Verfahren zur Herstellung von 4-Oestren-3,17-diolen und deren Estern Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von hochaktiven, den Auf baustoffwechsel beeinflussenden Steroiden der fol genden Formel:

in der R Wasserstoff, den Benzoylrest oder einen von einer aliphatischen Carbonsäure mit 1 bis 8 Kohlen stoffatomen abgeleiteten Acylrest und A ein Wasser stoffatom oder einen niederen gesättigten oder un gesättigten aliphatischen Kohlenwasserstoffrest be deutet.
Beispielsweise kann R den Formyl-, Acetyl-, Pro- pionyl-, Butyryl-, Pentanoyl-, Hexanoyl-, Cyclo- pentyl-acetyl-, a-Cyclopentyl-propionyl-, Cyclohexyl- acetylrest und A einen niederen Alkylrest, wie zum Beispiel den Methyl- oder Äthylrest,
oder einen grad- kettigen oder verzweigten Propyl- und Butylrest, einen Alkenylrest, zum Beispiel den Vinyl-, Allyl- oder Butenylrest, sowie einen Alkinylrest, zum Bei spiel den Äthinyl-, Propinyl-, Butinylrest oder der gleichen, bedeuten.
Das in 17-Stellung gegebenenfalls substituierte 4-Oestren-3,17-diol sowie seine Mono- und Diester sind verhältnismässig schwach androgene Mittel. Sie sind für die menschliche und veterinäre Therapie in folge ihrer hohen Wirksamkeit bei der Anregung des Aufbaustoffwechsels des Stickstoffes besonders wert voll. Die Verbindungen der eingangs angegebenen For mel werden erfindungsgemäss durch Reduktion der entsprechenden 3-Keto-Verbindungen mit die Kern doppelbindung nicht angreifenden Mitteln hergestellt.
Man kann als Reduktionsmittel zum Beispiel Alumi- niumalkoholate verwenden (Meerwein - Ponndorf - Verley-Reaktion). Besonders günstig sind Alkali- metallaluminiumhydride und -borhydride, wie zum Beispiel Lithiumaluminiumhydrid und Natriumbor- hydrid.
In 17-Stellung durch eine Alkylgruppe substitu ierte 2-Oestren-3,17-diole können auf bequeme Weise durch nachträgliche Hydrierung von erfindungsgemäss hergestellten 17a - Substitutionsprodukten des 4 - Oestren-3,17-diols hergestellt werden, bei denen der 17a-Substituent aus einem ungesättigten aliphati- schen Kohlenwasserstoffrest besteht.
Derartige Reste können Athylen- oder Acetylenbindungen enthalten; es kann sich beispielsweise um den Äthinyl-, Pro- pinyl- und Butinyl-, Vinyl-, Propenyl-, Allylreste und deren höheren Homologe handeln.
Zur Hydrierung beispielsweise von 17a-Äthinyl-4-oestren-3,17-diol kann man diese Verbindung in Lösung oder Suspen sion in einer Wasserstoffatmosphäre in Gegenwart eines Katalysators, beispielsweise eines Edelmetall- katalysators, wie Palladium auf Holzkohle, schütteln. Die Verbindung wird dabei stufenweise über 17a Vinyl-4-oestren-3,17-d'iol zu 17-Äthyl-4-oestren- 3,17-diol reduziert.
Unter diesen Bedingungen wird die im Kern befindliche Doppelbindung nur in ge ringfügigem Masse hydriert. 17-Äthyl-4-oestren-3,17- diol kann ferner vorteilhaft auch durch katalytische Reduktion von 17a-Vinyl-4-oestren-3,17-diol her gestellt werden.
Die zur Herstellung von 17a-Alkenyl- und Al- kinylderivaten von 4-Oestren-3,17-diol benötigten 17-Alkenyl- bzw. 17-Alkinyl-19-nortestosterone kön nen auf einfache Weise durch Anlagerung von Ace tylen, Acetylenhomologen oder ungesättigten organo- metallischen Verbindungen an entsprechende 17- Keto-steroide hergestellt werden.
Beispielsweise kann 17a-Äthinyl-19-nortestosteron durch Anlagerung von Acetylen an 3-Methoxy-2,5(10)-oestradien-17-on und anschliessende mit Säure katalysierte Hydrolyse und Isomerisierung des erhaltenen Produktes her gestellt werden. Die Anlagerung von Allylmagnesium- bromid an 3-Methoxy-2,5(10)-oestradien-17-on und anschliessende saure Hydrolyse und Isomerisierung er gibt 17-Allyl-19-nortestosteron.
In den folgenden Beispielen bedeuten die Teile Gewichtsteile.
<I>Beispiel 1</I> Eine Lösung von 7,2 Teilen 19-Nortesteron in 700 Teilen Äther wird unter Rühren einer Suspension von 6 Teilen Lithium-aluminium-hydrid in 210 Tei len Äther zugesetzt. Man erwärmt das Gemisch unter Rühren 45 Minuten lang auf 35 ; danach wird eine Menge Aceton zugesetzt, die zur Zersetzung des überschüssigen Lithiumaluminiumhydrids ausreicht.
Dann gibt man Wasser zu, trennt die organische Schicht ab und wäscht zuerst mit Wasser und an schliessend mit gesättigter Natriumchloridlösung. Nach dem Trocknen über wasserfreiem Natriumsulfat und dem Filtrieren wird das Lösungsmittel im Vakuum entfernt. Der Rückstand wird in 10 /oiger Lösung von Äthylacetat in Benzol auf eine chromatographi- sehe Kolonne aufgebracht, die 760 Teile Kieselsäure gel enthält.
