Das Verfahren wird beispielsweise so ausgeführt, dass 1-Methyl-4-oxy-piperidin mit einem substituierten Benzoesäure-alkylester in Gegenwart eines Kondensationsmittels, z. B. Natriumäthylat oder Natriummetall, auf höhere Temperatur, vorzugsweise 150-200 , erhitzt wird, wobei der im Verlauf der Umesterung freigewordene aliphatische Alkohol abdestilliert. Der Rückstand liefert bei der Fraktionierung im Vakuum das Endprodukt.
Ist das für die Acylierung des 1-Alkyl-4-oxy- piperidins verwendete substituierte Benzoesäure- derivat ein Säurehalogenid, z. B. Anissäurechlorid, dann gestaltet sich der Umsatz so, dass das 1-Alkyl4-oxy-piperidin-Derivat mit dem substituierten Ben zoylhalogenid zusammen in einem inerten organischen Lösungsmittel, z. B. Benzol, Toluol, Xylol, gelöst wird. Die Lösung der Reaktionspartner wird bei Zimmertemperatur undloder bei erhöhter Temperatur, z. B. in einem Dampfbad, stehengelassen ; schliesslich wird der gebildete Ester, der als Hydrohalogenid vorliegt, durch Zusatz eines Alkalis, z. B.
Soda, in Freiheit gesetzt und nach bekannten Methoden isoliert und gereinigt.
Die nach dem vorliegenden Verfahren hergestellten bisher unbekannten Piperidin-Derivate lassen sich im Vakuum ohne Zersetzung destillieren. Sie sind bei Zimmertemperatur flüssige oder feste, kristalline Basen, die mit Säuren beständige Salze bilden. Sie zeichnen sich durch therapeutisch verwertbare pharmakodynamische Eigenschaften aus. Sowohl die freien Basen als auch ihre Salze zeigen bei sehr guter lokaler Verträglichkeit und geringer Toxizität eine starke lokalanästhetische Wirkung, die sich sowohl bei der Oberflächen-als auch bei der Leitungsanästhesie nachweisen lässt. Die neuen Verbindungen sollen in der Therapie verwendet werden. In den nachfolgenden Beispielen sind alle Temperaturen in Grad Celsius angegeben.
Beispiel 1
Eine Mischung von 40,0 g p-n-Butoxy-benzoe säuremethylester, 88,5 g 1-Methyl-4-oxy-piperidin und 1,0 g Natriumäthylat (pulv.) wird während 2 Stunden am absteigenden Kühler in einem Ölbad von 200 erhitzt. Ist die Destillation von Methanol beendet, wird abgekühlt, mit 500 cm3 Aceton digeriert und abfiltriert. Das Filtrat wird im Vakuum bei 50 eingedampft und der Rückstand destilliert. Der p-n Butoxy-benzoesäure- (l-methyl-4-piperidyl)-ester siedet bei 0,05 mm Hg zwischen 160-165 und zeigt einen Smp. von 55-57 (nach dem Umkristallisieren aus AthanollWasser).
Das Monohydrochlorid wird hergestellt, indem man die Verbindung in der äquivalenten Menge 1-n.
Salzsäure löst, im Vakuum bei 50 eindampft, den Rückstand in der lOfachen Menge abs. Athanol löst und wieder im Vakuum bei gleicher Temperatur eindampft. Der Rückstand wird in der I Ofachen Menge siedendem abs. Athanol gelöst und nach dem Filtrieren mit der gleichen Menge Ather versetzt. Das kristalline Monohydrochlorid des p-n-Butoxy-benzoe säure- (1-methyl-4-piperidyl)-esters hat den Smp. 202 bis 2040.
Beispiel 2
43,0 g p-n-Butoxy-benzoylchlorid, gelöst in
100 cm3 Benzol, werden mit einer Lösung von 23,4 g 1-Methyl-4-oxy-piperidin in 100 cm3 Benzol bei Zimmertemperatur vermischt, über Nacht stehengelasse und dann eine Stunde auf 80 erwärmt.
