Verfahren zur Herstellung von Diphenylalkenderivaten Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Dialkylaminoalkyläthern von Diphenylalkenen oder von Salzen der genannten basischen Äther.
Das Verfahren gemäss der vorliegenden Erfin dung ist dadurch gekennzeichnet, dass man eine Ver bindung der Formel:

worin R und R1 Alkylenreste und R2 und R3 Alkyl- reste bedeuten, mit einer alkyl-, aralkyl- oder aryl- metallorganischen Verbindung zur Umsetzung bringt,
die entstandene Metallverbindung des tertiären Carbinols hydrolysiert und das entstandene tertiäre Carbinol einer intramolekularen Wasserabspaltung unterwirft. Das erhaltene Diphenylalken kann nach bekannten Methoden in Salze übergeführt werden.
Der in der obigen Formel verwendete Ausdruck Alkylen bezieht sich vorzugsweise auf niedere Alkylenreste. Diese Reste können eine gerade oder verzweigte Kette aufweisen. Die mit R2 und R3 be zeichneten Reste sind vorzugsweise niedere Alkyl- reste. Zur Durchführung des erfindungsgemässen Ver fahrens können beispielsweise die folgenden Ketone als Ausgangsmaterialien verwendet werden:
[p-(ss-Diäthylamino-äthoxy)-phenyl]- (phenäthyl)-keton, [p-(ss-Diäthylamino-äthoxy)-phenyl]-(ss-phenyl- butyl)-keton, 1-[p-(ss-Diäthylamino-äthoxy)-phenyl]-2-methyl- 3-phenyl-propanon-(1), [p-(y-Dipropylamino-propoxy)-phenyl]- (phenäthyl)-keton, [p-(y-Diäthylamino-propoxy)-phenyl]- (phenäthyl)-keton, [p-(y-Diäthylamino-propoxy)-phenyl]-(benzyl)- keton, [p-(ss-Diäthylamino-äthoxy)
-phenyl]-(ss-phenyl- propyl)-keton, [p-(ss-Diäthylarnino-äthoxy)-phenyl]-(phenäthyl)- keton usw.
Zur Durchführung des erfindungsgemässen Ver fahrens verwendbare Ketone können durch Umsetzung eines Alkalimetallsalzes eines Hydroxyphenyl-styryl- ketons oder eines im aliphatischen Teil des Moleküls alkylsubstituierten Hydroxyphenyl-styryl-ketons mit einem Dialkyl-, vorzugsweise mit einem Di(nieder- alkyl)-aminoalkylhalogenid nach folgender Gleichung:

und katalytische Hydrierung des erhaltenen unge sättigten Ketons erhalten werden.
In den obigen Formeln bedeutet M ein Alkali metall und X ein Halogen.
Als Ausgangsstoffe verwendbare [p-(Dialkyl- aminoalkoxy)-phenyl]-(phenylalkyl)-ketone können zwischen den beiden Phenylkernen eine unverzweigte oder verzweigte Kette enthalten.
Überdies kann man an das C-Atom in der a-Stel- lung, bezogen auf die Ketogruppe, Alkylgruppen an knüpfen, z. B. indem man ein gesättigtes- Keton mit Natriumamid zur Reaktion bringt und das erhaltene Natriumderivat des Ketons mit einem Alkylhalogenid zur Umsetzung bringt.
Die Einführung von Alkyl- gruppen in die ss-Stellung, bezogen auf die Keto- gruppe, kann durch Umsetzung eines ungesättigten Ketons mit einem Grignard-Reagens, z. B. einem Alkylmagnesiumhalogenid, wie Methylmagnesium- jodid, Äthylmagnesiumbromid usw., bewerkstelligt werden.
Das Verfahren gemäss der vorliegenden Erfin dung kann z. B. wie folgt durchgeführt werden: Die gesättigten Ausgangsketone werden mit Gri- gnard-Reagenzien, beispielsweise Alkyl-, Aralkyl- oder Arylmagnesiumhalogeniden, wie Methyl-, Äthyl-, Propyl-, Butyl-, Benzyl- oder Phenylmagnesium- chlorid oder -bromid, oder mit alkyl-,
aralkyl- oder aryl-alkalimetallorganischen Verbindungen, wie Me- thyl-, Äthyl-, Propyl-, Butyl-, Benzyl- oder Phenyl- lithium, zur Umsetzung gebracht. Diese Reaktion wird vorzugsweise in Diäthyläther bei niedriger Tempe ratur, z. B. zwischen -10 und 0 C, durchgeführt. Die entstandenen Metallverbindungen der tertiären Carbinole werden dann hydrolysiert, z. B. durch Be handlung mit kaltem Wasser.
Aus den tertiären Car- binolen spaltet man hierauf intramolekular Wasser ab, z. B. durch Behandlung mit heissem Essigsäure anhydrid.
