Verfahren zur Herstellung neuer Steroidverbindungen Vorliegende Erfindung bezieht sich auf die Her stellung von neuen :II-3-Keto-9a-halogen-11ss,17ss- dioxy-17a-alkyl-steroiden, deren 17-Alkylgruppe weniger als drei Kohlenstoffatome enthält, sowie von ihren an der 17ss-Oxygruppe mit einer Carbonsäure mit 1 bis 12 Kohlenstoffatomen veresterten Derivaten. Das erfindungsgemässe Verfahren ist dadurch gekenn zeichnet, dass man die entsprechenden 9,llss-Epoxyde mit einem Halogenwasserstoff umsetzt.
Das Verfahren kann durch die folgenden Formeln dargestellt werden:

In diesen Formeln bedeutet R einen niedrigen Alkylrest mit weniger als 3 Kohlenstoffatomen, das heisst Methyl oder Äthyl; R' Wasserstoff oder den Acylrest einer Carbonsäure mit 1 bis 12 Kohlenstoff atomen und X Fluor, Chlor, Brom oder Jod.
Die Herstellung der als Ausgangsstoffe zu verwen denden 9,11-Epoxyverbindungen kann ausgehend von dem bekannten 3-Pyrrolidylenamin des 4,9(11)- Androstadien-3,17-dions auf folgendem Weg erfolgen:
Man grignadiert das 3-Pyrrolidylenamin unter Bildung des entsprechenden 9(11)-Dehydro-17-alkyl-testo- steron-3-enamins und hydrolysiert die Enamingruppe alkalisch zur 3-Ketogruppe unter Bildung eines 9(11)- Dehydro-17-alkyltestosterons. 9(1 1)-Dehydro-17-alkyl- testosterone erhält man auch ausgehend von 11 a(oder ss)-Oxy-17-alkyl-testosteron, indem man z.
B. durch Umsetzung mit einem N-halogenierten Säureamid in trockenem Pyridin unter Wasserabspaltung in 9(11)- Stellung eine Doppelbindung einführt. Das 9(11)- Dehydro-17-alkyl-testosteron kann sodann in be kannter Weise, z.
B. mit Hilfe von N-Brom- oder N-Jodcarbonsäureamiden oder -imiden, in die ent sprechende 9a-Brom (oder -Jod) -1 lss-oxy-17 - alkyl- steroidverbindung und anschliessend durch Einwir kung einer wässrigen Base in das als Ausgangsstoff zu verwendende 9,11ss-Epoxy-17-alkyl-testosteron überführt werden:
Die nach dem erfindungsgemässen Verfahren erhält lichen 44-3-Keto-9a-halogen-11ss,17ss-dioxy-17a-alkyl- steroide können durch Oxydation, z. B. mit Cr03 in Eisessig, in die entsprechenden 11-Ketoverbindungen überführt werden.
Die neuen 9a-Halogen-llss-oxy-17-alkyl-testo- sterone und deren 17-Ester und die 9a-Halogen-11- keto-17-alkyltestosterone sowie deren 17-Ester be sitzen ausgesprochene androgene Wirksamkeit und beeinflussen den Anabolismus bei oraler Verabrei chung. So besitzt z.
B. 9a-Fluor-1 lss-oxy-17-methyl- testosteron bei oraler Verabreichung eine um 17- bis 19mal höhere anabolische Wirkung und eine etwa 10mal höhere androgene Wirksamkeit, verglichen mit derjenigen des 17-Methyl-testosterons. Ausserdem ist es beträchtlich weniger toxisch und zeigt überraschen derweise nicht die unerwünschte Nebenwirkung der Salzretention, die man von einer 9a-Fluorverbindung erwarten könnte.
Das 9a-Fluor-11-keto-17-methyl- testosteron besitzt oral verabreicht eine androgene Wirksamkeit, die mehr als zweimal so gross ist wie diejenige des 17-Methyl-testosterons,
und seine Wir kung auf den Anabolismus ist mehr als das Fünffache des letzteren. In gleicher Weise besitzen die 9a-Chlor- analogen hohe anabolische und androgene Wirksam keit und das gleiche trifft für die 17-Ester der 9a-Fluor- und 9a-Chlorverbindungen zu.
