Verfahren zur Herstellung von neuen Iminodibenzylderivaten Die vorliegende Erfindung betrifft neue N-hetero- cyclische Verbindungen mit wertvollen pharmakolo gischen Eigenschaften sowie Verfahren zu ihrer Herstellung.
In einem Benzolring durch einen Alkylsulfonyl- rest substituierte Iminodibenzylderivate sind bisher nicht bekanntgeworden. Es wurde nun gefunden, dass solche Verbindungen der Formel

Methyl- oder einen Äthylrest, Z einen geradkettigen oder verzweigten Alkylen- rest mit 2-4 Kohlenstoffatomen und Am eine niedere Dialkylaminogruppe bedeuten, wobei ein Alkylrest von, Am direkt mit dem Alkylenrest Z, oder beide Alkylreste einer Dialkylaminogruppe Am unter sich direkt oder über ein Sauerstoffatom, eine niedere Alkylimino-,
Hydroxyalkylimino- oder Al- kanoyloxy-alkyliminogruppe verbunden sein können, wertvolle pharmakologische Eigenschaften, besonders antiallergische, antiemetische und sedative Wirksam-. keit besitzen, sowie auch die Wirkung anderer Arz neistoffe, besonders von Narkotica, potenzieren.
In den Verbindungen der Formel I ist Z bei spielsweise ein Äthylen-, Propylen-, Trimethylen-, 1-Methyl-trimethylen, 2-Methyl trimethylen- oder ein Tetramethylenrest und Am z. B.
Dimethylamino-, Methyl äthylamino-, Diäthylamino-, Methyl-n-propylamino-, Methyl-isopropylamino-, Di-n-butylamino-, Di-isobutylamino-, 1-Pyrrolidinyl-, Piperidino-, Hexamethylenimino-, Morpholino-, 4-Methyl-l-piperazinyl-, 4-Isopropyl I-piperazinyl-, 4-(ss Hydroxyäthyl)
-1 piperazinyl-, 4-(ss Acetoxyäthyl)-1-piperazinyl- oder 4-(ss-Hydroxy-propyl)-1-piperazinyl-rest. Ferner kann Am zusammen mit Z z. B. den ss-(1-Methyl-2-pyrrolidinyl)-äthyl-, 1-Methyl-3 pyrrolidinylmethyl-, ss-(1-Methyl 2-piperidyl)-äthyl-, 1-Methyl-3-piperidylmethyl- oder 1-Methyl 4-piperidyl-rest bedeuten.
Zur Herstellung von .neuen Verbindungen der Formel I wird eine Verbindung der Formel