Darauf wird im Vakuum bei 50"zur Trockne ein geengt. Der Rückstand wird in 450 cm3 Wasser und 50 cm3 3-n. Salzsäure gelöst und unter Eiskühlung mit 120 g fester Soda versetzt. Die Lösung des ausgeschiedenen Öls in 600 cm3 Sither wird über Natriumsulfat getrocknet, filtriert, im Vakuum bei 30" vom Ather befreit, und der ölige Rückstand im Hochvakuum bei einem Druck von 0,05 mm Hg und einer Temperatur von 160-165"destilliert. Nach Umkristallisation aus ÄthanollWasser schmilzt der erhaltene p-n-Butoxy-benzoesäure- (l-methyl-4-piperi- dyl)-ester bei 5557 .
Beispiel 3
Eine Lösung von 46,0 g 1-Methyl-4-oxy-piperidin in 200 cm3 Benzol wird mit einer Lösung von 85,3 g Anissäurechlorid in 200 cm3 Benzol versetzt, bei einer Ölbadtemperatur von I00 2 Stunden am Rückfluss zum Sieden erhitzt und dann über Nacht bei Zimmertemperatur stehengelassen. Die ausgefallene Substanz wird abfiltriert, in 500 cm3 Wasser gelöst und nach Zusatz von Tierkohle filtriert. Dann wird unter Eiskühlung mit etwa 400 cm3 2-n. Sodalösung phenol phthaleinalkalisch gemacht und ausgeäthert. Nach dem Eindampfen der Ätherlösung wird der ölige Rückstand bei 11 mm Hg destilliert. Der Anissäure (1-methyl-4-piperidyl)-ester destilliert bei 11 mm Hg bei 195 .
Wird die Lösung des Esters in der äquivalenten Menge I-n. Salzsäure im Vakuum bei 50"zur Trockne eingeengt, und der Rückstand in der 4fachen Menge siedendem abs. Athanol gelöst, so fällt nach Zusatz der 10fachen Menge Äther das Hydrochlorid des Anissäure- (1-methyl-4-piperidyl)-esters kristallin aus.
Smp. 222-2240.
Beispiel 4
Eine Lösung von 23,0 g I-Methyl-4-oxy-piperidin in 100 cm3 Benzol wird mit einer Lösung von 40,4 g p-Athoxy-benzoesäurechlorid in 100 cm3 Benzol versetzt. Nach zweistündigem Erhitzen zum Sieden am Rückfluss wird das Reaktionsgemisch über Nacht bei Zimmertemperatur stehengelassen. Die Lösung der ausgefallenen Substanz in 400 cm3 Wasser wird nach Zugabe von Tierkohle filtriert, unter Eisküh- lung mit etwa 225 cm3 2-n. Sodalösung phenol phthalein-alkalisch gemacht und ausgeäthert. Nach dem Eindampfen der getrockneten Atherlösung wird der ölige Rückstand destilliert.
Der p-Athoxy-benzoe säure- (I-methyl-4-piperidyl)-ester siedet unter I I mm Hg bei 2000.
Das Hydrochlorid wird hergestellt, indem man den Ester in der äquivalenten Menge 1-n. Salzsäure löst, die Lösung im Vakuum bei 50 zur Trockne verdampft, den Rückstand in der dreifachen Menge siedendem abs. Athanol unter Zusatz von Tierkohle löst, filtriert und das Filtrat mit der 10fachen Menge Äther versetzt, wobei das Hydrochlorid des p-Äthoxy benzoesdure- (I-methyl-4-piperidyl)-esters auskristallisiert. Smp. 219-221 .
