Verfahren zur Herstellung von 2a-Methyl-dihydrotestosteronen Die vorliegende Erfindung betrifft ein neues Ver fahren zur Herstellung von 2a-Methyl-dihydrotesto- steronen. Die erfindungsgemäss hergestellten Produkte zeigen ein anabolisch-androgenes Verhältnis von günstiger Höhe und sind insbesondere wertvoll für die Behandlung solcher Leiden, bei welchen eine anabolische oder anti-östrogene Wirkung in Verbin dung mit einer geringeren androgenen Wirkung er zielt werden soll.
Nach einem bekannten Verfahren zur Herstellung von 2-Methyl-androstanen mit einer niedrigen Alkyl- gruppe in 17-Stellung werden die entsprechenden 2-Oxymethylen-Derivate hergestellt, diese Derivate in 2-Methyl-2'-formyl-Verbindungen übergeführt und aus letzteren zur Bildung des 2-Methyl-Produktes Kohlenmonoxyd abgespaltet.
Nunmehr wurde gefunden, dass sich 2-Methyl- androstan-Verbindungen oder Dihydrotestosteron- Derivate nach einem einfachen, einstufigen Verfahren herstellen lassen durch katalytische Hydrierung des entsprechenden 2-Oxymethylen-Ausgangsmaterials.

worin R' eine Acylgruppe einer Kohlenwasserstoff- Carbonsäure mit 2-l2 C-Atomen, vorzugsweise wie sie in veresterten Steroidalkoholen gewöhnlich auf treten, z. B. eine Acetoxy-, Propionoxy- oder Benzoyl- oxygruppe usw., oder aber Wasserstoff bedeutet, in Gegenwart eines Hydrierungskatalysators hydriert.
Die Ausgangsstoffe können erhalten werden, indem man Dihydrotestosteron oder ein 17-Nieder- alkyl-Derivat von Dihydrotestosteron, z.
B. 17-Me- thyl-dihydrotestosteron oder 17-Äthyl-dihydrotesto- steron (welches beispielsweise gewonnen werden kann durch Behandeln von Testosteron, 17-Methyl- testosteron oder 17 - Äthyltestosteron mit einem Alkalimetall in flüssigem Ammoniak), in einem inerten organischen Lösungsmittel, wie Benzol,
suspendiert und dann mit Äthylformiat und Natrium hydrid vermischt. Das Gemisch wird dann während etwa 5 Stunden bei Zimmertemperatur unter Stick stoffatmosphäre gerührt. Hierauf filtriert man die Suspension und behandelt das Gemisch des Natrium salzes der gewünschten Oxymethylen-Verbindung mit einer Säure, z. B. Salzsäure, um die Oxymethylen- Verbindung auszufällen.
Die so hergestellte Oxymethylen-Verbindung kann dann in üblicher Weise verestert werden, unter Bildung eines Diesters üblicher Art, wie schon er wähnt, wenn die 17-Oxygruppe des Ausgangsmate rials sekundär ist, oder eines Monoesters, wenn die 17-Oxygruppe tertiär ist (wie bei niedrigen 17-Alkyl- Derivaten). Die Oxymethylen-Verbindung bzw.
der Ester davon wird dann vorzugsweise, gelöst in einem organischen Lösungsmittel, in Gegenwart eines Hy- drierungskatalysators vorzugsweise bei Zimmer temperatur und Atmosphärendruck hydriert, bis die Wasserstoffabsorption aufhört.
Organische Lösungsmittel, welche sich für die Hydrierungsstufe eignen, sind beispielsweise niedrige aliphatische Alkohole, wie Methanol, Äthylacetat, Dioxan oder Essigsäure. Als Hydrierungskatalysa- toren bevorzugt man Palladium- oder Platinkatalysa toren, z. B. Palladium auf Holzkohle oder Palladium auf Bariumsulfat oder Platinoxyd.
Bei dieser Hydrie rung entsteht die entsprechende 2-Methyl-Verbindung entweder aus dem Ester oder der freien Oxymethylen- Verbindung, wobei eine allfällig vorhandene 17-Ester- gruppe intakt bleibt. Die entstehenden rohen 2-Me- thyl-Produkte können durch Chromatographieren ge reinigt werden.
Bei der Behandlung der freien Oxy- methylen-Derivate oder wenn ein freies 2a-Methyl- Produkt gewünscht wird, ist es zweckmässig, das rohe Hydrierungsprodukt vor dem Chromatographieren mit Alkali zu behandeln.
