Von beson derem Interesse wegen ihren therapeutischen Eigen schaften sind die Verbindungen der Formel

worin R4 für Wasserstoff, Chlor oder Niederalkyl seht, vor allem 1-(ss-Diäthylamino-äthyl)-2-benzyl-5-nitro- benzimidazol, 1-(ss-Diäthylamino-äthyl)-2-(p-chlorbenzyl)-5- nitro-benzimidazol, 1-(ssDiäthylamino-äthyl)-2-(p-methylbenzyl)-5- nitro-benzimidazol und ihre Salze.
Hervorragend in ihrer Wirkung sind Verbin dungen der obigen Formel, in denen R4 eine Nieder- alkoxygruppe ist, wie 1-(ss Diäthylamino-äthyl)-2- (p - methoxy - benzyl) - 5-nitro-benzimidazol, vorzugs weise 1-(ssDiäthylamino-ätlzyl)-2-(p-äthoxy-benzyl)
- 5 - nitro - benzimidazol. Diese Verbindungen sind zudem sehr gute Muskelrelaxantia. Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man ein 2-(R-A-NH)-5-nitro-anilin mit einer gegebenenfalls im Phenylrest substituierten Phenylessigsäure oder einem ihrer reaktionsfähigen funktionellen Derivate umsetzt.
Die verfahrensgemässe Umsetzung kann in An- oder Abwesenheit von Verdünnungs- und/oder Kon- densationsmitteln, wenn nötig, bei erhöhter Tempe ratur, im offenen oder im geschlossenen Gefäss unter Druck ausgeführt werden.
In den Verfahrensprodukten können Substituen- ten im Benzylrest durch andere Gruppen ersetzt, wie z. B. eine Hydroxygruppe in eine verätherte oder veresterte Hydroxylgruppe, wie eine Nieder- alkoxygruppe, oder eine Nitrogruppe in eine Amino- gruppe und diese in eine Niederalkoxygruppe oder ein Halogenatom übergeführt werden.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form der freien. Basen oder ihrer Salze. Aus den Salzen können in an sich bekannter Weise die freien Basen gewonnen werden. Von letz teren lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze ge eignet sind, Salze gewinnen, wie z.
B. der Halo genwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Bernstein säure, Äpfelsäure, Methansulfonsäure, Äthansulfon- säure, Oxyäthansulfonsäure, Benzol- oder Toluol- sulfonsäure.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden. Die neuen Verbindungen können als Arzneimit tel, z. B. in Form pharmazeutischer Präparate, Ver wendung finden, welche sie oder ihre Salze in Mi schung mit einem für die enterale, parenterale oder topicale Applikation geeigneten, pharmazeutischen, organischen oder anorganischen, festen oder flüssi gen Trägermaterial enthalten.
In den folgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben. <I>Beispiel 1</I> Zu einer aus 4,7 g Benzylcyanid und 2,3 ml Athanol, gelöst in 50 ml Chloroform, durch Einlei ten von trockenem Salzsäuregas bei -10 in üblicher Weise hergestellten, mit Salzsäure gesättigten Imino- ätherhydrochlorid-Lösung wird 5,04 g 2-(,d-Diäthyl- amino-äthylamino)-5-nitro-anilin zugegeben. Hierauf wird zunächst zwei Stunden bei 25 und dann wäh rend 20 Stunden am Rückfluss gerührt.
Das Reak tionsgemisch wird mit wässriger Salzsäure ausgezo gen, die nochmals mit Chloroform gewaschenen Auszüge mit Ammoniaklösung alkalisch gestellt und mit Chloroform extrahiert, und der mit Sodalösung gewaschene Chloroform Extrakt nach Trocknen mit Magnesiumsulfat eingedampft. Kristallisation des Rückstandes aus Äther ergibt 1-(B-Diäthylamino- äthyl)-2-benzyl-5 nitro-benzimidazol der Formel

Das durch Versetzen einer äthanolischen Lösung der Base mit einem Äquivalent äthanol'ischer Salzsäure, Ein engen und Kristallisation nach Zugabe von wenig Wasser erhaltene Hydrochlorid schmilzt bei 199 bis 201o. <I>Beispiel 2</I> Zu einer aus 5,65 g p-Chlorbenzyleyanid und 2,3 ml Äthanol, gelöst in 50 ml Chloroform, durch Einleiten von trockenem Salzsäuregas bei -10 in üblicher Weise hergestellten, mit Salzsäure gesättig ten Iminoäther-hydrochlorid-Lösung wird 5,04 g 2-(ss8 Diäthylamino-äthylamino)-5-nitro-anilin zugege ben.