Auch 9a- Brom- und 9a-Jod-1 1-keto-17-methyl-testosteron und deren 17-Ester besitzen anabolische und androgene Wirksamkeit.
Die neuen androgenen und anabolischen Stoffe eignen sich zur Ersatztherapie der männlichen Keim drüsen bei vor- und nachpuberalen Kastraten, bei durch den Hirnanhang bedingtem Zwergwuchs, bei der Simmondschen Krankheit und zur Unterdrük- kung der Milchbildung.- Ihre anabolische Wirkung macht sie geeignet zur Erhöhung des Gewichtes, der Muskelkraft sowie des Wohlbefindens und des posi tiven Stickstoffgleichgewichts bei Störungen der Funktionen des Hirnanhangs.
Durch angemessene Dosierung kann man gewünschtenfalls eine günstige Wirkung auf den Anabolismus erreichen, ohne die androgenen Funktionen zu beeinflussen.
Die neuen androgen und anabolisch wirksamen Stoffe werden normalerweise in Form von Tabletten, Kapseln oder flüssigen Präparaten oral verabreicht, die gewünschtenfalls zusätzlich noch östrogen wirk same Stoffe, wie östron, Östradiol, Östradiol-21ss- cyclopentylpropionat für die kombinierte androgen- östrogene Therapie enthalten können.
<I>Beispiel l</I> 9a-Fluor-1 lss-oxy-17a-methyl-testosteron.
Zu einer Lösung von 0,5g 9,llss-Epoxy-17a- methyl-testosteron in 10 cms Methylenchlorid gibt man 2 cm3 48 % ige wässrige Fluorwasserstoffsäure. Man rührt 5 Stunden bei Zimmertemperatur und giesst dann vorsichtig unter Rühren in einer Mischung von 6 g Natriumbicarbonat in Eis und Wasser.
Das aus gefallene Steroid wird mit Methylenchlorid extrahiert und der Extrakt mit Wasser gewaschen und getrock net. Man destilliert das Lösungsmittel ab und kristalli siert den Rückstand aus Methylenchlorid um, wobei man 148 mg 9a-Fluor-llss-oxy-17a-methyl-testo- steron vom Schmelzpunkt 265 (Zersetzung) erhält. [a]D+"0" in Chloroform.
Analyse: berechnet für C."H"903F: C71,40 H 8,69 - F 5,65 gefunden:<B>C71,71 H8,66 F5,75.</B> In gleicher Weise erhält man 9a-Fluor-1 lss-oxy- 17a-äthyl-testosteron, wenn man 9,l lf' Epoxy-17a- äthyl-testosteron als Ausgangsstoff verwendet.
<I>Beispiel 2</I> 9a-Fluor- 1 l /; - oxy -17a- methyl - testosteron-17- acetat.
Man arbeitet wie in Beispiel 1, ersetzt aber das Ausgangssteroid durch ein Moläquivalent 9,11ss Epoxy-17a-methyl-testosteron-17-acetat und erhält so das 9a-Fluor-llss-oxy-17a-methyl-testosteron-17- acetat.
Arbeitet man in gleicher Weise, jedoch unter Ver wendung entsprechender 17-Carbonsäureester des 9,llfl-Epoxy-17a-methyl-testosterons, so erhält man andere 17-Ester des 9a-Fluor-llss-oxy-17a-methyl- testosterons, in denen der Acylrest einer Kohlen- wasserstoffcarbonsäure mit 1 bis 12 Kohlenstoff atomen, beispielsweise einer niedrigen aliphatischen Säure, entspricht.
In gleicher Weise erhält man 9cc-Fluor-l 1ss-oxy- 17a-äthyl-testosteron-17-ester, indem man als Aus- gangssteroid einen 9,1lss-Epoxy-17a-äthyl-testosteron- 17-ester verwendet.