Die gemäss dem vorliegenden Verfahren erhält lichen Diphenylalkene können durch folgende Formel dargestellt werden:

In dieser Formel bezeichnet A einen Alkyl-, Aralkyl- oder Arylrest. R' stellt einen dreiwertigen Rest dar, der dem Rest R entspricht, jedoch ein Wasserstoff atom weniger aufweist als der Rest R.
Die nach dem vorliegenden Verfahren erhältlichen basischen Äther bilden mit anorganischen und orga nischen Säuren, wie z. B. Salzsäure, Bromwasserstoff säure, Jodwasserstoffsäure, Schwefelsäure, Phosphor säure, Oxalsäure, Citronensäure, Essigsäure, Milch säure, Weinsäure, Benzolsulfonsäure usw., Additions salze. Diese Salze können durch Umsetzung der Base mit der entsprechenden Säure, vorzugsweise in einem Lösungsmittel, erhalten werden. Da die Bildung von Additionssalzen mit Säuren eine zweckmässige Me thode zur Isolierung der reinen Verbindung in kristal liner Form darstellt, ist es häufig von Vorteil, die erhaltene rohe Base mit einer Säure, wie z. B.
Salz säure oder Citronensäure, zu behandeln, um das kristalline Säure-Additionssalz auszufällen, aus wel chem die reine freie Base durch Neutralisieren mit einer Base, wie z. B. Natriumhydroxyd, erhalten wer den kann. Die rohe Base kann auch durch Destilla tion gereinigt werden.
Die freien Basen bilden auch quaternäre Ammo- niumsalze mit acyclischen und cyclischen Ouaterni- siermitteln, wie z. B. niederen Alkylhalogeniden (z. B. Methylbromid, Äthyljodid, n-Butylchlorid, Di(nieder- alkyl)-sulfaten (z. B. Dimethylsulfat), Aralkylhalo- geniden (z.
B. Benzylbromid) usw.
Die nach dem Verfahren gemäss der vorliegenden Erfindung erhältlichen basischen Äther und ihre Salze sind als Chemotherapeutika, z. B. als fungicide oder fungistatische Mittel (z. B. bei der Behandlung von durch die Organismen Trichophyton mentagrophytes und Microsporon lanosum hervorgerufenen Infektio nen) und als protozoentötende Mittel (z. B. bei der Behandlung von durch Trichomonas vaginalis hervor gerufenen Infektionen) verwendbar.
In den folgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Einer ätherischen Lösung von Methyllithium [her gestellt aus 200 cm3 Äther, 2,8 g (0,405 Mol) Lithiumdraht und 28 g (0,198 Mol) frisch destillier tem Methyljodid (J. A. C. S., 55, 1252 [1933], Me thode C)] wurde eine Lösung von 40,7 g (0,125 Mol) [p - (ss - Diäthylamino- äthoxy) - phenyl] - (phenäthyl)- keton in 60 cm3 Äther zugesetzt.
Die Temperatur wurde mittels eines Kohlendioxyd-Aceton-Bades zwi schen -10 und -5 gehalten. Die für die Zugabe der Ketonlösung benötigte Zeit betrug 30 Minuten. Die so erhaltene Suspension wurde bei - 5 bis 0 während einer weiteren Stunde gerührt, worauf das gerührte Reaktionsgemisch mit 125 cm3 kaltem Was ser behandelt wurde, das anfänglich sehr vorsichtig aus einem Tropftrichter zugegeben wurde. Während der Zugabe des Wassers wurde die Temperatur durch äusserliche Kühlung unterhalb 10 gehalten. Das zwei- phasige Gemisch wurde gerührt, während es sich auf Raumtemperatur erwärmte. Es entstanden zwei klare Phasen, die hierauf getrennt wurden.
Die wässrige Schicht wurde mit 125 cm3 Äther extrahiert. Der Ätherextrakt wurde mit der abgetrennten Äther schicht vereinigt. Hierauf wurde die Ätherlösung über Natriumsulfat getrocknet und im Vakuum einge dampft, wodurch 2 - [p - (P-Diäthylamino - äthoxy)- phenyl]-4-phenyl-butanol-(2) als farbloses Öl erhal ten wurde.
Die ölige Base wurde in 60 em3 Aceton gelöst und mit einer Lösung von 32 g Citronensäure-monohydrat in 120 cm3 Aceton behandelt. Das auf diese Weise erhaltene Produkt, nämlich 2-[p-(ss-Diäthylamino- äthoxy)-phenyl]-4-phenyl-butanol-(2)-citrat, wurde aus Methyläthylketon umkristallisiert und wies dann einen Smp. von 118-119 auf.
Eine Lösung von 103 g (0,302 Mol) 2-[p-(ss- Diäthylamino-äthoxy)-phenyl]-4-phenyl-butanol-(2) in 500 cm3 Essigsäure-anhydrid wurde während 18 Stunden unter Rückfluss gekocht.
Das Essigsäure anhydrid wurde im Vakuum abdestilliert. Das zurück bleibende Öl wurde mit einem Gemisch von 700 cm- Äthanol, 150 em3 50 % igem Natriumhydroxyd und 150 cm3 Wasser während 8 Stunden unter Rückfluss gekocht.