Hierauf wird zunächst zwei Stunden bei 25 , dann 20 Stunden bei Rückfluss gerührt. Das Reak tionsgemisch wird mit wässriger Salzsäure ausgezo gen, die mit Chloroform gewaschenen Auszüge mit Ammoniaklösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung gewaschene Chloroformextrakt nach Trocknen mit Magnesium sulfat eingedampft. Kristallisation des Rückstandes aus Äther-Pentan gibt 1-(f-Diäthylamino-äthyl)-2-(p- chlor-benzyl)-5-nitro-benzimidazol in Massgelben Blättchen vom F. = 75-76 .
Das durch Versetzen einer äthanolischen Lösung der Base mit einem Äquivalent Salzsäure in Äthanol erhaltene kristalli sierte Hydrochlorid schmilzt bei 238-240 . Das auf analoge Weise hergestellte Methansulfonat zeigt Smp. 163-166 .
<I>Beispiel 3</I> Zum Iminoäther-hydrochlorid der m-Methoxy- phenyl-essigsäure, der erhalten wird aus 7,3 g m-Methoxy-benzylcyanid und 2,9 ml absolutem Al kohol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenlassen bei 25 während 16 Stunden und Eindampfen bei redu ziertem Druck, werden bei Raumtemperatur 7,2 g 2 - (ss -Diäthylamino-äthylamino)-5-nitro-anilin-hydro- chlorid in 90 ml Eisessig zugegeben.
Hierauf wird 20 Stunden bei 40-4.5 gerührt. Das Reaktions gemisch wird bei reduziertem Druck eingedampft, der Rückstand in wässriger Salzsäure aufgenommen, die mit Chloroform gewaschene, saure Lösung mit Ammoniaklösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung gewaschene und mit Magnesiumsulfat getrocknete Chloroform-Extrakt eingedampft.
Das so erhaltene, rohe 1-(ss-Diäthyl- amino - äthyl) -# 2 -,(m -methoxy-benzyl)-5-nitrobenz- imidazol kann durch Lösen in Äthanol und Zugabe eines Äquivalentes äthanolischer Salzsäure in das farblose, kristallisierte Hydrochlorid vom F.<B>186</B> bis 188 übergeführt werden.
<I>Beispiel 4</I> Zum Iminoäther-hydrochlorid der 3,4-Dimeth- oxy-phenylessigsäure, der erhalten wird aus 8,85 g 3,4 Dimethoxy-benzylcyanidund 2,9 ml absolutemÄthanol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenlassen während 16 Stunden bei 25 und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 7,2 g 2-(ss-Di- äthylamino - äthylamino) - 5 - nitro-ahilin - hydrochlorid in 90 ml Eisessig zugegeben.
Hierauf wird 20 Stun den bei 40-45 gerührt. Das Reaktionsgemisch wird bei reduziertem Druck eingedampft, der Rückstand in wässriger Salzsäure aufgenommen, die mit Chloro form gewaschene saure Lösung mit Ammoniaklösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung gewaschene und mit Magnesium- sulfat getrocknete Chloroform-Extrakt eingedampft.
Das so erhaltene, rohe 1-(ss Diäthylamino-äthyl)-2- (3',4'-dimethoxy benzyl)-5-nitro-benzimidazol kann durch Lösen in Athanol und Zugabe der berechneten Menge äthanolischer Salzsäure ins blassgelbe, kristal lisierte Hydrochlorid vom F. 192-194 übergeführt werden.
