösung) ab gefüllt und 20-30 Minuten bei 1 atü sterilisiert. Dann impft man mit bis zu 10%,
einer teilweise spo#rulie- renden, vegetativen Kultur des Streptomyces-Stammes A 20388 an und inkubiert unter gutem Schütteln bzw. Rühren und in den Fernentern unter Belüftung (mit etwa 1 Vol. steriler Luft pro Vol. Nährlösung pro Minute) bei 27 .
Nach 70-120 Stunden Wachstum filtriert man die Kulturen unter Zusatz eines Filter- hilfsmittels je nach Volumen durch eine Nutsche oder durch .eine Filterpresse oder einen rotierenden Filter und befreit so die antibiotisch wirksame wäss- rige Lösung vom Mycel und anderen: festen Bestand teilen.
<I>Beispiel 2</I> Verwendest man .anstelle des in. Beispiel 1 ange gebenen Mediums beispielsweise die im folgenden beschriebenem. Nährlösungen<I>a)</I> bis f), so erhält man nach analoger Sterilisation, Beimpfung mit dem Streptomyces-Stamm <B>A20388,</B> Inkubation bei 27 und Filtration wässrige antibiotisch wirksame Lösun gen.
a) 20 g Mannit, 20 g Distillers solubles, 1 g Natriunmitrat, 3 g Natriumchlorid und 1 Liter Lei tungswasser; pH vor der Sterilisation 7,3.
b) 20 g Malzextrakt, 20 g Distillers solubles, 1 g Natriumnitrat, 5 g Natriumchlorid und 1 Liter Leitungswasser; pH vor der Sterilisation 7,8.
c) 10 g Rohglukose, 10 g Distillers solubles, 1 g Natriumnitrat, 1 g Natriumchlorid, 10 g Calcium- carbonat und 1 Liter Leitungswasser; pH vor der Sterilisation 7,5.
d) 5 g Dis:tillers salubles, 40 g Sojamehl, 20 g Rohglukose, 20g Natriumchlorid, 10 g Calciumear- bonat und 1 Liter Leitungswasser; pH vor der Steri lisation 7,5.
e) 20 g Malzextrakt, 20 g Maisquellwasser, 5 g Natriumchlorid, 0,2 g sek. Kaliumphosphat, 20 g Calciumcarbonat und 1 Liter Leitungswasser; pH vor der Sterilisation 7,5.
f) 20 g Mannit, 20 g Sojamehl und 1 Liter Leitungswasser; pH vor der Sterilisation 7,3. <I>Beispiel 3</I> Das Kulturfiltrat eines gemäss Beispiel 1 oder 2 erhaltenen 150-Liter-Ansatzes wird mit 70 Liter Äthylacetat extrahiert, wobei die gesamte antibakte- rielle Aktivität in die organische Phase übergeht.
Der Extrakt wird mit Wasser gewaschen, im Vakuum auf 5 Liter eingedampft und dann mehrere Male mit 0,5n, Essigsäure und mit 2n Natronlauge ausgeschüt telt. Schliesslich trocknet man die Äthylacetatlösumg über Natriumsulfat und dampft sie im Vakuum ein, wobei ein öliger Rückstand erhalten wird. Durch Behandlung mit P.etroläther gewinnet man daraus das rohe Antibiotikum A 20388.
<I>Beispiel 4</I> 5,3 g des gemäss Beispiel 3 erhaltenen rohen Antibiotikums A 20388 löst man in 100 cm3 abso- lutem Chloroform und trägt die. Lösung auf eine Säule aus 150 g Aluminiumoxyd (Aktivität III nach Brockmann) auf. 400 cm3 absolutes Chloroform lösen nur inaktive Begleitstoffe heraus.
Anschliessend wird mit 450 cm3 Chloroform, das 3 % Methylalkohol enthält, eluiert, wobei die gesamte wirksame Substanz im Eluat erhalten wird. Reichlich braune bis schwarze RTI ID="0004.0233" WI="27" HE="4" LX="1173" LY="2195"> Verunreinigungen bleiben in der Säule haften.
