Verfahren zur Herstellung von Zitronensäure Vorliegende Erfindung bezieht sich auf ein sub- merses vegetatives Fermentationsverfahren zur Her stellung von Zitronensäure aus Kohlenhydrat und Eisen enthaltenden Materialien unter Verwendung eines Kulturmediums, das Kupferionen enthält.
Aus früheren Untersuchungen (u. a. USA-Patent Nr. 2492667) ist hervorgegangen, dass kleine Mengen Eisen im Kohlenhydratmedium die Erzeugung von grossen Mengen eines die Bildung von Zitronensäure hemmenden Zelltypus des fermentierenden Organis mus begünstigt. Es wurde auch gezeigt, dass bei einer konstant gehaltenen Fe+++-Konzentration von etwa 0,0002-0,0008 /0o die unerwünschte Wirkung des Eisens durch die Verwendung von Zn-I-+ vermindert werden kann.
Zweck der Erfindung ist nun die Verminderung der Bildung unproduktiver vegetativer Zellen und die Eliminierung der nachteiligen Wirkung des Eisens auf die Zellen, ohne das Eisen aus dem Kulturmedium entfernen zu müssen.
Das erfindungsgemässe Verfahren zur Herstellung von Zitronensäure durch submerse Fermentation mittels Aspergillus niger eines Kohlehydrat ent haltenden Mediums, das einen für die Zitronensäure produktion an sich ungünstigen Gehalt von 0,2 Teilen oder mehr Eisen pro Million Teile Medium sowie einen für die Zitronensäureproduktion ungünstigen Gehalt an andern Kationen aufweist, ist dadurch ge kennzeichnet,
dass durch Einverleibung von Kupfer ionen abgebenden Verbindungen in der Fermenta- tionsbrühe am Aspergillus niger während der Haupt wachstumsperiode eine Änderung der Zellstruktur und des Metabolismus herbeigeführt wird, welche sich in einer vermehrten Zitronensäurebildung auswirkt.
Unter der Hauptwachstumsperiode des Aspergillus niger wird diejenige Zeit verstanden, in welcher die Mikroorganismen ihr Hauptwachstum entwickeln, was ungefähr in den ersten 50 Stunden nach Inocu- lierung eintritt. Nach dieser Periode ist nur noch ein unbedeutendes Wachstum zu beobachten.
Man hat die Wirkung von Spuren Cu++ auf das allgemeine Sporen- und vegetative Wachstum bei optimalen Zuchtbedingungen, so wie sie bei Ober flächen-Fermentationsmethoden bestehen, eingehend studiert. Sogar unter diesen Umständen war ein be stimmtes Ansprechen der Pilze auf das Kupfer schwie rig zu erreichen. Noch weniger kennt man den Ein fluss des Cu++ auf das Wachstum und den Stoff- wechsel solcher Pilze unter Bedingungen, die ihren natürlichen Verhältnissen fremd sind, wie z.
B. bei submerser Zucht, wobei die normale Zellmorphologie in hohem Grade verändert wird und dadurch bio chemische Reaktionen auf bestimmte Ziele gelenkt werden.
An Hand der Beispiele wird nun der spezielle, dem Eisen entgegenwirkende Einfluss, welcher durch die Verwendung von Kupferionen im Fermentations- substrat hervorgerufen wird, erläutert. Fermentations- bedingungen und Analysenmethoden sind dieselben wie die im USA-Patent Nr. 2492667 beschriebenen. Es wurde dabei eine ausgewählte Aspergillus-niger-Art als Vergleichsbasis zur Bestimmung der Wirkung des Kupfers verwendet. Die typischen oder optimalen Zuchtmerkmale sind die gleichen, wie im oben ge nannten Patent beschrieben.
Man gibt Kupfer vorzugsweise in Form von Kupfersulfat zu, jedoch kann jedes im Kulturmedium lösliche, weder ein giftiges noch ein irgendwie un erwünschtes Anion enthaltende Kupfersalz verwendet werden. Ist die erforderliche Kupfermenge bereits aus früheren Versuchen bekannt, so kann die ganze Menge bereits vor der Inoculierung dem Medium zugesetzt werden. Andernfalls kann das Kupfer in Portionen während der Hauptwachstumsperiode des Aspergillus niger zugesetzt werden, wobei der Bedarf durch die Entwicklung einer fadenähnlichen, nicht säurebildenden Zellstruktur angezeigt wird.
<I>Beispiel 1</I> Nicht kationenfrei gemachter, Asche und Eisen enthaltender Maiszucker wurde für ein die nach folgende Zusammensetzung aufweisendes Gärmedium verwendet, welchem ausserdem die in Tabelle 1 an gegebenen wechselnden Mengen Cu++ und Fe<B>...</B> neben 0,05 /0o Zn++ zugefügt wurden:
Ammoniumcarbonat 0,2% KHZP04 0,014% M9S04 0,1 Kohlenhydrat 12-15% Wasser (kationenfrei) q. s. 4000 cm3 Das Medium wurde mit HZSO4 von Pharma- kopöe-Qualität auf px 2,5-2,65 gestellt, in einem Autoklav bei 0,7 kg/cm2 Überdruck während 10 Mi nuten sterilisiert und in einen Fermentierungszylinder eingeführt.
Die Säuren sind als Zitronensäuremono- hydrat und der umgesetzte Zucker auf die nach der Analysenmethode von Somogyi bestimmte Anfangs konzentration der fermentierbaren Kohlenhydrate berechnet.

