Verfahren zur Herstellung von neuen Bis-carbaminsäureestern
Es ist bekannt, dass bestimmte synthetisch gewonnene Carbaminsäureester von ein quartäres Stickstoffatom enthaltenden aromatischen bzw. hetero cyclischen Oxyverbindungen, die sich beispielsweise vom m-Dimethylamino-phenol bzw. vom 3-Oxypyridin ableiten, starke Cholinesterase-Hemmkörper sind und anstelle des natürlich vorkommenden Alkaloids Eserin medizinisch verwendet werden können.

Die pharmakologischen Eigenschaften dieser neuen Verbindungen sind teilweise von denen der bisher medizinisch verwendeten Substanzen verschieden. So zeigt beispielsweise die erfindungsgemäss hergestellte Verbindung der Formel

im Vergleich mit dem Brommethylat des Dimethyl carbaminsäure-m-dimethylamino-phenylesters ( Neo- stigmin , eingetragene Marke) bei geringerer Toxizität eine stärkere und länger anhaltende Wirkung.
Ausserdem ist die bei manchen Anwendungen unerwünschte, muscarinartige Wirkung (Wirkung auf die Darmmotorik, Speichelsekretion, Herztätigkeit usw.) wesentlich schwächer als bei Neostigmin und ähnlichen Verbindungen.
Die gemäss vorliegender Erfindung hergestellten, am Carbaminsäure-Stickstoff alkylierten Verbindungen sind in wässeriger Lösung bestandig, so dass eine Hitzesterilisation und ein Aufbewahren solcher Substanzen in wässeriger Lösung ohne Wirkungsverlust möglich ist. Dies ist für die praktische Anwendung von grosser Bedeutung.
Ahnliche Eigenschaften zeigen auch nach dem erfindungsgemässen Verfahren hergestellte, vom 3 Oxy-pyridin abgeleitete Verbindungen, zum Beispiel die Verbindung der Formel

Die erfindungsgemässeUmsetzung desN, N'. disub- stituierten Dicarbaminsäuredichlorids obiger Formel mit einer Verbindung der Formel Z-OH bzw.
[RZ. OH] +X-wird vorteilhaft bei Anwesenheit von indifferenten Lösungsmitteln, wie zum Beispiel Toluol oder Xylol, durchgeführt ; es kann aber auch ohne derartige Lösungsmittel gearbeitet werden. Bei dieser Reaktion ist es ausserdem oft zweckmässig, in Anwesenheit von säurebindenden Stoffen, wie zum Beispiel tertiären Aminen, zu arbeiten. Besonders gute Ergebnisse können auch mit den Alkalisalzen der Oxyverbindungen der Formel Z-OH, wie zum Beispiel dem Natriumsalz des m-Dimethylamino-phenols bzw. des 3-Oxy-pyridins, erzielt werden.

erfolgt überraschenderweise bei verhältnismässig sehr niedrigen Temperaturen, meist schon bei Zimmertemperatur, so dass ein häufig schädlicher Einfluss höherer Temperatur vermeidbar ist und sehr reine Produkte erhalten werden. Auch die Quaternisierung wird zweckmässigerweise in indifferenten Lösungs- mitteln, wie zum Beispiel Aceton, Methyläthylketon, Methanol, Athanol oder auch in Lösungsmittel- gemischen solcher Art durchgeführt.
Die in nachfolgenden Beispielen angegebenen Teile sind Gewichtsteile.
Beispiel 1
15 Teile N, N, N', N'-Tetramethyl-hexamethylen- diamin werden bei Raumtemperatur unter Rühren in 100 Teile einer 40 ouzo Phosgen enthaltenden Toluollösung eingetropft, wobei das mit dem gebildeten Chlormethyl durch einen aufsteigenden Kühler entweichende Phosgen in einer Kaltefalle kondensiert wird. Nach Beendigung des Zutropfens wird auf 100 C erwarmt und bei dieser Temperatur unter Rühren das Phosgen der Kältefalle im gasförmigen Zustand durch das Reaktionsgemisch geleitet ; das nicht umgesetzte, aus dem Reaktionsgemisch entweichende Phosgen wird neuerdings in einer Kältefalle kondensiert. Das wiederholte Durchleiten des nicht zur Reaktion gelangten Phosgens durch das Reaktionsgemisch wird auf diese Weise 7 Stunden lang fortgesetzt.
