Abstract
<PICT:0949473/C2/1> <PICT:0949473/C2/2> The antibiotics lankavamycin or lankavacidin or a mixture thereof are produced by cultivating Streptomyces violaceo-niger NRRL 2834 or a lankavamycin and/or lankavicidin-producing mutant thereof under aerobic conditions in an aqueous nutrient solution containing inorganic salts and a source of carbon and of nitrogen until the nutrient solution shows a substantial antibiotic activity, and the antibiotic lankavamycin and/or the antibiotic lankavacidin is/are then isolated. The antibiotic(s) may be extracted from the culture filtrate with an organic solvent immiscible with water and purified by adsorption and extraction from the adsorbate, by distribution between an alcoholic aqueous solution and an organic solvent immiscible with water or by chromotography. Lankavamycin forms colourless crystals which show a double melting point at 147-150 DEG C. and at 181-182 DEG C., [a ]20D=-93.8 DEG (C=1.23 in ethanol), elementary analysis C=60.18%, H=8.75%, O=31.06%, CH3CO=11.49%, OCH3=8.67%, (C)CH3=20.54%. The ultra-violet absorption spectrum shows a maximum at 289 mm (log e=1.50) and the infrared spectrum in potassium bromide is shown in Fig. 1. Lancavacidin forms pale yellow crystals which melt at 165-168 DEG C. and give a green colour reaction with ferric chloride solution. Elementary analysis gives C=62.71%, H=7.11%, N=2.87%, (C)CH3=12.60%. [a ]20D=-161 DEG (C=0.967 in absolute ethanol). The ultraviolet absorption spectrum shows a maximum at 227 mm (log e 1%=2.95) and the infrared spectrum 1 cm. in potassium bromide is shown in Fig. 2. Pharmaceutical preparations comprise lankavamycin and/or lankavacidin in admixture or conjunction with a pharmaceutical carrier in forms such as tablets, dragees, ampoules, salves, creams, powders, suppositories, solutions, suspensions or emulsions.