Die 17-Oxygruppe kann z. B. noch mit folgenden Säuren verestert sein: Ameisen-, Propion-, Butter-, Isobutter-, Valerian-, Isovalerian-, Trimethylessig-, 2-Methyl-butter-, 3-Äthyl-butter-, Capron-, Diäthyl- essig-, Triäthylessig-, Heptyl-, Capryl-, a-Äthyliso- valeriansäure,
Cyclopropylidenessig-, Cyclopentyl- ameisen-, Cyclopentylessig-, ss-Cyclopentyl-propion-, Cyclohexylameisen-, Cyclohexylessig-, ss-Cyclohexyl- propionsäure, Benzoe-säure, 2-, 3- oder 4-Methyl- benzoe-, 2,3-, 2,4-, 2,5-, 2,6-, 3,4- und 3,5-Di- methyl-benzoe-, Äthyl-benzoe-, 2,4,
6-Trimethyl- benzoe-, a-Naphthoe-, 3-Methyl-a-naphthoe-, Phenyl- essigsäure und dergleichen.
<I>Beispiel 3</I> 9a-Chlor-11 ss-oxy-17a-methyl-testosteron.
Man arbeitet wie in Beispiel 1, ersetzt aber die Fluorwasserstoffsäure durch wasserfreien Chlor wasserstoff und erhält so 9a-Chlor-llss-oxy-17a- methyl-testosteron.
In gleicher Weise erhält man 9a-Chlor- 1lss'-oxy- 17-methyl-testosteron-17-carbonsäureester aus den entsprechenden 9,llss-Epoxy-17cc-methyl-testosteron- 17-estern.
Ersetzt man das Ausgangssteroid durch 9, l I ss Epoxy-17a-äthyl-testosteron oder eines seiner 17- Ester, so erhält man 9a-Chlor-1 1ss-oxy-17-äthyl-testo- steron bzw. dessen 17-Ester.
Die 9a-Chlor- und 9a-Fluor-llss-oxy-17a-alkyl- testosteron-17-ester können auch erhalten werden durch Veresterung der entsprechenden freien Verbin dungen.
Analog den vorstehenden Beispielen 1 bis 3 kann man durch Ersatz von Fluor- und Chlorwasserstoff durch Brom- oder Jodwasserstoff zu den entsprechen den 9a-Broin oder -Jodverbindungen gelangen. Zu dem in den Beispielen 1 bis 3 verwendeten Ausgangsmaterial kann man wie folgt gelangen:
Eine Lösung von 59 g 3-Enamin des 4,9(11) Androstadien- 3,17-dions in 1 Liter wasserfreiem Tetrahydrofuran tropft man im Verlaufe von 15 Minuten unter Rühren und in einer Stickstoffatmosphäre zu 700 cm3 einer viermolaren Lösung von Methylmagnesiumbromid in Diäthyläther. Nach beendigtem Zusatz wird die Mi schung erwärmt und das Lösungsmittel so lange daraus abdestilliert, bis die Destillationstemperatur 60 erreicht.
Die Mischung wird dann unter Aus schluss von Feuchtigkeit und atmosphärischem Sauer stoff 18 Stunden am Rückfluss erhitzt. Dann wird im Eis-Wasserbad gekühlt und vorsichtig mit 430 cm3 Wasser versetzt. Immer noch unter Stickstoffatmo sphäre gibt man 260 cm3 Eisessig und dann 1 Liter Methanol zu und erhitzt, bis sich alles ausgeschiedene 3-Enamin des 9(11)-Dehydro-17a-methyl-testosterons gelöst hat.
Die das 3-Enamin des 9(11)-Dehydro-17a-methyl- testosterons enthaltende Lösung wird nun mit 700 cm3 10 % iger wässriger Natronlauge versetzt und dann 30 Minuten am Rückfluss erhitzt,
um die Enamin- gruppe unter Rückbildung der 44-3-Ketogruppe zu hydrolysieren. Man kühlt auf Zimmertemperatur ab, säuert mit Essigsäure an und destilliert unter vermin dertem Druck, bis der Rückstand etwa 1,5 Liter be trägt. Nach Zusatz einer Mischung von<B>100</B> cm3 kon zentrierter Salzsäure und 100g Eis wird der Rück stand mit Methylenchlorid extrahiert und der Extrakt in üblicher Weise aufgearbeitet.