<I>Beispiel 5</I> Zum Iminoäther-hydrochlorid der p-Methoxy- phenylessigsäure, das erhalten wird aus 7,3 g p-Meth- oxy-benzylcyanid und 2,9 ml absolutem Äthanol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenlassen während 16 Stunden bei 25 und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 7,
2 g 2-(ss Di- äthylamino-äthyl amino@ - 5 - nitro-anilin-hydrochl'orid in 90 ml Eisessig zugegeben. Hierauf wird 20 Stun den bei 40-45 gerührt.
Das Reaktionsgemisch wird bei reduziertem Druck eingedampft, der Rückstand in wässriger Salzsäure aufgenommen, die mit Chloro form gewaschene saure Lösung mit Ammoniaklösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung gewaschene und mit Magnesiumsulfat getrocknete Chloroformextrakt eingedampft.
Das so erhaltene, rohe 1-(l-Diäthylamino-äthyl')-2-(p-meth- oxy-benzyl)-5-nitro-benzimidazol kann durch Lösen in Äthanol und Zugabe der berechneten Menge äthanolischer Salzsäure ins farblose kristallisierte Hy- drochlorid vom F. 197-198 übergeführt werden.
<I>Beispiel 6</I> Zum Iminoäther-hydrochlorid der p-Methyl- phenylessigsäure, das erhalten wird aus 6,55 g 4 Me- thyl-benzylcyanid und 2,9 ml absolutem Alkohol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenlassen während 16 Stunden bei 25 und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 7,2 g 2-(ss Di- äthylamino-äthyl'amino)
- 5 -initro-anilin-hydrochlori;d in 90 ml Eisessig zugegeben. Hierauf wird 20 Stun den bei 40-4.5 gerührt. Das Reaktionsgemisch wird bei reduziertem Druck eingedampft, der Rück- stand in wässriger Salzsäure aufgenommen, die mit Chloroform gewaschene saure Lösung mit Ammo- niaklösung alkalisch gestellt, mit Chloroform extra hiert und der mit Sodalösung gewaschene und mit Magnesiumsulfat getrocknete Extrakt eingedampft.
Das so erhaltene rohe 1-(l-Diäthylamino-äthyl)-2- (p-methyl-benzyl)-5-nitro-benzimidazol kann durch Lösen in Äthanol und Zugabe der berechneten Menge äthanolischer Salzsäure ins farblose, kristalli sierte Hydrochlorid vom F. 216 übergeführt werden.
<I>Beispiel 7</I> Zum Iminoäther-hydrochlorid der p-Athoxy- phenyl-essigsäure, das erhalten wird aus 8,05 g p-Äthoxybenzylcyanid und 2,9 ml absolutem Alkohol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenlassen bei 25 wäh rend 16 Stunden und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 7,2 g 2-(ss Di- äthylamino-Whylamino)
- 5 - nitro"aniln'n. hydrochlorid in 90 ml Eisessig zugegeben. Hierauf wird 20 Stun den: bei 40-45 gerührt. Das Reaktionsgemisch wird bei reduziertem Druck eingedampft, der Rückstand in wässriger Salzsäure aufgenommen, die mit Chloro form gewaschene, saure Lösung mit Ammoniak lösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung gewaschene und,
mit Magne- siumsulfat getrocknete Chloroformextrakt einge dampft. Das so erhaltene, rohe 1-(ss-Diäthylamino- äthyl)-2-(p-äthoxy-benzyl')-5-nitro-benzimidazol kann durch Lösen in Äthanol und Zugabe eines Äquiva lentes äthanolischer Salzsäure in das farblose kristal- lisierte Hydrochlorid mit Doppelschmelzpunkt<B>105</B> bis 107 J162-163 übergeführt werden.
Auf analoge Weise kann auch 1-(B-Diäthylamino-äthyl)-2-(p-tert.-butyl-benzyl)- 5-nitro-benzimidazol-hydrochlorid vom F. 208 210 , 1-(B Diäthylamino-äthyl)-2-(p fluor-benzyl)-5-nitro- benzimidazol-methansulfonat vom F. 165-167 und 1-(B Diäthylamino-äthyl)-2-(p-hydroxy-benzyl)- 5-nitro benzimidazol vom F. 188-190 . Hydro- chlorid F. 246-248 , hergestellt werden.