Die Kolonne wird dann mit Benzol und 5-, 10- und 15ohigen Lösungen von Äthylacetat in Benzol gewaschen. Eluieren mit einer 20o/aigen Lö sung von Äthylacetat in Benzol und wiederholtes Umkristallisieren des Rückstandes aus einem Ge misch von Äthylacetat und Cyclohexan ergibt 4- Oestren-3,17-diol, das bei etwa 202 bis 204 schmilzt. Das Infrarotabsorptionsspektrum zeigt eine breite Bande bei 3,0 und eine Bande bei 6,

Es handelt sich augenscheinlich um das 4-Oestren-3 a,17f-diol. <I>Beispiel 2</I> Einer Lösung von 14,4 Teilen 19-Nortestosteron in 400 Teilen Methanol und 50 Teilen Äthylacetat wird eine Lösung von 10 Teilen Natriumborhydrid in 200 Teilen Methanol zugesetzt. Das Gemisch wird etwa eine Stunde lang bei Raumtemperatur gehalten und zuerst mit 30 Teilen Wasser und dann mit einer zur Herabsetzung des pH-Wertes auf 6,0 ausreichen den Menge Essigsäure behandelt.
Das Reaktions gemisch wird auf einem Dampfbad in einem Stick stoffstrom auf etwa 50 Teile eingeengt. Nach dem Ver dünnen mit 600 Teilen Wasser wird der Nieder schlag abfiltriert und mit Wasser gewaschen. Chro- matographieren über Kieselsäuregel wie im vor stehenden Beispiel ergibt 4-Oestren-3,17-diol mit den gleichen physikalischen Eigenschaften wie das im vorstehenden Beispiel erhaltene Produkt.
<I>Beispiel 3</I> Ein Gemisch aus 10 Teilen 4-Oestren-3,17-diol (Smp. 169 bis 172 , erhalten durch Reduktion von 19-Nortestosteron), 50 Teilen Essigsäureanhydrid, 100 Teilen Eisessig und 2 Teilen Natriumacetat wird 2 Stunden lang bei Raumtemperatur gehalten und dann mit Eis behandelt und 2 weitere Stunden ste hengelassen. Der Niederschlag wird abfiltriert und mit Wasser gewaschen.
Durch Umkristallisieren aus wäss- rigem Methanol und anschliessend aus Cyclohexan erhält man 3,17-Diacetoxy-4-oestren, das bei etwa 135 bis 137,5 schmilzt.
<I>Beispiel 4</I> Einem Gemisch aus 35 Teilen 19-Nortestosteron- propionat, 100 Teilen Methanol und 10 Teilen Äthyl- acetat werden 20 Teile Natriumborhydrid in 50 Tei len Methanol zugesetzt. Nach einstündigem Rühren bei Raumtemperatur werden 6 Teile Wasser zugesetzt und der pH-Wert wird durch Zugabe von Essigsäure auf 6 gesenkt. Das Gemisch wird durch Einengen auf einem Dampfbad im Stickstoffstrom auf ein Drittel seines ursprünglichen Volumens gebracht, und dieser Rest wird mit Wasser auf das Doppelte seines Volu mens verdünnt.
Der Niederschlag wird abfiltriert, mit Wasser gewaschen und nach dem Verfahren des Bei spiels 1 über einer Kolonne mit Kieselsäuregel chro- matographiert.
Das Eluieren mit einer 15o/aigen Lösung von Äthylacetat in Benzol ergibt 17-Propionyloxy-4- oestren-3-ol. Die Infrarotabsorption zeigt Maxima bei 2,9 und 5,9,u.
<I>Beispiel 5</I> Ein Gemisch aus 1 Teil 4-Oestren-3,17-diol, er halten durch Reduktion von 19-Nortestosteron, 10 Teilen Benzoesäureanhydrid und 1 Teil fein gepulver- tem Kaliumbenzoat wird 24 Stunden lang bei Raum temperatur gerührt, dann mit Eis gekühlt und weitere 3 Stunden bei Raumtemperatur stehengelassen.
Der erhaltene Niederschlag wird abfiltriert, mit Wasser gewaschen und nacheinander aus wässrigem Äthanol und Cyclohexan umkristallisiert. Das erhaltene 3,17 Dibenzoyloxy-4-oestren zeigt Infrarotmaxima bei 5,8 und 6,1 ,u.
<I>Beispiel 6</I> Einem Gemisch von 22 Teilen 19-Nortestosteron- f-cyclopentyl-propionat in 1000 Teilen Methanol werden 50 Teile Äthylacetat und 10 Teile Natrium borhydrid in 200 Teilen Methanol zugesetzt. Das Ge misch wird 10 Minuten lang bei 20 gerührt und dann mit 30 Teilen Wasser und einer Menge Eisessig be handelt, die ausreicht, um den p,1-Wert auf etwa 6,0 herabzusetzen. Durch Einengen auf dem Dampfbad in einer Stickstoffatmosphäre wird das Gemisch auf 300 Teile konzentriert und dann mit seinem zwei fachen Volumen Wasser verdünnt.