bis zur Abspaltung von einem Mol Kohlendioxyd er hitzt. Die Verbindungen der Formel II sind ihrer seits z. B. durch Einwirkenlassen von Phosgen auf 3-Methyl- oder Athylsulfonyl-iminodibenzyle und Umsetzung der entstandenen 3-Methyl- oder Äthyl- sulfonyl-5-chlorcarbonyl-iminodibenzyle mit Amino- alkoholen der Formel HO-Z-AM III worin Am und Z die
oben angegebene Bedeutung haben, erhältlich. Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthandisulfonsäure, ss-Hydroxy-äthansulfonsäure, Essigsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure,
Salicylsäure und Mandelsäure bilden die teritären Basen Salze, welche zum Teil wasserlöslich sind.
Im nachfolgenden Beispiel bedeuten Teile Ge wichtsteile, die sich zu Volumteilen wie g zu cm3 verhalten. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel</I> a) 25,2 Teile 3 - Amino - 5 - acetyl-iminodibenzyl werden in 150 Volumteilen Wasser und 30 Volum- teilen konzentrierter Schwefelsäure gelöst und bei 0 unter Rühren mit 7 Teilen Natriumnitrit in 20 Vo- lumteilen Wasser diazotiert. Hierauf wird die Lösung mit Schwefeldioxyd unter Kühlung in einem Eis Kochsalzbad gesättigt. Unter weiterem schwachem Durchleiten von Schwefeldioxyd wird Kupferpulver in kleinen Portionen zugegeben.
Nach Beendigung der Stickstoffentwicklung wird die Sulfinsäure mit Chloroform extrahiert und der getrocknete Extrakt eingeengt. Aus Aceton-Äther umkristallisiert, schmilzt die 5 Acetyl-iminodibenzyl-3-sulfinsäure unter Zer setzung bei 154 .
b) 30,1 Teile 5 - Acetyl- iminodibenzyl-3-sulfin säure werden zur Lösung von 2,3 Teilen Natrium in 250 Volumteilen absolutem Athanol gegeben und mit 16 Teilen Methyljodid versetzt. Nach vierstün digem Erhitzen unter Rückfluss wird das Athanol ab- destilliert. Der Rückstand wird in Chloroform auf genommen und mit wenig Wasser gewaschen. Die getrocknete Chloroformlösung wird eingeengt und mit Petroläther versetzt.
Die abgeschiedenen Kristalle schmelzen bei 153-154 und sind reines 3-Methyl sulfonyl-5-acetyl-iminodibenzyl.
c) 31,5 Teile 3 Methylsulfonyl-5-acetyl imino- dibenzyl werden in 500 Volumteilen Äthylenglykol, enthaltend 25 Teile Kaliumhydroxyd, durch 16stün diges Kochen unter Rückfluss hydrolysiert. Das Re- aktionsgemisch wird auf Wasser gegossen,
und die Kristalle werden abgenutscht. Reines 3 -Methyl- sulfonyl-iminodibenzyl schmilzt, aus Äther umkristal lisiert, bei 175-176 .
d) 27,3 Teile 3 - Methylsulfonyl - iminodibenzyl werden in 1000 Volumteilen absolutem Toluol ge löst. Bei 85-95 wird während 4 Stunden Phosgen eingeleitet. Anschliessend wird das überschüssige, in Toluol gelöste Phosgen entfernt, indem während 1 Stunde bei 95 ein kräftiger Stickstoffstrom durch die Reaktionslösung geblasen wird.
Nach dem Ein engen kristallisiert das 3 -Methylsulfonyl 5-chlor- carbonyl-iminodibenzyl. Aus wenig Benzol umkristal lisiert, schmilzt das reine Produkt bei 164-165 . Analog wird aus 28,7 Teilen 3-Äthylsulfonyl- iminodibenzyl das 3-Äthylsulfonyl-5-chlorcarbonyl- iminodibenzyl erhalten, Smp. 142,5-144 .
e) 33,5 Teile 3-Methylsulfonyl-5-chlorcarbonyl iminodibenzyl werden mit 20,6 Teilen 3-Dimethyl- amino-l-propanol in Toluol während 15 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch mit 70 Volumteilen 2n Na tronlauge geschüttelt, abgetrennt und mit Wasser ge waschen. Die getrocknete Toluollösung wird einge engt.
Der Rückstand, das rohe 3-Methylsulfonyl-5- (3-dimethylamino-l-propoxy-carbonyl)-iminodibenzyl, wird nun unter Wasserstrahlvakuum bis zur Beendi gung der Kohlendioxydentwicklung auf 220 erhitzt. Das Pyrolysat wird in heisses Hexan aufgenommen. Beim Abkühlen kristallisiert das 3-Methylsulfonyl-5- (3-dimethylamino-l-propyl)-iminodibenzyl. Falls not wendig, wird es ein zweites Mal umkristallisiert. Das reine Produkt schmilzt bei 110-111 .
Analog wird der basische Ester aus 33,5 Teilen 3 -MethylsulfonylL-5-chlorcarbonyl-iminodibenzyl und 23,4 Teilen 3-Dimethylamino - 2 - methyl-1 propanol hergestellt und pyrolysiert. Das Pyrolysat wird an neutralem Alox (Markenprodukt) chromatogra- phiert (oder unter Hochvakuum destilliert) und die Base mit ätherischer Salzsäure in das Hydrochlorid übergeführt.
Das reine 3-Methylsulfonyl-5-(3'-di methylamino-2-methyl-propyl-1)-iminodibenzyl-hydro- chlorid schmilzt (aus Methyläthylketon umkristalli siert) bei 229-231 .
Analog wird aus 3 Äthylsulfonyl-5-chlorcarbonyl- iminodibenzyl und 23,4 Teilen 3-Dimethylamino-2- methyl-l-propanol das Hydrochlorid von 3-Äthyl- sulfonyl - 5 - (3 -dimethylamino- 2 -methyl-l-propyl')- irninodibenzyl vom Schmelzpunkt 209-210,5 er halten.

c) 31.5 parts of 3-methylsulfonyl-5-acetyl imino dibenzyl are hydrolyzed in 500 parts by volume of ethylene glycol containing 25 parts of potassium hydroxide by refluxing for 16 hours. The reaction mixture is poured onto water,
and the crystals are sucked off. Pure 3-methylsulfonyl-iminodibenzyl melts, recrystallized from ether, at 175-176.
d) 27.3 parts of 3 - methylsulfonyl - iminodibenzyl are dissolved in 1000 parts by volume of absolute toluene. At 85-95 phosgene is passed in for 4 hours. The excess phosgene dissolved in toluene is then removed by blowing a vigorous stream of nitrogen through the reaction solution at 95% for 1 hour.

The residue, the crude 3-methylsulfonyl-5- (3-dimethylamino-1-propoxycarbonyl) -iminodibenzyl, is now heated to 220 under a water-jet vacuum until the evolution of carbon dioxide has ended. The pyrolysate is taken up in hot hexane. On cooling the 3-methylsulfonyl-5- (3-dimethylamino-1-propyl) -iminodibenzyl crystallizes. If necessary, it is recrystallized a second time. The pure product melts at 110-111.
The basic ester is prepared analogously from 33.5 parts of 3-methylsulfonyl-L-5-chlorocarbonyl-iminodibenzyl and 23.4 parts of 3-dimethylamino-2-methyl-1-propanol and pyrolyzed. The pyrolysate is chromatographed on neutral Alox (branded product) (or distilled under high vacuum) and the base is converted into the hydrochloride with ethereal hydrochloric acid.
The pure 3-methylsulfonyl-5- (3'-dimethylamino-2-methyl-propyl-1) -iminodibenzyl hydrochloride melts (recrystallized from methyl ethyl ketone) at 229-231.
Analogously, 3-ethylsulfonyl-5-chlorocarbonyl-iminodibenzyl and 23.4 parts of 3-dimethylamino-2-methyl-1-propanol are converted into the hydrochloride of 3-ethylsulfonyl -5 - (3-dimethylamino-2-methyl-1-propyl ') - keep irninodibenzyl from melting point 209-210.5.
