Verfahren zur Herstellung von Axerophten
Im Schrifttum sind verschiedene Versuche zur Synthese des Axerophtens (Desoxy-Vitamin A) dargelegt [vergl. O. Isler, Chimia 4 (1950), 107; J. G.
Baxter, Fortschritte der Chemie Organischer Naturstoffe Band 9 (1952), 78, sowie P. Karrer und J. Benz, Helv. Chim. Acta 31 (1948), 1048, und 32 (1949), 232]. Das in diesen Arbeiten als Axerophten bezeichnete Produkt wird als hellgelbes Öl beschrieben, das Absorptionsmaxima bei 331, 346 und 364 mjt zeigt.
Diese im Vergleich zum VitamSin-A- Alkohol unerwartet langwellige Absorption konnte erst später dahin aufgeklärt werden, dass das früher als Axerophten angesehen Produkt in Wirklichkeit das dem eigentlichen Axerophten I isomere Retro-Axerophten der Formel II ist:

Es war bei den erwähnten Syntheseversuchen also eine Verschiebung des konjugierten Systems von 5 Doppelbindungen erfolgt [vergl. P. Karrer und J. Kebrle, Helv. Chim. Acta 35 (1952), 2570, sowie H. O. Huisman u. Mitarb., Rec. Travaux Chimiques Pays-Bas, 71 (1952), 911].
Die Synthese des Axerophtens der Formel list erstmals in der Schweiz. Patentschrift Nr. 342946 beschrieben worden. Es zeigt, wie erwartet, ein im kürzeren Wellenbereich, nämlich bei etwa 320 my, liegendes Absorptionsmaximum.
Vorliegende Erfindung betrifft nun ein neues Ver- fahren zur Herstellung von Axerophten der Formel I, -welches dadurch gekennzeichnet ist, dass man eine Carbonylverbindung der Formel

umsetzen, wobei die R gleiche oder verschiedene aromatische Reste, vorzugsweise Benzolreste, be deuten.
Die unter a) bis e) genannten Phosphinylide lassen sich herstellen, indem man auf ein Triarylphosphin, vorzugsweise Triphenylphosphin, das betreffende Alkylhalogenid einwirken lässt und das entstandene quartäre Phosphoniumsalz mit halogenwasserstoffbindenden metallorganischen Verbindungen, wie Phenyl- oder Butyllithium, Natriumacetylid oder Alkylmagnesiumhalogeniden, oder mit Alkalimetallamiden oder -alkoholaten, behandelt.
Die Umsetzungen gemäss a) bis e) werden zweckmässig in Lösungs- oder Verdünnungsmitteln, die gegen die Phosphinylide indifferent sind, ausgeführt.
Genannt seien beispielsweise Ather, Tetrahydrofuran, Dimethyltetrahydrofuran, Dioxan, Benzol, Toluol, Xylol, Cyclohexan, Methylcyclohexan, Heptan, Isooctan, ferner Alkohole, Glykole, Acetonitril und Dimethylformamid. Die günstigen Temperaturen sind von Fall zu Fall verschieden; oft verläuft die Umsetzung bereits bei tiefen Temperaturen, z. B.
-20"C, in anderen Fällen wird sie durch Erwärmen gefördert.
Das so in jeweils einem einzigen Schritt und ohne Retro-Umlagerung erhältliche Axerophten kann aus den vom Phosphinoxyd abgetrennten Lösungen in üblicher Weise, z. B. durch Verteilung zwischen verschiedenen Lösungsmitteln, chromatographisch oder durch Destillation unter vermindertem Druck, in reiner Form gewonnen werden. Es siedet unter 0,01 Torr bei 140-145"C. Man erhält es im allgemeinen als öliges Gemisch verschiedener stereomerer Formen, die sich nach den in der Vitamin-A-Chemie üblichen Methoden, z. B. durch Behandeln mit Jod oder Säuren und bzw. oder Belichten, zu anderen stereomeren Formen bzw. Stereomerengemischen umlagern lassen.
Das reine all-trans-Axerophten bildet, aus Acetonitril umkristallisiert, gelbe Kristalle vom Schmp.
760 C und zeigt, in Tetrahydrofuran gelöst, eine intensive Absorptionsbande bei 325 m, (e = 50000). Die Absorptionskurve deckt sich praktisch vollständig mit der von Standard-Vitamin-A-Acetat. In Chloro formlösung gibt Axerophten mit Antimontrichlorid eine blauviolette Farbreaktion.
Das Axerophten der Formel I soll als Heilmittel verwendet werden, weil es beträchtliche, bisher unbekannte Vitamin-A-Wirksamkeit zeigt. Diese ist im Rattentest mindestens halb so gross, wie dies des Vitamins A selbst.
Die in den Beispielen genannten Teile sind Ge wichtsteile.
