Zusatzpatent zum Hauptpatent Nr. 329194 Verfahren zur Herstellung eines Progesteronderivates Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung des neuen lla-Oxy- 16,17-oxido-progesterons und dessen Ester.

in der R Wasserstoff oder ein Acylradikal darstellt, insbesondere den Rest einer Fettsäure mit vorzugs weise bis zu 8 Kohlenstoffatomen oder einer Toluol- sulfosäure.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man 16,17-Oxido-progesteron einer biochemischen Oxydation mittels Enzyme unterwirft, die von einer Pilzart der Gattung Rhizopus erzeugt werden, wonach man das erhal tene 1 la-Oxy-16,17-oxido - progesteron verestern kann.
Das neue 1 I a-Oxy-16,17-oxido-progesteron und seine Ester sind leicht kristallisierende feste Stoffe, die wertvolle pharmakologische und physiologische Eigenschaften aufweisen. Sie sind auch wertvolle Zwischenprodukte für die Herstellung von in 11-Stellung ein Sauerstoffatom aufweisenden Adreno- corticalhormonen. So stellt man beispielsweise Cor- tisonacetat her, indem man I 1 a-Oxy-16,17-oxido- progesteron mit Lithiumhydrid zum 3ss,lla,17a- 20-tetraoxy-4-pregnen reduziert,
das durch Oxyda tion mit Chromsäure in das 17a-Oxy-4-pregnen- 3,11,20-trion übergeht. Behandelt man letztere Ver bindung mit Bleitetraacetat, so erhält man 17a-Oxy- 21-acetoxy-4-pregnen-3,11,20-trion (Cortisonacetat).
Das als Ausgangsstoff verwendete 16,17-Oxido- progesteron kann nach Percy L. Julian und Mitarb. J. Am. Chem. Soc. 71, 756 (1949) hergestellt wer den. Dieses wird in der bereits angegebenen Weise an der 11-StelIung oxydiert. Das entstandene lla- Oxy-16,17-oxido-progesteron kann durch Extraktion oder Chromatographie isoliert werden.
Die Ver- esterung kann durch Behandlung des ll:a-Oxy-16,17- oxido-progesterons mit einem acylierenden Mittel, wie z. B.. einem der gewählten Säure entsprechenden Keten, einer Säure, einem Säurechlorid bzw.
Säure anhydrid oder sonstigen bekannten Acylierungsmit- teln erfolgen, wobei man gewöhnlich in einem Lö sungsmittel, wie Pyridin oder dergleichen oder in. einem inerten Lösungsmittel, wie Benzol, Toluol, Äther und dergleichen arbeitet, wobei man erhitzt oder bei Temperaturen zwischen etwa 0 und dem Siedepunkt der Mischung, in der Regel bei Zimmer temperatur (25-30 C) etwa 30 Minuten bis 96 Stunden stehenlässt. Die Reaktionsdauer, die Tempe ratur, das Acylierungsmittel und das Mengenverhält nis der Reaktionsteilnehmer können verschieden sein.
Das Reaktionsgemisch wird zweckmässig auf Eis oder in kaltes Wasser gegossen, das Produkt daraus mit einem geeigneten Lösungsmittel extra hiert, worauf man den Extrakt nacheinander mit schwach basischen Lösungen und Wasser praktisch neutral wäscht. In gewissen Fällen kann das Pro dukt aus dem Reaktionsgemisch auskristallisieren, so dass man es vorteilhaft durch Filtration oder an- fiere Massnahmen abtrennt, mit Wasser wäscht und dann in üblicher Weise, z. B. durch Umkristallisieren aus geeigneten Lösungsmitteln oder durch Chromato- graphie reinigt.
Als Acylierungsmittel kommen in Frage: Amei sensäure, Anhydride oder Säurehalogenide der Essig-, Propion-, a- und f-Cyclopentylpropion-, Butter-, Isobutter-, Valerian-, Isovalerian-, Ca- pron-, Heptyl-, Capryl-, Benzoe-, Phenylessig-, Tolyl-, Mandel-, Salicyl-, Gallus-, Anis-, Chloressig-,
Di- und Trichloressig-, Trimethylessig-, Bromessig-, Thioglykol-, Vinylessib , Milch-, Bernstein-, Malein-, m- oder p-Toluolsulfo-, Weinsäure und andere ähn liche Säuren, insbesondere mit weniger als 9 Kohlen stoffatomen. Die Säurehalogenide oder -anhydride können auch Substituenten tragen, die unter den. Reaktionsbedingungen nicht reagieren, wie z. B.
