Das Verfahren wird beispielsweise so ausgeführt, dass l-Methyl-4-hydroxy-piperidin mit einem substituierten Benzoesäure-alkylester in Gegenwart eines Kondensationsmittels, z. B. Natriumäthylat oder Natriummetall, auf höhere Temperatur, vorzugsweise 150-200 , erhitzt wird, wobei der im Verlauf der Umesterung freigewordene aliphatische Alkohol abdestilliert. Der Rückstand liefert bei der Fraktionierung im Vakuum das Endprodukt. Ist das für die Acylierung des l-Alkyl-4-hydroxy-piperidins verwendete substituierte Benzoesäurederivat ein Säurehalogenid, dann gestaltet sich der Umsatz so, dass das l-Alkyl-4-hydroxy-piperidin-Derivat mit dem substituierten Benzoylhalogenid zusammen in einem inerten organischen Lösungsmittel, z. B. Benzol, Toluol, Xylol, gelöst wird. Die Lösung der Reaktionspartner wird bei Zimmertemperatur undloder bei erhöhter Temperatur, z.
B. in einem Dampfbad, stehengelassen, schliesslich wird der gebildete Ester, der als Hydrohalogenid vorliegt, durch Zusatz eines Alkalis, z. B. Soda, in Freiheit gesetzt und nach bekannten Methoden isoliert und gereinigt.
Die nach dem vorliegenden Verfahren hergestellten, bisher unbekannten Piperidin-Derivate lassen sich im Vakuum ohne Zersetzung destillieren. Sie sind bei Zimmertemperatur flüssige oder feste kristalline Basen, die mit Säuren beständige Salze bilden.
Sie zeichnen sich durch therapeutisch verwertbare pharmakodynamische Eigenschaften aus. Sowohl die freien Basen als auch ihre Salze zeigen bei sehr guter lokaler Verträglichkeit und geringer Toxizität eine starke lokalanästhetische Wirkung, die sich sowohl bei der Oberflächen- als auch bei der Leitungsanästhesie nachweisen lässt. Die neuen Verbindungen sollen in der Therapie verwendet werden.
In den nachfolgenden Beispielen sind die Schmelz- und Siedepunkte in Grad Celsius und unkorrigiert angegeben.
Beispiel 1 p-(Phenoxyäthyl-amino)-benzoesäure-( 1 -methyl-
4pipendyl)-ester
Eine Mischung von 50,0 g p-(Phenoxyäthylamino)-benzoesäure-äthylester (Smp. 108-110 , hergestellt aus p-Amino-benzoesäure-äthylester durch Alkylieren mit Bromäthylphenyl-äther in n-Butanol bei Anwesenheit von Pottasche), 81,0 g l-Methyl- 4-hydroxy-piperidin (Sdp. 2000) und 4,0 g Natrium äthylat (pulv.) wird während 2 Stunden am absteigenden Kühler in einem Ölbad von 2000 erhitzt.
Ist die Destillation von Äthanol beendet, wird abgekühlt, mit 500 cm3 Aceton digeriert und abfiltriert Das Filtrat wird eingedampft und der Rückstand destilliert Nach Abtrennen des überschüssigen l-Me- thyl-4-hydroxy-piperidins bei 10 mm Hg wird ein im Hochvakuum unter 0,1 mm bis 210 übergehender Vorlauf abgetrennt. Die unter dem gleichen Druck bei 210-2300 übergehende Hauptfraktion wird aufgefangen. Aus Essigester kristallisiert, erhält man den analysenreinen p-(Phenoxy-äthyl-amino)-benzoe säure-(l-methyl-4-piperidyl)-ester vom Smp. 130 bis 132".
Zur Herstellung des Fumarats wird eine Lösung von 10,0 g Base in 50 cm3 abs. Äthanol mit einer Lösung von 3,5 g Maleinsäure in 50 cm3 abs.
Äthanol versetzt und noch 200 cm3 Äther hinzugefügt. Das auskristallisierte Fumarat wird aus
100 cm3 abs. Äthanol und 300 cm3 Äther umkristallisiert Das analysenreine Fumarat des p-(Phenoxy äthyl-amino)-benzoesäure- (1- methyl-4-piperidyl)- esters hat den Smp. 139-1410 (Zers.).
Beispiel 2
N-(p-Chlor)-phenoxyäthyl-anthranilsäure (1-methyI-4-piperidyl)-ester