<I>Beispiel 8</I> Zum Iminoäther-hydrochlorid der o-Chlorphenyl- essigsäure, das erhalten wird aus 7,6 g o-Chlorbenzyl- 'cyanid und 2,9 ml absolutem Alkohol in 50 ml Chloroform durch Einleiten von trockenem Salz säuregas bei -10 , Stehenlassen bei 25 während 16 Stunden und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 7,2 g 2-(8-Diäthylamino- äthylamino)-5-nitro-anilin-hydrochlorid in 90 ml Eisessig zugegeben.
Hierauf wird 20 Stunden bei 40-45 gerührt. Das Reaktionsgemisch wird bei reduziertem Druck eingedampft, der Rückstand in wässriger Salzsäure gelöst, die mit Chloroform ge waschene saure Lösung mit Ammoniaklösung alka lisch gestellt, mit Chloroform extrahiert und der mit :Sodalösung gewaschene und über Magnesiumsulfat getrocknete Chloroformextrakt eingedampft.
Kristal lisation aus Aceton-Äther gibt 1-(B Diäthylamino- 'äthyl) - 2 - (o-chlor-benzyl')-5-nitro-benzimidazol vom F. 104-107 . Das durch Lösen in Äthanol und Zu gabe von einem Äquivalent äthanolischer Salzsäure erhaltene Hydrochlorid zeigt einen F. 200-203 .
Auf genau analoge Weise kann 1-(f-Diäthylamino-äthyl)-2-(2',4'-dichlor-benzyl)- 5-nitro-benzimidazol vom F. 105-107 , Hydro- chlorid F. l98-199 , 1-(fj-Diäthylamino-äthyl)-2-(o-methyl-benzyl)-5-nitro- benzimidazol-hydrochlorid vom F. 198-l99 , 1-(ii-Diäthylamino-äthyl)-2-(2',5'-dimethyl-benzyl)- 5-nitro-benzimidazol-hydrochlorid vom F. 210 bis 212 , 1-(ss-Piperidino-äthyl)-2-(p-chlor-benzyl)-5-nitro- benzimidazol vom F. 109-110 , Hydrochlorid F.

Die Ausgangsstoffe können in folgender Weise dargestellt werden: a) 1-Methyl-4-(B-amino-äthyl)-piperazin.
38,6 g Methyl-di-(P-chlor-äthyl)-amin-hydro- chlorid werden in 300 ml absolutem Alkohol und 60 g trockenem Athylendiamin während 16 Stunden am Rückfluss gerührt. Dann wird abgekühlt und eine NatriumalkoholatLösung, hergestellt aus 14 g Na trium und 250 ml absolutem Alkohol, zugefügt, vom ausgeschiedenen Kochsalz abfiltriert und nach Ein dampfen der verbleibende Rückstand fraktioniert destilliert. Die bei 81-83 (11 mm Hg) siedende Fraktion besteht aus 1-Methyl-4-(fl-amino-äthyl)- piperazin.
b) N-[,- (4-Methyl-piperazino)-äthyl]-2,4-dinitro- anilin.
60,7 g 2,4-Dinitrochlorbenzol und 50 g 1-Methyl- 4-(d-amino-äthyl)-piperazin werden in 0,4 1 Alkohol während 4 Stunden am Rückfluss gekocht.
Dann wird mit wässrigem Ammoniak stark alkalisch gestellt und das ausgeschiedene N-[ss-(4-Methyl-piperazino)-äthyl]- 2,4-dinitro-anilin vom F. 120-121 abgenutscht. Auf analoge Weise kann N-(8 Dimethylamino@-äthyl) 2,4-dinitro-anifin-hydro- chlorid vom F. 224-225 , N-(fl-Piperidino-äthyl)-2,4-dinitro-anilin vom F.
103 bis 105 , N-(B-Morpholino-äthyl)-2,4-dinitro-anilin vom F.111 bis 112 und N - (y - Diäthylamino - propyl)-2,4-dinitro-anilin vom F. l70-175 hergestellt werden.
c) 2-f-(4'-Methyl-piperazino)-äthylamino-5-nitro- anilin.
