Verfahren zur Herstellung neuer 2-substituierter Pyrimidine
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Pyrimidinen der Formel

worin R1 eine mindestens monosubstituierte Aminogruppe, R2 eine freie odersubstituierteAminogruppe, Rs eine freie oder funktionell abgewandelte Carboxylgruppe und R4 Wasserstoff oder einen niederen Koh- lenwasserstoffrest bedeuten.
Als mindestens mono-substituierte Aminogruppen R1 sowie als substituierte Aminogruppe R2 kommen besonders durch niedere aliphatische, cycloaliphatische, aromatische, heterocycl, ische, araiiphatische oder heterocyclyl-aliphatische Reste mono-oder di- substituierte Aminogruppen in Betracht.
Beispiele hierfür sind Mono-oder Dialkylaminogruppen, wie Mono-oder Di-methyl-, -Ïthyl-, -propyl- oder -butylaminogruppen ; Alkylenaminogruppen, in denen der Alkylenrest auch durch Heteroatome, wie Sauerstoff, Schwefel-oder Stickstoff, unterbrochen sein kann, wie Pyrrolidino-, Piperidino-, Morpholino- oder Piperazinogruppen ;
Amino-alkylaminogruppen, in denen die endständige Aminogruppe vorzugsweise eine der oben angegebenen Aminogruppen darstellt, und worin der Alkylenrest mit Vorteil 2-6 Kohlen stoffatome aufweist, wie die Dimethylamino-oder Di äthylamino-äthyl-oder-propylgruppe, oder eine Pyrrolidino-, Piperidino-, Morpholinot odler Pipera zino-athyl-oder-propylgruppe ; Oxyalkylaminogrup- pen, wie die Mono-oder DioxyÏthylaminogruppe, Cycloalkylaminogruppen, Benzyl- oder Phenylaminogruppen, oder eine Furfurylaminogruppe.
Die Aminogruppe R2 kann au¯erdem auch eine e anorganisch substituierte Aminogruppe, wie z. B. eine Hydrazinogruppe, sein.
Als funktionell abgewandelte Carboxylgruppen R3 kommen besonders Carbonsäureestergruppen, wie Carbalkoxygruppen, z. B. CarbÏthoxygruppen, Amid-, Thioamid-oder Nitrilgruppen in Frage.
Die neuen Verbindungen besitzen wertvolle Eigenschaften. Sie sind Antimetaboliten des Purins. Ferner besitzen sie tumorhemmende, diuretische, coronar erweibernde, analgetische, antirheumatiscbe, antibakterielle und fungistatische Eigenschaften. Ferner sind sie gegen Viren wirksam. Sie können dementspre- chend als Heilmittel Verwendung finden.
Auch als Zwischenprodukte sind die neuen Verbindungen von Wert.
Besonders wertvoll. sind 1, 3, 6-unsubstituierte 5 Cyano-pyrimidine, die in 2- und 4-Stellung eine Amino gruppe gemäss den obigen Ausfüllrungen besitzen, und besonders das 2, 4-Bis-dimethylamino-5-cyan-pyrimidin der Formel

Diese Verbindungen bes. itzen eine bemerkens- werte Wirkung bei Virusinfektionen, wie z. B. Ektromeliavirus.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen besteht darin, d : ass man in einer Verbindung der Formel

