Verfahren zur Herstellung von neuen Derivaten der Essigsäure Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Derivaten der Essigsäure, die als geriatrische Arzneimittel zur Verhütung und Behandlung von senilen Degenerationen verwendbar sind.
Es sind verschiedene senile Degenerationserscheinungen bekannt, wie z. B. Arteriosklerose, erhöhter Blutdruck sowie Leberfunktionsstörungen, beispielsweise Lebercirrhose, denen ältere Personen unterworfen sind. In neuerer Zeit ist nun nach chemischen Verbindungen gesucht worden, die zur Verhütung und Behandlung solcher senilen Degenerationserkrankun- gen verwendet werden könnten. So ist z. B. berichtet worden, das Phenyl- oder Biphenylylbuttersäure und deren Salze den Cholesteringehalt des Blutes herabsetzen [siehe z. B.

Es wurde nun gefunden, dass man durch Umsetzung von a-Phenyl- oder a-Biphenylyl-buttersäure mit Aminosäuren oder Aminozuckern zu neuen Verbindungen gelangt, die auf den Lipoidstoffwechsel eine normalisierende Wirkung ausüben, indem sie den Cholesteringehalt des Blutes herabsetzen, und überdies regulierend auf die Leberfunktionen einwirken. Diese neuen Verbindungen sind deshalb zur Verhütung und Behandlung von senilen Degenerationen, wie z. B. Arteriosklerose, Lebercirrhose usw., verwendbar.

Zur Umsetzung mit a-Phenyl- oder a-Biphenylyl- buttersäure können verschiedene Aminosäuren oder Aminozucker verwendet werden, z. B. Methionin, Taurin usw., die, wie angenommen wird, an den Leberfunktionen beteiligt sind und am Lipo.idstoff- wechsel teilnehmen. Als Aminozucker kann man z. B. Glucosamin, Chondrosamin usw., von denen erwartet werden kann, dass sie senile Degenerationserscheinungen verhüten, verwenden.
Einige der nach dem erfindungsgemässen Verfahren hergestellten N-[a-Phenyl- (oder a-Biphenylyl)- butyroyl]-aminosäuren bzw. -aminozucker wurden in Tierversuchen mit Kaninchen geprüft. Bei diesen Prüfungen zeigte es sich, dass diese Verbindungen tatsächlich den Cholesteringehalt des Blutes herabsetzen und die Arteriosklerose in der Aorta verhindern.
Bei diesen Versuchen wurden unter anderem N-(a-Phenyl-butyroyl)-DL-methionin, N-(a-Phenyl- butyroyl)-taurin und N - (a - Phenyl - butyroyl) - D - glucosamin geprüft.

Für je eine der zu prüfenden Verbindungen wurden 10 männliche Kaninchen mit einem Körpergewicht von etwa 2,5 kg verwendet, denen mit der Nahrung täglich 7 cm-3 einer Mischung von wasserfreiem Lanolin und Baumwollsamenöl (2: 1) sowie 0,5 g der zu prüfenden Verbindung verfüttert wurden. Den Vergleichstieren wurde mit der Nahrung nur Lanolin verfüttert.
Die Versuche wurden während 16 Wochen durchgeführt, wobei Messungen des Cholesterinspiegels im Blut nach 5, 10, 14 bzw. 16 Wochen vorgenommen wurden. Die Verabreichung von N-(a-Phenyl-butyroyl)-D-glucosamin wurde von der 14ten Woche an unterbrochen.
Die erzielten Resultate sind in der Tabelle I zusammengefasst.

Am Schluss der Versuche wurden die Tiere getötet und seziert. Die Tiere wurden hinsichtlich des arteriosklerotischen Zustandes der Aorta verglichen. Die makroskopische Beobachtung der Sklerose in der Nähe des Aortenbogens bei 6 Tieren je einer Gruppe ergab die in der Tabelle 1I zusammengefassten Resultate.