<I>Beispiel 1</I> Man vermischte eine Suspension von 10 g Di- hydrotestosteron in 500 cm3 wasserfreiem und thio- phenfreiem Benzol mit 10 cm3 Äthylformiat und 3 g Natriumhydrid und rührte das Gemisch 5 Stunden lang unter Stickstoffatmosphäre bei einer Tempera tur von ungefähr 25 C.
Die entstehende Suspension wurde filtriert, und das gewonnene Gemisch aus dem Natriumsalz der Oxymethylen-Verbindung und dem Überschuss aus Natriumhydrid wurde mit Benzol ge waschen und getrocknet. Dieses Gemisch wurde lang sam einer kräftig gerührten Lösung von 20 cm3 kon zentrierter Salzsäure in 500 cm3 Wasser zugesetzt, und das Rühren wurde 30 Minuten lang fortgesetzt, worauf der Niederschlag gesammelt und mit destillier tem Wasser gut gewaschen wurde.
Nach dem Trock nen im Vakuum erhielt man 9,7 g 2-Oxymethylen- dihydrotestosteron.
Man löste 7 g 2-Oxymethylen-dihydrotestosteron in 300 cm3 Methanol und vermischte mit 2,511/o eines Katalysators aus 1011/o Palladium auf Holzkohle. Das Gemisch wurde bei ungefähr 25 C und bei Atmosphärendruck hydriert, bis die Wasserstoff absorption aufhörte. Man entfernte den Katalysator durch Filtrieren, setzte der Lösung 1 g Kalium hydroxyd in 5 cm3 Wasser zu und liess sie dann 1 Stunde bei Zimmertemperatur stehen.
Nach Zusatz von 2 cm?, Essigsäure entfernte man das Lösungs mittel vollständig unter vermindertem Druck, setzte dem Rückstand Wasser zu und extrahierte das Pro dukt mit Methylendichlorid. Der Auszug wurde mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter Vakuum zur Trockne einge dampft. Der Rückstand wurde in Benzol gelöst und auf eine chromatographische Säule mit 125 g alka lischer Tonerde aufgetragen. Das Eluieren erfolgte mit mehreren Fraktionen von je 100 cm- Benzol. Das gewünschte Produkt wurde aus den Fraktionen 2-6 gewonnen.
Nach dem Abdampfen des Lösungs mittels kristallisierte man das Produkt aus einem Aceton-Hexan-Gemisch, wobei man 3,3 g reines 2a-Methyl-dihydrotestosteron erhielt.
<I>Beispiel 2</I> Man löste 2 g 2-Oxymethyl2n-dihydrotestosteron in 80 cm?, Essigsäure und hydrierte mit 1,0 g eines Katalysators aus 10% Palladium auf Holzkohle unter den im vorhergehenden Beispiel beschriebenen Bedingungen. Nach dem Abfiltrieren des Katalysa- tors wurde das Lösungsmittel unter reduziertem Druck zur Trockne abgedampft. Der Rückstand wurde mit 100 cm-' Methanol und 1 g Kalium- hydroxyd vermischt.
Die Lösung wurde 30 Minuten auf Rückfluss erhitzt und dann mit Wasser verdünnt und mit Methylendichlorid extrahiert. Der Auszug wurde mit Wasser neutralgewaschen, über wasser freiem Natriumsulfat getrocknet und unter Vakuum zur Trockne eingedampft. Der Rückstand wurde in Benzol gelöst und in gleicher Weise wie in Beispiel 1 chromatographiert. Man erhielt 2a-Methyl-dihydro- testosteron.
<I>Beispiel 3</I> Ein Gemisch aus l g 2-Oxymethylen-dihydro- testosteron, 10 cm3 Pyridin und 2 cm3 Acetanhydrid wurde bei Zimmertemperatur 16 Stunden reagieren gelassen und dann in Wasser gegossen. Man extra hierte das Produkt mit Methylendichlorid und wusch nacheinander mit verdünnter Salzsäure, Na- triumbicarbonatlösung und Wasser, trocknete und dampfte unter reduziertem Druck zur Trockne ein.
Man erhielt in dieser Weise das Diacetat von 2-Oxy- methylen-dihydrotestosteron.
Das Diacetat wurde gleich wie in den vorherigen Beispielen hydriert und aufgearbeitet, wobei man 2a-Methyl-dihydrotestosteron erhielt, welches mit dem Produkt dieser Beispiele identisch war.
<I>Beispiel 4</I> Nach dem in den vorhergehenden Beispielen be schriebenen Verfahren wurde 17a-Methyl-2-oxy- methylen - dihydrotestosteron in 20,17a - Dimethyl- dihydrotestosteron übergeführt.