Nach dem Erkalten wird filtriert, das vorhandene Phosgen aus der klaren Lösung mit dem Toluol abdestilliert und der Rückstand im Vakuum destilliert. Das Hexamethylen-bis- (N-methyl-carbaminsäurechlorid) geht bei einem Druck von 0, 2 mm Hg bei 169 bis 170 C als farbloses, ziemlich viskoses 01 über, das bald vollständig kristallisiert. Die Ausbeute beträgt 10, 6 Teile.
Eine Lösung von 11, 9 Teilen m-Dimethylamino- phenol in 90 Teilen Xylol (Isomerengemisch) wird zu einer Natriummethylatlösung von 2, 0 Teilen Na in 25 Teilen Methanol hinzagefügt ; hierauf wird so weit abdestilliert, bis das Methanol vollständig entfernt und der Siedepunkt des Xylols erreicht ist. Zu dem so erhaltenen Rückstand, in dem sich das Natriumsalz des m-Dimethylamino-phenols in fester, kristallisierter Form befindet, werden 9, 9 Teile Hexa methylen-bis-(N-methyl-carbaminsäurechlorid) hinzugefügt.
Das Reaktionsgemisch wird unter Rühren 3 Stunden auf 100 C erhitzt, nach dem Erkalten 3mal mit Wasser, 3mal mit 5 o/siger Natronlauge und anschliessend noch 3mal mit Wasser gewaschen ; sodann wird das Xylol im Vakuum abgedampft und der ölige Rückstand durch Stehen an der Luft von. den letzten Resten Xylol befreit, wobei das Produkt vollständig kristallisiert. Es werden so 15, 6 Teile Hexamethylen-bis- (N-methyl-carbaminsäure-m-dimethyl- amino-phenylester) erhalten. Durch Umkristallisieren aus Methanol oder aus Aceton-Petroläther wird die zuvor meist etwas gefärbte Substanz in Form eines weissen, feinkristallinen Pulvers mit einem Mikroschmelzpunkt von 88, 5 bis 90 C gewonnen.
33, 4 Teile Hexamethylen-bis-(Nmethyl-carbamin säure-m-dimethylamino-phenylester) werden in 85 Teilen Methyläthylketon gelöst, 100 Teile absoluten Athanols und 50 Teile Jodmethyl hinzugefügt. Nach 4tägigem Stehen bei Raumtemperatur wird bis zur beginnenden Trübung Methyläthylketon hinzugefügt und 8 Tage bei Raumtemperatur belassen. Das feinkristallin ausgefallene Produkt wird abgesaugt, mit Methyläthylketon gewaschen und im Vakuum getrocknet. Es werden so 32, 6 Teile Hexamethylenbis- (N-methyl-carbaminsäure-m-dimethylamino- phenylester-jodmethylat) erhalten. Durch weitere Zugabe von Methyläthylketon ist noch mehr von der gleichen Substanz zu gewinnen.
Sie kann durch Umfällen aus einer Lösung in absolutem Athanol mit Methyläthylketon gereinigt werden und stellt ein wei sses, feinkristallines Pulver mit einem Mikroschmelzpunkt von 138 bis 139, 5 C. dar.
Durch Quaternisierung des Hexamethylen-bis (N-methyl-carbaminsäure-m-dimethylamino-phenyl- esters) mit andern Alkylierungsmitteln, wie zum Beispiel einem Athylhalogenid, Allylhalogenid oder Benzylhalogenid, sind die entsprechenden bis-quar tären Verbindungen erhältlich. Am leichtesten reagieren dabei die Jodide und Bromide, so dass die quar tären Verbindungen schon bei Raumtemperatur hergestellt sind. Bei den Chloriden ist vielfach Erwärmen notwendig, um lange Reaktionszeiten bei der Quater nisierung zu vermeiden.