Es bildet sich Nie derschlag, der abfiltriert und mit Wasser gewaschen wird. Beim Umkristallisieren aus Äthylacetat und Cyclohexanon erhält man 17-(f-Cyclopentyl-pro- pionyloxy)-4-oestren-3-ol, das durch Chromatogra- phieren über Kieselsäuregel weiter gereinigt werden kann. Das Produkt zeigt Infrarotmaxima bei 3,0, 5,8 und 6,05,u.
<I>Beispiel 7</I> Ein Gemisch aus 10 Teilen 17a - Äthinyl - 3 - methoxy-1,3,5(10) - oestratrien -17 - o1, 2,5 Teilen 51/aigem Palladium auf Holzkohle und 50 Teilen Dioxan wird in einer Wasserstoffatmosphäre geschüt telt, bis 2 Moläquivalente Wasserstoff verbraucht worden sind.
Der Katalysator wird durch - Filtrieren entfernt und der nach dem Konzentrieren des Filtrats erhaltene Rückstand wird durch Umkristallisieren aus einem Gemisch von Äther und Petroläther gereinigt. Auf diese Weise wird 17a-Athyl-3-methoxy-1,3,5(10)- oestratrien-17-ol erhalten, das bei etwa 88 bis 89 schmilzt. Eine Lösung von 50 Teilen 17a-Äthyl-3- methoxy-1,3,5(10)-oestratrien-17-ol in 710 Teilen Äther wird mit 2500 Teilen flüssigem Ammoniak verdünnt.
Während der folgenden Arbeitsgänge, bis zur Verdünnung mit Wasser, wird das Reaktions gemisch ständig gerührt und durch die Zugabe von insgesamt 2100 Teilen Äther in kleinen Portionen zum Ersatz des kontinuierlich durch Verdampfung verlorengehenden Ammoniaks bei konstantem Volu men gehalten. Der Lösung des 17a-Äthyl-3-methoxy- 1,3,5(10)-oestratrien-17-ols in Äther und flüssigem Ammoniak werden innerhalb 35 Minuten 35 Teile Lithium in kleinen Mengen zugegeben. Nach einer weiteren Stunde werden 240 Teile Äthanol in kleinen Mengen innerhalb 3 Stunden zugesetzt, und das Rüh ren wird einige Stunden lang fortgesetzt.
Dann werden 1000 Teile Wasser zugegeben, und die ätherische Phase wird mehrmals mit Wasser gewaschen, getrock net und konzentriert. Der durch Verdampfen der Atherlösung erhaltene Rückstand wird mit einem Ge misch aus 200 Teilen Methanol und einem Teil Pyridin gerührt, und die erhaltene Suspension wird mehrere Stunden lang gekühlt. Das kristalline Pro dukt wird abfiltriert und mit 120 Teilen kaltem Me thanol gewaschen. Man erhält so 17a -Äthyl - 3 - methoxy-2,5(10)-oestradien-17-ol, das bei etwa 127 bis 128 schmilzt.
Ein Gemisch aus 1,25 Teilen 17a -Äthyl - 3 - methoxy-2,5(10)-oestradien-17-ol, 24 Teilen Me thanol und 4 Teilen Essigsäure wird 5 Minuten lang am Rückflusskühler erhitzt. Es werden 150 Teile Wasser zugesetzt, und das Gemisch wird mehrere Stunden lang gekühlt. Das ausgefällte Produkt wird auf einem Filter gesammelt und getrocknet. Umkri- stallisieren aus einem Gemisch von Aceton und Pe- troläther ergibt 17a-Äthyl-3-keto-5(10)-oestren- 17-ol, das bei etwa 134 bis 136 schmilzt.
Ein Gemisch aus 10 Teilen 17a-Äthyl-3-keto- 5(10)-oestren-17-ol, 160 Teilen Methanol, 20 Teilen Wasser und 7 Teilen konzentrierter Salzsäure wird 2 Stunden lang bei Raumtemperatur gerührt, dann gekühlt und 2 Stunden lang bei 0 stehengelassen. Aus dem Gemisch wird eine kleine Menge unlöslicher Stoffe abfiltriert und das Filtrat unter ständigem Rühren mit 500 Teilen Wasser verdünnt, dann auf etwa 0 bis 5 gekühlt und filtriert. Das kristalline Pro dukt kann durch aus wässrigem Methanol oder aus einem Gemisch von Aceton und Petroläther gereinigt werden.
Auf diese Weise erhält man 17-Äthyl-19- nortestosteron, das bei 140 bis 141 schmilzt.
Einer Lösung von 2 Teilen 17a-Äthyl-19-norte- stosteron in 25 Teilen Methanol werden 2 Teile Na- triumborhydrid in 1Z Teilen Methanol zugesetzt. Das Gemisch wird 60 Minuten lang bei Raumtemperatur gehalten und dann mit Essigsäure langsam angesäuert. Das Gemisch wird mit 150 Teilen Wasser verdünnt und auf 0 gekühlt. Der erhaltene Niederschlag wird abfiltriert, mit Wasser gewaschen und unter Verwen dung von Holzkohle zur Entfärbung aus wässrigem Methanol umkristallisiert.