<I>Beispiel 5</I> Nach dem in den Beispielen 1, 2 und 3 beschrie benen Verfahren wurde 17a-Äthyl-2-oxymethylen- dihydrotestosteron in 2a-Methyl-17a-äthyl-dihydro- testosteron übergeführt.
<I>Beispiel 6</I> Man liess ein Gemisch aus 1 g 2-Oxymethylen- dihydrotestosteron, 10 cm3 Pyridin und 2 cm3 Pro- pionsäureanhydrid 16 Stunden bei Zimmertemperatur reagieren, worauf es in Wasser gegossen wurde.
Man erhitzte die entstehende Suspension 1 Stunde auf dem Dampfbad, um den überschuss an Propionsäure- anhydrid zu hydrolysieren, kühlte und extrahierte mit Methylendichlorid. Der Auszug wurde nacheinan der mit verdünnter Salzsäure, Natriumbicarbonat- lösung und Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter Vakuum zur Trockne eingedampft.
Man erhielt das Dipropionat von 2-Oxymethylen-dihydrotestosteron, welches in methanolischer Lösung unter den in Beispiel 1 be schriebenen Bedingungen mit Wasserstoff behandelt wurde. Nach dem Aufhören der Wasserstoffaufnahme filtrierte man vom Katalysator ab und dampfte die Lösung unter Vakuum zur Trockne ein.
Den Rück stand löste man in einem Benzol-Hexan-Gemisch, leitete dieses auf eine chromatographische Säule aus neutraler Tonerde und eluierte das Produkt mit Benzol-Hexan-Gemischen mit allmählich zunehmen dem Benzolanteil. Durch Kristallisieren des Eluates aus Aceton-Hexan erhielt man das Propionat von 2a-Methyl-dihydrotestosteron.
Process for the production of 2a-methyl-dihydrotestosterones The present invention relates to a new process for the production of 2a-methyl-dihydrotestosterones. The products prepared according to the invention show an anabolic-androgenic ratio of a favorable level and are particularly valuable for the treatment of those conditions in which an anabolic or anti-estrogenic effect in conjunction with a lower androgenic effect is to be achieved.
According to a known process for the preparation of 2-methyl-androstanes with a lower alkyl group in the 17-position, the corresponding 2-oxymethylene derivatives are prepared, these derivatives are converted into 2-methyl-2'-formyl compounds and from the latter to Formation of the 2-methyl product carbon monoxide is split off.
It has now been found that 2-methyl androstane compounds or dihydrotestosterone derivatives can be prepared by a simple, one-step process by catalytic hydrogenation of the corresponding 2-oxymethylene starting material.

The starting materials can be obtained by adding dihydrotestosterone or a 17-lower alkyl derivative of dihydrotestosterone, e.g.
B. 17-methyl-dihydrotestosterone or 17-ethyl-dihydrotestosterone (which can be obtained, for example, by treating testosterone, 17-methyl-testosterone or 17-ethyl testosterone with an alkali metal in liquid ammonia), in an inert organic solvent like benzene,
suspended and then mixed with ethyl formate and sodium hydride. The mixture is then stirred for about 5 hours at room temperature under a nitrogen atmosphere. The suspension is then filtered and the mixture of the sodium salt of the desired oxymethylene compound is treated with an acid, e.g. B. hydrochloric acid to precipitate the oxymethylene compound.
The oxymethylene compound prepared in this way can then be esterified in the usual way, with the formation of a diester of the usual type, as already mentioned if the 17-oxy group of the starting material is secondary, or a monoester if the 17-oxy group is tertiary (such as for lower 17-alkyl derivatives). The oxymethylene compound or
the ester thereof is then preferably, dissolved in an organic solvent, hydrogenated in the presence of a hydrogenation catalyst, preferably at room temperature and atmospheric pressure, until the absorption of hydrogen ceases.
Organic solvents which are suitable for the hydrogenation stage are, for example, lower aliphatic alcohols such as methanol, ethyl acetate, dioxane or acetic acid. As hydrogenation catalysts, palladium or platinum catalysts are preferred, e.g. B. palladium on charcoal or palladium on barium sulfate or platinum oxide.
During this hydrogenation, the corresponding 2-methyl compound is formed either from the ester or from the free oxymethylene compound, with any 17-ester group that may be present remaining intact. The resulting crude 2-methyl products can be purified by chromatography.
When treating the free oxymethylene derivatives or when a free 2a-methyl product is desired, it is advisable to treat the crude hydrogenation product with alkali before chromatography.
