Verfahren zur Herstellung von Streptomycin- und Dihydrostreptomycin-Komplexsalzen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen kristallinischen Caleium- glucoheptonat-Komplexsalzen von Streptomycinsulfat und Dihydrostreptomycinsulfat.
Es ist bekannt, dass Streptomycin und Dihydro- streptomycin, in verhältnismässig hohen Dosen und über längere Zeitabschnitte verabreicht, neuro- toxische Erscheinungen verursachen, die ihre thera peutische Verwendung begrenzen. Es ist deshalb von grosser Bedeutung, Präparate herzustellen, die eine Erhöhung der Dosen und eine Verlängerung der Therapie gestatten.
Es wurde gefunden, dass, wenn man 2 Mol Strepto- mycin- oder Dihydrostreptomycinsulfat mit 3 Mol Caloiumglucoheptonat in Wasser versetzt, sich kein Calciumsulfat abscheidet, wie es in Anbetracht der niedrigen Wasserlöslichkeit dieses Salzes zu erwarten wäre. In Gegenteil bildet sich ein lösliches Komplex salz, das aus seiner wässrigen Lösung durch Zusatz eines mit Wasser mischbaren organischen Lösungs mittels, z. B. Methanol oder Aceton, isoliert werden kann.
Das erfindungsgemässe Verfahren zur Herstellung eines kristallinischen Calciumglucoheptonat-Komplex- salzes von Streptomycin- bzw. Dihydrostreptomycin- sulfat ist dadurch gekennzeichnet, dass man Strepto- mycin- oder Dihydrostreptomyoinsulfat mit Calcium- glucoheptonat in einem molaren Verhältnis von 2 Mol Streptomycin bzw.
Dihydrostreptomycin zu 3 Mol Calciumglucoheptonat in Wasser umsetzt, und aus der wässrigen Lösung des entstandenen Komplexsalzes dieses mit mindestens einem organischen, mit Wasser mischbaren Lösungsmittel ausfällt.
Als Ausgangsstoff kann eine technische Gluco- heptonsäure verwendet werden, die aus D-Glucose durch ihr Cyanhydrin dargestellt wurde und aus p-Gluco-D-,gulo-heptonsäure (a-D-Glucoheptonsäure) neben kleinen Mengen von D-Gluco-D-ido-hepton- säure (ss-D-Glucoheptonsäure) besteht.
Die Durchführung des erfindungsgemässen Ver fahrens kann folgendermassen erfolgen: Eine wässrige Lösung von 2 Mol Streptomycin- oder Dihydrostreptomycinsulfat und eine wässrige Lö sung von 3 Mol Calciumglucoheptonat werden mit einander vermischt, und die dabei entstandene Lösung wird im Vakuum bis zu einer annähernden Konzen tration von 1 Mol Komplexsalz je 6-8 Liter Wasser eingeengt, ohne 40 C zu überschreiten.
Zur konzen trierten Komplexsalzlösung werden dann 4 bis 6 Raumteile .eines wasserlöslichen organischen Lö sungsmittels, wie z. B. eines niederen aliphatischen Alkohols, Dioxans, Acetons oder deren Mischungen schnell zugegeben. Das Produkt scheidet sich ab und wird abgesaugt und im Vakuum getrocknet.
Abwechselnd kann man zu einer konzentrierten wässrigen Lösung von 3 Mol Calciumglucoheptonat eine Menge von 2 Mol Streptomycin- oder Dihydro- streptomycinsulfat zugeben.. Die Wassermenge ist in der Weise berechnet, dass am Ende eine Lösung von 1 Mol Komplexsalz in 6-8 Litern Wasser entsteht. Das Komplexsalz wird dann wie oben angegeben isoliert.
Es kann auch eine wässrige Lösung von Gluco- heptonsäure einer dichten wässrigen Suspension von Calciumhydroxyd bis zu neutralem pH (5 bis 7) zugegeben werden. Zur dabei entstandenen Lösung wird Streptomycin- oder Dihydrostreptomycinsulfat zugegeben.
Auch in diesem Falle wird die Wasser- menge in der Weise berechnet, dass eine Endkonzen- tration von 1 Mol Komplexsalz in 6-8 Litern Was ser entsteht. Die Isolierung wird wie oben beschrie ben ausgeführt.
