Verfahren zur Herstellung von N'-substituierten N-Arylsulfonylharnstoffen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N'-substituierter N-Arylsulfonyl-harnstoffe mit wertvollen pharmakologischen Eigenschaften.
Als blutzuckersenkende Wirkstoffe haben sich der N-Sulfanilyl-N'-n-butyl-harnstoff und der N-(4-Methyl-benzolsulfonyl)-N'-n-butyl-harnstoff in umfangreicher klinischer Erprobung für die perorale Behandlung von Diabetes mellitus geeignet erwiesen.
Die erstgenannte Verbindung wurde auch bereits in die Praxis eingeführt.
Überraschenderweise wurde nun gefunden, dass N'-substituierte N-Arylsulfonyl-harnstoffe der Formel

worin R einen Alkyl-, Alkenyl- oder Cycloalkylrest und X Fluor, Chlor oder Brom bedeuten, eine entsprechende Senkung des Blutzuckerspiegels schon in wesentlich geringen Dosen bewirken und sich überdies durch eine gleichmässige, langanhaltende Wirkung auszeichnen.
Man kann die vorstehend definierten Verbindungen herstellen, indem man ein Arylsulfonamid der Formel

worin X die oben angegebene Bedeutung hat, oder ein Alkalisalz eines solchen, mit einem Alkyl-, Alkenyl- oder Cycloalkylisocyanat der Formel
R-N=C=O III oder mit einem reaktionsfähigen funktionellen Derivat einer Alkyl-, Alkenyl- oder Cycloalkylcarbaminsäure der Formel
R-NH-COOH IV gegebenenfalls in Gegenwart eines Kondensationsmittels, umsetzt und zum entsprechenden Harnstoff hydrolysiert. Geeignete funktionelle Derivate von Carbaminsäuren sind insbesondere deren Ester, z.
B. niedermolekulare Alkylester oder deren Halogenide, sowie die Amide, das heisst Harnstoffe der Formel R-NH-CO-NH2. Anstelle von Isocyanaten können auch die leicht zu solchen zersetzbaren Azide, N-Brom-amide und N-Chlor-amide von Carbonsäuren der Formel R-COOH verwendet werden.
Als Kondensationsmittel können basische Kondensationsmittel, wie z. B. Alkalialkoholate, Pyridin oder Triäthylamin, Verwendung finden und kommen insbesondere dann in Betracht, wenn die Reaktion unter Freisetzung einer Säure vor sich geht. Man kann die Arylsulfonamide auch vor der Umsetzung durch Behandlung mit einer Alkaliverbindung in ein Alkalisalz überführen und, anstelle der freien Sulfonamide in Gegenwart eines basischen Kondensa tionsmittels, deren vorgebildete Alkalisalze zur Umsetzung verwenden. Anderseits kommt als Kondensationsmittel für die Umsetzungen von Arylsulfonamiden mit Alkyl-, Alkenyl- oder Cycloalkylcyanamiden insbesondere Chlowasserstoff in Betracht.
Bei den N-Arylsulfonamiden der Formel II handelt es sich um das 4-Fluor-, das 4-Chlor-und das 4-Brombenzolsulfonamid. Diese lassen sich beispielsweise mit dem Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek. Butyl-, n-Amyl-, Isoamyl-, n-Hexyl-, ss-Methyl-pentyl-, n-Octyl-, p-Sithyl- hexyl-,-n-Dodecyl-, Allyl-, Crotyl-, Methallyl-, Cyclopentyl- und Cyclohexyl-isocyanat umsetzen.
Die erfindungsgemäss herstellbaren Verbindungen der Formel I lassen sich in üblicher Weise in beständige wasserlösliche Alkalisalze überführen.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile, diese verhalten sich zu Volumteilen wie Gramm zu Kubikzentimeter. Die Temperaturen sind in Centigraden angegeben.
Beispiel 1
19,15 Teile p-Chlor-benzolsulfonamid werden in einer Lösung von 4 Teilen Natriumhydroxyd in 20 Teilen Wasser und 60 Teilen Alkohol suspendiert, auf 0O gekühlt und unter Rühren bei 020O 8,5 Teile Äthylisocyanat zutropfen gelassen. Nach Abklingen der Reaktion wird die entstandene Lösung noch 2 Stunden unter Rückfluss gekocht, dann abkühlen gelassen und in 75 Teile 2-n. Salzsäure und 75 Teile Eis eingerührt. Die Ausfällung wird abgenutscht, der Niederschlag in 75 Teilen 2-n. Ammoniak gelöst, von geringen unlöslichen Nebenprodukten abfiltriert und der N-(p-Chlor-benzolsulfonyl) -N'-äthyl-harnstoff durch Eingiessen in überschüssige, verdünnte Salzsäure wieder ausgefällt.
Das durch Nutschen abgetrennte Produkt lässt sich aus Alkohol umkristallisieren und zeigt einen Schmelzpunkt von 124-125,5".
Ersetzt man in obigem Beispiel das Alkylisocyanat durch 6,8 Teile Methylisocyanat oder 9,5 Teile Isopropylisocyanat oder 9,1 Teile Allylisocyanat oder 11 Teile n-Butylisocyanat, so erhält man bei gleicher Ausführung und Umkristallisation aus verdünntem Alkohol den N-(p-Chlor-benzolsulfonyl)-N'-methyl- harnstoff vom Schmelzpunkt 158,5-160,5" bzw. den N - (p - Chlor - benzolsulfonyl) - N'-isopropyl-harn- stoff bzw. den N-(p-Chlor-benzolsulfonyl)-N'-allylharnstoff vom Schmelzpunkt 156-159" bzw. den N-(p-Chlor-benzolsulfonyl)-N'-n-butyl-harnstoff vom Schmelzpunkt 106,5-109".
Beispiel 2
23,6 Teile p-Brom-benzolsulfonamid werden in 60 Teilen Alkohol suspendiert, 20 Teile 200/obiger Natronlauge zugegeben und unter Rühren 11 Teile n-Butylisocyanat derart zutropfen gelassen, dass die Temperatur nicht über 40 steigt. Anschliessend wird noch 2 Stunden auf dem kochenden Wasserbad erwärmt, dann abgekühlt und in überschüssige verdünnte Essigsäure eingerührt. Der bereits kristallin anfallende N-(p-Brom- benzolsulfonyl)- N' -n-butyl- harnstoff wird aus verdünntem Ammoniak mittels verdünnter Salzsäure umgefällt und anschliessend aus 100 Teilen heissem Alkohol unter Zugabe von 3 Teilen Tierkohle umkristallisiert; Schmelzpunkt 121 bis 123".

23.6 parts of p-bromo-benzenesulphonamide are suspended in 60 parts of alcohol, 20 parts of 200 / above sodium hydroxide solution are added and 11 parts of n-butyl isocyanate are added dropwise with stirring so that the temperature does not exceed 40.
