EMI2.1
Vertreter von Verbindungen der Formel I.
Man erhielt die folgenden Resultate: Verbindung B Verbindung C
EMI2.2
je nach Herkunft, (a) und (b) benannt.
A B (a) B (b) C Kontrastwirkung ++++ ++ + + + Ausscheidung 6 Stunden 8-11 Tage 4-6 Tage 12 Tage unvollständig
Use of iodized acid amides as X-ray contrast media
The present invention relates to the use of iodinated acid amides of the formula
EMI1.1
<tb> <SEP> 0
<tb> <SEP> ÜÜ <SEP> 1
<tb> N
<tb> <SEP> alkylene-CO-NH-alkylene-C <SEP> OOH
<tb> wherein <SEP> alkylene <SEP> e.g. <SEP> B. <SEP> -CH2-, <SEP> -CH.2-CH.ff, <SEP> -CH-,
<tb> <SEP> CH3
<tb> -CH.ffCH2-CH <SEP> and <SEP> -CH2-CH-CH2- <SEP> mean
<tb> <SEP> CH3
<tb> can, as an X-ray contrast medium, whereby the new iodinated acid amides can be used in free form or in the form of inorganic or organic salts.
Compounds of this type are particularly suitable for bronchography and salpingography. For these purposes, they can be in the form of finely divided aqueous suspensions with thickeners, e.g. B.
Carboxymethyl cellulose, carboxymethyl starch, polyethylene glycol, dextran, etc., can be applied.
The new compounds have significant advantages over the previously known unsubstituted amide of 3,5-diiodo-4-pyridonyl- (l) -acetic acid, such as: B. deeper shading, better suspension capacity, very low toxicity, but especially rapid excretion.
Compared to the X-ray contrast media already in use for bronchography [3,5-Diiodo-4-pyridonyl (1) acetic acid and its propyl ester], they have the following advantages:
1. Slightly irritating to the bronchial mucous membrane.
2. Good adhesion or wall affinity. In the case of pralifilling, a complete spout pattern or an excellent fitting pattern and after partial
Expiration or absorption to achieve a clear mucous skin relief display.
3. The viscosity, adjusted to the consistency of a toothpaste that is not too thin, allows good unimpeded instillation through the
Catheter probe, avoids alveolar filling, i.e. easy drainage into the periphery of the lungs, and enables targeted and gentle filling of individual segments with rapid elimination.
A very good contrast effect and very fast elimination must be mentioned as the most important advantage.
With regard to these two properties, the following three compounds, for example, were tested comparatively in rabbits: Compound A
EMI2.1
Representatives of compounds of the formula I.
The following results were obtained: Compound B Compound C
EMI2.2
named according to origin, (a) and (b).
A B (a) B (b) C Contrast effect ++++ ++ + + + excretion 6 hours 8-11 days 4-6 days 12 days incomplete
The advantage that can be achieved with compounds of the formula I as X-ray contrast media is therefore obvious.
example
38 g of chemically pure grape sugar are dissolved in 721 g of freshly distilled water and the resulting solution is filtered. 4 g of pure (medium-viscosity grade M) sodium carboxymethyl cellulose are then introduced into the filtrate with vigorous turbination. The sodium carboxymethyl cellulose is moistened with 11 g of twice distilled benzyl alcohol before being added. The clear solution obtained is filtered through a Jena G1 or Jena G2 glass filter (branded products), filled into closed containers and sterilized while hot. After cooling, 2 g of sorbitan polyoxyethylene lauric acid ester (Tweens 20, branded product) and 500 g of microcrystalline contrast substance of the formula are added to the solution
EMI2.3
given.
An ultra-high-speed stirrer is used to distribute the contrast substance evenly; but you can also let the whole thing flow through a homogenizer under sterile conditions.
A 50/0 (vol. E / o) contrast suspension is thus obtained, which is ready for use for performing bronchographies. The amount of thickener is reduced for performing salpingographies.