Bei fehlenden Kupferionen entwickelte sich in der Aspergillus-niger- Art eine fadenähnliche, keine Säure bildende Zell- form. Mit der eventuellen Ausnahme des Versuchs Nr. 6, in welchem 0, t 0100 Kupfer die Bildung einer Zucht mit vermindertem Säurebildungsvermögen nicht verhinderte, fermentierten alle Versuche mit 0,1 bis 0,20/00 Cu++ befriedigend.

Bei 0,320/00 Zn++ zeigte sich eine Neigung zur Bildung von säurebildendem Zellmaterial. Im Vergleich dazu ist es interessant festzustellen, dass Kupfer schon bei 0,02%0 zur Er- zeugung einer ganz typischen Zeltform imstande war.
<I>Beispiel 4</I> Auch in den Fällen, bei denen es wegen des vermin derten Aschengehaltes vorteilhafter war, mit kationen- freien, Kohlenhydrat enthaltenden Rohstoffen zu arbeiten, war die hemmende Wirkung des Kupfers noch immer deutlich erkennbar. Die nachfolgende Tabelle zeigt die dem Eisen entgegenwirkenden Eigen schaften des Cu++ und Zn++ in einem Nährmedium, welchem entkationisierter Maiszucker und 0,010/00 Fe... zugegeben wurden.
Das für diesen Vergleich verwendete Maiszuckermedium wies die nachfolgende Zusammensetzung auf: Ammoniumcarbonat 0,2 /o KH,P04 0,014 /o MgS0, 0,10/0 CaC12 0,0060/00 Mo03 0,050/00 H20 0,0060/00 Morpholin 0,50/00 Kohlenhydrat (kationenfrei) 12,0-15,00/0 Wasser (kationenfrei) q. s.
4000 cm3 Dieses Kulturmedium wurde mit Salzsäure von Pharmakopöe-Qualität auf pH 2,5-2,65 gestellt, in einem Autoklav bei 0,7 kg/cm3 Überdruck während 10 Minuten sterilisiert und in den Fermentierungs- zylinder eingeführt.
Die in Tabelle 4 gezeigten Versuchergebnisse dieses Kulturmediums können mit den Resultaten des nicht kationenfrei gemachten Maiszuckers in Tabelle 3 verglichen werden. Das Zink versagte in jeder nor malen Konzentration bei der Bildung von gutem säurebildendem Zellmaterial, während ein beinahe normales Zellwachstum bei nur 0,0250/00 Kupfer auftrat.

Die grosse Bedeutung dieser entdeckten Wirkung des Kupfers liegt darin, dass man durch Benützung dieser Eigenschaft das teure Entionisieren des Zuckermediums vermeiden kann.
Dieses Ionenfreimachen war bei den früheren Me thoden für die Entfernung des Hauptteils der Eisen ionen und folglich auch für die Verminderung der Konzentration daraus in dem zu fermentierenden Kulturmedium unerlässlich. Überdies ermöglicht der entdeckte, dem Eisen entgegenwirkende Einfluss des Kupfers die Verwendung von Reaktionsteilnehmern von technischer Qualität, die bis jetzt wegen ihres verhältnismässig hohen Schwermetallgehaltes (Eisen inbegriffen) nicht in Frage kommen.
Zudem bedarf es geringer Sorgfalt hinsichtlich der Wahl des Reak tionsgefässes und der Zeit der Lagerung in diesen Gefässen, weil aufgenommene Verunreinigungen wenig Einfluss auf die Art der Zellform haben werden, falls Kupfer vorhanden ist.

Using the examples, the special influence that counteracts iron, which is caused by the use of copper ions in the fermentation substrate, will now be explained. Fermentation conditions and analytical methods are the same as those described in US Patent No. 2492667. A selected Aspergillus niger species was used as a basis for comparison to determine the effect of copper. The typical or optimal breeding traits are the same as described in the patent mentioned above.
Copper is preferably added in the form of copper sulphate, but any copper salt which is soluble in the culture medium and contains neither a poisonous nor any undesirable anion can be used. If the required amount of copper is already known from previous experiments, the entire amount can be added to the medium before inoculation. Otherwise, the copper can be added in portions during the main growth period of the Aspergillus niger, the need being indicated by the development of a thread-like, non-acidic cell structure.
<I> Example 1 </I> Ashes and iron-containing corn sugar that has not been depleted of cations was used for a fermentation medium with the following composition, which also contains the alternating amounts of Cu ++ and Fe given in Table 1. B> in addition to 0.05 / 0o Zn ++ were added:
Ammonium carbonate 0.2% KHZP04 0.014% M9S04 0.1 carbohydrate 12-15% water (cation-free) q. s. 4000 cm3 The medium was adjusted to px 2.5-2.65 with HZSO4 of pharmaceutical quality, sterilized in an autoclave at 0.7 kg / cm2 overpressure for 10 minutes and introduced into a fermentation cylinder.
The acids are calculated as citric acid monohydrate and the converted sugar based on the initial concentration of fermentable carbohydrates determined by Somogyi's analytical method.

The test results of this culture medium shown in Table 4 can be compared with the results of the corn sugar in Table 3 which has not been made cation-free. The zinc failed to produce good acid-producing cell material at any normal concentration, while almost normal cell growth occurred with only 0.0250 / 00 copper.

The great importance of this discovered effect of copper lies in the fact that by using this property one can avoid the expensive deionization of the sugar medium.
In the earlier methods, this ion-freeing was essential for removing the majority of the iron ions and consequently also for reducing the concentration thereof in the culture medium to be fermented. In addition, the discovered influence of copper, which counteracts iron, enables the use of reactants of technical quality, which up to now have not been possible due to their relatively high heavy metal content (including iron).
In addition, little care is required with regard to the choice of the reaction vessel and the time of storage in these vessels, because absorbed impurities will have little influence on the type of cell shape if copper is present.