Nach Umkristallisie- ren des Rohproduktes aus verdünntem Aceton erhält man 26,8 g 9(11)-Dehydro-17-methyl-testosteron vom Schmelzpunkt 167 bis 170 .
Verwendet man anstelle von Methylmagnesium- bromid Athylmagnesiumbromid, so erhält man das 9(11)-Dehydro-17-äthyl-testosteron.
9(11)-Dehydro-17a-methyl-testosteron erhält man auch, indem man zu einer Lösung von 1 g l lss-Oxy- 17-methyl-testosteron in 10 cm3 trockenem Pyridin unter Rühren bei 25 und in einer Stickstoffatmo sphäre nach und nach 0,5 g N-Brom-acetamid gibt. Man rührt weitere 15 Minuten, kühlt auf 10 und leitet so lange SO.-Gas ein, bis die Jodstärkepapier- reaktion negativ ist. Dann setzt man 10 cm3 Wasser und eine Mischung von 15 cm3 konzentrierter Salz säure mit 25g Eis zu.
Die Ausfällung wird abge trennt, mit Wasser gewaschen, getrocknet, dann zuerst aus Methylenchlorid und Skellysolve B (Hexan- kohlenwasserstoffe) und hernach aus verdünntem Aceton umkristallisiert. Das so erhaltene 9(11)-De- hydro-17a-methyl-testosteron schmilzt bei 170 bis 172 .
Ferner gelangt man zu der 9(11)-Dehydroverbin- dung, indem man zu einer warmen Lösung von 1 g lla-Oxy-17-methyl-testosteron in 2 cm3 trockenem Pyridin 1 g p-Toluolsulfonylchlorid gibt. Man hält die Mischung 18 Stunden bei Zimmertemperatur, giesst dann in 25 cm3 Wasser und rührt, bis das aus geschiedene Öl fest wird. Man filtriert, wäscht mit Wasser und trocknet. Man erhält 1,41 g lla-(p Toluolsulfonyloxy)-17a-methyl-17ss-oxy-4-androsten- 3-on vom Schmelzpunkt 144 bis 148 (unter Zer setzung).
Nach dem Umkristallisieren aus einer Mi schung von Methylenchlorid und Hexankohlenwasser- stoffen ist der Schmelzpunkt 141 bis 144 (Zerset zung).
Eine. Mischung von 1 g des so erhaltenen 11 a-(p- Toluolsulfonyloxy) -17a-methyl-17ss-oxy-4-androsten- 3-ons, 0,2 g Natriumformiat, 0,57 cm3 Wasser und 14 cm3 absolutem Alkohol wird 19 Stunden am Rück fluss gekocht. Man lässt die Lösung abkühlen und giesst sie auf eine Mischung von 50 g Eis und Wasser, wobei man rührt.
Der Niederschlag wird abfiltriert und getrocknet und man erhält 0,59 g 9(11),Dehydro- 17-methyl-testosteron, dessen Schmelzpunkt nach dem Umkristallisieren aus einem Gemisch von Methylen- chlorid und Hexankohlenwasserstoffen 167 bis 170 beträgt.
Das 9(11)-Dehydro-17-methyl-testosteron-17- acetat erhält man, wenn man eine Lösung von 9(11)- Dehydro-17-methyl-testosteron in trockenem Pyridin mit Essigsäureanhydrid behandelt, wobei das Molver- hältnis von Steroid zu Essigsäureanhydrid etwa 1 : 3 ist. Die Mischung wird 6 Stunden am Rückfluss erhitzt. Man kühlt ab, versetzt unter Rühren mit- Wasser und filtriert vom festen Niederschlag ab.