Auf diese Weise wird 17a-Äthyl-4-oestren-3,17-diol erhalten. Es scheint, dass dieses Produkt vorwiegend aus dem 3ss,17ss-Iso- meren besteht, während die 3a-Verbindung nur in geringer Menge vorhanden ist. Auf einem Schmelz block schmilzt das Produkt bei etwa 80 , erstarrt teil weise wieder und schmilzt erneut bei l25 . Das vor stehende Material zeigt keine spezifische Absorption im ultravioletten Bereich zwischen 220 und 360 my. <I>Beispiel 8</I> Spuren Jod werden einem Gemisch von 1,7 Tei len Magnesium und 18 Teilen Äther zugesetzt.
Der gesamte Äther wird abdestilliert, und das Magnesium wird mit einer frischen Menge von 70 Teilen Äther bedeckt. Eine Lösung von 9 Teilen Allylbromid in 70 Teilen Äther wird langsam unter Rühren dem Reaktionsgemisch zugegeben, und danach wird all mählich eine Lösung von 2 Teilen 3-Methoxy-2,5(10)- oestradien-17-on in 70 Teilen Äther zugesetzt.
Das Reaktionsgemisch wird eine weitere Stunde am Rückflusskühler erhitzt, dann wird eine Lösung von 10 Teilen Natriumkaliumtartrat in 100 Teilen Was ser vorsichtig unter wirksamem Rühren zugesetzt. Die Ätherschicht wird abgetrennt, mit Wasser ge waschen, über Natriumsulfat getrocknet und filtriert. Der durch Konzentrieren des Filtrats erhaltene Rück stand wird aus einem Gemisch von Äther und Me thanol umkristallisiert, wobei ein bei etwa 88 bis 96 schmelzendes Produkt erhalten wird, das vorwiegend aus 17a-Allyl-3-methoxy-2,5(10)-oestradien-17-ol be steht.
Eine Mischung aus 1,5 Teilen dieses Produk tes, 32 Teilen Methanol, 5 Teilen Wasser und 1,8 Tei len konzentrierter Salzsäure wird 5 Minuten lang am Rückflusskühler erhitzt. Das gummiartige Produkt, das durch Verdünnung des Reaktionsgemisches mit Wasser erhalten wird, wird mehrere Male durch De kantieren mit Wasser gewaschen und getrocknet. Es wird durch Kristallisieren aus einem Gemisch von Aceton und Petroläther oder durch chromatographi- sche Fraktionierung über Kieselsäuregel gereinigt.
Auf diese Weise wird 17-Allyl-19-nortestosteron er halten, das bei etwa 95,5 bis 96,5 schmilzt.
Ein Gemisch aus 5 Teilen 17-Allyl-19-nortesto- steron, 100 Teilen Dioxan und 1,5 Teilen eines aus 5 II/o Palladium auf Holzkohle bestehenden Katalysa- tors wird in einer Wasserstoffatmosphäre geschüttelt, bis etwa ein Moläquivalent Wasserstoff verbraucht worden ist. Der Katalysator wird abfiltriert und das Filtrat wird unter vermindertem Druck konzentriert.
Kristallisiert man den durch Verdampfung des Lö sungsmittels erhaltenen Rückstand wiederholt aus einem Gemisch von Aceton und Petroläther um, so erhält man 17-Propyl-19-nortestosteron, das bei etwa 120 bis 122 schmilzt.
Einer Lösung von 28 Teilen 17a-Propyl-19- nortestosteron in 200 Teilen Methanol wird eine Lö sung von 17,5 Teilen Natriumborhydrid in 120 Tei len Methanol zugesetzt. Das Gemisch wird 45 Minu ten lang bei Raumtemperatur gehalten und dann mit 5 Teilen Wasser und 15 Teilen Essigsäure behandelt. Das Reaktionsgemisch wird mit Wasser verdünnt und dann gekühlt.
Der Niederschlag wird abfiltriert und aus wässrigem Methanol umkristallisiert. Das auf diese Weise erhaltene Gemisch von 17a-Propyl-4- oestren-3fl-17fl-diol und 17a-Propyl-3a,17f-diol zeigt Infrarotmaxima bei 2,9 und 6,0 /1.
<I>Beispiel 9</I> Eine Lösung von einem Teil 17a-Äthyl-4-oestren- 3,17-.diol (erhalten durch Reduktion von 17a-Äthyl- 19-nortestosteron) in 4 Teilen Propionsäureanhydrid und 4 Teilen Pyridin wird auf einem Dampfbad in einer Stickstoffatmosphäre eine Stunde lang erhitzt und dann mit Eis behandelt. Der feste Niederschlag wird auf einem Filter gesammelt und nacheinander aus wässrigem Methanol und Petroläther umkristalli siert.
Das auf diese Weise erhaltene 17a-Äthyl-3-pro- pionyloxy-4-oestren-17-ol schmilzt bei etwa 87 bis 88,5 und zeigt Infrarotmaxima bei 2,79; 5,82; 6,87; 8,40; 9,25 und 9,92,u.
<I>Beispiel 10</I> Ein Gemisch aus 253 Teilen 17a-Äthyl-4-oestren- 3,17-diol (erhalten durch Reduktion von 17a-Äthyl- 19-nortestosteron), 852 Teilen Bernsteinsäureanhydrid und 2000 Teilen Pyridin wird 90 Minuten auf einem Dampfbad erhitzt und dann mit Eis behandelt.
Nach 30 Minuten wird der Niederschlag abfiltriert, mit Wasser gewaschen und zuerst aus wässrigem Methanol, dann aus einem Gemisch von Äthylacetat, Cyclo- hexan und Petroläther umkristallisiert. Das 3-He- misuccinat des 17a-Äthyl-4-oestren-3,17-diols, das auf diese Weise erhalten wird, schmilzt bei etwa 168 bis 170 .