Beispiel 5
Eine Lösung von 46,0 g l-Methyl-4-oxy-piperidin in 200 cyme Benzol wird mit einer Lösung von 74,0 g p-Toluylsäure-chlorid in 200 cm3 Benzol versetzt, 2 Stunden zum Sieden am Rückfluss erhitzt und über Nacht stehengelassen. Die ausgefallene Substanz wird abfiltriert, in 700 cm3 Wasser gelöst und unter Eiskühlung mit etwa 400 cm3 2-n. Sodalösung phenolphthaleinalkalisch gemacht. Der Ester wird ausge- äthert, der Atherauszug über Natriumsulfat getrocknet und im Vakuum eingedampft. Der ölige Rück- stand wird destilliert. p-Toluylsäure- (I-methyl-4- piperidyl)-ester siedet unter 11 mg Hg bei 172.
Die Lösung des Esters in der äquivalenten Menge e 1-n. Salzsäure wird bei 11 mm und 50 zur Trockne eingedampft, der Rückstand in der 6fachen Menge siedendem abs. Athanol gelöst, filtriert, und das Filtrat mit der 30fachen Menge Ather versetzt, wobei das Hydrochlorid des p-Toluylsäure-(l-methyl-4- piperidyl)-esters auskristallisiert. Smp. 211-213".
Beispiel 6
Eine Lösung von 46,0 g 1-Methyl-4-oxy-piperidin in 200 cms Benzol wird mit einer Lösung von g p-Chlor-benzoesäurechlorid in 200 cm :' Benzol ver setzt, 2 Stunden zum Sieden am Rückfluss erhitzt und über Nacht bei Zimmertemperatur stehengelassen.
Man filtriert ab, löst den Niederschlag bei 40'in 700 cm3 Wasser, filtriert nach Zugabe von Tier kohle und macht die Lösung mit etwa 400 cm3 2-n.
Sodalösung alkalisch. Der ausgefallene p-Chlor- benzoesäure- (l-methyl-4-piperidyl)-ester wird abfil triert und aus der lOfachen Menge 57proz. Athanol umkristallisiert. Smp. 77-79 .
Die Lösung des Esters in der vierfachen Menge l-n. Salzsäure wird im Vakuum bei 50 zur Trockne verdampft, der Rückstand in der dreifachen Menge siedendem abs. Athanol unter Zusatz von Tierkohle gelöst, filtriert, und das Filtrat mit der lOfachen Menge Ather verdünnt, wobei das Hydrochlorid des p-Chlor-benzoesäure- (1-methyl-4-piperidyl)-esters kristallin ausfällt. Smp. 222-224 .
Beispiel 7
Eine Lösung von 5,6 g 1-Methyl-4-oxy-piperidin in 75 cm3 Benzol wird unter Rühren oder Schütteln mit einer Suspension von 13,2 g p-Butylaminobenzoesäurechlorid-hydrochlorid in 75 cm3 Benzol versetzt und 2 Stunden zum Sieden am Rückfluss erhitzt. Von aussen wird mit Eiswasser auf 20 abgekühlt, und das Benzol abdekantiert. Der Rückstand wird in 150 cm3 Wasser, dem einige Kubikzentimeter verdünnte Salzsäure zugesetzt sind, gelöst, nach Zusatz von Tierkohle filtriert, und das Filtrat mit etwa 250 cm3 2-n. Sodalösung phenolphthalein-alkalisch gemacht. Die Lösung der ausgefallenen Substanz in Ather wird über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird destilliert.
Der p Butylamino-benzoesäure- (1-methyl-4-piperidyl)-ester destilliert bei 0,05 mm Hg bei 185-195 . Das kristallin erstarrte Destillat wird aus der lOfachen Menge 50proz. Athanol umkristallisiert. Smp. 109 bis 111o.