In der obigen Tabelle bedeuten -:
- @- + eine wesentliche Veränderung, + + eine verhältnismässig starke Veränderung, -i- eine schwache Veränderung, + eine sehr schwache Veränderung und - keine Veränderung.
Die Toxizität der geprüften Verbindungen wurde an Mäusen eines durch Züchten von (aus Deutschland in Japan eingeführten) Albinomäusen in geschlossener Kolonie erhaltenen homogenen Stammes geprüft. Die Dosis letalis LD.o überstieg 1000 mg/kg und betrug in einigen Fällen mehrere -/kg.
Wie aus der Formel I hervorgeht, sind die neuen Verbindungen Säureamide. Sie lassen sich nach den bekannten allgemeinen Methoden der Darstellung von Säureamiden herstellen. So kann man z. B. a-Phenyl- oder a-Biphenylylbuttersäure in ein reaktionsfähiges Derivat überführen und das letztere mit einer Aminosäure oder einem Ester derselben oder mit einem Aminozucker zur Reaktion bringen. Als reaktionsfähige Derivate kann man Säurehalogenide, Säureanhydride oder Ester verwenden.
Die Reaktion zwischen diesen reaktionsfähigen Derivaten und den Aminosäuren bzw. den Estern derselben oder den Aminozuckern wird zweckmässigerweise in einem wässerigen Lösungsmittel in Gegenwart eines säurebindenden Mittels durchgeführt. Als säurebindendes Mittel wird mit Vorteil eine anorganische basische Verbindung, z. B. ein Hydroxyd oder Carbonat eines Alkali- oder Erdalkalimetalls verwendet. An Stelle von reaktionsfähigen Derivaten der a-Phenyl- oder a-Biphenylyl-buttersäure kann man auch die freie Säure oder deren Alkalimetallsalze verwenden.
In diesem Fall ist es zweckmässig, die freie Säure oder deren Salz mit Aminosäureestern oder Aminozuckern in einem zweckentsprechenden Lösungsmittel in Gegenwart eines Katalysators umzusetzen. Als Lösungsmittel kann man z. B. Benzol, Toluol oder Tetra- hydrofuran verwenden.
Werden Aminosäuren in Form ihrer Ester verwendet, so sind die Reaktionsprodukte häufig ebenfalls Ester, die sich jedoch in üblicher Weise durch alkalische Hydrolyse in freie Säuren oder in deren Alkalimetall- oder Erdalkalimetallsalze überführen lassen.
In den folgenden Ausführungsbeispielen bedeuten Teile Gewichtsteile.

Beispiel 1 Einer Lösung von 15 Teilen DL-Methionin in 60 Teilen 2n Natronlauge werden unter Kühlen und unter kräftigem Rühren tropfenweise 19 Teile a- Phenyl-butyroylchlorid zugegeben. Gleichzeitig wird tropfenweise 2n Natronlauge zugesetzt, um die Reaktionslösung während der Reaktion alkalisch zu halten. Die Lösung, die zu Beginn der Reaktion trübe ist, wird im Verlaufe von etwa 3 Stunden klar. Beim Ansäuern der Lösung mit Salzsäure scheidet sich eine ölige Substanz ab, die beim Kühlen erstarrt.
Die feste Substanz besteht aus rohem N-(a-Phenyl- butyroyl)-DL-methionin. Durch Umkristallisieren dieses Produktes aus verdünntem Äthanol erhält man kleine Nadeln vom Smp. 129-130 C. Die Ausbeute beträgt 25 Teile. Das Produkt ist in wässerigen Säuren und in Wasser unlöslich, hingegen in wässerigen Alkalien, Alkohol, Aceton usw. leicht löslich.
Analyse: für Ci,,H"1NO3S: berechnet: C 61,00 %, H 7,16 %, N 4,74 0/0 gefunden: C 60,98 0/0, H 7,18 %, N 4,92 0/0 Beispiel 2 Man löst 6 Teile Taurin in 10 Teilen 5n Natronlauge und kühlt die Lösung unter kräftigem Rühren ab. Der Lösung werden dann tropfenweise 9 Teile ,-c-Phenyl-butyroylchlorid zugegeben.
Gleichzeitig gibt man dem Gemisch tropfenweise 5 Teile 5n Natronlauge zu, um die Reaktionslösung während der Reaktion alkalisch zu halten. Nachdem die durch das Chlorid verursachte Trübung verschwunden ist, wird die Lösung unter vermindertem Druck nahezu bis zur Trockne eingeengt. Man gibt dem Rückstand Alkohol zu, um das Natriumchlorid abzuscheiden, und dampft die alkoholische Lösung unter alkalischen Bedingungen bei -einem pH von 8-9 zur Trockne ein. Durch Umkristallisieren des Rückstandes aus einer kleinen Menge Wasser erhält man das Natriumsalz von N(a-Phenyl-butyroyl)-taurin mit 2 Mol Kristallwasser in Form von Schuppen, die man zwecks Entfernung des Kristallwassers trocknet.
Das getrocknete Produkt schmilzt bei 210'C und gibt eine negative Ninhydrinreaktion.
Analyse: für C12H"N04SNa - 2H20: berechnet: C 43,760/ü" H 6,001/o gefunden: C 43,5711/9, H 5,8811/o Beispiel 3 Einer Lösung von 11 Teilen D-Glucosamin- hydrochlorid in 20 Teilen 5n Natronlauge werden unter Rühren und Kühlen 9 Teile a-Phenyl-butyroyl- chlorid zugesetzt. Die Reaktionsmischung wird allmählich klar. Es bildet sich ein weisses öliges oder halbfestes Produkt, das später erstarrt.
Das erhaltene Produkt wird aus 90%igem Alkohol umkristallisiert. Man erhält auf diese Weise 12 Teile N-(a-Phenyi- butyroyl)-D-glucosamin in Form kleiner Nadeln vom Smp. 172 C.
Analyse: für C16H2306N - H20: berechnet: C 56,000/u, H 7,30%, N 4,1011/o gefunden: C 56,4511/o, H 7,381/o, N 3,86% Beispiel 4 Einer aus 1,6 Teilen a-Phenyl-buttersäure, 0,9 Teil Triäthylamin und 30 Teilen wasserfreiem Benzol bestehenden Lösung wird .eine Lösung von 0,7 Teilen Äthyldichlorphosphit