Das Äthanol wurde im Vakuum abdestilliert, worauf der Rückstand nach Zugabe von 750 cm3 Wasser dreimal mit je 250 cm3 Äther extrahiert. Der Ätherextrakt wurde über Natriumsulfat getrock net und dann im Vakuum eingedampft, wobei die Base, 2-[p-(ss-Diäthylamino-äthoxy)-phenyl]-4-phenyl- buten-(2) als Öl zurückblieb.
Die auf diese Weise erhaltene Base wurde in 250 cm3 Aceton gelöst und mit einer Lösung von 70 g (0,33 Mol) Citronensäure-monohydrat in 250 crn3 Aceton behandelt. Das 2-[p-(ss-Diäthyl- amino-äthoxy)-phenyl]-4-phenyl-buten-(2)-citrat wurde aus 2600 cm3 Methyläthylketon umkristallisiert; Smp. 117-119 .
<I>Beispiel 2</I> Aus 34 g (0,105 Mol) [p-(ss-Diäthylamino- äthoxy)-phenyl]-(phenäthyl)-keton und Äthyl-lithium wurde in der im Beispiel 1 beschriebenen Weise 3-[p-(ss-Diäthylamino-äthoxy)-phenyl]-1-phenyl-pen- tanol-(3) erhalten, dessen Citrat nach Umkristallisie- ren aus Methyl-äthyl-keton bei 87-90 schmolz.
Aus 38,8 g (0,11 Mol) 3-[p-(ss-Diäthylaniino- äthoxy)-phenyl]-1-phenyl-pentanol-(3) und 250 cm3 Essigsäureanhydrid wurde in der im Beispiel 1 be schriebenen Weise 3 - [p- (ss-Diäthylamino-äthoxy)- phenyl]-1-phenyl-penten-(2) erhalten, dessen Citrat nach Umkristallisieren aus Methyl-äthyl-keton bei 110-112 schmolz.
<I>Beispiel 3</I> Aus 46,7 g (0,143 Mol) [p-(ss-Diäthylamino- äthoxy)-phenyl]-(phenäthyl)-keton und Propyl-lithium wurde in der im Beispiel 1 beschriebenen Weise 3-[p (ss-Diäthylamino-äthoxy)-phenyl]-1-phenyl-hexanol- (3) erhalten, dessen Citrat nach Umkristallisieren aus Methyl-äthyl-keton bei 83-86 schmolz.
Aus 37,5 g (0,102 Mol) 3-[p-(ss-Diäthylamino- äthoxy)-phenyl]-1-phenyl-hexanol-(3) und 250 cm3 Essigsäureanhydrid wurde in der im Beispiel 1 be schriebenen Weise 3-[p-(ss-Diäthylamino-äthoxy)- phenyl]-1-phenyl-hexen-(2) erhalten, dessen Citrat nach Umkristallisieren aus Methyl-äthyl-keton bei 110-112 schmolz.
<I>Beispiel 4</I> Aus 19 g (0,059 Mol) [p-(ss-Diäthylamino- äthoxy)-phenyl]-(phenäthyl)-keton und Butyl-lithium wurde in der im Beispiel 1 beschriebenen Weise <B>3-[p-</B> (ss -Diäthyl amino . äthoxy) - phenyl] -1- phenyl- heptanol-(3) erhalten, dessen Citrat nach Umkristalli- sieren aus Methyl-äthyl-keton bei 91-94 schmolz.
Aus 42 g (0,11 Mol 3-[p-(ss-Diäthylamino- äthoxy)-phenyl]-1-phenyl-heptanol-(3) und 250 cm3 Essigsäureanhydridwurde in der im Beispiel 1 beschrie benen Weise 3-[p-(ss-Diäthylamino-äthoxy)-phenyl]- 1-phenyl-hepten-(2) erhalten, dessen Citrat nach Um- kristallisieren aus Methyl-äthyl-keton bei 112-113 schmolz.
<I>Beispiel 5</I> Aus 47,8 g (0,147 Mol) [p-(ss-Diäthylamino- äthoxy)-phenyl]-(phenäthyl)-keton und Phenyl-lithium wurde in der im Beispiel 1 beschriebenen Weise 1-[p (ss-Diäthylamino-äthoxy)-phenyl]-1,3-diphenyl-propa- nol-(1) erhalten, dessen Citrat nach Umkristallisieren aus Aceton-Äther bei 85-87 schmolz.
Aus 40 g (0,099 Mol) 1-[p-(ss-Diäthylamino- äthoxy)-phenyl]-1,3-diphenyl-propanol-(1) und 250cm3 Essigsäureanhydrid wurde in. der im Beispiel 1 be schriebenen Weise 1-[p-(ss-Diäthylamino-äthoxy)- phenyl]-1,3-diphenyl-propen-(1) erhalten, dessen Citrat nach Umkristallisieren aus Methyläthyl-keton bei 123-125 schmolz.