<I>Beispiel 11</I> Einer Suspension von 6 Teilen Lithium-Alumi- niumhydrid in 2100 Teilen Äther werden unter Rüh ren 7,2 Teile 19-Nortestosteron in 700 Teilen Äther zugesetzt. Das Gemisch wird unter Erhitzen auf einem Dampfbad 45 Minuten lang gerührt; danach wird das nicht umgesetzte Lithium Aluminiumhydrid durch Zugabe von Aceton zersetzt. Das Gemisch wird mit Wasser verdünnt, die organische Schicht wird ab getrennt und nacheinander mit Wasser und gesättig ter, wässriger Natriumchloridlösung gewaschen.
Nach dem Trocknen über wasserfreiem Natriumsulfat wird die Ätherlösung unter Vakuum eingedampft, der Rückstand wird in Benzol gelöst und so auf eine chromatographische Kolonne aufgebracht, die 760 Teile Kieselsäuregel enthielt. Die Kolonne wird mit Benzol und dann nacheinander mit 5- und l0o/oigen Lösungen von Athylacetat in Benzol entwickelt.
Wei teres Eluieren mit einer 15o/nigen Lösung von Äthyl- acetat in Benzol und die Konzentration des Eluats ergibt einen Rückstand, der, aus Aceton und Wasser, Athylacetat und Petroläther, und wiederum aus Aceton und Wasser umkristallisiert, ein bei etwa 169 bis 172 schmelzendes 4-Oestren-3,17-diol ergibt.
Diese Verbindung ist augenscheinlich das 4-Oestren- 3ss,17,8-diol. Diese Verbindung zeigt Infrarotmaxima bei 2,91; 6,0; 6,90; 8,26; 8,78; 9,36; 9,61; 11,21; 11,43 und 11,62 ,u und kein Ultraviolettabsorptions- spektrum zwischen 220 und 360 mu.
Die weitere Entwicklung der Kolonne mit einer 20o/oigen Lösung von Äthylacetat in Benzol und das Konzentrieren des Eluats ergibt eine relativ kleinere Menge einer Verbindung, die vermutlich das 3a- Epimere des vorstehenden 4-Oestren-3,17-diols dar stellt. Nacheinander erfolgende Umkristallisierungen aus wässrigem Methanol und dann aus Äthylacetat und Cyclohexan ergeben ein bei etwa 202 bis 204 schmelzendes Produkt.
Das Infrarotabsorptionsspek- trum zeigt Maxima bei 3,0; 6,05; 7,01; 7,23; 7,42; 7,84; 8,80; 9,19; 9,46; 10,05;<B>11,06</B> und 11,64 @i. <I>Beispiel 12</I> Eine Lösung von einem Teil 17a-Äthyl-19-nor- testosteron in 120 Teilen absolutem 2-Propanol wird mit 11 Teilen frisch destilliertem Aluminiumiso- propylat behandelt und dann langsam destilliert, bis das Destillat kein Aceton mehr enthält.
Das zurück bleibende 2-Propanol wird abdestilliert und der Rückstand wird unter Rühren in 200 Teile kalten Wassers gegossen. Das Gemisch wird mit Äther extrahiert. Dieser Extrakt wird mit Wasser ge waschen, über wasserfreiem Natriumsulfat getrock net, filtriert und eingedampft. Der Rückstand wird wie in Beispiel 7 umkristallisiert und ergibt ebenfalls 17-Äthyl-4-oestren-3,17-diol, das die gleichen Eigen schaften hat wie das Produkt des Beispiels 7.
Bei Verwendung von 17a-Methyl-19-nortesto- steron erhält man 17-Methyl-4-oestren-3,17-diol. Das Infrarotabsorptionsspektrum dieser Verbindung zeigt Maxima bei 2,92; 6,03; 6,9; 7,3 und 8,7 @@.

in which R is hydrogen, the benzoyl radical or an acyl radical derived from an aliphatic carboxylic acid having 1 to 8 carbon atoms and A is a hydrogen atom or a lower saturated or unsaturated aliphatic hydrocarbon radical.
For example, R can be the formyl, acetyl, propionyl, butyryl, pentanoyl, hexanoyl, cyclopentyl-acetyl, α-cyclopentyl-propionyl, cyclohexyl-acetyl radical and A can be a lower alkyl radical, for example the methyl or ethyl radical,
or a straight-chain or branched propyl and butyl radical, an alkenyl radical, for example the vinyl, allyl or butenyl radical, and an alkynyl radical, for example the ethynyl, propynyl, butynyl radical or the like.
4-Oestren-3,17-diol, which is optionally substituted in the 17-position, and its mono- and diesters are relatively weakly androgenic agents. They are particularly valuable for human and veterinary therapy as a result of their high effectiveness in stimulating the metabolism of nitrogen. The compounds of the formula given at the outset are prepared according to the invention by reducing the corresponding 3-keto compounds with agents which do not attack the double bond.
Aluminum alcoholates, for example, can be used as reducing agents (Meerwein - Ponndorf - Verley reaction). Alkali metal aluminum hydrides and borohydrides, such as, for example, lithium aluminum hydride and sodium borohydride, are particularly favorable.