Der Rückstand wird mit 2a/oiger wässriger Salzsäure und Wasser ge waschen, dann im Vakuum getrocknet. Man erhält so 9(11)-Dehydro-17-methyl-testosteron-17-acetat. In gleicher Weise kann 9(11)-Dehydro-17-methyl-testo- steron durch Veresterung der 17-Oxygruppe in andere Ester umgewandelt werden, z. B. durch Reaktion mit dem betreffenden Säureanhydrid, Säurechlorid oder -bromid, durch Umesterung mit Säure in Gegenwart eines Veresterungskatalysators usw.
17-Carbonsäureester des 9(11)-Dehydro-17-äthyl- testosterons erhält man in analoger Weise aus 9(11), Dehydro-17-äthyl-testosteron.
Zu einer Lösung von 1 g 9(11)-Dehydro-17- methyl-testosteron in 50 cm3 Aceton gibt man tropfen weise unter Rühren bei 15 eine Lösung von 1 g Bromacetamid in 25 cms Wasser. Dann setzt man bei der gleichen Temperatur 20 cm3 einer 0,5n Lösung von Perchlorsäure in Wasser langsam zu. Nach 20 Mi nuten gibt man genügend gesättigte wässrige Natrium sulfitlösung zu, um die ,gelbe Farbe der Lösung zum Verschwinden zu bringen.
Die Mischung wird nun mit 100 cm3 Wasser verdünnt, wobei 1 g 9a-Brom- llss-oxy-17-methyl-testosteron in Form von bei 153 bis 155 schmelzenden Nadeln ausfällt.
In analoger Weise lassen sich 17-Carbonsäure- ester von 9(11)-Dehydro-17-methyl-testosteron, vor zugsweise das Acetat, in die 17-Carbonsäureester des 9a-Brom-llss-oxy-17-methyl-testosterons überführen.
Setzt man 9a-Brom-llss-oxy-17-methyl-testo- steron oder einen seiner 17-Ester in an sich bekannter Weise mit Natriumjodid in Aceton um, so erhält man 9a-Jod-llss-oxy-17-methyl-testosteron und- seine Ester. 9a-Brom-1 1/3-oxy-17a-äthyltestosteron und dessen Ester lassen sich in analoger Weise herstellen, indem man vom 9(11)-Dehydro-17a-äthyl-testosteron und dessen 17-Estern ausgeht.
Eine Suspension von 1 g 9a-Brom-llss-oxy-17a- methyltestosteron in 30 cm3 Methanol wird mit einem Moläquivalent 0,1m wässriger Natronlauge behandelt. Die Mischung wird mit 50 cm3 Wasser verdünnt und auf etwa 0 gekühlt, wobei 0,64 g 9,11 ss-Epoxy- 17a-methyl-testosteron vom Schmelzpunkt 170 bis l76 ausfällt, das nach Umkristallisieren aus verdünn tem Methanol bei 165 bis 172 (unter Sublimation) schmilzt.
In gleicher Weise lassen sich 17-Ester des 9a Brom-llss-oxy-17-methyl-testosterons in die 17-Car- bonsäureester des 9,11 ss-Epoxy-17-methyl-testosterons überführen.
9,llss-Epoxy-17-methyl-testosteron und seine 17-Carbonsäureester können auch erhalten werden, wenn man als Ausgangssteroid das 9a-Jod-llss Oxy- 17-methyl-testosteron und dessen 17-Ester verwendet. <I>Beispiel 4</I> 9a-Fluor-1 1-keto-17a-methyl-testosteron.
Zu einer Lösung von 0,457 g 9a-Fluor-1 lss-oxy- 17a-methyl-testosteron (erhalten z. B. nach Beispiel 1) in 20 cm3 Eisessig gibt man eine Lösung von 0,25 g Chromtrioxyd und 1 cm3 Wasser in 20 cm3 Essig säure. Die Mischung wird 41:Stunden bei Zimmer temperatur gehalten und dann mit 5 cm3 Methanol vermischt. Das Lösungsmittel wird unter verminder tem Druck abdestilliert und der nahezu trockene Rückstand mit 20 cm3 Wasser versetzt.