Beispiel 2
In gleicher Weise wie das im Beispiel 1 beschriebene Hexamethylen-bis- (N-methyl-carbaminsäure- chlorid) wird das Octamethylen-bis- (N-methyl-carbaminsäurechlorid) hergestellt. Das nach dem Abdestillieren des Toluols erhaltene Rohprodukt kann ohne weitere Reinigung für die weitere Umsetzung verwendet werden.
Das aus 19 Teilen N, N, N', N'-Tetramethyl-octamethylen-diamin hergestellte rohe Octamethylen-bis (N-methyl-carbaminsäurechlorid) wird mit 30 Teilen in Xylol suspendiertem Natriumsalz von m-Dimethyl- amino-phenol durch 4stündiges Rühren bei 100 C zur Reaktion gebracht. Das erkaltete Reaktionsprodukt wird mit Wasser, mit 5"der Natronlauge, hierauf wieder mit Wasser gewaschen und anschliessend das Xylol im Vakuum abgedampft. Es werden so 26 Teile Octamethylen-bis- (N-methyl-carbaminsäure-m-di- methylamino-phenylester) erhalten.
10, 9 Teile Octamethylen-bis- (N-methyl-carbamin säure-m-dimethylamino-phenylester) werden in 40 Teilen Benzol gelöst und unter Zusatz von Tierkohle 1 Stunde unter Rückfluss gekocht, dann filtriert und das Benzol abdestilliert. Der Rückstand wird in 100 Teilen acetonischer Methylbromidlösung mit einem Gehalt von 13 Teilen Methylbromid gelöst und 4 Tage bei Raumtemperatur stehengelassen. Die ausgefallenen Kristalle werden abgesaugt, mit Aceton gewaschen und im Vakuum getrocknet. Es werden so 12, 4 Teile Octamethylen-bis- (N-methyl-carbamin säure-m-d'unethylamino-phenylester-brommethylat) erhalten.
Das aus einer Lösung in Eisessig mit Me thyläthylketon umgefällte Produkt ist ein weisses, feinkristallines Pulver und zeigt einen Schmelzpunkt von 150 bis 154 C.
Beispiel 3
Das Decamethylen-bis-(N-methyl-carbaminsäure m-dimethylamino-phenylester-brommethylat) wird auf gleiche Weise wie das im Beispiel 2 beschriebene Octamethylen-bis-(N-methyl-carbaminsäure-m-dime- thylamino-phenylester-brommethylat) hergestellt. Das dabei als weisses, feinkristallinisches Pulver erhaltene Decamethylen-bis- (N-methyl-carbaminsäure-m-di methylamino-phenylester-brommethylat) zeigt aus einer Lösung in Eisessig mit Methyläthylketon umgefällt einen Mikroschmelzpunkt von 164 bis 170 C.
Beispiel 4
15 Teile N, N, N', N'-Tetramethyl-hexamethylendiamin werden unter Rühren in 100 Teile einer 35 /o Phosgen enthaltenden Toluollösung eingetropft und das Reaktionsgemisch 6 Stunden bei 100 C gehalten, wobei das entweichende, nicht umgesetzte Phosgen kondensiert und immer wieder gasförmig durch das Reaktionsgemisch geleitet wird. Nach 12stündigem Stehen bei Raumtemperatur wird filtriert ; von der klaren Toluollösung werden 60 Teile abdestilliert und zu der verbleibenden Toluollösung des Hexamethylen bis-(N-methyI-carbaminsäurechlorids) 40 Teile 8-Di- methylamino-äthanol zugesetzt, wobei unter Erwär- mung eine Fällung entsteht.