2-estrene-3,17-diols substituted in the 17-position by an alkyl group can be conveniently prepared by subsequent hydrogenation of 17a-substitution products of 4-estrene-3,17-diol prepared according to the invention in which the 17a-substituent consists of an unsaturated aliphatic hydrocarbon radical.
Such radicals can contain ethylene or acetylene bonds; it can be, for example, ethynyl, propynyl and butynyl, vinyl, propenyl, allyl radicals and their higher homologues.
To hydrogenate, for example, 17a-ethynyl-4-oestren-3,17-diol, this compound can be shaken in solution or suspension in a hydrogen atmosphere in the presence of a catalyst, for example a noble metal catalyst, such as palladium on charcoal. The compound is gradually reduced to 17-ethyl-4-oestren-3,17-diol via 17a vinyl-4-oestren-3,17-d'iol.
Under these conditions, the double bond in the core is hydrogenated only to a small extent. 17-Ethyl-4-estrene-3,17-diol can also advantageously be made by catalytic reduction of 17a-vinyl-4-estrene-3,17-diol.
The 17-alkenyl or 17-alkynyl-19-nortestosterones required for the preparation of 17a-alkenyl and alkynyl derivatives of 4-estrene-3,17-diol can easily be added by addition of acetylene, acetylene homologues or unsaturations organometallic compounds to corresponding 17-keto-steroids are produced.
For example, 17a-ethynyl-19-nortestosterone can be produced by adding acetylene to 3-methoxy-2,5 (10) -oestradien-17-one and then acid-catalyzed hydrolysis and isomerization of the product obtained. The addition of allyl magnesium bromide to 3-methoxy-2,5 (10) -oestradien-17-one and subsequent acidic hydrolysis and isomerization gives 17-allyl-19-nortestosterone.
In the following examples, the parts mean parts by weight.
<I> Example 1 </I> A solution of 7.2 parts of 19-nortesterone in 700 parts of ether is added to a suspension of 6 parts of lithium aluminum hydride in 210 parts of ether while stirring. The mixture is heated to 35 for 45 minutes with stirring; then an amount of acetone is added which is sufficient to decompose the excess lithium aluminum hydride.
Water is then added, the organic layer is separated off and washed first with water and then with saturated sodium chloride solution. After drying over anhydrous sodium sulfate and filtering, the solvent is removed in vacuo. The residue is applied in a 10% solution of ethyl acetate in benzene to a chromatographic column which contains 760 parts of silica gel.
The column is then washed with benzene and 5-, 10- and 15ohigen solutions of ethyl acetate in benzene. Eluting with a 20o / aigen solution of ethyl acetate in benzene and repeated recrystallization of the residue from a mixture of ethyl acetate and cyclohexane gives 4-estrene-3,17-diol, which melts at about 202-204. The infrared absorption spectrum shows a broad band at 3.0 and a band at 6,
05 / t. It is evidently the 4-oestren-3 a, 17f-diol. <I> Example 2 </I> A solution of 10 parts of sodium borohydride in 200 parts of methanol is added to a solution of 14.4 parts of 19-nortestosterone in 400 parts of methanol and 50 parts of ethyl acetate. The mixture is kept at room temperature for about an hour and treated first with 30 parts of water and then with an amount of acetic acid sufficient to reduce the pH to 6.0.
The reaction mixture is concentrated to about 50 parts in a stream of nitrogen on a steam bath. After thinning with 600 parts of water, the precipitate is filtered off and washed with water. Chromatography over silica gel as in the previous example gives 4-estrene-3,17-diol with the same physical properties as the product obtained in the previous example.
Example 3 A mixture of 10 parts of 4-estren-3,17-diol (melting point 169 to 172, obtained by reducing 19-nortestosterone), 50 parts of acetic anhydride, 100 parts of glacial acetic acid and 2 parts of sodium acetate is kept at room temperature for 2 hours and then treated with ice and left to stand for 2 more hours. The precipitate is filtered off and washed with water.
Recrystallization from aqueous methanol and then from cyclohexane gives 3,17-diacetoxy-4-oestrene, which melts at about 135 to 137.5.
<I> Example 4 </I> 20 parts of sodium borohydride in 50 parts of methanol are added to a mixture of 35 parts of 19-nortestosterone propionate, 100 parts of methanol and 10 parts of ethyl acetate. After stirring for one hour at room temperature, 6 parts of water are added and the pH is lowered to 6 by adding acetic acid. The mixture is brought to one third of its original volume by concentration on a steam bath in a stream of nitrogen, and this remainder is diluted to twice its volume with water.
The precipitate is filtered off, washed with water and chromatographed according to the method of Example 1 over a column with silica gel.
Elution with a 150 / aigen solution of ethyl acetate in benzene gives 17-propionyloxy-4-oestren-3-ol. The infrared absorption shows maxima at 2.9 and 5.9, u.
<I> Example 5 </I> A mixture of 1 part of 4-estrene-3,17-diol, obtained by reducing 19-nortestosterone, 10 parts of benzoic anhydride and 1 part of finely powdered potassium benzoate is left in the room for 24 hours stirred temperature, then cooled with ice and left to stand for a further 3 hours at room temperature.
The precipitate obtained is filtered off, washed with water and successively recrystallized from aqueous ethanol and cyclohexane. The 3.17 dibenzoyloxy-4-oestrene obtained shows infrared maxima at 5.8 and 6.1, u.