worin X einen gegen die Aminogruppe R2 austausch- baren Rest, wie z. B. ein Halbgenatom ader eine freie oder verätherte Mercaptogruppe, insbesondere ein Chlor-oder Bromatom, oder eine Alkyl-oder Aralkylmercaptogruppe bedeutet, den Rest X gegen den Rest R2 austauscht. Dieser Austausch kann in be kannter Weise, beispielsweise durch Umsetzung mit einer Verbindung der Formel R2H erfolgen, vorzugsweise in Anwesenheit von basischen Kondensationsmitteln.
In den nach dem erfindungsgemässen ! Verfahren hergestellten Verbindungen k¯nnen die Substituenten in 2-, 4-undloder 5-Stellung in üblicher Weise abgewandelt werden. So lässt sich z. B. eine freie Carb oxylgruppe in 5-Stellung funktionelI abwandeln, z. B. amidieren oder verestern. Anderseits lassen sich funk tionelle Carbonsäurederivate in andere Derivate überf hren, z. B. Nitrile in Amide oder Thioamide oder Ester in Amide, oder funktionelle Carbonsäurederivate in die freien Carbonsäuren umwandeln.
Die genannten Reaktionen können in An-oder Abwesenheit von Verdünnungsmitteln undloder Kon- densationsmitteln, wie z. B. basischen Kondensations- mitteln, gegebenenfalls bei erhöhter Temperatur, im offenen oder im geschlossenen Gefäss unter Druck vorgenommen werden.
Je nach der Ausführungsform des Verfahrens werden die neuen Verbindungen in freier Form oder in Form ihrer Salze erhalten. Besitzen die neuen Verbindungen Säurecharakter, so können daraus Metallsalze oder Salze mit Basen gebildet werden. Insbesondere sind hier die Alkali-und Erdalkalisalze zu er wähnen. Aus den neuen Verbindungen können femer Additionssalze mit anorganischen oder organischen Säuren hergestellt werden.
Als salzbildende Säuren kommen hierbei beispielsweise in Frage : HalogenwasserstoffsÏuren, Schwefelsäuren, PhosphorsÏuren, Salpetersäure, PerchlorsÏure ; aliphatische, alicyclische, aromatische oder heterocyclische Carbon-oder Sul fonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milchs, Apfel-, Wein-, Zitronen-, Ascorbin-, Oxymalein-, Dioxymalein-oder Brenz traubensäure ; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Oxybenzoe-, Salicyl-oder p Aminosalicylsäure ;
Methansulfon-, ¯thansulfon-, Oxyäthansulfon-, ¯thylensulfonsÏure ; Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure ; Methionin, Triptophan, Lysin oder Arginin.
Die neuen Verbindungen, ihre Salze oder ent sprechende Gemische davon können wie schon gesagt als Arzneimittel Verwendung finden. Diese enthalten die genannten Verbindungen in Mischung mit einem f r die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen arganischen oder anorganischen Träge. rmaterial.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
18, 2 g 2-Dimethylamino-4-chlor-5-cyan-pyrimidin werden zusammen mit einer 8n benzolischen Dimethylaminlösung wahrend 2 Stunden auf 70-800 erhitzt. Man lässt erkalten, nutscht von dem ausgefallenen Dimethylamin-hydrochlorid ab, dampft das Filtrat zur Trockne ein und kristallisiert den Rückstand aus Petroläther. 2, 4-Bis-dimethylamino-5-cyan-pyri- midin der Formel

Beispiel 2
Eine Lösung von 18, 2 g 2-Dimethylamino-4 chlor-5-cyan-pyrimidin in 500 cm3 Toluol wird mit 20 g Furfurylamin versetzt und während 10 Stunden am Rückfluss erhitzt. Man nutscht heiss von wenig Ungelöstem ab und dampft im Vakuum auf 200 cm3 ein. Nach dem Erkalten wird abgenutscht. Man erhÏlt so 2-Dimethylamino-4-furfurylamino-5-cyan- pyrimidin der Formel

Eine Losung von 18, 2 g 2-Dimethylamino-4- chlor-5-cyan-pyrimidin und 20 g N-Meth-piperazin in 150 cm3 Aceton wird im geschlossenen Rohr wÏhrend 4 Stunden auf 120-13 () erhitzt. Man dampft dann im Vakuum zur Trockne ein und kristallisiert den Rückstand aus PetrolÏther. 2-Dimethylamino-4 (4'-methyl-piperazino)-5-cyan-pyrimidin der Formel

Beispiel 4
18, 2 g 2-Dimethylamino-4-chlor-5-cyan-pyrimi- dm werden zusammen mit 50 cm3 Diäthylamino- äthylamin wÏhrend 1 Stunde auf 150-160¯ erhitzt.
Man destilliert dann im Vakuum das überschüssige Diäthylamino-äthylamin ab, versetzt den Rückstand mit Wasser, stellt mit 2n Natronlauge stark alkalisch und nutscht vom ausgefallenen Niederschlag ab. Letzterer wird aus Petroläther umkristallisiert. Man erhält so 2-Dimethylamino-4- (¯-diÏthylamino-Ïthylamino) 5-cyan-pyrimidin der Formel

Eine Lösung von 18, 2 g 2-Dimethylamino-4chlor-5-cyan-pyrimidin und 30 g p-Chlor-anilin in 500 cm3 Toluol wird während 10 Stunden zum Sieden erhitzt.