<I>Beispiel 6</I> Aus 65,5 g (0,185 Mol) [p-(ss-Diäthylamino- äthoxy)-phenyl]-(ss-phenyl-butyl)-keton und Methyl- lithium wurde in der im Beispiel 1 beschriebenen Weise 2-[p-(ss-Diäthylamino-äthoxy)-phenyl]-4-phe- nyl-hexanol-(2) erhalten, dessen Citrat nach Um kristallisieren aus Methyl-äthyl-keton und anschlie ssend aus Aceton-Äther bei 91-93 schmolz.
Aus 44 g (0,12 Mol) 2-[p-(ss-Diäthylamino- äthoxy)-phenyl]-4-phenyl-hexanol-(2) und 250 cm3 Essigsäureanhydrid wurde in der im Beispiel 1 be schriebenen Weise 2-[p-(ss-Diäthylamino-äthoxy)- phenyl]-4-phenyl-hexen-(2) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril bei 114-116 schmolz.
Das in diesem Beispiel verwendete Ausgangs material wurde wie folgt erhalten: Eine Lösung von 18 g (0,056 Mol) [p-(ss-Diäthyl- amino-äthoxy) phenyl]-(styryl)-keton in Äther wurde unter Rühren langsam dem aus 2,5 g (0,1 Mol) Magnesium, 15,6 g (0,1 Mol) Athyljodid und 200 cm3 Äther hergestellten Grignard-Reagens zugesetzt.
Das Gemisch wurde während 5 Stunden gerührt und unter Rückfluss gekocht und dann in eine kalte wässrige Lösung von Ammoniumehlorid gegossen. Die ätherische Schicht wurde abgetrennt und die wässrige Schicht wurde nochmals mit Äther ausge zogen. Die ätherischen Lösungen wurden vereinigt, über Natriumsulfat getrocknet und im Vakuum ein gedampft. Die Base, nämlich [p-(ss-Diäthylamino- äthoxy)-phenyl]-(ss-phenyl-butyl)-keton, . wurde in Form eines Öls erhalten.
Diese Base wurde durch Behandlung mit 11g Citronensäure-monohydrat in Aceton in das entsprechende Citrät vom Smp. 122 bis 123 übergeführt. . <I>Beispiel 7</I> Aus 25 g (0,074 Mol) 1-[p-(ss-Diäthylamino- äthoxy)-phenyl]-2-methyl-3-phenyl-propanon-(1) und Methyl-lithium wurde in der im Beispiel 1 beschrie benen Weise 2-[p-(ss-Diäthylamino-äthoxy)-phenyl]-3- methyl-4-phenyl-butanol-(2) erhalten,
dessen Citrat nach Umkristallisieren aus Acetonitril-Äther bei<B>101</B> bis 103 schmolz.
Aus 22,7 g (0,065 Mol) 2-[p-(ss-Diäthylamino- äthoxy)-phenyl]-3-methyl-4-phenyl-butanol-(2) und 200 cm3 Essigsäureanhydrid wurde in der im Bei spiel 1 beschriebenen Weise 2-[p-(ss-Diäthylamino- äthoxy)-phenyl]-3-methyl-4-phenyl-buten-(2) erhal ten, dessen Citrat nach Umkristallisieren aus Aceto- nitril bei 130-132 schmolz.
Das in diesem Beispiel verwendete Ausgangs material wurde wie folgt erhalten: Ein Gemisch von 100 cm3 trockenem Benzol, 9,35 g (0,243 Mol) Natriumamid (in Form einer Suspension in Mineralöl) und 68,88 g (0,211 Mol) [p-(ss-Diäthylamino-äthoxy)-phenyl]-(phenäthyl)-keton wurde während 3 Stunden gerührt und unter Rück fluss gekocht. Das Gemisch wurde auf 15 abgekühlt, und hierauf wurde innerhalb von 15 Minuten eine Lösung von 30 g (0,21 Mol) Methyljodid in 100 cm3 Benzol zugegeben.
Durch gelegentliches Kühlen wurde die Temperatur der exothermen Reaktion zwischen 35 und 50 gehalten. Nach beendeter Zugabe der Lösung wurde das Reaktionsgemisch während 1 Stunde gerührt und unter Rückfluss gekocht. Dem abgekühlten Reaktionsgemisch wurden dann vorsich tig 30 cm3 Alkohol zugesetzt. Hierauf wurde das Reaktionsgemisch mit 150 cm3 Wasser gerührt. Die benzolische Schicht wurde abgetrennt, über Natrium sulfat getrocknet und schliesslich im Vakuum destil liert.
Das auf diese Weise erhaltene Produkt, 1-[p-(ss- Diäthylamino-äthoxy) -phenyl] - 2 - methyl - 3 - phenyl- propanon-(1), destillierte bei 182 bis 190 /0,01 mm über.