Durch Eindampfen der aktiven Fraktionen gewinnt man 2,75 g des angereichterten Antibiotikums A 20388 in Form eines dickflüssigen blass4bräunlichen Öls.
<I>Beispiel 5</I> 2,75 g des gemäss Beispiel 4 angereicherten Anti biotikums A 20388 werden einer 100stufigen Gegen- stromverteilung unterworfen, wobei man folgendes Lösungsmittelgemisch verwendet: 3 Volumteile Tetra chlorkoh:
lenstoff, 2 Volumteile Chloroform, 4 Volum- teile Methanol und 1 Volumteil Wasser. Die einzel- nen Fraktionen werden auf ihre antibakterielle Akti- vität geprüft, wobei als Testorganismen Bacillus sub- tilis und Micrococcus pyogenes,
var. aureus verwen det werden. Mit dem ersteren ergibt sich ein Aktivi- tätsmaximu.m bei der Stufe 27 (Komponente N 1), während mit dem zweiten Teistorganismus ein. solches bei Stufe 80 (Komponente N 2) beobachtet wird. Darauf vereinigt man einerseits die Stufen 20-37 und anderseits die .Stufen 70-91 und arbeitet sie ge trennt auf folgende Art auf: Nach dem Abtrennen der unteren Phase wird die obere Phase mit Wasser verdünnt und dreimal mit frischem Chloroform aus geschüttelt.
Man vereinigt diese Extrakte mit der abgetrennten unteren Phase, trocknest .diese Lösung über Natriumsulfat und dampft sie im Vakuum ein. Man erhält so die Komponente N 1 und die Kompo nente N 2 des Antibiotikums A 20388 in einheit l-icher Form.
<I>Beispiel 6</I> 9,63 g der gemäss Beispiel 5 erhaltenen Kompo nente N 1 werden an einer Säule aus 200 g Alumi niumoxyd (Aktivität 111 nach Brockmann) chromato- graphi:ert. Die mit 800 cm3 Benzol eluierten Anteile sind unwirksam und werden verworfen. 400 cm3 Chloroform, das man.
durch Destillation über Phos- phorsäureanhy drill völlig von Alkohol befreit hat, lösen 2,0 g einer blass-bräunlichen, honigartigen Sub stanz mit mittelmässiger Aktivität heraus. Durch wei tere Elution mit 300 cm3 Chloroform-Methylalkohol 99: 1 gewinnt man die Hauptmenge des Antibioti- kums als weisses amorphes Pulver (5,5 g).
Die weite ren Eluate enthalten nur noch geringe Mengen .an Aktivität.
4,0 g der Hauptfraktion löst man in wenig Aceton und verdünnt diese Lösung mit der fünffachen Menge Äther. Nun wird die Lösung konzentriert und das verdampfte Lösungsmittel sukzessive durch Petrol äther ersetzt. Beim Stehen in der Kälte kristallisiert die Komponente Lankavamycin in unregelmässigen derben Kristallen aus.
Die farblosen Kristalle sind bei 1301 im Hochvakuum nicht su blimierbar. Einer Lösung in Äthylacetat kann Lankavamycin weder mit verdünnter Essigsäure noch mit verdünntem Na triumcarbonat entzogen werden.
Ein viermal um kristallisiertes und 20 Stunden im Hochvakuum bei 80 getrocknetes Präparat zeigt folgende Eigenschaf ten: F. 147-150 , beim weiteren Erhitzen wachsen aus der Schmelze lange Nadeln, die. endgültig bei 181-182y schmelzen;