Beispiel I
Man stellt zunächst auf folgende Weise eine Lösung von 1- (Triphenylphosphinyliden -(2,6)-di- methyl-octatrien-(2,4,6) der oben unter a) angegebenen Formel her:
76 Teile 1 -Oxy-2,6-dimethyl-octatrien - (2,4,6) vom Kp. 80-82"C unter 0,04 Torr (das durch Reduzieren von 2,6-Dimethyl-octatrien-(2,4,6)-ai-(1) nach Meerwein-Ponndorf in sehr guter Ausbeute erhältlich ist) werden in 200 Teilen absolutem Äther gelöst und bei -5"C unter Rühren allmählich mit einer Lösung von 50 Teilen Phosphortribromid in 80 Teilen absolutem Ather versetzt. Nach dreistündigem Rühren giesst man auf Eis, trennt die Ätherschicht ab, wäscht sie mit Wasser, trocknet sie mit Calciumchlorid und verdampft dann den Äther bei vermindertem Druck.
Der Rückstand, der aus dem 1 -Brom-2,6-dimethyl-octatrien-(2,4,6) besteht, wird in 350 Teilen Dimethylformamid gelöst und mit 110 Teilen Triphenylphosphin versetzt. Nach 12stündigem Rühren gibt man zu der entstandenen Lösung des quartären Phosphoniumsalzes 70 Teile einer 300/obigen methanolischen Lösung von Natriummethylat und rührt weitere 30 Minuten.
Zu der so entstandenen roten Lösung des Phos phinylids lässt man unter Rühren eine Lösung von 67 Teilen p-Cyclocitral in 80 Teilen Dimethylformamid fliessen, wobei sich das Gemisch etwas erwärmt.
Man rührt weitere 12 Stunden bei gewöhnlicher Temperatur und 20 Minuten bei 600 C, worauf man abkühlt und mit Petroläther, in dem sich das Triphenylphosphinoxyd nicht löst, erschöpfend extrahiert. Die gelbe Petrolätherlösung wird mit Wasser gewaschen, über Natriumsulfat bei -5"C getrocknet und bei vermindertem Druck vom Lösungsmittel befreit. Der ölige Rückstand liefert bei der fraktionierten Destillation nach einem geringen Vorlauf 85 Teile Axerophten vom Kpo ot = 138-1 45o C. Beim Stehenlassen bei -5" C beginnt allmählich die all-trans-Form auszukristallisieren. Die gelben Kristalle werden nach Verreiben mit eiskaltem Methanol abgesaugt und aus Acetonitril umkristallisiert, worauf sie bei 760 C schmelzen. Die Ausbeute an all-trans-Form beträgt 32-35 Teile.
Aus den Mutterlaugen gewinnt man 50 Teile eines öligen Gemisches cis-trans-isomerer Formen, das die gleiche analytische Zusammensetzung besitzt wie die Kristalle.
Beispiel 2
Man stellt zunächst auf die folgende Weise eine
Lösung des oben unter Formel b) aufgeführten Ylids, 1 (Triphenylphosphinyiiden)-4-methyl-hexadien- (2,4) her:
110 Teile l-Oxy-4-methyl-hexadien-(2,4) vom Kpil = 86870 C, das in guter Ausbeute durch Reduzieren von 4-Methylhexadien-(2,4)-al-(1) nach Meerwein-Ponndorf erhältlich ist, werden in 250 Tel- len absolutem Äther gelöst und bei -5"C unter Rühren allmählich mit der Lösung von 110 Teilen Phosphortribromid in 150 Teilen absolutem Äther versetzt.
Man giesst nach dreistündigem Rühren auf Eis, wäscht und trocknet die Ätherschicht, verdampft den Äther und löst den Rückstand der aus dem 1-Brom-4-methylhexadien-(2,4) besteht, in 400 Teilen trockenem Tetrahydrofuran. Nach Zugabe von 250 Teilen Triphenylphosphin rührt man 12 Stunden bei gewöhnlicher Temperatur und saugt dann das in einer Ausbeute von 340 Teilen auskristallisierte quartäre Phosphoniumsalz (Sclrmp. 174-176" C) ab.
200 Teile dieses Salzes werden in 600 Teilen Dimethylformamid gelöst, worauf man unter Rühren 82 Teile einer 300/eigen methanolischen Natriummethylatlösung hinzufügt. Dabei entsteht unter Temperaturabfall eine orangerote Lösung von 1-(Triphenylphosphinyliden)-4-methyl-hexadien- (2,4).
Zu dieser Phosphinylidlösung gibt man 95 Teile jB-Jonon und rührt 16 Stunden bei gewöhnlicher Temperatur. Die erhaltene gelbe Lösung wird wie im Beispiel 1 aufgearbeitet und liefert 105 Teile Axerophten, davon 44 Teile all-trans-Form.
