Pour l'usage pharmaceutique, les phénothiazines obtenues par le procédé selon l'invention sont utili- sées sous forme de sels thérapeutiquement accepta- bles, tels que:
chlorhydrate, phosphate, nitrate, sul fate, maléate, fumarate, citrate, tartrate, oxalate, méthanesulfonate, éthanedisulfonate ou de composés d'ammonium quaternaire thérapeutiquement accep tables, tels que chloro-, bromo- ou iodométhylate ou -éthylate,
chloro- ou bromobenzylate ou -allylate. Dans les exemples qui suivent, les points de fusion, sauf indication contraire, ont été déterminés au banc Kofler.
Les exemples ci-après illustrent le procédé selon l'invention <I>Exemple 1</I> On dissout 0,80 g de cyano-3-(méthylamino-3'- méthyl-2'-propyl-l')-10-phénothiazine base, libérée à partir du maléate correspondant dans 30 cm3 de méthanol avec 3 em3 d'acide chlorhydrique aqueux normal,
5 cm3 d'une solution aqueuse de formol à 30 % et 0,40 g de chlorure d'aluminium cristallisé. On ajoute 0,10 g de catalyseur au platine selon Adams et agite à température ordinaire sous une faible surpression d'hydrogène.
Quand la quantité théorique d'hydrogène a été absorbée, on filtre le catalyseur, évapore le solvant, reprend par de l'eau. La solution aqueuse est ensuite alcalinisée, on extrait à l'éther et sèche la solution éthérée.
Par évaporation de l'éther, il reste 0,8 g d'un résidu huileux qui donne en solution éthanolique avec de l'acide maléi que 0,65 g d'un maléate présentant les caractéristi- ques suivantes<B>:
</B> P.F. = 200-204 . La base cyano-3- (diméthylamino - 3' - méthyl - 2'- propyl - l')-10-phéno- thiazine libérée à partir de ce maléate est résineuse ;
elle présente le pouvoir rotatoire suivant: [a] D = + 16 (c = 41%, chloroforme).
Le maléate de la phénothiazine de départ peut être préparé comme suit On dissout 41 g de cyano-3-(N-formyl-N-méthyl- amino-3'-méthyl-2'-propyl-l')-10-phénothiazine opti- quement active dans 410cm3 de méthanol. On ajoute 82 cm3 d'acide chlorhydrique 4 N et chauffe pendant 5 heures à reflux. On évapore ensuite le méthanol et dilue avec 200 cm3 d'eau.
On extrait à l'éther puis alcalinise la solution aqueuse restante avec un excès de carbonate de potassium. Il se sépare une huile qu'on extrait à l'éther.
Par évaporation de l'éther, il reste 11,5 g d'une huile foncée qu'on purifie par pas sage sur colonne d'alumine en solution benzénique puis cristallisation du maléate dans l'acétate d'éthyle on obtient ainsi le maléate de cyano-3-(méthylamino- 3'-méthyl-2'-propyl-l')-10-phénothiazine qui présente les caractéristiques suivantes: P.F. =l70 [10]D = -12,70 (c = 1,2 0/0 ; chloroforme).
La cyano-3-(N-formyl-N-méthylamino-3'-méthyl- 2'-propyl-l')-10-phénothiazine optiquement active est obtenue par condensation sur la cyano-3-phénothi- azine sodée dans le xylène à reflux du N-formyl-N- méthylamino-3-méthyl-2-chloro-l-propane de P.E.23 = 141-143.
Cet amide halogéné est obtenu par action du chlorure de thionyle, au sein du chloro forme, en présence de pyridine anhydre, sur le N- formyl-N-méthylamino-3-méthyl-2-propanol-1 de P.E.o,O = 124-127 . L'amide-alcool est obtenu par action du formamide à reflux sur le méthyl- amino-3-méthyl-2-propanol de P.E.1g = 80-890 [10]D =-3,50 (substance pure).
Cet amino-alcool enfin est obtenu par réduction à l'hydrure de lithium- aluminium du monométhylamino-3-méthyl-2 pro pianate de benzyle qui présente un pouvoir rotatoire [a]20 = + 18,40 (c = 5 0/0, CHC13). L'es ter optiquement actif est obtenu par résolution du produit racémique à l'aide d'acide camphosulfo- nique (acide de Reyçhler)
. Le produit racémique est obtenu par transestérification die l'ester méthylique connu.