Das nach dem erfindungsgemässen Verfahren dar gestellte Streptomycinglucoheptonat-Komplexsalz ist ein weisses, kristallinisches Pulver. Seine Brutto formel ist Cs4Ht62Ca3Nt40,4Ss, mit Molekulargewicht 2928,76. Der theoretische Streptomycingehalt ist ungefähr 397 y/mg. Der pH- Wert einer 200000 y/mg Streptomycin enthaltenden wässrigen Lösung ist 5,95 0,1.
Das Komplexsalz ist in Wasser sehr löslich, in niederen aliphatischen Alkoholen, Aceton und Dioxan fast unlöslich.
Das Dihydrostreptomycinglucoheptonat-Komplex- salz ist ebenfalls ein weisses, kristallinisches Pulver. Seine Bruttoformel ist Ce4HissCasNt40s4Ss, mit Molekulargewicht 2932,77.
Der theoretische Dihydrostreptomycingehalt ist ungefähr 398 y/mg. Der pH-Wert einer 200000 y/mg Dihydrostreptomycin enthaltenden wässrigen Lösung ist 6,0 0,1. Das Komplexsalz ist in Wasser sehr löslich, in niederen aliphatischen Alkoholen, Aceton und Dioxan fast unlöslich.
Die wässrigen Lösungen der neuen Komplexsalze färben sich nicht braun beim Stehen. Das unterschei- det sie von den üblichen wässrigen Lösungen von Streptomycin- bzw. Dihydrostreptomycinsulfat oder -hydröchlorid.
Es ist bekannt, dass die chronische Toxizität von Streptomycin und Dihydrostreptomycin durch schwere Schädigung des achten Gehirnnervenpaares gekenn zeichnet ist, mit Gleichgewichtsstörungen und Taub heit.
Die äusserst niedrige chronische Toxizität der Streptomycin- und Dihydrostreptomycinglucoheptonat- Komplexsalze wurde durch die von Ballard et a1., J. Amer. Pharmac. Assoc. 45, S. 181 (1956), beschrie bene Goldfisch-Methode untersucht.
Vier Gruppen von je 10 Goldfischen wurden in wässrige Lösungen von Streptomycin-, Dihydrostrepto- mycinsulfat, Streptomycin- und Dihydrostrepto- mycinglucoheptonat-Komplexsalz eingetaucht; eine fünfte Gruppe von 10 Goldfischen diente als Kon trolle.
Die folgende Tabelle gibt die mit Konzentrationen von 200, 500 und<B>1000</B> mg/1 der vier Substanzen in deionisiertem Wasser erhaltenen Ergebnisse. Wenn nicht anders angegeben, werden in der Tabelle die nach 10 Tagen beobachteten Werte wiedergegeben.

Diese gleichzeitige Verabreichung kann nicht dadurch erfol gen, dass man z. B. eine Lösung bereitet, die die allgemein verwendeten anorganischen Streptomycin- bzw. Dihydrostreptomycinsalze, wie z. B. das Sulfat, und ein wasserlösliches Calciumsalz, wie z. B. das Chlorid, enthält, da sich Calciumsulfat sofort ab scheiden würde.
Die neuen Streptomycin- und Dihydrostrepto- mycinglucoheptonat-Komplexsalze können allein oder in Verbindung mit anderen Streptomycin- oder Di- hydrostreptomycinsalzen, wie z. B. dem Sulfat, dem p-Aminosalicylat, dem Pantothenat und/oder dem Salz mit Brenztraubensäureisonicotinylhydrazon ver abreicht werden. In jedem Falle erniedrigen sie die chronische Toxizität dieser bekannten Salze.
Sie können auch mit anderen Calciumsalzen, mit anderen Antibiotika, wie z. B. Penicillinsalzen, und/ oder mit anderen pharmakologisch wirksamen Sub stanzen vereinigt werden. Ihren pharmazeutischen Präparaten können auch verdickende, stabilisierende, benetzende oder puffernde Stoffe zugesetzt werden.
In Anbetracht ihrer niedrigen Toxizität können die neuen Komplexsalze in Tagesdosen. verwendet werden, die vielgrösser als die gewöhnlichen Strepto- mycin- und Dihydrostreptomycin-Tagesdosen sind. Tagesdosen von z.
B. 5 g, die einem Gehalt von etwa 2 g Streptomycin oder Dihydrostreptomycin entspre chen, können an Menschen durch intramuskuläre Einspritzung abgegeben werden, ohne die neuro- toxischen Erscheinungen zu beobachten, die mit der Streptomycin- bzw. DihydrostreptomycinAbgabe ver bunden sind.