Beispiel 3
Man stellt zunächst eine ätherische Lösung von 1-(Triphenylphosphinyliden)-4-methyl-hexadien- (2,4) her, indem man eine Aufschlämmung von 44 Teilen des nach Beispiel 2, Absatz 1, hergestellten 4-Methylhexadien- (2,4) -yl- (1) -triphenylphosphoniumbromids in 300 Teilen absolutem Äther allmählich mit einer absolut-ätherischen Lösung von n-Butyllithium, die einem Lithiumgehalt von 0,69 Teilen entspricht, versetzt und 4 Stunden bei gewöhnlicher Temperatur unter Luftabschluss rührt.
Zu dieser Phosphinylidlösung lässt man allmählich eine Lösung von 19 Teilen ss-Ionon in 100 Teilen absolutem Äther fliessen. Nach zweistündigem Rühren saugt man das ausgefallene Triphenylphosphinoxyd ab, verdampft den Äther und destilliert den Rückstand unter vermindertem Druck. Man erhält 22 Teile Axerophten.
Beispiel 4
Man stellt zunächst eine benzolische Lösung von 1 (Triphenylphosphinyliden)-4-methyl-hexadlen- (2,4) her, indem man in eine Suspension von 44 Teilen des nach Beispiel 2, Absatz 1 hergestellten 4-Methyl hexadien-(2,4) -yl - (1) - triphenylphosphoniumbromids in 200 Teilen trockenem Benzol 40 Teile einer
300/oigen benzolischen Suspension von fein gepul vertem Natriumamid einträgt und 50 Stunden unter
Luftausschluss bei gewöhnlicher Temperatur rührt.
In der unter Feuchtigkeitsausschluss filtrierten orangeroten Lösung des Phosphinylids gibt man 19 Teile B-Jonon, worauf man 20 Stunden bei gewöhnlicher Temperatur rührt. Dann wird das Benzol unter vermindertem Druck abgedampft und der Rückstand mit Petroläther ausgezogen, wobei das Triphenylphosphinoxyd ungelöst bleibt. Die gelbe petrolätherische Lösung wird 8 Stunden bei - 50 C mit Natriumsuifat getrocknet. Man erhält 13 Teile Axerophten.
Beispiel 5
Man stellt zunächst eine ätherische Lösung von Äthylidentriphenylphosphin (s. obige Formel unter d) her, indem man zu einer Aufschlämmung von 40 Tei- len Athyl-triphenyl-phosphoniumbromid in 100 Teilen absolutem Tetrahydrofuran eine 0,7 Teile Lithium enthaltende absolut-ätherische Lösung von n-Butyllithium hinzusetzt und 6 Stunden unter Luftausschluss rührt.
Zu der tiefgelben Lösung gibt man eine Lösung von 25 Teilen 8-[(2', 6', 6'-Trimethylcyclohexen-(1')- yl-(1')]-6-methyl-octatrien-(3, ,5,7)-on-(2), das nach den Angaben von Arens und van Dorp (s. Recueil des Travaux Chimiques Pays-Bas, 65, 1946, 338) erhältlich ist, in 50 Teilen absolutem Tetrahydrofuran. Nach 12stündigem Rühren bei gewöhnlicher Temperatur wird wie im Beispiel 4 aufgearbeitet.
Man erhält 14 Teile Axerophten.
Beispiel 6
Man stellt zunächst eine Lösung des oben unter e) angegebenen ss-Jonyliden-äthyliden-triphenylphos- phins her, indem man 44 Teile ss-Jonylidenäthanol wie im Beispiel 1 mit der berechneten Menge Phosphortribromid in absolutem Äther bei - 5o C umsetzt, das erhaltene ss-Jonylidenäthylbromid in 150 Teilen Dimethylformamid mit einer Lösung von 40 Teilen Triphenylphosphin in 150 Teilen Dimethylformamid 48 Stunden rührt, dann bei vermindertem Druck ungefähr 100 Teile Dimethylformamid abdestilliert und schliesslich zu der braungelben Lösung von ss-Jonylidenäthyltriphenylphosphoniumbromid 20 Teile einer 300/eigen methanolischen Natriummethylatlösung hinzufügt.
Zu der braunroten Lösung des Phosphinylids gibt man 10 Teile Tiglinaldehyd und rührt 8 Stunden bei gewöhnlicher Temperatur. Dann arbeitet man wie im Beispiel 1 auf. Die Ausbeute an Axerophten beträgt 9 Teile.

Various attempts to synthesize axerophthalene (deoxy-vitamin A) are presented in the literature [cf. O. Isler (1950) Chimia 4: 107; J. G.

The product referred to as axerophten in this work is described as a light yellow oil, which shows absorption maxima at 331, 346 and 364 mjt.
