Le procédé suivant l'invention est caractérisé en ce qu'on fait réagir sur le 6-méthoxy-indole un halogénure de magnésium primaire,
condense l'ha- logénure de 6-méthoxy-indolylmagnésium obtenu avec l'a-bromo-propionitrile et réduit le 2-[6'-mé- thoxy-indolyl-(3')]-propionitrile formé.
Un mode de mise en aeuvre préféré du procédé selon l'invention est le suivant: on fait réagir (K. Ei- ter et O. Svierak, Monatsh. 83, 1453-76, 1952) un halogénure d'éthylmagnésium, par exemple l'iodure, sur le 6-méthoxy-indole (formule I) au sein d'un sol vant, notamment d'un éther phénolique comme l'ani- sole,
condense à température modérée l'halogénure de 6-méthoxy-indolylmagnésium formé avec l'a-bromo- propionitrile, extrait le 2-[6'-méthoxy-indolyl-(3')]- propionitrile (formule II) résultant et purifie ce der nier par les procédés habituels. La réduction du pro duit de condensation est effectuée par hydrogéna tion en solution alcoolique ammoniacale en présence d'un catalyseur au nickel. On fait cristalliser sous forme de picrate peu soluble la 2-[6'-méthoxy-indo- lyl-(3')]-propylamine-(1) (formule III) ainsi obtenue.
On décompose le picrate par l'action d'un hydroxyde alcalin, extrait l'amine libre par le chlorure de mé thylène et l'isole par les procédés habituels. Les formules de quelques-uns des composés men tionnés ci-dessus sont développées dans le schéma de réaction qui suit

Le nouveau composé obtenu suivant l'invention trouve son application en tant qu'intermédiaire dans la synthèse de composés physiologiquement actifs de la famille de la réserpine.
<I>Exemple</I> A une solution glacée d'iodure d'éthylmagnésium dans l'anisole, préparée à partir de 20,5 g d'iodure d'éthyle, de 2,85 g de magnésium et de 32,5 emi3 d'anisole, on ajoute goutte à goutte une solution à environ 40,1 C de 9,5 g de 6-méthoxy-indole (for mule I) dans 20 cms d'anisole. On agite pendant une demi-heure à température ambiante,
refroidit la solu tion à 00 C et l'additionne de 12 g d'a-bromo-propio- nitrile et de 10 ce d'anisole. Le mélange est soumis à l'agitation pendant 3 heures, en refroidissant au début à 00 C. Après addition de 200 ce d'acide acétique dilué à 10 %, on extrait au chlorure de méthylène et lave l'extrait à l'eau, avec une solution de bicarbonate de sodium, puis à l'eau. Le chlorure de méthylène est évaporé sous vide et l'anisole dis tillé sous 0,2 mm de mercure.
Le résidu de distilla tion est un produit visqueux constitué en grande par tie par le 2-[6'-méthoxy-indolyl-(3')]-propionitrile et sera utilisé sous cette forme pour la suite de la syn thèse. Ce produit n'est pas décrit dans la littérature.
On dissout le résidu "précédent dans 150 cms d'éthanol, ajoute 80 ce, d'une solution alcoolique saturée d'ammoniac et 12 g de nickel de Raney. Après avoir chassé l'air par un courant d'azote puis d'hydrogène, la solution est soumise à la réduction pendant 7 heures, en procédant au bout de 3 heures à une nouvelle introduction de 12 g de nickel de Raney. On filtre pour éliminer le catalyseur et chasse le solvant sous vide.
Le résidu est repris par le chlorure de méthylène, on extrait par 200 ce d'acide acétique dilué à 10 %, puis par 50<I>ce</I> d'eau, lave l'extrait aqueux au chlorure de méthylène, alca linise la phase aqueuse par la soude concentrée et extrait la tryptamine substituée, au chlorure de mé thylène. La phase organique est ensuite lavée avec une solution de chlorure de sodium et évaporée à sec. Le résidu obtenu est repris dans l_'éthanol, addi tionné d'acide picrique en solution dans l'éthanol et refroidi. Il se forme un précipité qu'on sépare par filtration.
On recueille ainsi le picrate de 2-[6'-mé- thoxy-indolyl-(3')]-propylamine-(1), produit bien cris tallisé et stable, de couleur rouge orange. Ce com posé qui est nouveau, est insoluble dans l'éther, soluble dans l'alcool et l'acétone à chaud et dans le diméthylformamide à froid. F. = 242e C (déc.), (fusion instantanée, déterminée sur bloc de Ma- quenne).
Le rendement global à partir du 6-méthoxy-in- dole est de 20 %.

Les extraits organiques réunis sont lavés d'abord avec un mélange à<B><I>50%</I></B> d'une solution saturée de lithine et d'une solution saturée de chlorure de sodium, puis avec une solu tion saturée de chlorure de sodium. On filtre, éva pore sous vide et recueille 680 mg de 2-[6'-méthoxy- indolyl-(3')]-propylamine-(1) (formule III) se présen tant sous forme d'une huile visqueuse (rendement <B>96%).</B> Traité au nitroprussiate de sodium, ce pro duit donne la coloration violette caractéristique des amines primaires aliphatiques.
Il est insoluble dans l'eau, peu soluble dans l'éther, soluble dans l'alcool, le chloroforme et le chlorure de méthylène. Le spec tre I.R. est en accord avec la structure du produit. Ce composé n'est pas décrit dans la littérature.

The new compound obtained according to the invention finds its application as an intermediate in the synthesis of physiologically active compounds of the reserpine family.
<I> Example </I> Has an ice-cold solution of ethylmagnesium iodide in anisole, prepared from 20.5 g of ethyl iodide, 2.85 g of magnesium and 32.5 emi3 of anisole is added dropwise a solution at about 40.1 C of 9.5 g of 6-methoxy-indole (formula I) in 20 cms of anisole. Stirred for half an hour at room temperature,
the solution is cooled to 00 ° C. and the addition of 12 g of α-bromopropionitrile and 10 cc of anisole is added. The mixture is stirred for 3 hours, cooling at the start to 00 ° C. After addition of 200 cc of dilute 10% acetic acid, extraction is carried out with methylene chloride and the extract is washed with water, with sodium bicarbonate solution, then with water. The methylene chloride is evaporated off in vacuo and the anisole distilled under 0.2 mm of mercury.

The above residue is dissolved in 150 cms of ethanol, 80 cc of a saturated alcoholic solution of ammonia and 12 g of Raney nickel are added. After having expelled the air with a stream of nitrogen and then of hydrogen. , the solution is subjected to reduction for 7 hours, proceeding after 3 hours to a further introduction of 12 g of Raney nickel, filtering to remove the catalyst and removing the solvent in vacuo.
The residue is taken up in methylene chloride, extracted with 200 cc of acetic acid diluted to 10%, then with 50 <I> cc </I> of water, the aqueous extract washed with methylene chloride, alca linins the aqueous phase with concentrated sodium hydroxide and extracts the substituted tryptamine with methylene chloride. The organic phase is then washed with sodium chloride solution and evaporated to dryness. The residue obtained is taken up in ethanol, added with picric acid dissolved in ethanol and cooled. A precipitate forms which is separated by filtration.
The 2- [6'-methoxy-indolyl- (3 ')] - propylamine- (1) picrate is thus collected, a well crystallized and stable product, which is orange red in color. This compound, which is new, is insoluble in ether, soluble in alcohol and acetone when hot and in dimethylformamide when cold. F. = 242nd C (dec.), (Instantaneous fusion, determined on a Maquenne block).
The overall yield from 6-methoxyindole is 20%.