in 10 Teilen wasserfreiem Benzol zugesetzt.
Das ausgeschiedene Triäthylamin-hydrochlorid wird abfiltriert. Das Filtrat wird mit 1,6 Teilen DL-Methionin- methylester versetzt, worauf man das Gemisch während 30 Minuten unter Rückfluss zum Sieden erhitzt und anschliessend über Nacht stehen lässt. Die Benzollösung wird mit verdünnter Alkalilösung gewaschen, worauf das Lösungsmittel abdestilliert wird. Es bleibt der Äthylester von N-(a-Phenyl-butyroyl)-DL- methionin als Rohprodukt zurück, das auf dem Wasserbad mit einer verdünnten Natriumhydroxydlösung erhitzt wird, bis die Lösung klar wird.
Durch Ansäuern der Lösung mit Salzsäure wird ein öliges Produkt ausgeschieden. Durch Umkristallisieren dieses Produktes aus verdünntem Alkohol erhält man 2,5 Teile N-(a-Phenyl-butyroyl)-DL-methionin vom Smp. 128-130 C.
Beispiel 5 Einer aus 1,6 Teilen a - Phenyl - buttersäure, 1,6 Teilen DL-Methionin-äthylester und 30 Teilen Tetrahydrofuran bestehenden Lösung gibt man 3 Teile N,N'-Dicyelohexylcarbodiimid zu und lässt das Gemisch während 5 Stunden bei Raumtemperatur stehen. Man extrahiert die Reaktionslösung mit Essig- säu.reäthylester und destilliert den letzteren aus dem Extrakt ab. Man erhitzt den Rückstand mit verdünnter Natriumhydroxydlösung auf dem Wasserbad, bis die Lösung klar wird.
Nach Abkühlung wird die Lösung mit verdünnter Salzsäure angesäuert. Es scheidet sich dabei eine ölige Substanz ab, die nach kurzer Zeit fest wird. Durch Umkristallisieren des erhaltenen Produktes aus verdünntem Äthanol erhält man 2,1 Teile N-(a-Phenyl-butyroyl)-DL-methionin, Smp. 129 C.
Beispiel 6 Einer Lösung von 2,4 Teilen Taurin in 5 Teilen 5n Natronlauge wird tropfenweise unter Kühlen und Rühren eine Lösung von 5 Teilen p-Biphenylyl- butyroylchlorid in 70 Teilen Äther zugesetzt. Gleichzeitig werden 5 Teile 5n Natronlauge zugesetzt, um zu verhindern, dass die Lösung sauer wird.
Während der Reaktion scheiden sich in der wässerigen Schicht Kristalle ab. Im Verlaufe von 3 Stunden entsteht ein Kristallbrei. Diesem werden 20 Teile Wasser zugesetzt, worauf das Gemisch