<I>Beispiel 8</I> Aus 83 g (0,245 Mol) [p-(y-Diäthylamino- propoxy)-phenyl]-(phenäthyl)-keton und Phenyl- lithium wurde in der im Beispiel 1 beschriebenen Weise 1-[p-(y-Diäthylamino-propoxy)-phenyl]-1,3- diphenyl-propanol-(1) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril-Äther bei 82-84 schmolz.
Aus 99 g (0,237 Mol) 1-[p-(y-Diäthylamino- propoxy) - phenyll - 1,3-diphenyl - propanol- (1) und 500 cm3 Essigsäureanhydrid wurde in der im Bei spiel 1 beschriebenen Weise 1-[p-(y-(Diäthylamino- propoxy)-phenyl]-1,3-diphenyl-propen-(1) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril bei 93-95 schmolz.
Das in diesem Beispiel verwendete Ausgangsmate rial wurde wie folgt erhalten: Eine Lösung von 10,58 g (0,02 Mol) [p-(y- Diäthylamino-propoxy)-phenyl]-styryl)-keton-citrat in wässrigem Äthanol wurde mit 1,5 g eines Palladium- Kohlenstoff-Katalysators (10 ah, Palladium) und Was serstoff bei atmosphärischem Druck gerührt, bis 0,02 Mol Wasserstoff verbraucht waren. Der Kata lysator wurde alsdann abfiltriert. Die Mutterlösung wurde im Vakuum eingedampft, wobei ein Rückstand verblieb, der aus Aceton-Wasser umkristallisiert wurde.
Das [p-(y-Diäthylamino-propoxy)-phenyl]- (phenäthyl)-keton-citrat schmolz bei<B>136</B> bis 137 . Die freie Base wurde durch Neutralisieren des Ci- trates mit verdünntem wässrigem Natriumhydroxyd erhalten.
<I>Beispiel 9</I> Aus 62 g (0,2 Mol) [p-(j3-Diäthylamino-äthoxy)- phenyl]-(benzyl)-keton und Methyl-lithium wurde in der im Bespiel 1 beschriebenen Weise 2-[p-(ss-Di- äthylamino-äthoxy)-phenyl] - 1-phenyl - propanol-(2) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril-Äther bei 105-107 schmolz.
Aus 47,4 g (0,145 Mol) 2-[p-(ss-Diäthylamino- äthoxy)-phenyl]-1-phenyl-propanol-(2) und 500 cm3 Essigsäureanhydrid wurde in der im Beispiel 1 be schriebenen Weise 2-[p-(ss-Diäthylamino-äthoxy)- phenyl]-1-phenyl-propen-(1) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril bei 117 bis 120 schmolz.
<I>Beispiel 10</I> Aus 79,5 g (0,245 Mol) [p-(.-Diäthylamino- propoxy)-phenyl]-(benzyl)-keton und Methyl-lithium wurde in der im Beispiel 1 beschriebenen Weise 2-[p (y-Diäthylamino-propoxy)-phenyl]-1-phenyl-propanol- (2) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril-Äther bei 110-112 schmolz.
Aus 63 g (0,184 Mol) 2-[p-(" -Diäthylamino- propoxy)-phenyl]-1-phenyl-propanol-(1) und 460 cm3 Essigsäureanhydrid wurde in der im Beispiel 1 be schriebenen Weise 2-[p-(y-Diäthylamino-propoxy)- phenyl]-1-phenyl-propen-(1) erhalten, dessen Citrat nach Umkristallisieren aus Acetonitril bei 140 bis 142 schmolz.
Das in diesem Beispiel verwendete Ausgangs material wurde wie folgt erhalten: Ein Gemisch von 127 g (0,6 Mol) [p-Hydroxy- phenyl]-(benzyl)-keton, 35 g (0,65 Mol) Natrium- methoxyd und 1400 cm3 Chlorbenzol wurde gerührt und zum Sieden erhitzt. Man liess dann die auf diese Weise erhaltene Suspension des Natriumsalzes von [p-Hydroxy-phenyl]-(benzyl)-keton auf 40 abkühlen, worauf man 92,5 g (0,65 Mol) ;,-Diäthylamino- propyl-chlorid zugab.
Hierauf wurde das Gemisch während 6 Stunden gerührt und unter Rückfluss ge kocht. Nach Abfiltrieren des Natriumchlorids wurde das Filtrat im Vakuum eingedampft, wobei man [p-(y-Diäthylamino-propoxy)-phenyl] - (benzyl) -keton in Form einer öligen Base vom Smp. 207a,#0,06 mm erhielt.
10 g der auf diese Weise erhaltenen Base wurden in 30 cm3 Aceton gelöst und durch Zugabe einer Lösung von 7,2 g Citronensäure-monohydrat in 30 cm3 Aceton in das Citrat übergeführt. Das [p-(. Diäthylamino-propoxy)-phenyl]-(benzyl) - keton- citrat wurde aus Acetonitril umkristallisiert und schmolz dann bei 135-136 .