<I> Example 1 </I> A suspension of 10 g of dihydrotestosterone in 500 cm3 of anhydrous and thiophene-free benzene was mixed with 10 cm3 of ethyl formate and 3 g of sodium hydride and the mixture was stirred for 5 hours under a nitrogen atmosphere at a temperature of about 25 C.
The resulting suspension was filtered, and the obtained mixture of the sodium salt of the oxymethylene compound and the excess sodium hydride was washed with benzene and dried. This mixture was slowly added to a vigorously stirred solution of 20 cm3 of concentrated hydrochloric acid in 500 cm3 of water and stirring was continued for 30 minutes after which the precipitate was collected and washed well with distilled water.
After drying in vacuo, 9.7 g of 2-oxymethylene dihydrotestosterone were obtained.
7 g of 2-oxymethylene dihydrotestosterone were dissolved in 300 cm3 of methanol and mixed with 2.511 / o of a catalyst composed of 1011 / o palladium on charcoal. The mixture was hydrogenated at about 25 C and at atmospheric pressure until hydrogen absorption ceased. The catalyst was removed by filtration, 1 g of potassium hydroxide in 5 cm3 of water was added to the solution and it was then left to stand for 1 hour at room temperature.
After adding 2 cm ?, acetic acid, the solvent was completely removed under reduced pressure, water was added to the residue and the product was extracted with methylene dichloride. The extract was washed with water, dried over anhydrous sodium sulfate and evaporated to dryness in vacuo. The residue was dissolved in benzene and applied to a chromatographic column with 125 g of alkaline clay. The elution was carried out with several fractions of 100 cm benzene each. The desired product was obtained from fractions 2-6.
After evaporation of the solution, the product was crystallized from an acetone-hexane mixture, 3.3 g of pure 2a-methyl-dihydrotestosterone being obtained.
<I> Example 2 </I> 2 g of 2-oxymethyl2n-dihydrotestosterone were dissolved in 80 cm ?, acetic acid and hydrogenated with 1.0 g of a catalyst composed of 10% palladium on charcoal under the conditions described in the previous example. After filtering off the catalyst, the solvent was evaporated to dryness under reduced pressure. The residue was mixed with 100 cm- 'methanol and 1 g of potassium hydroxide.
The solution was heated to reflux for 30 minutes and then diluted with water and extracted with methylene dichloride. The extract was washed neutral with water, dried over anhydrous sodium sulfate and evaporated to dryness in vacuo. The residue was dissolved in benzene and chromatographed in the same way as in Example 1. 2a-methyl-dihydro-testosterone was obtained.
<I> Example 3 </I> A mixture of 1 g of 2-oxymethylene dihydro testosterone, 10 cm 3 of pyridine and 2 cm 3 of acetic anhydride was allowed to react at room temperature for 16 hours and then poured into water. The product was extracted with methylene dichloride and washed successively with dilute hydrochloric acid, sodium bicarbonate solution and water, dried and evaporated to dryness under reduced pressure.
The diacetate of 2-oxymethylene dihydrotestosterone was obtained in this way.
The diacetate was hydrogenated and worked up in the same way as in the previous examples, 2a-methyl-dihydrotestosterone being obtained, which was identical to the product of these examples.
<I> Example 4 </I> According to the process described in the preceding examples, 17a-methyl-2-oxymethylene - dihydrotestosterone was converted into 20.17a - dimethyldihydrotestosterone.
<I> Example 5 </I> Using the method described in Examples 1, 2 and 3, 17a-ethyl-2-oxymethylene-dihydrotestosterone was converted into 2a-methyl-17a-ethyl-dihydrotestosterone.
Example 6 A mixture of 1 g of 2-oxymethylene dihydrotestosterone, 10 cm3 of pyridine and 2 cm3 of propionic anhydride was allowed to react for 16 hours at room temperature, after which it was poured into water.
The resulting suspension was heated on a steam bath for 1 hour in order to hydrolyze the excess propionic anhydride, cooled and extracted with methylene dichloride. The extract was washed one after the other with dilute hydrochloric acid, sodium bicarbonate solution and water, dried over anhydrous sodium sulfate and evaporated to dryness under vacuum.
The dipropionate of 2-oxymethylene dihydrotestosterone was obtained, which was treated with hydrogen in methanolic solution under the conditions described in Example 1. After the uptake of hydrogen had ceased, the catalyst was filtered off and the solution was evaporated to dryness in vacuo.
The residue was dissolved in a benzene-hexane mixture, this was passed onto a chromatographic column of neutral clay and the product was eluted with benzene-hexane mixtures with gradually increasing benzene content. The propionate of 2a-methyl-dihydrotestosterone was obtained by crystallizing the eluate from acetone-hexane.