Der Nieder schlag wird filtriert, mit Wasser gewaschen und ge trocknet. Man erhält 0,362 g 9a-Fluor-11-keto-17a- methyl-testosteron vom Schmelzpunkt 208 bis 212 . Nach Umkristallisieren aus verdünntem Methanol und dann aus einer Mischung von Aceton und Hexan- kohlenwasserstoffen steigt der Schmelzpunkt auf 213 bis 220'; [a]D + 144 (in Chloroform). Analyse: 40 berechnet für CzpHz703F: C 71,83 H 8,14 F 5,68 gefunden: C 72,13 H 8,03 F 5,83.
Verwendet man als Ausgangssteroid einen 9a- Fluor-11/3-oxy-17a-methyl-testosteron-17-ester, so erhält man den entsprechenden Ester des 9a-Fluor- 4s 11-keto-17a-methyl-testosterons.
In gleicher Weise erhält man aus 9a-CWor-llss- oxy-17a-methyl-testosteron das 9a-Chlor-11-keto- 17a-methyl-testosteron.

The solvent is distilled off and the residue is recrystallized from methylene chloride, 148 mg of 9a-fluoro-llss-oxy-17a-methyl-testosterone having a melting point of 265 (decomposition). [a] D + "0" in chloroform.
Analysis: calculated for C. "H" 903F: C71.40 H 8.69 - F 5.65 found: C71.71 H8.66 F5.75. 9a-fluoro is obtained in the same way -1 lss-oxy-17a-ethyl-testosterone, if one uses 9.1 lf 'epoxy-17a-ethyl-testosterone as a starting material.
<I> Example 2 </I> 9a-fluoro-1 l /; - oxy -17a- methyl - testosterone-17- acetate.
The procedure is as in Example 1, but the starting steroid is replaced by a molar equivalent of 9.11ss epoxy-17a-methyl-testosterone-17-acetate and the 9a-fluoro-llss-oxy-17a-methyl-testosterone-17-acetate is obtained.
If you work in the same way, but using the corresponding 17-carboxylic acid esters of 9, llfl-epoxy-17a-methyl-testosterone, then other 17-esters of 9a-fluoro-llss-oxy-17a-methyl-testosterone are obtained in to which the acyl radical of a hydrocarbon carboxylic acid having 1 to 12 carbon atoms, for example a lower aliphatic acid, corresponds.
In the same way, 9cc-fluoro-1ss-oxy-17a-ethyl-testosterone-17-ester is obtained by using a 9.11ss-epoxy-17a-ethyl-testosterone-17-ester as the starting steroid.

<I> Example 3 </I> 9a-chloro-11ss-oxy-17a-methyl-testosterone.
The procedure is as in Example 1, but the hydrofluoric acid is replaced by anhydrous hydrogen chloride and 9a-chloro-llss-oxy-17a-methyl-testosterone is obtained.
In the same way, 9α-chloro-1lss'-oxy-17-methyl-testosterone-17-carboxylic acid esters are obtained from the corresponding 9, 11ss-epoxy-17cc-methyl-testosterone-17-esters.
If the starting steroid is replaced by 9.15 epoxy-17a-ethyl-testosterone or one of its 17-esters, 9a-chloro-11s-oxy-17-ethyl-testosterone or its 17-ester is obtained.
The 9a-chloro and 9a-fluoro-llss-oxy-17a-alkyl-testosterone-17-esters can also be obtained by esterification of the corresponding free compounds.
Analogously to the above Examples 1 to 3, the corresponding 9a-bromine or -iodine compounds can be obtained by replacing fluorine and hydrogen chloride with hydrogen bromide or hydrogen iodide. The starting material used in Examples 1 to 3 can be obtained as follows:
A solution of 59 g of 3-enamine of 4,9 (11) androstadiene-3,17-dione in 1 liter of anhydrous tetrahydrofuran is added dropwise over the course of 15 minutes with stirring and in a nitrogen atmosphere to 700 cm3 of a four molar solution of methyl magnesium bromide in diethyl ether . When the addition is complete, the mixture is heated and the solvent is distilled off from it until the distillation temperature reaches 60.