Chlor-, Brom-, Jodatome oder Methoxy-, Äthoxy-, Hydroxyl-, Vinyl- oder Thiogruppen.
Die folgenden Beispiele zeigen, wie die Erfindung durchgeführt werden kann.
<I>Beispiel 1</I> Man stellt ein Nährmedium her, das 20 g Edamine (enzymatisches Abbauprodukt des Lact- albumins), 3 g Maiseinweichflüssigkeit und 50 g technische Dextrose, mit Leitungswasser auf 1 Liter verdünnt und auf PH 4,3-4,5 eingestellt, enthält.
Drei Liter dieses sterilisierten Mediums werden mit einem Rhizopus nigricans minus (American Type Culture Collection Nr.<I>6227b)</I> geimpft und 24 Stun den bei 28 C bebrütet, wobei man so belüftet und, rührt, dass die Sauerstoffaufnahme 7 Millimol/1 NazSO-, nach der Methode von Cooper, Fernstrom und Miller [lud. Eng. Chem. 36, 504 (l944)] be trägt.
Zu diesem, die 24stündige Kultur des Pilzes enthaltenden Medium gibt man 1 g 16,17-Oxido- progesteron in 30 cm@ absolutem Alkohol, so dass eine Suspension des Steroids in der Kultur entsteht. Nach weiteren 24 Stunden Bebrütung bei gleicher Temperatur und den gleichen Belüftungsbedingungen; wird die Flüssigkeit und das Mycelium extrahiert. Das Mycelium wird durch Filtration abgetrennt, zweimal mit je etwa seinem Volumen Aceton ge waschen und dann zweimal mit je etwa seinem Vo lumen Methylenchlorid extrahiert.
Die Mischung der Extrakte und das Filtrat der Nährlösung werden nacheinander mit je dem halben Volumen Methylen- chlorid und dann zweimal mit je einem Viertel des Volumens Methylenchlorid extrahiert.
Die vereinig ten Methylenehloridextrakte werden zweimal mit je einem Zehntel ihres Volumens 2 %iger wässriger Na- triumbikärbonatlösung und dann zweimal mit je einem Zehntel ihres Volumens Wasser gewaschen. Nach dem Trocknen der Extrakte mit 3-5 g wasser freiem Natriumsulfat pro Liter Lösungsmittel und Filtrieren destilliert man das Lösungsmittel ab. Der Rückstand wiegt 2,5492 g.
Eine teilweise Entfärbung des rohen Rückstandes erreicht man durch Auflösen in 3.5 cm3 Methylen- chlorid, Zusatz von 3 g Magnesol (synthetisches Magnesiumsilikat) und Filtrieren. Nach Verdampfen des Lösungsmittels wird der Rückstand in über schüssigem Aceton aufgenommen, mit 0,2 g Celite Nr. 545 (Diatomeenerde) versetzt, filtriert und zur Trockne eingedampft.
Man erhält so insgesamt 3,23 g durch Aceton solvatisiertes festes Produkt, das in 100 cms Benzol gelöst und über 50 g Alumi niumoxyd mit den folgenden 100 cm-' Fraktionen der angegebenen Lösungsmittel eluiert wird. Die in der Tabelle angegebene Summe der Eluatfeststoffe erscheint wegen geringer Mengen von Aluminium oxyd höher als die Menge des Rohproduktes.

Nach nochmaligem Umkristallisieren erhält man 330 mg lla-Oxy-16,17-oxido-progesteron vom Schmelzpunkt 247-249 C. Die Infrarotanalyse be stätigt die für lla-Oxy-16,17-oxido-progesteron an genommene Struktur.
<I>Beispiel 2</I> Man stellt ein Medium her aus 0,5 g Sojamehl, 2 g Dextrose und je 0,5 g entbittertem Brauereihefe extrakt, Natriumchlorid und Monokaliumphosphat, verdünnt mit Leitungswasser auf 100 cms und stellt auf pH 4,5.