<I>Exemple 2</I> En opérant comme dans l'exemple précédent, mais au départ de maléate de cyano-3-(méthylamino- 3'-méthyl-2'-propyl-l')-10-phénothiazine, on obtient la cyano- 3 - (diméthylamino - 3'-méthyl-2'-propyl-l')- 10-phénothiazine base lévogyre de pouvoir rotatoire suivant [U] <B>D</B> =-160 (c=4%; chloroforme).
Le maléate de départ peut être obtenu de la même façon que le maléate de cyano-3-(méthyl- amino-3'-méthyl-2'-propyl-l')-10-phénothiazine dé crit à l'exemple précédent, mais au départ de méthyl- amino-3-méthyl-2-propionate de benzyle lévogyre, dans le chloroforme.
Les caractéristiques de ce maléate de cyano-3 (méthylamino-3'-méthyl-2'-propyl-l')-10-phénothi- azine de départ sont les suivantes: P.F.: 1701, [a]1 = + 12 (c = 1 % ; chloroforme).
On montre dans (exemple suivant comment les phénothiazines obtenues par mise en oeuvre du pro cédé selon (invention peuvent être oxydées sur l'atome de soufre.
<I>Exemple 3</I> On chauffe à 60o, jusqu'à dissolution complète, 5 g de cyano-3-(diméthylamino-3'-méthyl-2'-propyl)- 10-phénothiazine, obtenue de manière analogue aux exemples précédents, dans 25 cml d'acide acétique glacial;
après refroidissement à 20o, on coule en 15 minutes une solution de 2,16 g d'acide sulfurique pur (d =1,83) dans 25 cm3 d'acide acétique glacial, puis une solution de 3,
9 cm3 d'eau oxygénée à 34 % dans 6,5 cm3 d'acide acétique glacial, en maintenant la température vers 200 ; on chauffe ensuite à 600 pendant 16 heures.
Après refroidissement, le liquide réactionnel est versé sur 200 g de glace et la solution obtenue est alcalinisée avec 12.0 cm-' de lessive de soude (d = 1,33) ; la base qui précipite est extraite avec 200 cm3 d'acétate d'éthyle, la base organique est séchée sur carbonate de potassium et concentrée à sec; le résidu solide, qui pèse 4,5 g, est finalement recristallisé dans l'éthanol.
On obtient ainsi la cyano- 3 -dioxo-9,9-(diméthylamino-3'-m6thyl-2'-propyl)-10- phénothiazine, poudre cristalline blanc crème, fon dant à 166o.
A une solution refroidie à 101, de<B>10 g</B> de la même cyano-3-(diméthylamino-3'-méthyl-2'-propyl)- 10-phénothiazine dans 100 cm3 d'acide acétique, on verse goutte à goutte une solution de 3,1 cm3 d'eau oxygénée à 34 % et 1,
1 cm3 d'acide sulfurique dans 31 cm3 d'acide acétique. Après une nuit d'agitation, on ajoute 154 cm3 d'eau et 225 em3 de soude 10 N. On extrait par 500 cm3 d'acétate d'éthyle, sèche la solution obtenue sur carbonate de potassium et éva pore le solvant au bain-marie.
On recueille 12 g de cyano-3-oxo-9-(diméthylamino-3'-méthyl-2'-propyl)= 10 ghénothiazine brute que l'on peut purifier par chromatographie sur alumine. Le produit obtenu fond vers 135,1.

For pharmaceutical use, the phenothiazines obtained by the process according to the invention are used in the form of therapeutically acceptable salts, such as:
hydrochloride, phosphate, nitrate, sulfate, maleate, fumarate, citrate, tartrate, oxalate, methanesulfonate, ethanedisulfonate or of therapeutically acceptable quaternary ammonium compounds, such as chloro-, bromo- or iodomethylate or -ethoxide,
chloro- or bromobenzylate or -allylate. In the examples which follow, the melting points, unless otherwise indicated, were determined on the Kofler bench.