Eine Mischung von 20 g N-(p-Chlor)-phenoxy äthyl-anthranilsäure-methylester (Smp. 85O, hergestellt aus Anthranilsäure-methylester durch Alkylieren mit p-Chlorphenyl-ss-bromäthyläther in Dimethylformamid bei Anwesenheit von wasserfreier Soda), 85 g 1-Methyl-4-hydroxy-piperidin und 500 mg Natriumäthylat wird während 2 Stunden am absteigenden Kühler in einem Ölbad von 2000 erhitzt. Nach dem Abkühlen wird das Reaktionsprodukt in 200 cm3 Äther aufgenommen, diese Lösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und nach Entfernen des Äthers fraktioniert.
Die Hauptfraktion der Base destillierte bei 212 bis 2130/0,07 Torr. Das stark gelb gefärbte viskose Ö1 wird in wenig Benzol aufgenommen und diese Lösung durch eine Schicht Aluminiumoxyd filtriert. Die aus Petroläther kristallisierte Base schmilzt bei 70 bis 73 .
Das Chlorhydrat der so erhaltenen Base wird hergestellt, indem man obige Lösung mit einem geringen Überschuss alkoholischer Salzsäure versetzt, im Vakuum eindampft und das Chlorhydrat aus Isopropanol umkristallisiert. Smp. 189, 0-189, 5 .
Beispiel 3
N-(p-Methoxy)-phenoxyäthyl-anthranilsäure (1 -methyl-4-piperidyl)-ester
Eine Mischung von 9,9 g N - (p - Methoxy)- phenoxyäthyl - anthranilsäure - methylester (Smp. 92 bis 94O, hergestellt aus Anthranilsäure-methylester durch Alkylieren mit p-Methoxyphenyl-ss-bromäthyl- äther in Dimethylformamid bei Anwesenheit von wasserfreier Soda), 43 g 1-Methyl-4-hydroxy-pipe- ridin und 250 mg Natriummethylat wird während 2 Stunden am absteigenden Kühler im Ölbad von 2000 erhitzt. Überschüssiges Methylpiperidinol wird im Wasserstrahlvakuum abdestilliert, der Rückstand in Benzol aufgenommen und mit verdünnter Salzsäure extrahiert.
Die salzsaure Lösung wird nochmals mit Benzol ausgezogen und schliesslich mit Natronlauge alkalisch gestellt. Die freiwerdende Base wird mit Benzol extrahiert und nach dem Trocknen und Einengen der Lösung durch eine kurze Aluminiumoxydsäule filtriert. Die Base kristallisiert aus Petrol äther in feinen Nadeln, Fp. 86-880.
Zum Chlorhydrat gelangt man durch Fällen der Lösung der Base in Ather mit ätherischer Salzsäure und Kristallisation aus Isopropanol. Smp. 174,0 bis 174,50.
Beispiel 4 N-Phenoxypropyl-anthranilsäure-( 1 -methyl-
4-piperidyl)-ester
Eine Mischung von 13,5 g N-Phenoxypropylanthranilsäure-methylester (Sdp. 2300/0,05 Torr, hergestellt aus Anthranilsäure-methylester durch Alkylieren mit Phenol-y-brompropyläther in Dimethylformamid bei Anwesenheit wasserfreier Soda), 57 g l-Methyl-4-hydroxy-piperidin und 335 mg Natriummethylat wird während 2 Stunden im Ölbad von 2000 erhitzt. Überschüssiges Methyl-piperidinol wird im Wasserstrahlvakuum abdestilliert, der Rückstand in Benzol aufgenommen und mit verdünnter Salzsäure extrahiert. Die salzsaure Phase wird nochmals mit Benzol geschüttelt, von letzterem abgetrennt und mit Natronlauge alkalisch gestellt. Man zieht wiederum mit Benzol aus, engt die Lösung der Base im Vakuum weitgehend ein und filtriert durch eine kurze Aluminiumoxydsäule.
Nach dem Eindampfen der gereinigten Basenlösung kristallisiert das Amin und weist nach dem Umkristallisieren aus Petroläther den Smp. 73-74O auf.
Das Chlorhydrat wird erhalten, wenn man die obige Base in ätherischer Lösung mit einem geringen Überschuss von ätherischer Salzsäure fällt und das rohe Salz aus Isopropanol umkristallisiert. Smp.