Dieses sterilisierte Medium wird mit Sporen von Rhizopus reflexus (ATCC Nr. 1225) von einer kleinen Agarplatte beimpft und 24 Stun den bei 28 C bebrütet, wobei man so rührt und be lüftet, dass pro Stunde und Liter Na2S03 nach der Methode von Cooper, Fernstrom und Miller Und, Eng. Chem. 36, 504 (1944)] 8 Millimol Sauerstoff aufgenommen werden.
Zu diesem Medium, das die 24stündige Kultur von Rhizopus reflexus enthält, gibt man eine Lösung von 20 mg 16,17-Oxido- progesteron in 1 cm3 Aceton und erzeugt so eine Suspension des Steroids in der Kultur. Nach einer weiteren 24stündigen Bebrütung unter den gleichen Bedingungen der Temperatur und Belüftung wird die Flüssigkeit und das Mycelium wie in Beispiel 1 extrahiert.
Der konzentrierte Extrakt wird durch Papierchromatographie unter Verwendung eines Propylenglykol-Toluol-Systems zerlegt und man er hält lla-Oxy-16,17-oxido-progesteron.
Ähnliche Ergebnisse erhält man durch Fermen tation von Rhizopus reflexus in einem Medium aus 2 g Mais-Einweichwasser, 2 g Dextrin, 0,1g Mono kaliumphosphat, 0,2 g Natriumnitrat, 0,5g Magne- siumsulfat-heptahydrat, 0,02 g Kaliumehlorid, 0,001g Ferrosulfatheptahydrat und 0,25g Natrium acetat mit Leitungswasser auf 100 cm3 verdünnt und auf pH 7,0 eingestellt.
Das neue Oxyprogesteron kann wie folgt in Ester übergeführt werden: A. Zu 94 mg l l a-Oxy-16,17-oxido-progesteron in 2 cm 3, mit Bariumoxyd getrocknetem Pyridin gibt man 1 cm3 frisch destilliertes Essigsäureanhydrid und lässt 16 Stunden bei Zimmertemperatur stehen. Dann wird mit Eiswasser auf 80 cm3 verdünnt und 24 Stunden im Kühlschrank aufbewahrt.
Der so er haltene kristalline Niederschlag von 11 a-Acetoxy- 16,17-oxido-progesteron wird abfiltriert, viermai mit je 1 cm3 Eiswasser gewaschen und im Vakuum bei 70 C getrocknet. Das trockene Produkt wird mehrmals aus Äthylacetat umkristallisiert und man erhält 19,7mg lla-Acetoxy-16,17-oxido-progesteron vom Schmelzpunkt 225-226 C. B.
Durch Umsetzung von 11a-Oxy-16,17-oxido- progesteron mit überschüssiger Ameisensäure wäh rend 12 Stunden bei Zimmertemperatur erhält man das 11a-Formyloxy-16,17-oxido-progesteron.
C. In gleicher Weise wie unter A erhält man durch Umsetzung von 11a - Oxy - 16,17 - oxido- progesteron mit Propionsäureanhydrid in Pyridiü das lla-Propionyloxy-16,17-oxido-progesteron.
<I>D.</I> In gleicher Weise wie unter<I>A</I> erhält man durch Umsetzung von 11a - Oxy - 16,17 - oxido- progesteron mit Buttersäureanhydrid in Pyridin das 11 a Butyroxy-16,17-oxido-progesteron.
E. In gleicher Weise wie unter A erhält man durch Umsetzung von 11a - Oxy - 16,17 - oxido- progesteron mit fl-Cyclopentyl-propionylchlorid in Pyridin das 11 a-(ss-cyclopentyl-propionyloxy)-16,17- oxido-progesteron.
F. In gleicher Weise wie unter A erhält man durch Umsetzung von 11a - Oxy - 16,17 - oxido- progesteron mit Benzoylchlorid in Pyridin das 11 a-Benzoyloxy-16,17-oxid'o-progesteron.
In gleicher Weise wie unter A-F erhält man durch Behandlung von l 1,a - Oxy - 16,17 - oxido- progesteron mit organischen Säuren, Anhydriden, Ketenen organischer Säuren oder Acylhalogeniden die entsprechenden 11a-Ester.

The combined methylene chloride extracts are washed twice with a tenth of their volume of 2% aqueous sodium bicarbonate solution and then twice with a tenth of their volume of water each time. After drying the extracts with 3-5 g of anhydrous sodium sulfate per liter of solvent and filtering, the solvent is distilled off. The residue weighs 2.5492 g.