The mixture is then refluxed for 18 hours with the exclusion of moisture and atmospheric oxygen. Then it is cooled in an ice-water bath and 430 cm3 of water are carefully added. Still under a nitrogen atmosphere, 260 cm3 of glacial acetic acid and then 1 liter of methanol are added and the mixture is heated until all the 3-enamine of 9 (11) -dehydro-17a-methyl-testosterone that has separated out has dissolved.
The solution containing the 3-enamine of 9 (11) -dehydro-17a-methyl-testosterone is now mixed with 700 cm3 of 10% aqueous sodium hydroxide solution and then refluxed for 30 minutes,
to hydrolyze the enamine group with regression of the 44-3-keto group. It is cooled to room temperature, acidified with acetic acid and distilled under reduced pressure until the residue is about 1.5 liters. After adding a mixture of 100 cm3 of concentrated hydrochloric acid and 100 g of ice, the residue is extracted with methylene chloride and the extract is worked up in the usual way.
After recrystallization of the crude product from dilute acetone, 26.8 g of 9 (11) -dehydro-17-methyl-testosterone with a melting point of 167 to 170 are obtained.
If ethylmagnesium bromide is used instead of methylmagnesium bromide, 9 (11) -dehydro-17-ethyl-testosterone is obtained.
9 (11) -Dehydro-17a-methyl-testosterone is also obtained by gradually adding to a solution of 1 gl lss-oxy-17-methyl-testosterone in 10 cm3 of dry pyridine with stirring at 25 and in a nitrogen atmosphere 0.5 g of N-bromo-acetamide are there. The mixture is stirred for a further 15 minutes, cooled to 10 and SO gas is passed in until the iodine starch paper reaction is negative. Then 10 cm3 of water and a mixture of 15 cm3 of concentrated hydrochloric acid with 25g of ice are added.
The precipitate is separated off, washed with water, dried, then first recrystallized from methylene chloride and Skellysolve B (hexane hydrocarbons) and then from dilute acetone. The 9 (11) -dehydro-17a-methyl-testosterone obtained in this way melts at 170 to 172.
The 9 (11) -dehydrocompound is also obtained by adding 1 g of p-toluenesulfonyl chloride to a warm solution of 1 g of IIIa-oxy-17-methyl-testosterone in 2 cm3 of dry pyridine. The mixture is kept at room temperature for 18 hours, then poured into 25 cm3 of water and stirred until the separated oil solidifies. It is filtered, washed with water and dried. 1.41 g of lla- (p toluenesulfonyloxy) -17a-methyl-17ss-oxy-4-androsten-3-one of melting point 144 to 148 (with decomposition) are obtained.
After recrystallization from a mixture of methylene chloride and hexane hydrocarbons, the melting point is 141 to 144 (decomposition).
A. Mixture of 1 g of the 11 a- (p-toluenesulfonyloxy) -17a-methyl-17ss-oxy-4-androsten-3-one obtained in this way, 0.2 g of sodium formate, 0.57 cm3 of water and 14 cm3 of absolute alcohol is 19 Cooked for hours on the reflux. The solution is allowed to cool and is poured onto a mixture of 50 g of ice and water, while stirring.
The precipitate is filtered off and dried and 0.59 g of 9 (11), dehydro-17-methyl-testosterone, whose melting point is 167 to 170 after recrystallization from a mixture of methylene chloride and hexane hydrocarbons, is obtained.
The 9 (11) -dehydro-17-methyl-testosterone-17-acetate is obtained when treating a solution of 9 (11) -dehydro-17-methyl-testosterone in dry pyridine with acetic anhydride, the molar ratio of Steroid to acetic anhydride is about 1: 3. The mixture is refluxed for 6 hours. It is cooled, water is added while stirring and the solid precipitate is filtered off.
The residue is washed with 2 a / o aqueous hydrochloric acid and water, then dried in vacuo. This gives 9 (11) -dehydro-17-methyl-testosterone-17-acetate. In the same way, 9 (11) -Dehydro-17-methyl-testosterone can be converted into other esters by esterification of the 17-oxy group, e.g. B. by reaction with the relevant acid anhydride, acid chloride or bromide, by transesterification with acid in the presence of an esterification catalyst, etc.