Process for the Production of Diphenylalkene Derivatives The present invention relates to a process for the production of dialkylaminoalkyl ethers of diphenylalkenes or of salts of the stated basic ethers.
The method according to the present invention is characterized in that a compound of the formula:

where R and R1 are alkylene radicals and R2 and R3 are alkyl radicals, reacts with an alkyl, aralkyl or aryl organometallic compound,
the resulting metal compound of the tertiary carbinol is hydrolyzed and the resulting tertiary carbinol is subjected to intramolecular dehydration. The diphenylalkene obtained can be converted into salts by known methods.
The term alkylene used in the above formula preferably refers to lower alkylene radicals. These residues can have a straight or branched chain. The radicals identified by R2 and R3 are preferably lower alkyl radicals. For example, the following ketones can be used as starting materials to carry out the process according to the invention:

In the following examples, the temperatures are given in degrees Celsius.
<I> Example 1 </I> An ethereal solution of methyllithium [made from 200 cm3 of ether, 2.8 g (0.405 mol) of lithium wire and 28 g (0.198 mol) of freshly distilled methyl iodide (JACS, 55, 1252 [1933 ], Method C)] a solution of 40.7 g (0.125 mol) of [p - (ss - diethylaminoethoxy) - phenyl] - (phenethyl) - ketone in 60 cm3 of ether was added.
The temperature was kept between -10 and -5 by means of a carbon dioxide-acetone bath. The time required for the addition of the ketone solution was 30 minutes. The suspension thus obtained was stirred at -5 to 0 for a further hour, after which the stirred reaction mixture was treated with 125 cm3 of cold water, which was initially added very carefully from a dropping funnel. During the addition of the water, the temperature was kept below 10 by external cooling. The two-phase mixture was stirred while warming to room temperature. Two clear phases emerged, which were then separated.
The aqueous layer was extracted with 125 cm3 of ether. The ether extract was combined with the separated ether layer. The ether solution was then dried over sodium sulfate and evaporated in vacuo, whereby 2 - [p - (P-diethylamino - ethoxy) - phenyl] -4-phenyl-butanol- (2) was obtained as a colorless oil.
The oily base was dissolved in 60 cubic meters of acetone and treated with a solution of 32 g of citric acid monohydrate in 120 cm3 of acetone. The product obtained in this way, namely 2- [p- (ss-diethylamino-ethoxy) -phenyl] -4-phenyl-butanol- (2) -citrate, was recrystallized from methyl ethyl ketone and then had a melting point of 118-119 on.
A solution of 103 g (0.302 mol) of 2- [p- (s-diethylamino-ethoxy) -phenyl] -4-phenyl-butanol- (2) in 500 cm3 of acetic anhydride was refluxed for 18 hours.
The acetic anhydride was distilled off in vacuo. The oil that remained was refluxed for 8 hours with a mixture of 700 cm3 of ethanol, 150 cm3 of 50% sodium hydroxide and 150 cm3 of water.
The ethanol was distilled off in vacuo, whereupon the residue, after the addition of 750 cm3 of water, was extracted three times with 250 cm3 of ether each time. The ether extract was dried over sodium sulfate and then evaporated in vacuo, the base, 2- [p- (ss-diethylamino-ethoxy) -phenyl] -4-phenyl-butene (2) remaining as an oil.
The base obtained in this way was dissolved in 250 cm 3 of acetone and treated with a solution of 70 g (0.33 mol) of citric acid monohydrate in 250 cm 3 of acetone. The 2- [p- (ss-diethylamino-ethoxy) phenyl] -4-phenyl-butene (2) citrate was recrystallized from 2600 cm3 of methyl ethyl ketone; 117-119.
<I> Example 2 </I> From 34 g (0.105 mol) [p- (ss-diethylamino-ethoxy) -phenyl] - (phenethyl) -ketone and ethyl-lithium, in the manner described in Example 1, 3- [ p- (ß-diethylamino-ethoxy) -phenyl] -1-phenyl-pentanol- (3), the citrate of which, after recrystallization from methyl-ethyl-ketone, melted at 87-90.
From 38.8 g (0.11 mol) of 3- [p- (ss-diethylaniino-ethoxy) -phenyl] -1-phenyl-pentanol- (3) and 250 cm3 of acetic anhydride, in the manner described in Example 1, 3 - [p- (ss-Diethylamino-ethoxy) -phenyl] -1-phenyl-pentene- (2) obtained, the citrate of which melted after recrystallization from methyl-ethyl-ketone at 110-112.
Example 3 From 46.7 g (0.143 mol) of [p- (ss-diethylamino-ethoxy) -phenyl] - (phenethyl) -ketone and propyl-lithium, 3 was obtained in the manner described in Example 1 - [p (ss-Diethylamino-ethoxy) -phenyl] -1-phenyl-hexanol- (3) obtained, the citrate of which, after recrystallization from methyl-ethyl-ketone, melted at 83-86.