60 g N-[fj-(4-Methyl-piperazino)-äthyl]-2,4-di- nitro-anilin werden in 400 ml Alkohol und 107 cm- konz. wässrigem Ammoniak gelöst und Hss bis zur Sättigung eingeleitet. Der Alkohol wird abdestilliert, der Rückstand in 5n HCl aufgenommen, vom aus geschiedenen Schwefel abfiltriert, das Filtrat wieder mit Ammoniaklösung alkalisch gestellt und mit Benzol extrahiert.
Das mit Magne@siumsulfat getrock nete und eingeengte Benzol gibt 2-f-(4'-Methyl- piperazino)-äthylamino-5-nitro-anilin in roten Kri stallen vom F. 148-151 , Monohydrochlorid F. 180 bis 185".
Auf analoge Weise kann 2-(ss-Dimethylaniino-äthylamino)-5-nitro-anilin- hydrochlorid vom F. 222-223 , 2-(fl-Piperidino-äthylamino)-5-nitro-anilin vom F. 106-108 , Hydrochlorid F. 220-222 , 2-(f-Morpholino-äthylamino)-5-nitro-anilin vom F.
144-145 , Hydrochlorid F.225-227 und 2-(y-Diäthylamino-gropylamino)-5-nitro-anilin-hydro- chlorid vom F. 199-200 hergestellt werden.
<I>Beispiel 9</I> Zum Iminoäther-hydrochlorid der p-Butoxy- phenylessigsäure, das erhalten wird aus 5,66 g p-Butoxy-benzylcyanid und 1,75 ml absolutem Äthanol in 50 ml Chloroform durch Einleiten von trockenem Salzsäuregas bei -10 , Stehenl'assen während 16 Stunden bei 25 und Eindampfen bei reduziertem Druck, werden bei Raumtemperatur 4,3 g 2-(p-Diäthylamino-äthylamino)-5-nitro-anilin- hydrochlorid in 60 ml Eisessig zugegeben. Hierauf wird 20 Stunden bei 40-45 gerührt.
Das Reak tionsgemisch wird bei reduziertem Druck einge dampft, der Rückstand in wässriger Salzsäure auf genommen, die mit Chloroform gewaschene saure Lösung mit Ammoniaklösung alkalisch gestellt, mit Chloroform extrahiert und der mit Sodalösung ge waschene und mit Magnesiumsulfat getrocknete Chloroformextrakt eingedampft. Das so erhaltene, rohe 1-(ss-Diäthylamino-äthyl)-2-(p-butoxy-benzyl)- 5-nitro-benzimidazol kann durch Lösen in Äthanol und Zugabe der berechneten Menge äthanolischer Salzsäure ins farblose kristallisierte Hydrochlorid vom F. 150-152 übergeführt werden.
Auf analoge Weise kann 1-(i3-Diäthylamino-äthyl)-2-(p-propyloxy-benzyl)- 5-nitro-benzimidazol-hydrochlorid vom F. 115 bis 116 und 1-(ss-Diäthylamino-äthyl)-2-(p-isopropyloxy-benzyl)- 5-nitro-benzimidazol-hydrochlorid vom F.<B>172</B> bis 173 hergestellt werden.
Das als Ausgangsmaterial benötigte p-Butoxy- benzylcyanid kann wie folgt erhalten werden: Zu 13,3 g p-Hydroxybenzylcyanid in 100 ml Alkohol werden unter Rühren gleichzeitig 37 g Butyl- jodid und 20 ml lOn NaOH zugetropft. Nach 16 Stunden Rückfluss wird im Vakuum eingedampft, der Rückstand in Äther und Wasser aufgenommen,
die mit Natronlauge gewaschene und über MgS04 ge trocknete Ätherlösung eingedampft und das erhaltene p-Butoxy-benzylcyanid bei 100-110 und 0,18 Torr destilliert.
Die auf ähnliche Weise hergestellten p-Propyloxy- benzylcyanid und p-Isopropyl-benzylcyanid destillie- ren beide bei 100 und 0,2 Torr.