Eine Mischung von 19,0 g p-Butylamino-benzoe säure-äthylester, 39,5 g 1-Methyl-4-oxy-piperidin und 1, 0 g pulverisiertes Natriumäthylat wird während 2 Stunden mit absteigendem Kühler in einem Olbad von 200"erhitzt. Ist die Destillation von Athanol beendet, wird abgekühlt, mit 250 cm3 Aceton digeriert und abfiltriert. Das Filtrat wird im Vakuum bei 50 eingedampft, und der Rückstand destilliert. Der p Butylamino-benzoesäure- (1-methyl-4-piperidyl)-ester siedet bei 0,05 mm Hg bei 185195 und zeigt einen Smp. von 109-111 .
Das Monohydrochlorid wird hergestellt, indem man die Lösung der Verbindung in der äquivalenten Menge l-n. Salzsäure im Vakuum bei 50 eindampft, die Lösung des Rückstandes in der 10fachen Menge siedendem Aceton filtriert und das Filtrat mit der lOfachen Menge Ather versetzt. Das kristalline Monohydrochlorid des p-Butylamino-benzoesäure-(1-me- thyl-4-piperidyl)-esters hat den Smp. 177-179 .
Beispiel 9
Eine Lösung von 3,1 g 1-Methyl-4-oxy-piperidin in 40 cm3 Benzol wird mit einer Suspension von 7,3 g p-Methoxyäthylamino-benzoesäurechlorid-hydro- chlorid in 40 cm3 Benzol versetzt und 2 Stunden zum Sieden am Rückfluss erhitzt. Nach dem Abkühlen wird das Benzol abdekantiert, der Rückstand in 100 cm3 Wasser gelost, nach Zusatz von Tierkohle filtriert, unter Eiskühlung mit etwa 100 cm3 2-n. Sodalösung phenolphthalein-alkalisch gemacht und die ölige Substanz in Chloroform aufgenommen. Der Chloroformauszug wird mit Natriumsulfat getrocknet, das Chloroform im Vakuum bei 50 verdampft und der Rückstand im Hochvakuum destilliert. Der p Methoxyäthylamino-benzoesäure- (1-methyl-4-piperidyl)-ester siedet unter 0,15 mm Hg bei 203 .
Beispiel 10
Eine Mischung von 28,6 g p-Methoxyäthylamino benzoesäure-äthylester, 50,0 g 1-Methyl-4-oxy-pipe- ridin und 1, 5 g Natriumäthylat (pulv.) wird während 2 Stunden mit absteigendem Kühler in einem Ölbad von 200 erhitzt. Destilliert kein Athanol mehr ab, wird abgekühlt, mit 350 cm3 Aceton digeriert und abfiltriert. Das Filtrat wird im Vakuum bei 50 eingedampft und der Rückstand destilliert. p-Methoxy äthylamino-benzoesäure- (l-methyl-4-piperidyl)-ester siedet unter 0,15 mm Hg bei 203 .
Das Monohydrochlorid wird hergestellt, indem die Lösung der Verbindung in der äquivalenten Menge l-n. Salzsäure im Vakuum bei 50 zur Trockne eingedampft wird. Der Rückstand wird in der 20fachen Menge siedendem Athanol/Aceton (1 : 3) gelöst und mit der 20fachen Menge Ather versetzt, wobei das Monohydrochlorid des p-Methoxyäthylamino-benzoe säure- (1-methyl-4-piperidyl)-esters ausfällt. Smp. 169 bis 171 .
Beispiel 11 N-a-Butyl-antlzranilsaure- (l-metlayl-4-piperidyl)-ester
Eine Mischung von 65,2 g N-n-Butyl-anthranil säure-äthyl-ester (Sdp. 160 112 mm Hg, hergestellt aus N-n-Butyl-anthranilsäure (Smp. 80-81 ) durch Verestern mit abs. Athanol in Anwesenheit von konz.