Eine 0,25 o/aaige Lösung des Antibiotikums in optisch reinem Feinsprit zeigt ein schwaches Ab sorptionsmaximum bei 289 m,ct (log E = 1,50). Im Infrarot Spektrum, aufgenommen in Kaliumbromid, vgl. Fig. 1, sind Banden u. a. bei folgenden Wellen längen sichtbar:

Bei der Hydrolyse des Lankavamycins mit 0,5n .Schwefelsäure (1 Stunde, 100 ) wird ein redur zierender Zucker abgespalten, dar bei der Papier- chroanatographie im System n Butanol-Eisessig- Wasser (4.- 1 : 1) einen Rf-Wert von 0,68 zeigt.
Wird Lankavamycin mit Alkali behandelt, so entsteht Essigsäure, wie durch Papierchromatogra- phie .sowie durch Mischschmelzpunkt und Infrarot- Absorptionsspektrum des kristallirren p-Phenyl- phen@acylesters (F. 112,5-114 ) .gezeigt werden kann.
Eine Lösung von Lankavamycin in, oxydations- beständigem Aceton wird durch eine 5 o/oige wässrige Kaliumpermanganatlö.sung bei Zimmertemperatur während mehreren Stunden nicht angegriffen. Bei: 95 .erfolgt dagegen rasche Abscheidung von Braun stein.
<I>Beispiel 7</I> 600 mg der gemäss Beispiel 5 erhaltenen Kom ponente Lankavacidin werden zur weiteren Reini gung an einer Säule aus 15g Aluminiumoxyd chro- matographiert, wobei mit Chloroform und mit Chloroform-Methanol-Gemischen eluiert wird.
Die gesamte Aktivität ist in den Fraktionen, die mit Chloroform Methanol<B>99:</B> 1 und 49: 1 eluiert wer den, enthalten.
Die aktiven Fraktionen .geben beim Eindampfen im Vakuum 280 mg eines ;gelblichen Öls, das beim Stehen teilweise kristallin, erstarrt. Durch Kristallisation aus wenig Äthylacetat kann man das Lankavacidin als blass cremefarbiges Kristall- pulver gewinnen. Ausbeute etwa 90 mg.
Die Mutter laugen sind nahezu so aktiv wie die Kristalle und erstarren. beim Stehen teilweise. Lankavacidinschmilzt nach Umkristallisieren ,aus Äthylacetat Äther bei 165-168 . Mit 5 %iger alkoholischer Ferrichlorid- lösung .erhält man eine grüne Farbreaktion.
Die Elementaranalyse liefert die folgenden Werte: C = 62,71'%, H = 7,11%, N = 2,87'%, (C)CH3 = 12,60 %.

5 mg Lankavacidin werden in 1 ml 6n Salzsäure über Nacht bei 110 hydrolysiert. Die paperchroma- tographische Untersuchung des Hydrolysates ergibt, ,dass keine Niuhydrin-positiven Substanzen darin. ent halten sind.

As will be shown below, the new antibiotic <B> A20388 </B> and its components N 1 and N 2 are neutral or phenolic substances, so that identity with nibericin can be ruled out.
For the production of the antibiotic A 20388, variants such as those described in e.g. B. can be obtained by selection or mutation, in particular under the action of ultraviolet or X-rays or nitrogen mustard oils, are used.

A good cleaning process for the new antibiotic as well as for its components is again the distribution between an alcoholic aqueous solution and a water-immiscible solvent. The distribution is expediently carried out according to the countercurrent process in appropriate apparatus. Chromatography, for example on aluminum oxide, is also very suitable for cleaning.
These cleaning methods can be used individually or in combination with one another.

A 0.25 o / aaige solution of the antibiotic in optically pure fine spirit shows a weak absorption maximum at 289 m, ct (log E = 1.50). In the infrared spectrum, recorded in potassium bromide, cf. Fig. 1, bands are u. a. visible at the following wavelengths:

During the hydrolysis of lankavamycin with 0.5N sulfuric acid (1 hour, 100) a reducing sugar is split off, which in paper chromatography in the system n butanol-glacial acetic acid-water (4-1: 1) has an Rf value of 0.68 shows.
If lankavamycin is treated with alkali, acetic acid is formed, as can be shown by paper chromatography and by the mixed melting point and infrared absorption spectrum of the crystalline p-phenylphenyl acyl ester (m.p. 112.5-114).
A solution of lankavamycin in oxidation-resistant acetone is not attacked by a 5% aqueous potassium permanganate solution at room temperature for several hours. At: 95., On the other hand, there is rapid deposition of brown stone.
<I> Example 7 </I> 600 mg of the component lankavacidin obtained according to Example 5 are chromatographed for further purification on a column of 15 g of aluminum oxide, eluting with chloroform and with chloroform-methanol mixtures.
All of the activity is contained in the fractions which are eluted with chloroform, methanol 99: 1 and 49: 1.
The active fractions give on evaporation in vacuo 280 mg of a yellowish oil, which solidifies in part crystalline on standing. Lankavacidin can be obtained as a pale cream-colored crystal powder by crystallization from a little ethyl acetate. Yield about 90 mg.
The mother liquors are almost as active as the crystals and solidify. when standing partially. Lankavacidin melts after recrystallization, from ethyl acetate, ether at 165-168. A green color reaction is obtained with a 5% alcoholic ferric chloride solution.
The elemental analysis gives the following values: C = 62.71%, H = 7.11%, N = 2.87%, (C) CH3 = 12.60%.

The ultraviolet absorption spectrum in fine spirits shows a maximum at 227 my (log E% = 2.95) im and an inflection at 285 mu (log E% n - 1.54).

5 mg of lankavacidin are hydrolyzed in 1 ml of 6N hydrochloric acid at 110 overnight. The paper chromatographic examination of the hydrolyzate shows that there are no Niuhydrin-positive substances in it. are included.
