48, 72 und 96 Stunden nach intmaperitonealer Injektion von 100 MLD von Diplococcus pneumoniae, Type 1, wurde bei dner Dosis von 1 und 0,5 mg pro Gramm Körpergewicht ein Prozentsatz von 80,04 bzw.
10"/o an Überlebenden festgestellt, während das bekannte Sulfisoxazol bei einer Dosierung von 2 und 1 mg unter analogen Versuchsbedingungen, nur 50 und 0 % ergab. Bei allen unbehandelten Tieren wirkten die Injektionen tödlich bei ST 100 = 2 Tagen.
Bei diesem Experiment zeigte sich die neue Verbindung bei Verabreichung per os mehr als zweimal so wirksam als S@ulfiis.oxazol.

Bei dem vorliegenden Verfahren wird die Art des angewandten Lösungsmittels durch seine Nicht- Reaktionsfähigkeit bestimmt. Wenn Sulanilhalogenide als leicht reagierende Derivate der Benzolsulfosäure Anwendung finden, werden vorzugsweise basische und bzw. oder basebilidende Lösungsmittel;
, wie Pynidin, Picolin oder Aceton, mit Natriumbicarbonat angewandt. Wenn auch gewöhnlich als leicht reagie- rende Derivate der Benzolsulfosäure Benzolsulfonyl- halogenid'e, insbesondere das Chlorid, Verwendung finden, s,
o können doch auch Derivate der Benzol-' mit anderen Funktionsgruppen, so z. B.
der Säureester, angewandt werden. Als in die pri märe Amimogruppe überführbarer Rest in der 4-Stel- lung der reaktionsfähigen Benzolsulfosäune-Derivate kommt insbesondere eine Nitro-,
Acylamino- oder Carbalkoxyamino-Gruppe in Betracht. Die Art der Reaktion in der Stufe B richtet sich nach der Natur des Substituenten R. Wenn R ein Acylamino-Radikal, z. B. eine Acetiamino-Grupp,e, oder ein Carbalkoxy- amino-Rad'ikal, z.
B. eine Carbäthoxyamino-Gruppe, ist, so muss das Produkt verseift werden; wenn da gegen R eine Nitro-Gruppe ist, so muss das Produkt reduziert werden. Diese Reaktionen kann man durch- führen, wie es in; der chemischen Technik üblich ist, so z.
B. eine Hydrolyse unter Erhitzen mit wässriger Natriumhydroxyd-Lösung oder eine Reduktion durch Behandeln mit Zinkstaub und Chlorwasserstoffsäure.
Das 3-Arnino-5-merhyl-isoxazol, das bei der vor- liegenden Erfindung als Ausgangsmaterial dient, ist gleichfalls eine neue Verbindung. Man, erhält dieselbe durch Hydrolyse von Äthyl-5-methyl-isoxazo;L#-3- carbamat und Curtiuswhen Abbau von: 5-Methyl- isoxazoil,-3-camboxylsäure nach der Frerischen Me thode (CL CA 26, 5953 [1932]).
Die Verbindung knistalfislert in Prismen mit dem Schmelzpunkt 61 bis 62 C.
<I>Beispiel</I> <I>a) Herstellung von</I> 3-Amino-5-methyl-isoxazol 1,7 g Äthyl5-methy14soxazol-3-carbamat wer den auf einem siedenden Wasserbad mit 5 cm3 10 A/o iger wässriger Natriumhydroxyd-Lösung 8 Stun- den lang erhitzt. Hierauf wird das Reaktionsgemisch mehrere Male mit Äther oder Benzol extrahiert und der Extrakt zwecks Abtrennung aus dem Lösungs mittel gekühlt und dann <RTI
ID="0003.0008"> getrocknet. Der Rückstand wird nach einiger Zeit fest und gibt nach Umkristalli- sieren aus Benzol das 3 Amino-5-methyl-isoxazol in prisma#tische.n Kristallen vom Schmelzpunkt 61 bis 62 C. Elementananalysie für die empirische Formel C4H6ON2.
Errechnet: C 48,97; H 6,16; N 28,57. Gefunden: C 49,42; H 6,50; N 28,28. Diese Substanz ist eine neue Verbindung und wird in ähnlicher Weise auch durch Hydrolyse von Benzyl@5-methyl-isoxazol-3-carlbamat (Schmelzpunkt 80-81 C) erhalten.