Nach 12stündigem Stehen bei Raumtemperatur werden die gebil. deten hygroskopischen Kristalle (Hydrochlorid des Dimethyl amino-äthanols) abgetrennt. Die Lösung wird im Vakuum durch schwaches Erwärmen vom Toluol befreit und der erhaltene Rückstand zur Entfernung des überschüssigen Dimethylamino-äthanols eine Woche in einer Schale im Vakuum über konzentrierter Schwefelsäure stehengelassen. Auf diese Weise werden 21 Teile Hexamethylen-bis- (N-methyl-carbamin säure-dimethylamino-äthylester) als praktisch geruchloses 0l erhalten.
21 Teile. des so erhaltenen Hexamethylen-bis- (Nmethyl-carbaminsäure-dimethylamino-äthylesters) werden in 125 Teilen Aceton gelöst und 40 Teile Methyljodid hinzugefügt. Unter Erwärmen tritt Reaktion ein ; es scheiden sich bald feine weisse Kristalle ab, die nach 12stündigem Stehen bei Raumtemperatur abgesaugt, mit Aceton gewaschen und getrocknet werden. Es werden so 35, 3 Teile Hexamethylen-bis (N-methyl-carbaminsäure-cholinjodidester) erhalten, die nach dem Umfällen aus einer Lösung in Methanol mit Aceton einen Mikroschmelzpunkt von 169 bis 171 C zeigen.
Beispiel S
2 Teile Natrium werden in 24 Teilen Methanol gelöst, zu der gebildeten Natriummethylatlösung 8, 25 Teile 3-Oxy-pyridin und 90 Teile Xylol (Isomerengemisch) hinzugefügt. Nun wird unter Stickstoff als Schutzgas so weit abdestilliert, bis der Siedepunkt des Xylols erreicht und das gesamte Methanol entfernt ist. Der Rückstand wird mit einer Lösung von 11, 7 Teilen Hexamethylen-bis- (N-methyl-carb aminsäure-chlorid) in 45 Teilen Xylol versetzt und unter starkem Rühren 4 Stunden bei 80 C gehalten.
Nach dem Erkalten wird 3mal mit Wasser, 3mal mit 5 íoiger Natronlauge und dann noch 3mal mit Wasser gewaschen. Die Xylollösung wird über Natriumsulfat getrocknet und das Xylol im Vakuum vollständig abdestilliert. Es werden so 11, 0 Teile Hexamethylen-bis- (N-methyl-carbaminsäure-3- pyridylester) erhalten.
6 Teile Hexamethylen-bis-(N-methyl-carbamin säure-3-pyridylester) werden in 8 Teilen Aceton gelöst und diese Lösung mit 10 Teilen Jodmethyl vermischt, worauf bei Raumtemperatur stehengelassen wird. Sobald die Reaktion beendet ist, wird das kristallin abgeschiedene Hexamethylen-bis- (N-methyl- carbaminsäure-3-pyridylesterjodmethylat) abgesaugt, mit Aceton gewaschen und getrocknet. Das so erhaltene Rohprodukt wird aus 24 Teilen 96 tigern Athanol umgelöst und dabei 7, 5 Teile Hexamethylenbis- (N-methyl-carbaminsäure-3-pyridylester-jodme- thylat) mit einem Schmelzpunkt von 152 bis 157 C erhalten.
7, 3 Teile Hexamethylen-bis- (N-methyl-carbamin säure-3-pyridylester) werden in 120 Teilen Aceton gelöst, mit 22 Teilen Methylbromid versetzt und bis zur Beendigung der Reaktion bei Raumtemperatur stehengelassen, wobei Kristalle ausfallen. Das abgesaugte und getrocknete Reaktionsgut (9, 9 Teile) kann durch Lösen in Eisessig und Fällen mit Methyläthylketon gereinigt werden. Das Hexamethy len-bis-(N-methyl-carbaminsäure-3-pyridylester-brom- methylat) zeigt einen Mikroschmelzpunkt von 147 bis 150 C.
Beispiel 6
Wie im vorstehenden Beispiel für den Hexamethylen-bis- (N-methyl-carbaminsäure-3-pyridyl- ester) beschrieben, werden aus 10 Teilen Decamethy len-bis- (N-methyl-carbaminsäure-chlorid) 10, 1 Teile Decamethylen-bis-(N-methyl-carbaminsäure-3-pyridyl- ester) erhalten.