The examples below illustrate the process according to the invention <I> Example 1 </I> 0.80 g of cyano-3- (methylamino-3'-methyl-2'-propyl-1 ') - 10 is dissolved. -phenothiazine base, released from the corresponding maleate in 30 cm3 of methanol with 3 em3 of normal aqueous hydrochloric acid,
5 cm3 of a 30% aqueous formalin solution and 0.40 g of crystallized aluminum chloride. 0.10 g of Adams platinum catalyst is added and stirred at room temperature under low hydrogen pressure.
When the theoretical amount of hydrogen has been absorbed, the catalyst is filtered, the solvent evaporated and taken up in water. The aqueous solution is then basified, extracted with ether and the ethereal solution dried.
On evaporation of the ether, 0.8 g of an oily residue remains which, in ethanolic solution with male acid, gives 0.65 g of a maleate exhibiting the following charac- teristics <B>:

The starting phenothiazine maleate can be prepared as follows. 41 g of cyano-3- (N-formyl-N-methyl-amino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine opti are dissolved. - only active in 410cm3 of methanol. 82 cm3 of 4N hydrochloric acid are added and heated for 5 hours at reflux. The methanol is then evaporated off and diluted with 200 cm3 of water.
Extracted with ether then basified the remaining aqueous solution with an excess of potassium carbonate. An oil separates which is extracted with ether.
By evaporation of the ether, 11.5 g of a dark oil remain, which is purified by passing through a column of alumina in benzene solution and then crystallization of the maleate from ethyl acetate, the maleate of ethyl acetate is thus obtained. cyano-3- (methylamino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine which has the following characteristics: PF = 170 [10] D = -12.70 (c = 1.2 0 / 0; chloroform).
Optically active cyano-3- (N-formyl-N-methylamino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine is obtained by condensation on sodium cyano-3-phenothiazine in xylene at reflux of N-formyl-N-methylamino-3-methyl-2-chloro-1-propane from PE23 = 141-143.
This halogenated amide is obtained by the action of thionyl chloride, within the chloroform, in the presence of anhydrous pyridine, on N-formyl-N-methylamino-3-methyl-2-propanol-1 of PEo, O = 124- 127. The amide-alcohol is obtained by the action of formamide at reflux on methyl-amino-3-methyl-2-propanol of P.E. 1g = 80-890 [10] D = -3.50 (pure substance).
Finally, this amino alcohol is obtained by reduction with lithium aluminum hydride of the monomethylamino-3-methyl-2 pro benzyl pro pianate which has a rotatory power [a] 20 = + 18.40 (c = 5 0/0 , CHC13). The optically active ester is obtained by resolution of the racemic product using camphosulfonic acid (Reyçhler's acid)
. The racemic product is obtained by transesterification of the known methyl ester.
<I> Example 2 </I> By operating as in the previous example, but starting from cyano-3- (methylamino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine maleate, obtains the cyano- 3 - (dimethylamino - 3'-methyl-2'-propyl-1 ') - 10-phenothiazine base levorotatory with rotatory power according to [U] <B> D </B> = -160 (c = 4 %; chloroform).
The starting maleate can be obtained in the same way as the cyano-3- (methyl-amino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine maleate described in the previous example, but starting from levorotatory benzyl methyl-amino-3-methyl-2-propionate, in chloroform.
The starting characteristics of this cyano-3 (methylamino-3'-methyl-2'-propyl-1 ') - 10-phenothiazine maleate are as follows: PF: 1701, [a] 1 = + 12 (c = 1%; chloroform).

after cooling to 20o, a solution of 2.16 g of pure sulfuric acid (d = 1.83) in 25 cm3 of glacial acetic acid is poured in 15 minutes, then a solution of 3,
9 cm3 of 34% hydrogen peroxide in 6.5 cm3 of glacial acetic acid, maintaining the temperature around 200; then heated at 600 for 16 hours.
After cooling, the reaction liquid is poured onto 200 g of ice and the solution obtained is basified with 12.0 cm 3 of sodium hydroxide solution (d = 1.33); the base which precipitates is extracted with 200 cm3 of ethyl acetate, the organic base is dried over potassium carbonate and concentrated to dryness; the solid residue, which weighs 4.5 g, is finally recrystallized from ethanol.
There is thus obtained cyano-3 -dioxo-9,9- (dimethylamino-3'-methyl-2'-propyl) -10-phenothiazine, creamy white crystalline powder, melting at 166o.