From 37.5 g (0.102 mol) of 3- [p- (ss-diethylamino ethoxy) phenyl] -1-phenyl-hexanol- (3) and 250 cm3 of acetic anhydride, in the manner described in Example 1, 3- [ p- (ß-diethylamino-ethoxy) -phenyl] -1-phenyl-hexene (2) obtained, the citrate of which, after recrystallization from methyl-ethyl-ketone, melted at 110-112.
Example 4 From 19 g (0.059 mol) [p- (ss-diethylaminoethoxy) phenyl] - (phenethyl) ketone and butyl lithium, in the manner described in Example 1, <B> 3- [p- </B> (ss -diethylamino. Ethoxy) -phenyl] -1-phenyl-heptanol- (3), the citrate of which melted at 91-94 after recrystallization from methyl-ethyl-ketone.
From 42 g (0.11 mol of 3- [p- (ß-diethylamino-ethoxy) -phenyl] -1-phenyl-heptanol- (3) and 250 cm3 of acetic anhydride), in the manner described in Example 1, 3- [p- (ß-diethylamino-ethoxy) -phenyl] -1-phenyl-hepten- (2), the citrate of which melted at 112-113 after recrystallization from methyl-ethyl-ketone.
<I> Example 5 </I> From 47.8 g (0.147 mol) of [p- (ss-diethylaminoethoxy) phenyl] - (phenethyl) ketone and phenyllithium, 1 was obtained in the manner described in Example 1 - [p (ß-diethylamino-ethoxy) -phenyl] -1,3-diphenyl-propanol- (1) obtained, the citrate of which melted at 85-87 after recrystallization from acetone-ether.
From 40 g (0.099 mol) of 1- [p- (ss-diethylamino-ethoxy) -phenyl] -1,3-diphenyl-propanol- (1) and 250 cm3 of acetic anhydride, in the manner described in Example 1, 1- [ p- (ss-diethylamino-ethoxy) -phenyl] -1,3-diphenyl-propene (1) obtained, the citrate of which melted at 123-125 after recrystallization from methylethyl ketone.
<I> Example 6 </I> From 65.5 g (0.185 mol) [p- (ss-diethylaminoethoxy) phenyl] - (ss-phenyl-butyl) ketone and methyl lithium in the example 1 described manner 2- [p- (ss-diethylamino-ethoxy) -phenyl] -4-phenyl-hexanol- (2) obtained, the citrate of which crystallizes from methyl-ethyl-ketone and then from acetone-ether melted at 91-93.
From 44 g (0.12 mol) of 2- [p- (ss-diethylamino-ethoxy) -phenyl] -4-phenyl-hexanol- (2) and 250 cm3 of acetic anhydride, in the manner described in Example 1, 2- [ p- (ss-diethylamino-ethoxy) -phenyl] -4-phenyl-hexene (2) obtained, the citrate of which melted at 114-116 after recrystallization from acetonitrile.
The starting material used in this example was obtained as follows: A solution of 18 g (0.056 mol) [p- (ss-diethylamino-ethoxy) phenyl] - (styryl) ketone in ether was slowly stirred into the from 2 , 5 g (0.1 mol) of magnesium, 15.6 g (0.1 mol) of ethyl iodide and 200 cm3 of ether added Grignard reagent.
The mixture was stirred and refluxed for 5 hours and then poured into a cold aqueous solution of ammonium chloride. The ethereal layer was separated and the aqueous layer was again drawn out with ether. The ethereal solutions were combined, dried over sodium sulfate and evaporated in vacuo. The base, namely [p- (ss-diethylamino-ethoxy) -phenyl] - (ss-phenyl-butyl) ketone,. was obtained in the form of an oil.
This base was converted into the corresponding citrate with a melting point of 122 to 123 by treatment with 11 g of citric acid monohydrate in acetone. . <I> Example 7 </I> From 25 g (0.074 mol) 1- [p- (β-diethylaminoethoxy) -phenyl] -2-methyl-3-phenyl-propanone- (1) and methyl-lithium became obtained in the manner described in Example 1 2- [p- (ss-diethylamino-ethoxy) phenyl] -3-methyl-4-phenyl-butanol- (2),
the citrate of which, after recrystallization from acetonitrile ether, melted at <B> 101 </B> to 103.
From 22.7 g (0.065 mol) of 2- [p- (ss-diethylamino-ethoxy) -phenyl] -3-methyl-4-phenyl-butanol- (2) and 200 cm3 of acetic anhydride was described in the example 1 described Way 2- [p- (ss-diethylamino-ethoxy) -phenyl] -3-methyl-4-phenyl-butene (2) obtained, the citrate of which melted at 130-132 after recrystallization from acetonitrile.
The starting material used in this example was obtained as follows: A mixture of 100 cm3 of dry benzene, 9.35 g (0.243 mol) of sodium amide (in the form of a suspension in mineral oil) and 68.88 g (0.211 mol) [p- ( ß-diethylamino-ethoxy) phenyl] - (phenethyl) ketone was stirred for 3 hours and refluxed. The mixture was cooled to 15 and then a solution of 30 g (0.21 mol) of methyl iodide in 100 cm 3 of benzene was added over 15 minutes.
