worin R1 und R2 niedere Alkylenreste, die maximal je 3 Kohlenstoffatome in gerader Kette enthalten, und Am eine tertiäre Aminogruppe bedeuten, und R3 für Wasserstoff,
Chlor oder die Methylmercaptogruppe steht.
Die neuen tert.-Amino-alkylthioallcylphenothiazine sollen als Tranquilizer verwendet werden.
Wie schon erwähnt, bedeutet Am eine tertiäre Aminogruppe, also beispielsweise die Dimethylamino- gruppe, die Diäthylaminogruppe, die Pyrrolidino-, Piperidino-, Morpholino-oder Thiomorpholinogruppe.
R1 und R2 stellen vorzugsweise je einen Athylen- oder Isopropylenrest dar.
Das erfindungsgemässe Verfahren zur Herstellung der neuen tert.-Amino-alkylthioalkylphenothiazine ist dadurch gekennzeichnet, d'ass man ein entsprechendes Phenothiazin oder ein N-Metallsalz davon mit einem organischen Sulfonsäureester eines entsprechenden Halogenalkanols umsetzt und das erhaltene Halogen- alkylphenothiazin mit einem entsprechenden tert.-
Amino-alkanthiol zur Reaktion bringt.
Die neuen tert.-Amino-alkylthioal'kylphenothiazine der eingangs erwähnten Formel I können in: Form ihrer Salze mit anorganischen oder organischen Säu ren isoliert werden. Als anorganische Säuren können zur Salzbildung verwendet werden: Schwe- felsäure, Chlorwasserstoffsäure, Bromwasserstoffsäure und Phosphorsäure; als organische Säuren:
Essig säure, Glycolsäure, Citronens.äure, Bernsteinsäure, Fumarsäure, Maleinsäure, D,ioxymaleinsäure, Me- thansulfonsäure und Oxyäthansulfonsäure. <I>Beispiel 1</I> Zu 600 cm3 einer ätherischen Lösung von n- Butyl=lithium (enthaltend 0,7 Mol BuLi) werden 139 g gereinigtes Phenothiazin gegeben.
Eine Lösung von 155g p-Toluolsulfonsäure-(fl-chlor-äthyl)-ester in 100 em3 abs. Äther wird unter Rühren und Kühlen in die Phenothiazin-Li-Lösung eingetropft. Nach 2 Stunden wird mit Wasser versetzt, die Ätherschicht abgetrennt, der Äther getrocknet und verdampft. Der Rückstand, bestehende aus 10-(fl-Chlor-äthyl)-pheno- thiazin, wird aus Äthanol umkristallisiert. Man er hält zwischen 90 und 100 g des bei 97 C schmelzen den Produktes.
4 g Natrium werden in 100 cms abs. Athanol unter Stickstoffatmosphäre heiss gelöst und zu dieser Lösung eine Lösung von 20 g Dimethylaminoäthan- dol in 250 cm3 Äthanol gegeben. Unter Rühren werden nun 40 g 10@-(ss-Chlor-äthyl)-phenothiazin ein getragen.
Das Ganze wird 11 Stunden unter Stickstoff atmosphäre und unter Rühren zum Sieden erhitzt. Nach dem Abkühlen wird die Lösung im Vakuum eingeengt und der Rückstand mit Äther und 2n-Essig- säure gut geschüttelt. Die Schichten werden getrennt, wobei die Ätherlösung noch zweimal mit 2n-Essig- säure ausgezogen wird.
Die vereinigten sauren wäss- rigen Auszüge werden alkalisch gemacht und das sich abscheidende öl .in Äther aufgenommen. Nach dem Trocknen der ätherischen Lösung wird verdampft und der Rückstand im Hochvakuum destilliert. Man erhält 41 g 10-(P-Dimethylamino-äthylthioäthyl)-pheno- thiazin, das unter 0,
08 mm bei 200-205 C übergeht. Das neue Phenothiazin löst sich leicht in verdünnten Säuren.
Setzt man anstelle von Dimethylaminoäthanthiol Pyrrolidinoäthanthiol mit 10-(ss-Chlor -äthyl)-phena- thiazin um, so erhält man das unter 0,05 mm bei 215 C übergehende 10-(ss-Pyrrolidino-äthylthioäthyl)- phenothiazin.
<I>Beispiel 2</I> In gleicher Weise wie im Beispiel 1 beschrieben, erhält man aus 40 g 10-(ss-Chlor-äthyl)-phenothiazin, 25 g Diäthylaminoäthanthiol und 4 g Natrium in total 350 cm3 Athanol 45 g 10-(ss-Diäthylamino- äthylthioäthyl)-phenothiazin, das unter 0,07 mm bei 198-203 C siedet.
In gleicher Weise erhält man aus 10-(ss-Chlor- äthyl)-phenothiazin und Piperidinoäthanol das 10-(ss- Piperidino-äthyl'thioäthyl)-phenothiazin, das unter 0,05 nun bei 205-207 C übergeht.
<I>Beispiel 3</I> 40 g 10-(B-Chlor-äthyl)-phenothiazin, das wie im Beispiel 1 beschrieben erhalten wurde, werden in üblicher Weise mit 30 g Diisopropylaminoäthanthiol in Gegenwart von 4 g Natrium in total 350 cm'. Äthanol 12 Stunden unter Rühren zum Sieden erhitzt.
Nach dem Aufarbeiten, wie im Beispiel 1 beschrieben, erhält man 35 g 10-(B-Diisopropylamino-äthylthio- äthyl)-phenothiazin, das unter 0,04 mm bei 208 bis 212 C übergeht. Das neue Phenothiazin bildet vor erst ein schwach gelb gefärbtes 01, das nach einiger Zeit kristallin erstarrt.
Die Salze mit Chlorwasser stoffsäure, Essigsäure und Methansulfonsäure sind nicht mehr als etwa 2-4% in Wasser löslich.
Setzt man 10-(ss-Chlor-äthy'I)-phenothiazin mit ss-(2,6-Dimethyl-piperidyl)-äthanthiol um, so erhält man das 10-[ss-(2',6'-D:imethyl-piperidyl-)-äthylthio- äthyl]-phenothiazin, das unter 0,03 mm bei 214 bis 216 C übergeht.
Anstelle von 10-(ss-Chlor-äthyl)-phenothiazin kann man auch 10-(r-Chlor-propyl)-phenothiazin und die genannten tert: Amino-alkanthiole mit gleichem Erfolg zur Reaktion bringen.
Setzt man anstelle von 10-(ss-Chlor-äthyl)- bzw. 10-(y-Chlor-propyl)-phenothiazin 10-(Chlor-R1)-3- chlor- oder - 3 - methylmercapto-phenothiazine mit tert: Amino-R2-thiolen um, so erhält man die ent sprechenden 10-(tert: Amino-alkylthioalkyl)-3-chlor- bzw. -3-methylmercapto-phenothiazine.

