Verfahren zur Herstellung von N'-substituierten N-Arylsulfonyl-harnstoffen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N'-substituierter N-Arylsulfonyl-harnstoffe mit wertvollen pharmakologischen Eigenschaften. Die neuen Erzeugnisse bewirken eine Senkung des Blutzuckerspiegels schon in wesentlich geringeren Dosen als die bekannten Mittel und zeichnen sich überdies durch eine gleichmässige langanhaltende Wirkung aus.

Als Kondensationsmittel können basische Verbindungen, wie z. B. Alkalialkoholate, Pyridin oder Triäthylamin Verwendung finden; sie kommen insbesondere dann in Betracht, wenn die Reaktion unter Freisetzung einer Säure vor sich geht.
Zur Anwendung im erfindungsgemässen Verfahren geeignete Amine sind beispielsweise das Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek.-Butyl-, n-Amyl-, Isoamyl-, n-Hexyl-, jB-Methyl-pentyl-, n-Octyl-, B-Athyl-hexyl ;, n-Dodecyl-, Allyl-, Crotyl-, Methallyl-, Cyclopentyl-, Cyclohexyl- und Endomethylen-cyclohexylmethyl-amin.
Die erfindungsgemäss herstellbaren Verbindungen der Formel I lassen sich in üblicher Weise in beständige wasserlösliche Alkalisalze überführen.
In den nachfolgenden Beispielen bedeuten die Teile Gewichtsteile, diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Centigraden angegeben.
Beispiel 1
Zu einer Lösung von 21,8 Teilen p-Chlor-benzolsulfonylisocyanat in 100 Teilen absolutem Toluol werden unter Rühren und gutem Kühlen bei 10-20" 8 Teile n-Butylamin, gelöst in 10 Teilen Toluol, zugetropft. Nach Abklingen der Reaktion wird das ausgeschiedene Produkt abgenutscht und aus verdünntem Alkohol umkristallisiert. Man erhält den N-(p Chlor-benzolsulfonyl)-N'-butyl-harnstoff in Form farbloser Kristalle, die bei 106,5-108" schmelzen.
Beispiel 2
23,3 Teile N-(p-Fluor-benzolsulfonyl)-carbaminsäure-methylester werden mit 8 Teilen Isobutylamin in 50 Volumteilen Glykolmonomethyläther 4 Stunden unter Rückfluss gekocht. Anschliessend engt man das Reaktionsgemisch im Vakuum ein, löst den Rückstand in verdünntem wässrigem Ammoniak, fil triert, entfärbt das Filtrat mit Tierkohle und fällt daraus das Reaktionsprodukt durch Ansäuern mit verdünnter Salzsäure.
Nach Umkristallisieren aus 120 Volumteilen Benzol erhält man den N-(p-Fluor-benzolsulfonyl) N'-isobutyl-harnstoff als weisse Nadeln, die bei 152,5 bis 154,5" schmelzen.

The compounds of the formula I which can be prepared according to the invention can be converted into stable water-soluble alkali salts in the customary manner.

example 1
To a solution of 21.8 parts of p-chlorobenzenesulfonyl isocyanate in 100 parts of absolute toluene, 8 parts of n-butylamine dissolved in 10 parts of toluene are added dropwise with stirring and good cooling at 10-20 " The product is filtered off with suction and recrystallized from dilute alcohol. The N- (p chloro-benzenesulfonyl) -N'-butylurea is obtained in the form of colorless crystals which melt at 106.5-108 ".
Example 2
23.3 parts of methyl N- (p-fluoro-benzenesulfonyl) carbamate are refluxed with 8 parts of isobutylamine in 50 parts by volume of glycol monomethyl ether for 4 hours. The reaction mixture is then concentrated in vacuo, the residue is dissolved in dilute aqueous ammonia, filtered, the filtrate is decolorized with animal charcoal and the reaction product is precipitated therefrom by acidification with dilute hydrochloric acid.
After recrystallization from 120 parts by volume of benzene, N- (p-fluoro-benzenesulphonyl) N'-isobutylurea is obtained as white needles which melt at 152.5 to 154.5 ".