Schwefelsäure), 134,5 g 1-Methyl-4-oxy-piperidin (Sdp. 200 ) und 7,0g Natriumäthylat (pulv.) wird während 2 Stunden am absteigenden Kühler in einem Ölbad von 200 erhitzt. Ist die Destillation von Athanol beendet, wird abgekühlt, mit 700 cm3 Äther digeriert und abfiltriert. Das Filtrat wird eingedampft und der Rückstand destilliert. Nach Abtrennen des überschüssigen 1-Methyl-4-oxy-piperidins bei 15 mm Hg wird im Hochvakuum ein unter 0, 006 mm Hg bis 142 übergehender Vorlauf abgetrennt. Die unter dem gleichen Druck bei 142-144 übergehende Hauptfraktion wird aufgefangen.
Der analysenreine N-n-Butyl-anthranilsäure- (1-methyl-4-piperidyl)-ester hat den Sdp. 143 /0, 006 mm Hg.
Das Monohydrochlorid wird hergestellt, indem die Lösung von 40,62 g der freien Base in 140,5 cm3 1-n. Salzsäure im Vakuum zum Sirup eingeengt wird.
Der Rückstand wird in 250 cm3 Isopropanol gelöst und zur Entfernung des restlichen Wassers im Vakuum nochmals eingedampft. Der Rückstand wird aus 250 cm3 siedendem Isopropanol umkristallisiert.
Das analysenreine Monohydrochlorid des N-n-Butyl anthranilsäure- (l-methyl-4-piperidyl)-esters hat den konstanten Smp. 169-171 .
Beispiel 12
O-n-Butyl-salicylsäure- (l-methyl4-pperdyl)-ester
Eine Mischung von 54,3 g O-n-Butyl-salicylsäure- methylester (Sdp. 141 /10mm Hg, hergestellt aus Salicylsäure-methylester durch Veräthern mit n-Butylbromid in Aceton bei Anwesenheit von Pottasche), 119g 1-Methyl-4-hydroxy-piperidin (Sdp. 200 ) und 4,4 g Natriumäthylat (pulv.) wird während 2 Stunden am absteigenden Kühler in einem Ölbad von 200 erhitzt. Ist die Destillation von Methanol beendet, wird abgekühlt, mit 750 cm3 Aceton digeriert und abfiltriert. Das Filtrat wird eingedampft und der Rückstand destilliert.
Nach Abtrennen des überschüssigen 1-Methyl-4-hydroxy-piperidins bei 10 mm Hg wird im Hochvakuum ein unter 0,01 mm Hg bis 147 übergehender Vorlauf abgetrennt. Die unter dem gleichen Druck bei 147-153 übergehende Hauptfraktion wird aufgefangen. Der analysenreine O-n Butyl-salicylsäure- (1-methyl-4-piperidyl)-ester hat den Sdp. 150 l0, 01 mm Hg.
Das Hydrochlorid wird hergestellt, indem eine Lösung von 17,0 g der freien Base in 95 cm3 abs.
Athanol mit äthanolischem Chlorwasserstoff bis zur kongosauren Reaktion versetzt wird. Nach Zugabe von 150 cm3 Äther kristallisiert das Hydrochlorid aus. Durch Umkristallisieren aus 20 cm3 abs. Athanol und 100 cm3 Äther erhält man das analysenreine Hydrochlorid des O-n-Butylsalicylsäure-(1-methyl-4- piperidyl)-esters vom konstanten Smp. 1471495.

Ethanol is dissolved with the addition of animal charcoal, filtered and 10 times the amount of ether is added to the filtrate, the hydrochloride of p-ethoxy benzoic acid (I-methyl-4-piperidyl) ester crystallizing out. M.p. 219-221.
Example 5
A solution of 46.0 g of 1-methyl-4-oxy-piperidine in 200 cm of benzene is mixed with a solution of 74.0 g of p-toluic acid chloride in 200 cm3 of benzene, heated to reflux for 2 hours and overnight ditched. The precipitated substance is filtered off, dissolved in 700 cm3 of water and with ice-cooling with about 400 cm3 2-n. Soda solution made phenolphthalein alkaline. The ester is extracted with ether, the ether extract is dried over sodium sulfate and evaporated in vacuo. The oily residue is distilled. p-Toluic acid (I-methyl-4-piperidyl) ester boils below 11 mg Hg at 172.