Die Komplexsalze sind auch zur Injektion in den Liquor cephalo-rachidianum geeignet, wobei in, phy siologischer Lösung gelöste Tagesdosen bis zu 20 mg verwendet werden können. Auch die intrapleurale Verabreichung erwies sich als sehr nützlich, und in physiologischer Lösung gelöste Tagesdosen bis zu 3-4 g verursachten keine toxischen Folgen.
Lösungen von 200-300 mg in 1-2 cm physio logischer Lösung wurden auch mit Erfolg als Aerosole verwendet. <I>Beispiel 1</I> Zu einer Lösung von 145,75 g Streptomycin- sulfat in<B>1000</B> cm3 Wasser wird eine Lösung von 147,13g Calciumglycoheptonat in 1000 cm3 Wasser zugefügt.
Die dabei entstandene, leicht opalisierende Lösung wird durch ein Seitz-Filter steril filtriert und auf dem Wasserbad bis zu einem Volumen von 700 cms eingeengt, ohne die Temperatur von 40 C zu überschreiten. Die Mischung wird auf Raumtempe ratur gekühlt, dann werden 3500 cm3 sterilisiertes Methanol schnell zugefügt, wobei sich weisse Kristalle abscheiden.
Die Masse wird 30 Minuten gerührt, dann wird der Niederschlag abgesaugt, mit kaltem sterili- siertem Methanol gewaschen und im Vakuum bei 40 C bis zu konstantem Gewicht :getrocknet. Aus beute 280 g (95,5 4/o der Theorie) Streptomycin- glucoheptonatKomplexsalz.
Aus der Analyse ergibt sich ein Streptomycin- Gehalt von 376 ",/mg, der zu 94,7 4/o der Theorie ent spricht. Auf dieser Basis gerechnet beträgt die Aus beute 90,5 4/u der Theorie.
<I>Beispiel 2</I> Zu einer Lösung von 147,13 g Calcium-Gluco- heptonat in 700 cm3 Wasser werden portionsweise unter Rühren 145,95 g Dihydrostreptomycinsulfat zugefügt. Die dabei entstandene klare Lösung wird durch ein Seitz-Filter steril filtriert, dann werden 4 Liter Dioxan zugegeben, wobei sich ein weisser Niederschlag bildet. Die Masse wird 30 Minuten ge rührt, dann wird der Niederschlag abgesaugt, mit Dioxan gewaschen und im Vakuum bei 40 C ge trocknet.
Ausbeute 284 g (974/a) Dihydrostrepto- mycinglucoheptonat-Komplexsalz.
Aus der Analyse ergibt sich ein Dihydrostrepto- mycingehalt von 382 y/mg, der zu 96 4/o der Theorie entspricht. Auf dieser Basis gerechnet, beträgt die Ausbeute 934/4 der Theorie.
<I>Beispiel 3</I> Zu einer Suspension von 22,2 g Caleiumhydroxyd in 50 cm3 Wasser wird unter Rührung eine Lösung von 135,6g Glucoheptonsäure in 750 cm3 Wasser schnell zugefügt, der endgültige pH-Wert der dabei entstandenen klaren Lösung ist 7,5. Zu dieser Lösung werden portionsweise 145,75 g Streptomycinsulfat zugegeben. Die entstandene Lö sung wird durch ein Seitz-Filter steril filtriert und mit 3500 cm3 Aceton verdünnt, wobei sich ein schwerer, weisser Niederschlag bildet.
Die Masse wird 30 Minuten gerührt, dann wird der Nieder schlag abgesaugt und im Vakuum bei 40 C getrock net. Ausbeute 275 g. Streptomycinglucoheptonat- Komplexsalz mit 385 y/mg Streptomycin-Gehalt (90,8% der Theorie).

On the contrary, a soluble complex salt forms, which means from its aqueous solution by adding a water-miscible organic solvent, z. B. methanol or acetone can be isolated.
The method according to the invention for the production of a crystalline calcium glucoheptonate complex salt of streptomycin or dihydrostreptomycin sulfate is characterized in that streptomycin or dihydrostreptomyoin sulfate is mixed with calcium glucoheptonate in a molar ratio of 2 moles of streptomycin or
