Verfahren zur Herstellung neuer Piperazin-Verbindungen und ihrer Säureadditionssalze
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer heterocyclischer Verfahren der allgemeinen Formel I :

Die neuen Verbindungen werden gemäss der Erfindung durch Umsetzung eines Halogen-Derivates der allgemeinen Formel II :
Het-Z, (II) in der Z Cl oder Br und Het einen, wie oben definierten heterocyclischen Rest bedeutet, mit 1- (3', 4'Methylendioxy-benzyl)-piperazin der Formel III :

Die Reaktion wird in einem polaren Lösungsmittel, wie Alkoholen mit hohem Siedepunkt, z. B. Butanol oder Pentanol, einem Amid, wie Dimethylformamid oder in einem nicht polaren Lösungsmittel, besonders einem aromatischen Kohlenwasserstoff, z. B. Benzol, Toluol oder Xylol, durchgeführt. Es ist vorteilhaft, bei einer Temperatur zwischen 80 und 140o C in Gegenwart eines Akzeptors für die Säure, die im Verlauf der Reaktion gebildet wird, zu arbeiten. Dieser Säureakzeptor kann ein Alkali- oder Erdalkalicarbonat oder-hydrogencarbonat, besonders Natrium- oder Kaliumcarbonat, Calciumcarbonat oder eine tertiäre organische Base, wie Dimethylanilin, Pyridin oder Triäthylamin, sein. Man kann auch in gleicher Weise falls gewünscht, diese Salze oder diese tertiären Basen durch einen Überschuss am monosubstituiertem Piperazin ersetzen.
Es liegt ferner im Rahmen der Erfindung, die erhaltenen neuen heterocyclischen Verbindungen, die schwache Basen sind, mit Säuren in ihre Additionssalze überzuführen. Diese Additionssalze können durch Einwirkung der neuen Verbindungen auf die Säuren in einem geeigneten Lösungsmittel, z. B.
Wasser oder mit Wasser mischbaren Alkoholen, erhalten werden. Als Säuren, die für die Bildung der Additionssalze geeignet sind, können Mineralsäuren wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, oder organische Säuren, wie Essigsäure, Propion-, Malein-, Fumar-, Wein-, Zitronen-, Oxal-, Benzoe-, Methansulfon-Säure usw. genannt werden.
Die neuen Verbindungen können gegebenenfalls durch physikalische Methoden, wie Destillation, Kristallisation, Chromatographie oder durch chemische Methoden, wie Zersetzung der Additionssalze durch alkalische Mittel, gereinigt werden.

Die erfindungsgemäss herstellbaren neuen Verbindungen haben interessante pharmakologische und therapeutische Eigenschaften und können besonders als periphere vasodilatorische und bronchodilatorische Mittel verwendet werden.
Die akute Toxizität wurde an Mäusen bestimmt, wobei eine LD50 bei intraperitonaler Verabreichung von 50 bis 1500 mg/kg bei oraler Verabreichung dieser Verbindungen von 1200 bis 3000 mg/kg gefunden wurde.
Bei der Verabreichung von 5 mg/kg der neuen Verbindungen durch intravenöse Perfusion beim Kaninchen wurde eine bedeutende Vergrösserung des Blutvolumens in der Pfote, die sich zwischen 20 und 225% bei den verschiedenen Verbindungen bewegt, festgestellt, dies beweist eine bemerkenswerte vasodilatorische Wirkung.
Die bronchodilatorische Wirksamkeit wurde nach der Methode von Konzett und Rossler [Arch. exp.
Path. und Pharm. 195, 71 (1940)] bestimmt. Es wurde gefunden, dass die neuen Verbindungen im Stande sind, den durch Histamin, Serotonin oder Acetylcholin verursachten Bronchospasmen beim Meerschweinchen entgegenzuwirken. Diese Spasmen werden durch intravenös verabreichte Dosen von 1 bis 5 mg/kg zu 300-100% gehemmt.
Diese Eigenschaften erlauben die therapeutische Verwendung dieser Verbindungen, besonders bei der Behandlung von peripheren Kreislaufstörungen, z. B. Venen- oder Arterienentzündungen, sowie akuter oder chronischer Atmungsinsuffizienz, besonders von Bronchialasthma.
In diesen Fällen können 2-4mal täglich 10-200 mg verabreicht werden.
Die Verbindungen können oral, rectal oder parenteral verabreicht werden, wobei der aktive Bestandteil mit pharmakologischen Trägern, wie destilliertem Wasser, Stärke, Talk, Laktose, Äthylcellulose oder Kakaobutter vermischt oder an diese gebunden sein kann. Die gewünschten pharmazeutischen Verabreichungsformen können Tabeletten, Dragées, Kapseln, Suppositoren, Injizierbare oder trinkbare Lösungen sein.
Die folgenden Beispiele dienen zur Erläuterung des erfindungsgemässen Verfahrens.
Die Schmelzpunkte wurden auf der Heizbank nach Koffler unter dem Mikroskop bestimmt.
Beispiel 1 : 1- (5'-Chlor-2'-pyrimidinyl-)-4- (3", 4"-methylendioxybenzylpiperazin.
Zu einer Lösung von 21 g 1- (3', 4'-Methylendioxybenzyl) -piperazin und 14, 9 g 2, 5-Dichlorpyrimidin in 150 cm3 Dimethylformamid (DMF) gibt man 28 g Kaliumcarbonat und erhitzt die Mischung 8 h auf 1300 C.. Nach Ablauf dieser Zeit werden die gebildeten Salze abfiltriert und das DMF unter vermindertem Druck abdestilliert. Der warme Rückstand wird mit 100 cm3 siedendem Wasser behandelt und unter starkem Rühren abgekühlt, wobei das Öl kristallisiert. Nach dem Trocknen wird aus Äthanol umkristallisiert. Man erhält 25 g weisser Kristalle mit einem Schmelzpunkt von 99 bis 1010 C.
Entsprechend dem Beispiel l erhält man folgende Verbindungen :

Beispiel 2 : 1- (4'-Pyrimidinyl) -4- (3", 4"-methylendioxybenzyl) -piperazin erhält man nach der Verfahrensweise des Beispiels 1 beim Arbeiten in Toluol unter Rückfluss in Gegenwart von Kaliumcarbonat, ausgehend von 4-Chlorpyrimidin, Dichlorhydrat, F. 192-198 C (Zersetzung).
Entsprechend dem Beispiel 2 erhält man folgende Verbindung : l- (2'-Amino-4'-pyrimidinyl)-4- (3", 4"- methylendioxybenzyl)-piperazin aus 2-Amino-4-chlorpyrimidin ; freie Base, F. 171 C.
Beispiel 3 : l- (4', 6'-Dimethoxy-2'-s-triazinyl)-4- (5", 4"-methylendioxybenzyl)-piperazin erhält man nach der Verfahrensweise des Beispiels 1 beim Arbeiten mit Benzol unter Rückfluss in Gegenwart von Kaliumcarbonat, ausgehend von 2-Chlor-4, 6-dimethoxy-s-triazin, freie Base, F. 102 C, Chlorhydrat, F. 250 C (Zersetzung).
