Description
Die Erfindung betrifft Monophosphorsäureester von 2,2'-Biphenolen der allgemeinen FormelThe invention relates to monophosphoric acid esters of 2,2'-biphenols of the general formula
OH 0-P(OH)2 OH O-P (OH) 2
in der X Wasserstoff, Chlor oder Brom und Y Chlor oder Brom bedeutet, sowie deren Alkalisalze.in which X denotes hydrogen, chlorine or bromine and Y denotes chlorine or bromine, and also alkali metal salts thereof.
The invention also relates to a process for the preparation of these monophosphoric acid esters is characterized in that in each case known per se, the corresponding biphenol with phosphoryl chloride in the presence of pyridine to form the cyclic phosphoric acid ester chloride, this means aqueous sodium hydroxide solution over the cyclic phosphate and then by means of concentrated 'Reacts hydrochloric acid and optionally the 2,2'-biphenol monophosphoric acid ester into an alkali salt converts.
This procedure is for the monophosphoric acid ester of 2,2'-methylenebis (3,4,6-trichlorophenol) in Dutch patent specification 128,693. In the production of the invention Monophosphoric acid ester has been found to give considerably better results than according to the Dutch Patent can be obtained because the reaction is carried out with phosphoryl chloride in the presence of pyridine will. Those mentioned above as intermediates in the preparation of the monophosphoric acid esters cyclic phosphates are also new compounds and can be isolated if desired.
und mitunter sogar deren Tod.The monophosphoric acids of the formula given have an anti-parasitic effect. For example, they are effective against liver fluke or protect against liver fluke infestation (fascioliase). However, they are also suitable for combating other parasites.
•, Fascioliase or liver fluke infection is a disease that affects sheep and cattle in particular, but also affects other warm-blooded animals and in some cases even humans. This disease is caused by the liver fluke (Fasciola hepatica), a parasite that thrives in the liver and bile ducts of infected organisms and causes damage there, which disrupts and even prevents the normal functioning of these organs.
The end effect is a more or less serious damage to the general welfare of the animals and sometimes even their death.
A number of means have been used to combat this disease, for example 2,2'-methylene-bis (3,4,6-trichlorophenol). as well as the monophosphoric acid ester of 2,2'-methylene-bis- (3,4,6-trichlorophenol) (see Dutch patent specification 128 693). Tnsbe ^ is liable for these connections special the disadvantage that their toxicity is relatively high, so that a need for well-acting and especially less toxic compounds. The monophosphoric acid esters according to the invention of 2,2'-biphenols have a pronounced Activity against fascioliase and protect against liver fluke infestation, these compounds being a better one Have solubility than the above-mentioned known compounds.
This better solubility has an advantageous effect on the processing of the compounds pharmaceutical preparations and when administering these preparations, in particular to sheep. The alkali salts of the new monophosphoric acid esters are useful in combating infections by liver fluke particularly useful in sheep due to their ready solubility in water at a physiological level usable pH.
The active compounds according to the invention are used to produce preparations with an antiparasitic effect processed into tablets, pills, capsules and potions.
. : v For this purpose, the active ingredients can be mixed with the usual pharmaceutical carriers, such as. B. talc, magnesium stearate, starch, lactose and physiologically acceptable ^ solvents. . : v
.Particularly effective are tablets that 'additionally to the monophosphoric esters according to the invention one equivalent of acidic sodium carbonate or Contain sodium bicarbonate. It is also possible to mix the active ingredients with feed or feed components. .
It has been found that in general a single dose of about 10 to about 30 mg / kg is suitable to a long-lasting reduction in the number of eggs appearing in the faeces of the infected animals achieve. Animal studies have also found that this dose is generally all mature Liver fluke kills.

In addition to the experiments mentioned above, the compounds according to the invention when carried out have been tested under conditions very different from normal, so that the results are only of relative value also made experiments with naturally infected animals. So compounds C and D became in in a comparative experiment on the same farm with two groups of 33 cattle. Since the Compound C according to the invention acts more strongly than the known compound D, if the first-mentioned compound was administered in a lower dose (12 mg / kg) than the latter compound (18 mg / kg). The results are summarized in the table below. In it is the E. p. G. value is the number of liver fluke eggs per gram of faeces.
