Verfahren zur Herstellung neuer Azaverbindungen Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 1-Aza-2-niederalkyl-6- (tert. - amino - niederalkylamino)-9-methyl-thioxantho- nen, besonders von 1-Aza-2-niederalkyl-6-(di-nieder- alkylamino -niederalkylamino)-9 -methyl-thioxantho- nen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man 1-Aza-2-niederalkyl-9-methyl- 6-X-thioxanthone mit Verbindungen der Formel R-Y-Z umsetzt, wobei R einen in eine tertiäre Aminogruppe überführbaren Rest, Y einen niederen Alkylenrest und X und Z zwei unter Hinterlassung der in einem von ihnen enthaltenen Gruppe. -NH- unter den Reaktionsbedingungen abspaltbare Reste darstellen, und in den erhaltenen Verbindungen den Rest R in eine tertiäre Aminogruppe überführt. Dabei ist X z.
B. ein Halogenatom und Z eine Aminogruppe, oder X eine Aminogruppe und Z eine mit einer starken Säure reaktionsfähig veresterte Oxygruppe, wie ein Halogenatom. R ist z. B. eine freie oder mit starken Säuren reaktionsfähig veresterte Oxygruppe, die durch Umsetzung mit einem sekundären Amin, gegebenenfalls nach Veresterung mit einer starken Säure, wie einer Halogenwasserstoffsäure, in die tertiäre Aminogruppe übergeführt wird.
Die neuen Verbindungen können weitere Sub- stituenten enthalten, wie insbesondere eine niedere Alkylgruppe in 4-Stellung. In erster Linie werden nach der vorliegenden Erfindung 1-Aza-2-niederalkyl- 6-diäthylaminäthylamino-9-methyl-thioxanthone, z. B. 1-Aza-2-methyl-6-diäthylaminoäthylamino-9-methyl- thioxanthon hergestellt.
Die genannten Verbindungen weisen eine gegen- über Schistosomen und Amoeben ausgeprägte Wirkung auf und können bei humaner Schistosomiasis und Amoebiasis als Heilmittel Verwendung finden. Sie besitzen im Vergleich zum bekannten 1-Aza-6- diäthylaminoäthylamino-9-methyl-thioxanthon besonders wertvolle Eigenschaften.
Das erfindungsgemässe Verfahren kann in An- oder Abwesenheit von Verdünnungsmitteln undloder Kondensationsmitteln undloder Katalysatoren, bei gewöhnlicher oder erhöhter Temperatur, im offenen oder im geschlossenen Gefäss unter Druck durchgeführt werden.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form ihrer Basen oder Salze. Aus den Salzen können die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z.
B. der Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasser- stoffsäure, Essigsäure, Propionsäure, Oxalsäure, Ma- lonsäure, Bernsteinsäure, Äpfelsäure, Methansulfon- säure, Äthansulfonsäure, Oxyäthansulfonsäure, Benzol- oder Toluolsulfonsäure.
Die Ausgangsstoffe sind bekannt oder können in an sich bekannter Weise hergestellt werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden.
Die Temperaturen im folgenden Beispiel sind in Celsiusgraden angegeben.
Beispiel 2,75 g 1-Aza-2,9-dimethyl-6-chlor-thioxanthon werden mit 30 cms Äthanolamin unter Zusatz von 0,5 g Kupferpulver 15 Stunden am Rückfluss gekocht. Nach dem Erkalten wird der Kristallbrei mit 1/2 Liter Wasser verdünnt und das Reaktionsprodukt,

1-Aza-2,9 - dimethyl- 6 -oxyäthylamino-thioxanthon, abgenutscht, mit Wasser neutral gewaschen und getrocknet. Es wird aus viel Alkohol in gelben, glänzenden Nadeln vom F. 216 erhalten. Ausbeute 2,8 g.
2,8 g 1-Aza-2,9-dimethyl-6-oxyäthylamino-thio- xanthon werden mit 25 cm3 Phosphoroxychlorid auf dem Wasserbad 2 Stunden auf 90-100 erhitzt. Dann wird das überschüssige Phosphoroxychlorid im Vakuum abdestilliert, der Rückstand in konz. Salzsäure gelöst, die Lösung mit Kohle filtriert und aus dem Filtrat das Chlorierungsprodukt unter Eiszusatz mit konz. Ammoniak ausgefällt. Der Niederschlag wird abgenutscht, mit Wasser gewaschen, mit 300,
cm-' Äthanol kurz aufgekocht und die alkoholische Lösung von wenig Unlöslichem abfiltriert. Die alkoholische Lösung wird nun mit 25 cm3 Diäthylamin versetzt und 12 Stunden im Rohr auf 120-125 erhitzt. Die gelbbraune Lösung wird eingedampft, der Rückstand mit 100 cm3 10prozentiger Essigsäure erwärmt, die Lösung mit Kohle filtriert und mit Ammoniak basisch gestellt. Der Niederschlag wird abgenutscht, zur Reinigung nochmals in 5 proz. Essigsäure gelöst, die mit Kohle filtrierte Lösung mit 10-n. Natronlauge basisch gestellt und das ausfallende Produkt mit Äther extrahiert.
Die Ätherlösung hinterlässt beim Eindampfen das 1-Aza-2,9-dimethyl-6-diäthylaminoäthylamino- thioxanthon als gelbes Öl, welches aus Äther-Petrol- äther in gelben Nadeln kristallisiert.
Bei Verwendung von Äthanolamin und Morpho- lin bzw. 3-Amino-propanol und Dimethylamin erhält man das 1-Aza-2,9-dimethyl-6-(morpholino-äthyl- amino)-thioxanthon vom F. 135,5-136,5 bzw. das 1-Aza-2,9 - dimethyl - 6 - dimethylaminopropylamino- thioxanthon vom F.95-96 , deren Hydrochloride bei 280-282 bzw.
226-228 schmelzen. Geht man von 1-Aza-2-isobutyl-6-chlor-9-methyl-thioxanthon aus und verwendet Äthanolamin und Diäthylamin, so erhält man das 1-Aza-2-isobutyl-6-diäthylaminoäthyl- amino-9-methyl-thioxanthon vom F. 88 , dessen Hy- drochlorid bei l95-196 schmilzt.