A partial discoloration of the crude residue is achieved by dissolving it in 3.5 cm3 of methylene chloride, adding 3 g of magnesol (synthetic magnesium silicate) and filtering. After evaporation of the solvent, the residue is taken up in excess acetone, mixed with 0.2 g of Celite No. 545 (diatomaceous earth), filtered and evaporated to dryness.
This gives a total of 3.23 g of solid product solvated by acetone, which is dissolved in 100 cms of benzene and eluted over 50 g of aluminum oxide with the following 100 cm- 'fractions of the specified solvents. The sum of eluate solids given in the table appears to be higher than the amount of the crude product due to the small amounts of aluminum oxide.

<I> Example 2 </I> A medium is prepared from 0.5 g soy flour, 2 g dextrose and 0.5 g each of debittered brewer's yeast extract, sodium chloride and monopotassium phosphate, diluted with tap water to 100 cms and adjusted to pH 4, 5.
This sterilized medium is inoculated with spores of Rhizopus reflexus (ATCC No. 1225) from a small agar plate and incubated for 24 hours at 28 C, stirring and ventilating so that per hour and liter of Na2S03 according to the method of Cooper, Fernstrom and Miller And, Eng. Chem. 36, 504 (1944)] 8 millimoles of oxygen can be taken up.
A solution of 20 mg of 16,17-oxido progesterone in 1 cm3 of acetone is added to this medium, which contains the 24-hour culture of Rhizopus reflexus, and a suspension of the steroid is created in the culture. After a further 24 hours of incubation under the same conditions of temperature and aeration, the liquid and the mycelium are extracted as in Example 1.
The concentrated extract is separated by paper chromatography using a propylene glycol-toluene system and obtained IIIa-oxy-16,17-oxido-progesterone.
Similar results are obtained by fermentation of Rhizopus reflexus in a medium of 2 g of maize soaking water, 2 g of dextrin, 0.1 g of monopotassium phosphate, 0.2 g of sodium nitrate, 0.5 g of magnesium sulfate heptahydrate, 0.02 g of potassium chloride , 0.001g ferrous sulfate heptahydrate and 0.25g sodium acetate diluted to 100 cm3 with tap water and adjusted to pH 7.0.
The new oxyprogesterone can be converted into esters as follows: A. 1 cm3 of freshly distilled acetic anhydride are added to 94 mg of ll a-oxy-16,17-oxido-progesterone in 2 cm 3 of pyridine dried with barium oxide and left for 16 hours at room temperature stand. Then it is diluted to 80 cm3 with ice water and stored in the refrigerator for 24 hours.
The crystalline precipitate of 11α-acetoxy-16,17-oxido-progesterone obtained in this way is filtered off, washed four times with 1 cm3 of ice water each time and dried at 70 ° C. in vacuo. The dry product is recrystallized several times from ethyl acetate and 19.7 mg of IIIa-acetoxy-16,17-oxido-progesterone with a melting point of 225-226 C. B.
By reacting 11a-oxy-16,17-oxido-progesterone with excess formic acid for 12 hours at room temperature, 11a-formyloxy-16,17-oxido-progesterone is obtained.
C. In the same way as under A, the reaction of 11a-oxy-16,17-oxido-progesterone with propionic anhydride in pyridium gives Ila-propionyloxy-16,17-oxido-progesterone.
<I> D. </I> In the same way as under <I> A </I>, 11 a-butyroxy-16.17 is obtained by reacting 11a-oxy-16.17-oxido-progesterone with butyric anhydride in pyridine -oxido-progesterone.
E. In the same way as under A, the reaction of 11a-oxy-16.17-oxido progesterone with fl-cyclopentyl-propionyl chloride in pyridine gives 11 a- (ss-cyclopentyl-propionyloxy) -16.17-oxido- progesterone.
F. In the same way as under A, the reaction of 11a - oxy - 16,17 - oxido progesterone with benzoyl chloride in pyridine gives 11 a-benzoyloxy-16,17-oxy-progesterone.
In the same way as under A-F, the corresponding 11a-esters are obtained by treating l 1, a - oxy - 16, 17 - oxido progesterone with organic acids, anhydrides, ketenes of organic acids or acyl halides.