13, 1 Teile Decamethylen-bis- (N-methyl-carbamin säure-3-pyridylester) werden in 8 Teilen abs. Athylalkohol und 200 Teilen Aceton gelöst, diese Lösung mit 40 Teilen Methylbromid versetzt und bei Raumtemperatur stehengelassen. Nach beendeter Reaktion wird das kristallin ausgefallene Decamethylen-bis-(N- methyl-carbaminsäure-3-pyridylester-brommethylat) aus einem Gemisch von abs. Athylalkohol und Me thyläthylketon umgefällt, wobei 9, 4 Teile mit einem Mikroschmelzpunkt von 100 bis 103 C erhalten werden.
Beispiel 7
4, 7 Teile gemäss Beispiel 1 hergestellter Hexa methylen-bis-(N-methyl-carbaminsäure-m-dimethyl- amino-phenylester) werden in 40 Teilen Methyläthyl- keton gelöst und diese Lösung mit 3, 15 Teilen Dimethylsulfat versetzt. Nach mehrstündigem Stehen bei Raumtemperatur fällt das gebildete Hexamethy len-bis- (N-methyl-carbaminsäure-m-dimethylamino- phenylester-methosulfat) als dickes Ol aus.
Beispiel 8
4, 7 Teile gemäss Beispiel 1 hergestellter Hexa methylen-bis-(N-methyl-carbaminsäure-m-dimethyl- amino-phenylester) werden in 40 Teilen Methyläthyl- keton gelöst und diese Lösung mit 6, 3 Teilen Athyljodid versetzt. Nach mehrtägigem Stehen bei 25 bis 30 C lässt sich das gebildete Hexamethylen-bis- (N- methyl-carbaminsäure-medimethylamino-phenylester- jodäthylat) mit für derartige Quartärprodukte üblichen Fällungsmitteln, wie zum Beispiel Benzol, ausfällen.
Beispiel 9 5, 0 Teile gemäss Beispiel 2 hergestellter Octa methylen-bis- (N-methyl-carbaminsäure-m-dimethyl- amino-phenylester) werden in 35 Teilen Methyläthyl- keton gelöst. Diese Lösung wird mit einer Lösung von 3, 8 Teilen p-Toluolsulfosäuremethylester in 20 Teilen Methyläthylketon versetzt und das Reaktionsgemisch 9 Stunden bei 60 C gehalten. Das
Octamethylen-bis- (N zmethyl-carbaminsäureZm-di- methylamino-phenylester-p-toluolsulfat) fällt als dikkes 01 aus.
An Stelle des in den vorhergehenden Beispielen zur Anwendung gelangten m-Dimethylamino-phenols können mit ähnlichem Ergebnis äquimolekulare Mengen von o-Dialkylamino-phenolen oder p-Dialkylamino-phenolen bzw. auch andere m-Dialkylamino- phenole verwendet werden. Als Beispiele hierfür seien genannt : m-Diäthylamino-phenol, m-Dipropylamino-phenol, m-Diisobutylamino-phenol, m-Di-nbutylamino-phenol, m- [N-Methyl-N-äthyl-amino]- phenol sowie die entsprechenden o-bzw. p-Dialkyl amino-phenole, ferner Dicyclohexylamino-phenole, Dibenzylamino-phenole und ähnliche Verbindungen.
Beispiel 10
In gleicher Weise wie in Beispiel 1 beschrieben, wird durch Quaternisieren von Hexamethylen-bis (N-methyl-carbaminsäure-m-dimethylamino-phenyl- ester) mit Brommethyl an Stelle von Jodmethyl das Hexamethylen-bis- (N-methyl-carbaminsäure-m-di- methylamino-phenylester-brommethylat) erhalten.
Nach Reinigung durch Lösen in Eisessig und Fällen mit Methyläthylketon zeigt die Substanz einen Mikroschmelzpunkt von 170 bis 173 C.