The process according to the invention for preparing the new tert-amino-alkylthioalkylphenothiazines is characterized in that a corresponding phenothiazine or an N-metal salt thereof is reacted with an organic sulfonic acid ester of a corresponding haloalkanol and the haloalkylphenothiazine obtained is reacted with a corresponding tert.-
Amino-alkanethiol brings to reaction.
The new tert-amino-alkylthioal'kylphenothiazines of the formula I mentioned at the outset can be isolated in the form of their salts with inorganic or organic acids. The following inorganic acids can be used for salt formation: sulfuric acid, hydrochloric acid, hydrobromic acid and phosphoric acid; as organic acids:
Acetic acid, glycolic acid, citric acid, succinic acid, fumaric acid, maleic acid, D, ioxymaleic acid, methanesulfonic acid and oxyethanesulfonic acid. <I> Example 1 </I> 139 g of purified phenothiazine are added to 600 cm3 of an ethereal solution of n-butyl lithium (containing 0.7 mol of BuLi).
A solution of 155 g of p-toluenesulfonic acid (fl-chloro-ethyl) ester in 100 em3 abs. Ether is added dropwise to the phenothiazine-Li solution while stirring and cooling. After 2 hours, water is added, the ether layer is separated off, the ether is dried and evaporated. The residue, consisting of 10- (fl-chloro-ethyl) -phenothiazine, is recrystallized from ethanol. He holds between 90 and 100 g of the product that melts at 97.degree.

08 mm at 200-205 C. The new phenothiazine dissolves easily in dilute acids.
If, instead of dimethylaminoethane thiol, pyrrolidinoethane thiol is reacted with 10- (β-chloroethyl) -phenothiazine, 10- (β-pyrrolidino-ethylthioethyl) phenothiazine is obtained under 0.05 mm at 215 ° C.
<I> Example 2 </I> In the same way as described in Example 1, 45 g of 10 are obtained from 40 g of 10- (β-chloroethyl) phenothiazine, 25 g of diethylaminoethane thiol and 4 g of sodium in a total of 350 cm3 of ethanol - (ss-diethylamino-äthylthioäthyl) -phenothiazine, which boils below 0.07 mm at 198-203 C.

The salts with hydrochloric acid, acetic acid and methanesulfonic acid are not more than about 2-4% soluble in water.

If instead of 10- (ss-chloro-ethyl) - or 10- (y-chloro-propyl) -phenothiazine 10- (chloro-R1) -3-chloro- or -3-methylmercapto-phenothiazine with tert: amino -R2-thiols, the corresponding 10- (tert: amino-alkylthioalkyl) -3-chloro- or -3-methylmercapto-phenothiazines are obtained.