Beispiel 5 N-Phenoxyäthyl-m-aminobenzoesäure-(l-methyl-
4-piperidyl)-ester
Eine Mischung von 10 g N-Phenoxyäthyl-maminobenzoesäure-methylester (Smp. 78-790, hergestellt aus m-Aminobenzoesäure-methylester durch Alkylieren mit Phenyl-bromäthyläther in Dimethylformamid bei Anwesenheit von wasserfreier Soda), 43 g 1-Methyl-4-hydroxy-piperidin und 500 mg Natriummethylat wird während 4 Stunden am absteigenden Kühler im Ölbad von 2000 erhitzt. Nach dem Abkühlen wird in Äther aufgenommen, die Lösung mit Wasser gewaschen, über Natriumsulfat getrocknet und destilliert. Die Base geht als stark gefärbtes Ö1 bei 115-119"/0,0015 Torr über.
Das Tartrat wird erhalten durch Vereinigung von molaren Mengen Base und Weinsäure in alkoholischer Lösung. Das Salz wird aus 250/obigem Alkohol umkristallisiert und schmilzt bei 155-156 .

Example 1 p- (phenoxyethyl-amino) -benzoic acid- (1 -methyl-
4pipendyl) ester
A mixture of 50.0 g of ethyl p- (phenoxyethylamino) benzoate (melting point 108-110, prepared from ethyl p-amino-benzoate by alkylation with bromoethylphenyl ether in n-butanol in the presence of potash), 81, 0 g of 1-methyl-4-hydroxypiperidine (boiling point 2000) and 4.0 g of sodium ethylate (powder) are heated in a 2000 oil bath on a descending cooler for 2 hours.
When the distillation of ethanol has ended, the mixture is cooled, digested with 500 cm3 of acetone and filtered off. The filtrate is evaporated and the residue is distilled. After separating off the excess 1-methyl-4-hydroxy-piperidine at 10 mm Hg, a vacuum under 0 , 1 mm to 210 passing flow cut off. The main fraction passing over under the same pressure at 210-2300 is collected. Crystallized from ethyl acetate, the analytically pure p- (phenoxy-ethyl-amino) -benzoic acid (l-methyl-4-piperidyl) ester with a melting point of 130 to 132 "is obtained.

Example 2
N- (p-chloro) -phenoxyethyl-anthranilic acid (1-methyl-4-piperidyl) ester
A mixture of 20 g of N- (p-chloro) -phenoxy-ethyl-anthranilic acid methyl ester (melting point 85O, prepared from anthranilic acid methyl ester by alkylating with p-chlorophenyl-β-bromoethyl ether in dimethylformamide in the presence of anhydrous soda), 85 g 1-methyl-4-hydroxypiperidine and 500 mg of sodium ethylate are heated in a 2000 oil bath on a descending cooler for 2 hours. After cooling, the reaction product is taken up in 200 cm3 of ether, this solution is washed with water, dried over sodium sulfate and, after removal of the ether, fractionated.
The major fraction of the base distilled at 212-2130 / 0.07 torr. The strongly yellow colored viscous oil is taken up in a little benzene and this solution is filtered through a layer of aluminum oxide. The base crystallized from petroleum ether melts at 70 to 73.
The chlorohydrate of the base obtained in this way is prepared by adding a small excess of alcoholic hydrochloric acid to the above solution, evaporating in vacuo and recrystallizing the chlorohydrate from isopropanol. 189,0-189,5.
Example 3

The chlorine hydrate is obtained by precipitating the solution of the base in ether with ethereal hydrochloric acid and crystallization from isopropanol. M.p. 174.0 to 174.50.
Example 4 N-phenoxypropyl-anthranilic acid- (1 -methyl-
4-piperidyl) ester
A mixture of 13.5 g of N-phenoxypropylanthranilic acid methyl ester (bp. 2300 / 0.05 Torr, prepared from anthranilic acid methyl ester by alkylation with phenol-γ-bromopropyl ether in dimethylformamide in the presence of anhydrous soda), 57 g of 1-methyl-4 Hydroxypiperidine and 335 mg of sodium methylate are heated in a 2000 oil bath for 2 hours. Excess methyl piperidinol is distilled off in a water jet vacuum, the residue is taken up in benzene and extracted with dilute hydrochloric acid. The hydrochloric acid phase is shaken again with benzene, separated from the latter and made alkaline with sodium hydroxide solution. It is again extracted with benzene, the solution of the base is largely concentrated in vacuo and filtered through a short aluminum oxide column.
After evaporation of the purified base solution, the amine crystallizes and, after recrystallization from petroleum ether, has a melting point of 73-740.
The chlorohydrate is obtained if the above base is precipitated in an ethereal solution with a slight excess of ethereal hydrochloric acid and the crude salt is recrystallized from isopropanol. M.p.

4-piperidyl) ester
A mixture of 10 g of N-phenoxyethyl-maminobenzoic acid methyl ester (melting point 78-790, prepared from m-aminobenzoic acid methyl ester by alkylation with phenyl bromoethyl ether in dimethylformamide in the presence of anhydrous soda), 43 g of 1-methyl-4-hydroxy -piperidine and 500 mg of sodium methylate is heated for 4 hours on a descending cooler in an oil bath of 2000. After cooling, it is taken up in ether, the solution is washed with water, dried over sodium sulfate and distilled. The base passes over as a strongly colored oil at 115-119 "/ 0.0015 Torr.