<I> Example 6 </I> 50 parts of ethyl acetate and 10 parts of sodium borohydride in 200 parts of methanol are added to a mixture of 22 parts of 19-nortestosterone-f-cyclopentyl propionate in 1000 parts of methanol. The mixture is stirred for 10 minutes at 20 and then treated with 30 parts of water and an amount of glacial acetic acid sufficient to reduce the p, 1 value to about 6.0. The mixture is concentrated to 300 parts by concentration on the steam bath in a nitrogen atmosphere and then diluted with two times its volume of water.
A precipitate forms, which is filtered off and washed with water. Recrystallization from ethyl acetate and cyclohexanone gives 17- (f-cyclopentyl-propionyloxy) -4-oestren-3-ol, which can be further purified by chromatography on silica gel. The product shows infrared maxima at 3.0, 5.8 and 6.05, u.
<I> Example 7 </I> A mixture of 10 parts of 17a-ethynyl-3-methoxy-1,3,5 (10) -estratriene -17-o1, 2.5 parts of 51% palladium on charcoal and 50 parts Dioxane is shaken in a hydrogen atmosphere until 2 molar equivalents of hydrogen have been consumed.
The catalyst is removed by filtration and the residue obtained after concentrating the filtrate is purified by recrystallization from a mixture of ether and petroleum ether. In this way 17a-ethyl-3-methoxy-1,3,5 (10) - oestratrien-17-ol, which melts at about 88 to 89, is obtained. A solution of 50 parts of 17a-ethyl-3-methoxy-1,3,5 (10) -oestratrien-17-ol in 710 parts of ether is diluted with 2500 parts of liquid ammonia.
During the following operations, up to dilution with water, the reaction mixture is constantly stirred and kept at a constant volume by adding a total of 2100 parts of ether in small portions to replace the ammonia continuously lost through evaporation. 35 parts of lithium are added in small amounts to the solution of 17a-ethyl-3-methoxy-1,3,5 (10) -oestratrien-17-ol in ether and liquid ammonia within 35 minutes. After a further hour, 240 parts of ethanol are added in small amounts over 3 hours and stirring is continued for a few hours.
1000 parts of water are then added, and the ethereal phase is washed several times with water, dried and concentrated. The residue obtained by evaporation of the ether solution is stirred with a mixture of 200 parts of methanol and one part of pyridine, and the suspension obtained is cooled for several hours. The crystalline product is filtered off and washed with 120 parts of cold methanol. This gives 17a-ethyl -3-methoxy-2,5 (10) -oestradien-17-ol, which melts at about 127 to 128.
A mixture of 1.25 parts of 17a-ethyl -3-methoxy-2,5 (10) -oestradien-17-ol, 24 parts of methanol and 4 parts of acetic acid is refluxed for 5 minutes. 150 parts of water are added and the mixture is cooled for several hours. The precipitated product is collected on a filter and dried. Recrystallization from a mixture of acetone and petroleum ether gives 17α-ethyl-3-keto-5 (10) -oestren-17-ol, which melts at about 134 to 136.
A mixture of 10 parts of 17a-ethyl-3-keto-5 (10) -oestren-17-ol, 160 parts of methanol, 20 parts of water and 7 parts of concentrated hydrochloric acid is stirred for 2 hours at room temperature, then cooled and for 2 hours left at 0. A small amount of insoluble matter is filtered off from the mixture and the filtrate is diluted with 500 parts of water, with constant stirring, then cooled to about 0 to 5 and filtered. The crystalline product can be purified from aqueous methanol or from a mixture of acetone and petroleum ether.
In this way 17-ethyl-19-nortestosterone is obtained, which melts at 140 to 141.
2 parts of sodium borohydride in 1Z parts of methanol are added to a solution of 2 parts of 17α-ethyl-19-nortestosterone in 25 parts of methanol. The mixture is kept at room temperature for 60 minutes and then slowly acidified with acetic acid. The mixture is diluted with 150 parts of water and cooled to zero. The resulting precipitate is filtered off, washed with water and recrystallized from aqueous methanol using charcoal to decolorize.
In this way 17a-ethyl-4-oestren-3,17-diol is obtained. It appears that this product consists mainly of the 3ss, 17ss isomer, while the 3a compound is only present in small amounts. On a melting block, the product melts at around 80, partially solidifies again and melts again at 125. The above material shows no specific absorption in the ultraviolet range between 220 and 360 my. <I> Example 8 </I> Traces of iodine are added to a mixture of 1.7 parts of magnesium and 18 parts of ether.
All of the ether is distilled off and the magnesium is covered with a fresh amount of 70 parts of ether. A solution of 9 parts of allyl bromide in 70 parts of ether is slowly added to the reaction mixture with stirring, and then a solution of 2 parts of 3-methoxy-2,5 (10) -estradien-17-one in 70 parts of ether is gradually added.
The reaction mixture is refluxed for a further hour, then a solution of 10 parts of sodium potassium tartrate in 100 parts of water is carefully added with efficient stirring. The ether layer is separated off, washed with water, dried over sodium sulfate and filtered. The residue obtained by concentrating the filtrate is recrystallized from a mixture of ether and methanol to give a product which melts at about 88 to 96 and which consists mainly of 17a-allyl-3-methoxy-2,5 (10) -oestradien -17-ol exists.