17-carboxylic acid ester of 9 (11) -dehydro-17-ethyl-testosterone is obtained in an analogous manner from 9 (11), dehydro-17-ethyl-testosterone.
A solution of 1 g of bromoacetamide in 25 cm of water is added dropwise to a solution of 1 g of 9 (11) -dehydro-17-methyl-testosterone in 50 cm 3 of acetone while stirring at 15. Then 20 cm3 of a 0.5N solution of perchloric acid in water are slowly added at the same temperature. After 20 minutes, enough saturated aqueous sodium sulfite solution is added to make the yellow color of the solution disappear.
The mixture is then diluted with 100 cm3 of water, 1 g of 9a-bromine-llss-oxy-17-methyl-testosterone precipitating in the form of needles melting at 153 to 155.
In a similar manner, 17-carboxylic acid esters of 9 (11) -dehydro-17-methyl-testosterone, preferably the acetate, can be converted into the 17-carboxylic acid esters of 9a-bromo-llss-oxy-17-methyl-testosterone.
If 9a-bromo-llss-oxy-17-methyl-testosterone or one of its 17-esters is reacted in a manner known per se with sodium iodide in acetone, 9a-iodine-llss-oxy-17-methyl-testosterone is obtained and - its esters. 9a-bromo-1 1/3-oxy-17a-ethyltestosterone and its esters can be prepared in an analogous manner by starting from 9 (11) -dehydro-17a-ethyltestosterone and its 17-esters.
A suspension of 1 g of 9a-bromo-llss-oxy-17amethyltestosterone in 30 cm3 of methanol is treated with a molar equivalent of 0.1 M aqueous sodium hydroxide solution. The mixture is diluted with 50 cm3 of water and cooled to about 0, whereupon 0.64 g of 9.11 ss-epoxy-17a-methyl-testosterone with a melting point of 170 to 176 precipitates, which after recrystallization from dilute methanol at 165 to 172 ( under sublimation) melts.
In the same way, 17-esters of 9a bromo-llss-oxy-17-methyl-testosterone can be converted into the 17-carboxylic acid esters of 9,11 ss-epoxy-17-methyl-testosterone.
9, llss-epoxy-17-methyl-testosterone and its 17-carboxylic acid esters can also be obtained if the 9a-iodine-llss oxy-17-methyl-testosterone and its 17-ester are used as the starting steroid. <I> Example 4 </I> 9a-fluoro-1 1-keto-17a-methyl-testosterone.
A solution of 0.25 g of chromium trioxide and 1 cm3 of water in 20 cm3 of glacial acetic acid is added to a solution of 0.457 g of 9a-fluoro-1 lss-oxy-17a-methyl-testosterone (obtained for example according to Example 1) in 20 cm3 of glacial acetic acid cm3 acetic acid. The mixture is kept at room temperature for 41 hours and then mixed with 5 cm3 of methanol. The solvent is distilled off under reduced pressure and the almost dry residue is mixed with 20 cm3 of water.
The precipitate is filtered, washed with water and dried ge. 0.362 g of 9a-fluoro-11-keto-17a-methyl-testosterone with a melting point of 208 to 212 are obtained. After recrystallization from dilute methanol and then from a mixture of acetone and hexane hydrocarbons, the melting point rises to 213 to 220 °; [a] D + 144 (in chloroform). Analysis: 40 Calcd for CzpHz703F: C 71.83 H 8.14 F 5.68 Found: C 72.13 H 8.03 F 5.83.
If a 9a-fluoro-11/3-oxy-17a-methyl-testosterone-17-ester is used as the starting steroid, the corresponding ester of 9a-fluoro-4s 11-keto-17a-methyl-testosterone is obtained.
In the same way, 9a-chloro-11-keto-17a-methyl-testosterone is obtained from 9a-CWor-llssoxy-17a-methyl-testosterone.