unter vermindertem Druck zur Trockne eingedampft wird, um rohe Kristalle zu erhalten. Durch Umkri- stallisieren des erhaltenen Produktes aus einer kleinen Menge Wasser erhält man 5,5 Teile des Natriumsalzes von N-(a-p-Biphenylyl-butyroyl)-taurin vom Smp. 218 C. Das Produkt gibt eine negative Nin- hydrinreaktion.
Analyse: für C"HzoNO4SNa - H20: berechnet: C 55,80 %, H 5,72 %, N 3,610/0 gefunden: C 55,99 %, H 5,81 %, N 3,55 % Beispiel 7 Einer Lösung von 2,9 Teilen DL-Methionin in 10 Teilen 2n Natronlauge wird tropfenweise unter Kühlen und unter kräftigem Rühren eine Lösung von 5 Teilen p-Biphenylyl-butyroylchlorid in 50 Teilen Benzol zugesetzt.
Gleichzeitig werden 10 Teile 2n Natronlauge zugesetzt, um das Reaktionsgemisch alkalisch zu halten. Nach einer Reaktionszeit von 4 Stunden wird das Benzol abgetrennt und die wässerige Schicht mit verdünnter Salzsäure angesäuert, um eine ölige Substanz abzuscheiden. Diese Substanz wird nach kurzer Zeit fest. Man kristallisiert das erhaltene Produkt aus verdünntem Äthanol, um 4,5 Teile N-(a-p-Biphenylyl-butyroyl)-DL-methionin in Form kleiner farbloser Nadeln vom Smp. 138 < IC zu erhalten. Dieses Produkt ist in organischen Lösungsmitteln, wie z. B. Äthanol, Aceton und Benzol, löslich, hingegen in Wasser unlöslich.
Analyse: für C21H2503NS: berechnet: C 67,89 0/0, H 6,78 %, N 3,77 0/0 gefunden: C 67,90%, H 6,90%, N 4,08 0/0 Beispiel 8 Einer Lösung von 4,2 Teilen D-Glucosamin in 5 Teilen 5n Natronlauge werden 70 Teile einer ätherischen Lösung von 5 Teilen p-Biphenylyl- butyroylcWorid zugesetzt.
Gleichzeitig werden 5 Teile 5n Natronlauge zugesetzt, um das Reaktionsgemisch alkalisch zu halten. Nachdem die Lösung während 8 Stunden gerührt worden ist, wird die ätherische Schicht abgetrennt und das feste Produkt, das sich in der wässerigen Schicht gebildet hat, abfiltriert. Durch Umkristallisieren des erhaltenen Produktes aus 99%igem Äthanol erhält man 2,5 Teile N-(a-p-Bi- phenylyl-butyroyl)-D-glucosamin in Form von farblosen kleinen Nadeln vom Smp. 187 C.

It has now been found that the reaction of a-phenyl- or a-biphenylyl-butyric acid with amino acids or amino sugars leads to new compounds which have a normalizing effect on the lipoid metabolism by reducing the cholesterol content of the blood and, moreover, regulating it affect the liver functions. These new compounds are therefore used in the prevention and treatment of senile degenerations such as e.g. B. atherosclerosis, cirrhosis of the liver, etc., can be used.

For each of the compounds to be tested 10 male rabbits with a body weight of about 2.5 kg were used, to which 7 cm-3 of a mixture of anhydrous lanolin and cottonseed oil (2: 1) and 0.5 g of the diet were added daily test compound were fed. The comparison animals were fed only lanolin with their food.
The experiments were carried out for 16 weeks, with measurements of the cholesterol level in the blood being made after 5, 10, 14 and 16 weeks, respectively. The administration of N- (α-phenyl-butyroyl) -D-glucosamine was interrupted from the 14th week.

At the end of the experiments, the animals were killed and dissected. The animals were compared with regard to the arteriosclerotic condition of the aorta. The macroscopic observation of the sclerosis in the vicinity of the aortic arch in 6 animals in each group gave the results summarized in Table 1I.

The toxicity of the tested compounds was tested on mice of a homogeneous strain obtained by breeding albino mice (imported from Germany to Japan) in a closed colony. The lethal dose LD.o exceeded 1000 mg / kg and in some cases was several - / kg.
As can be seen from the formula I, the new compounds are acid amides. They can be produced by the known general methods of preparing acid amides. So you can z. B. convert a-phenyl- or a-biphenylylbutyric acid into a reactive derivative and react the latter with an amino acid or an ester of the same or with an amino sugar. Acid halides, acid anhydrides or esters can be used as reactive derivatives.
The reaction between these reactive derivatives and the amino acids or their esters or the amino sugars is expediently carried out in an aqueous solvent in the presence of an acid-binding agent. As an acid-binding agent, an inorganic basic compound, e.g. B. a hydroxide or carbonate of an alkali or alkaline earth metal is used. Instead of reactive derivatives of α-phenyl- or α-biphenylyl-butyric acid, it is also possible to use the free acid or its alkali metal salts.