This unexpectedly long-wave absorption compared to the VitamSin-A alcohol could only be clarified later to the effect that the product previously regarded as axerophten is in fact the retro-axerophten of formula II isomeric to the actual axerophten I:

The phosphinylides mentioned under a) to e) can be prepared by allowing the alkyl halide in question to act on a triarylphosphine, preferably triphenylphosphine, and the resulting quaternary phosphonium salt with organometallic compounds which bind hydrogen halide, such as phenyl- or butyllithium, sodium acetylide or alkylmagnesium halides, or with alkali metal halides or alcoholates.
The reactions according to a) to e) are expediently carried out in solvents or diluents which are indifferent to the phosphinylides.
Examples include ethers, tetrahydrofuran, dimethyltetrahydrofuran, dioxane, benzene, toluene, xylene, cyclohexane, methylcyclohexane, heptane, isooctane, and also alcohols, glycols, acetonitrile and dimethylformamide. The favorable temperatures vary from case to case; the implementation often takes place at low temperatures, e.g. B.
-20 "C, in other cases it is promoted by heating.
The axerophten thus obtainable in a single step and without retro-rearrangement can be prepared from the solutions separated from the phosphine oxide in the usual way, e.g. B. can be obtained in pure form by partitioning between different solvents, chromatographically or by distillation under reduced pressure. It boils below 0.01 torr at 140-145 "C. It is generally obtained as an oily mixture of various stereomeric forms which can be obtained by the methods customary in vitamin A chemistry, e.g. by treatment with iodine or acids and / or exposure to rearrange to other stereomeric forms or stereomeric mixtures.
The pure all-trans-axerophten forms, recrystallized from acetonitrile, yellow crystals of mp.
760 C and, when dissolved in tetrahydrofuran, shows an intense absorption band at 325 m (e = 50,000). The absorption curve is practically completely identical to that of standard vitamin A acetate. In chloroform solution, axerophten gives off a blue-violet color reaction with antimony trichloride.
The axerophten of formula I is intended to be used as a remedy because it shows considerable, previously unknown vitamin A activity. In the rat test, this is at least half as large as that of vitamin A itself.
The parts mentioned in the examples are parts by weight.
Example I.
A solution of 1- (triphenylphosphinylidene- (2,6) -dimethyl-octatriene- (2,4,6) of the formula given above under a) is first prepared in the following manner:
76 parts of 1-oxy-2,6-dimethyl-octatriene- (2,4,6) with a b.p. 80-82 "C under 0.04 torr (obtained by reducing 2,6-dimethyl-octatriene- (2, 4,6) -ai- (1) according to Meerwein-Ponndorf is obtainable in very good yield) are dissolved in 200 parts of absolute ether and a solution of 50 parts of phosphorus tribromide in 80 parts of absolute ether is gradually added at -5 "C with stirring . After stirring for three hours, it is poured onto ice, the ethereal layer is separated off, washed with water, dried with calcium chloride and then the ether is evaporated off under reduced pressure.
The residue, which consists of 1-bromo-2,6-dimethyl-octatriene- (2,4,6), is dissolved in 350 parts of dimethylformamide, and 110 parts of triphenylphosphine are added. After stirring for 12 hours, 70 parts of a 300 / above methanolic solution of sodium methylate are added to the resulting solution of the quaternary phosphonium salt and the mixture is stirred for a further 30 minutes.
A solution of 67 parts of p-cyclocitral in 80 parts of dimethylformamide is allowed to flow into the resulting red solution of phosphinylid with stirring, the mixture heating up somewhat.
The mixture is stirred for a further 12 hours at normal temperature and 20 minutes at 600 ° C., after which it is cooled and extracted exhaustively with petroleum ether, in which the triphenylphosphine oxide does not dissolve. The yellow petroleum ether solution is washed with water, dried over sodium sulfate at -5 ° C. and freed from the solvent under reduced pressure. In the fractional distillation, the oily residue yields 85 parts of axerophtene of 138-145 ° C. after a small forerun When left to stand at -5 "C, the all-trans form gradually begins to crystallize out. After trituration with ice-cold methanol, the yellow crystals are filtered off with suction and recrystallized from acetonitrile, whereupon they melt at 760.degree. Example 2

After adding 250 parts of triphenylphosphine, the mixture is stirred for 12 hours at normal temperature and the quaternary phosphonium salt (mp 174-176 ° C.) which has crystallized out in a yield of 340 parts is then filtered off with suction.
200 parts of this salt are dissolved in 600 parts of dimethylformamide, whereupon 82 parts of a 300% methanolic sodium methylate solution are added with stirring. As the temperature drops, an orange-red solution of 1- (triphenylphosphinylidene) -4-methyl-hexadiene- (2,4) is formed.