Example 6
A solution of 46.0 g of 1-methyl-4-oxy-piperidine in 200 cms of benzene is mixed with a solution of g of p-chloro-benzoic acid chloride in 200 cm: of benzene, heated to reflux for 2 hours and overnight left to stand at room temperature.
It is filtered off, the precipitate is dissolved in 700 cm3 of water at 40 ', filtered after adding animal charcoal and the solution is made with about 400 cm3 2-n.
Soda solution alkaline. The precipitated p-chlorobenzoic acid (l-methyl-4-piperidyl) ester is filtered off and from the 10-fold amount 57 percent. Ethanol recrystallized. M.p. 77-79.

Example 7
A solution of 5.6 g of 1-methyl-4-oxy-piperidine in 75 cm3 of benzene is mixed with a suspension of 13.2 g of p-butylaminobenzoic acid chloride hydrochloride in 75 cm3 of benzene while stirring or shaking and the mixture is refluxed for 2 hours heated. From the outside it is cooled to 20 with ice water, and the benzene is decanted off. The residue is dissolved in 150 cm3 of water to which a few cubic centimeters of dilute hydrochloric acid have been added, filtered after adding animal charcoal, and the filtrate with about 250 cm3 of 2-n. Soda solution made phenolphthalein-alkaline. The solution of the precipitated substance in ether is dried over sodium sulfate and evaporated. The residue is distilled.
The p-butylaminobenzoic acid (1-methyl-4-piperidyl) ester distills at 0.05 mm Hg at 185-195. The crystalline solidified distillate is 50 per cent from the tenfold amount. Ethanol recrystallized. M.p. 109 to 111o.
Example 8
A mixture of 19.0 g of ethyl p-butylamino-benzoate, 39.5 g of 1-methyl-4-oxy-piperidine and 1.0 g of powdered sodium ethylate is heated in a 200 "oil bath for 2 hours with a descending cooler When the distillation of ethanol has ended, the mixture is cooled, digested with 250 cm 3 of acetone and filtered off. The filtrate is evaporated in vacuo at 50 and the residue is distilled. The p-butylamino-benzoic acid (1-methyl-4-piperidyl) ester boils at 0.05 mm Hg at 185195 and shows a m.p. of 109-111.
The monohydrochloride is prepared by adding the solution of the compound in the equivalent amount of l-n. Hydrochloric acid was evaporated in vacuo at 50, the solution of the residue was filtered in 10 times the amount of boiling acetone and the filtrate was treated with 10 times the amount of ether. The crystalline monohydrochloride of p-butylaminobenzoic acid (1-methyl-4-piperidyl) ester has a melting point of 177-179.
Example 9
A solution of 3.1 g of 1-methyl-4-oxy-piperidine in 40 cm3 of benzene is mixed with a suspension of 7.3 g of p-methoxyethylaminobenzoic acid chloride hydrochloride in 40 cm3 of benzene and refluxed for 2 hours heated. After cooling, the benzene is decanted off, the residue is dissolved in 100 cm3 of water, after the addition of animal charcoal it is filtered, while cooling with ice with about 100 cm3 of 2-n. Soda solution made phenolphthalein alkaline and the oily substance was taken up in chloroform. The chloroform extract is dried with sodium sulfate, the chloroform is evaporated in vacuo at 50 and the residue is distilled in a high vacuum. The p methoxyethylamino-benzoic acid (1-methyl-4-piperidyl) ester boils below 0.15 mm Hg at 203.