A mixture of 1.5 parts of this product, 32 parts of methanol, 5 parts of water and 1.8 parts of concentrated hydrochloric acid is refluxed for 5 minutes. The rubbery product obtained by diluting the reaction mixture with water is washed several times by decanting with water and dried. It is purified by crystallization from a mixture of acetone and petroleum ether or by chromatographic fractionation over silica gel.
In this way, 17-allyl-19-nortestosterone is obtained, which melts at about 95.5 to 96.5.
A mixture of 5 parts of 17-allyl-19-nortestosterone, 100 parts of dioxane and 1.5 parts of a catalyst consisting of 5 II / o palladium on charcoal is shaken in a hydrogen atmosphere until about one molar equivalent of hydrogen has been consumed . The catalyst is filtered off and the filtrate is concentrated under reduced pressure.
If the residue obtained by evaporation of the solvent is crystallized repeatedly from a mixture of acetone and petroleum ether, 17-propyl-19-nortestosterone, which melts at about 120 to 122, is obtained.
A solution of 17.5 parts of sodium borohydride in 120 parts of methanol is added to a solution of 28 parts of 17a-propyl-19- nortestosterone in 200 parts of methanol. The mixture is kept at room temperature for 45 minutes and then treated with 5 parts of water and 15 parts of acetic acid. The reaction mixture is diluted with water and then cooled.
The precipitate is filtered off and recrystallized from aqueous methanol. The mixture of 17a-propyl-4-estren-3fl-17fl-diol and 17a-propyl-3a, 17f-diol obtained in this way shows infrared maxima at 2.9 and 6.0 / 1.
<I> Example 9 </I> A solution of one part of 17a-ethyl-4-oestren-3,17-.diol (obtained by reducing 17a-ethyl-19-nortestosterone) in 4 parts of propionic anhydride and 4 parts of pyridine is used heated on a steam bath in a nitrogen atmosphere for one hour then treated with ice. The solid precipitate is collected on a filter and successively recrystallized from aqueous methanol and petroleum ether.
The 17a-ethyl-3-propionyloxy-4-oestren-17-ol obtained in this way melts at about 87 to 88.5 and shows infrared maxima at 2.79; 5.82; 6.87; 8.40; 9.25 and 9.92, etc.
<I> Example 10 </I> A mixture of 253 parts of 17a-ethyl-4-oestren-3,17-diol (obtained by reducing 17a-ethyl-19-nortestosterone), 852 parts of succinic anhydride and 2000 parts of pyridine becomes 90 Heated for minutes on a steam bath and then treated with ice.
After 30 minutes the precipitate is filtered off, washed with water and recrystallized first from aqueous methanol and then from a mixture of ethyl acetate, cyclohexane and petroleum ether. The 3-hemisuccinate of 17a-ethyl-4-oestrene-3,17-diol obtained in this way melts at about 168-170.
Example 11 7.2 parts of 19-nortestosterone in 700 parts of ether are added to a suspension of 6 parts of lithium aluminum hydride in 2100 parts of ether. The mixture is stirred with heating on a steam bath for 45 minutes; then the unreacted lithium aluminum hydride is decomposed by adding acetone. The mixture is diluted with water, the organic layer is separated and washed successively with water and saturated aqueous sodium chloride solution.
After drying over anhydrous sodium sulfate, the ether solution is evaporated in vacuo, the residue is dissolved in benzene and thus applied to a chromatographic column which contained 760 parts of silica gel. The column is developed with benzene and then successively with 5 and 10% solutions of ethyl acetate in benzene.
Further elution with a 15% solution of ethyl acetate in benzene and the concentration of the eluate results in a residue which, from acetone and water, ethyl acetate and petroleum ether, and again from acetone and water, recrystallizes, melting at about 169 to 172 4-Oestren-3,17-diol gives.
This compound is apparently the 4-oestren-3ss, 17,8-diol. This compound shows infrared maxima at 2.91; 6.0; 6.90; 8.26; 8.78; 9.36; 9.61; 11.21; 11.43 and 11.62, u and no ultraviolet absorption spectrum between 220 and 360 mu.
Further development of the column with a 20% solution of ethyl acetate in benzene and concentration of the eluate yields a relatively smaller amount of a compound which is believed to be the 3a-epimer of the above 4-estrene-3,17-diol. Successive recrystallizations from aqueous methanol and then from ethyl acetate and cyclohexane give a product which melts at about 202-204.
The infrared absorption spectrum shows maxima at 3.0; 6.05; 7.01; 7.23; 7.42; 7.84; 8.80; 9.19; 9.46; 10.05; <B> 11.06 </B> and 11.64 @i. <I> Example 12 </I> A solution of one part of 17α-ethyl-19-nortestosterone in 120 parts of absolute 2-propanol is treated with 11 parts of freshly distilled aluminum isopropylate and then slowly distilled until the distillate does not contain acetone contains more.
The remaining 2-propanol is distilled off and the residue is poured into 200 parts of cold water with stirring. The mixture is extracted with ether. This extract is washed with water, getrock net over anhydrous sodium sulfate, filtered and evaporated. The residue is recrystallized as in Example 7 and also gives 17-ethyl-4-oestren-3,17-diol, which has the same properties as the product of Example 7.
If 17α-methyl-19-nortestosterone is used, 17-methyl-4-estren-3,17-diol is obtained. The infrared absorption spectrum of this compound shows maxima at 2.92; 6.03; 6.9; 7.3 and 8.7 @@.