Example 10
A mixture of 28.6 g of p-methoxyethylamino benzoic acid ethyl ester, 50.0 g of 1-methyl-4-oxy-pipe- ridin and 1.5 g of sodium ethylate (powder.) Is in an oil bath for 2 hours with a descending cooler 200 heated. If no more ethanol distills off, it is cooled, digested with 350 cm3 of acetone and filtered off. The filtrate is evaporated in vacuo at 50 and the residue is distilled. p-Methoxyethylaminobenzoic acid (l-methyl-4-piperidyl) ester boils below 0.15 mm Hg at 203.
The monohydrochloride is prepared by adding the solution of the compound in the equivalent amount of l-n. Hydrochloric acid is evaporated to dryness in vacuo at 50. The residue is dissolved in 20 times the amount of boiling ethanol / acetone (1: 3) and treated with 20 times the amount of ether, the monohydrochloride of p-methoxyethylamino-benzoic acid (1-methyl-4-piperidyl) ester precipitating. M.p. 169 to 171.
Example 11 N-α-Butyl-antlzranilic acid (1-metlayl-4-piperidyl) ester
A mixture of 65.2 g of N-n-butyl-anthranilic acid ethyl ester (bp. 160 112 mm Hg, prepared from N-n-butyl-anthranilic acid (melting point 80-81) by esterification with absolute ethanol in the presence of conc.
Sulfuric acid), 134.5 g of 1-methyl-4-oxy-piperidine (boiling point 200) and 7.0 g of sodium ethylate (powder) are heated in a descending cooler in an oil bath of 200 for 2 hours. When the distillation of ethanol is over, it is cooled, digested with 700 cm3 of ether and filtered off. The filtrate is evaporated and the residue is distilled. After the excess 1-methyl-4-oxy-piperidine has been separated off at 15 mm Hg, a forerun under 0.006 mm Hg to 142 is separated off in a high vacuum. The main fraction passing over under the same pressure at 142-144 is collected.
The analytically pure N-n-butyl-anthranilic acid (1-methyl-4-piperidyl) ester has a boiling point of 143/0, 006 mm Hg.
The monohydrochloride is prepared by dissolving the solution of 40.62 g of the free base in 140.5 cm3 of 1-n. Hydrochloric acid is concentrated in vacuo to a syrup.
The residue is dissolved in 250 cm3 of isopropanol and evaporated again in vacuo to remove the remaining water. The residue is recrystallized from 250 cm3 of boiling isopropanol.
The analytically pure monohydrochloride of N-n-butyl anthranilic acid (l-methyl-4-piperidyl) ester has a constant melting point of 169-171.
Example 12
O-n-Butyl-salicylic acid (1-methyl-4-pperdyl) ester
A mixture of 54.3 g of on-butyl salicylic acid methyl ester (bp. 141 / 10mm Hg, produced from salicylic acid methyl ester by etherification with n-butyl bromide in acetone in the presence of potash), 119 g 1-methyl-4-hydroxy- Piperidine (boiling point 200) and 4.4 g of sodium ethylate (powder) are heated in a 200 oil bath on a descending cooler for 2 hours. When the distillation of methanol has ended, it is cooled, digested with 750 cm3 of acetone and filtered off. The filtrate is evaporated and the residue is distilled.
After the excess 1-methyl-4-hydroxypiperidine has been separated off at 10 mm Hg, a forerun under 0.01 mm Hg to 147 is separated off in a high vacuum. The main fraction passing over under the same pressure at 147-153 is collected. The analytically pure O-n butyl-salicylic acid (1-methyl-4-piperidyl) ester has a boiling point of 150 l0.01 mm Hg.
The hydrochloride is prepared by adding a solution of 17.0 g of the free base in 95 cm3 abs.
Ethanol is mixed with ethanolic hydrogen chloride until the Congo acid reaction occurs. After adding 150 cm3 of ether, the hydrochloride crystallizes out. By recrystallization from 20 cm3 abs. Ethanol and 100 cm3 of ether give the analytically pure hydrochloride of O-n-butylsalicylic acid (1-methyl-4-piperidyl) ester of constant melting point 1471495.