The temperature of the exothermic reaction was kept between 35 and 50 by occasional cooling. After the addition of the solution had ended, the reaction mixture was stirred for 1 hour and refluxed. 30 cm3 of alcohol were then carefully added to the cooled reaction mixture. The reaction mixture was then stirred with 150 cm3 of water. The benzene layer was separated off, dried over sodium sulfate and finally distilled in vacuo.
The product obtained in this way, 1- [p- (s-diethylamino-ethoxy) -phenyl] -2-methyl-3-phenyl-propanone- (1), distilled over at 182 to 190 / 0.01 mm.
<I> Example 8 </I> From 83 g (0.245 mol) [p- (γ-diethylaminopropoxy) phenyl] - (phenethyl) ketone and phenyllithium, in the manner described in Example 1, 1- [ p- (γ-Diethylamino-propoxy) -phenyl] -1,3-diphenyl-propanol- (1), the citrate of which melted at 82-84 after recrystallization from acetonitrile ether.
From 99 g (0.237 mol) 1- [p- (γ-diethylamino-propoxy) -phenyl-1,3-diphenyl-propanol- (1) and 500 cm3 acetic anhydride was in the manner described in Example 1 1- [p - (y- (Diethylamino-propoxy) -phenyl] -1,3-diphenyl-propene (1) obtained, the citrate of which melted at 93-95 after recrystallization from acetonitrile.
The starting material used in this example was obtained as follows: A solution of 10.58 g (0.02 mol) [p- (γ-diethylamino-propoxy) phenyl] -styryl) ketone citrate in aqueous ethanol was mixed with 1.5 g of a palladium-carbon catalyst (10 ah, palladium) and hydrogen was stirred at atmospheric pressure until 0.02 mol of hydrogen were consumed. The catalyst was then filtered off. The mother solution was evaporated in vacuo, leaving a residue which was recrystallized from acetone-water.
The [p- (y-diethylamino-propoxy) -phenyl] - (phenethyl) -ketone citrate melted at 136-137. The free base was obtained by neutralizing the citrate with dilute aqueous sodium hydroxide.
<I> Example 9 </I> From 62 g (0.2 mol) [p- (j3-diethylamino-ethoxy) -phenyl] - (benzyl) -ketone and methyl-lithium, in the manner described in example 1, 2 - [p- (ss-Diethylamino-ethoxy) -phenyl] -1-phenyl-propanol- (2) obtained, the citrate of which melted at 105-107 after recrystallization from acetonitrile ether.
From 47.4 g (0.145 mol) of 2- [p- (ss-diethylamino ethoxy) phenyl] -1-phenyl-propanol- (2) and 500 cm3 of acetic anhydride, in the manner described in Example 1, 2- [ p- (ß-diethylamino-ethoxy) -phenyl] -1-phenyl-propen-1, the citrate of which melted at 117 to 120 after recrystallization from acetonitrile.
<I> Example 10 </I> From 79.5 g (0.245 mol) [p - (.- diethylaminopropoxy) phenyl] - (benzyl) ketone and methyl lithium, in the manner described in Example 1, 2 - [p (γ-Diethylamino-propoxy) -phenyl] -1-phenyl-propanol- (2) obtained, the citrate of which, after recrystallization from acetonitrile ether, melted at 110-112.
From 63 g (0.184 mol) of 2- [p- ("-Diethylamino-propoxy) -phenyl] -1-phenyl-propanol- (1) and 460 cm3 of acetic anhydride was in the manner described in Example 1, 2- [p- (γ-Diethylamino-propoxy) -phenyl] -1-phenyl-propen-1, the citrate of which melted at 140 to 142 after recrystallization from acetonitrile.
The starting material used in this example was obtained as follows: A mixture of 127 g (0.6 mol) of [p-hydroxyphenyl] (benzyl) ketone, 35 g (0.65 mol) of sodium methoxide and 1,400 cm3 chlorobenzene was stirred and heated to the boil. The suspension of the sodium salt of [p-hydroxyphenyl] - (benzyl) ketone obtained in this way was then allowed to cool to 40, after which 92.5 g (0.65 mol) of diethylaminopropyl chloride were added .
The mixture was then stirred and refluxed for 6 hours. After the sodium chloride had been filtered off, the filtrate was evaporated in vacuo, giving [p- (γ-diethylaminopropoxy) phenyl] - (benzyl) ketone in the form of an oily base with a melting point of 207a, # 0.06 mm.
10 g of the base obtained in this way were dissolved in 30 cm 3 of acetone and converted into the citrate by adding a solution of 7.2 g of citric acid monohydrate in 30 cm 3 of acetone. The [p- (. Diethylamino-propoxy) -phenyl] - (benzyl) - ketone citrate was recrystallized from acetonitrile and then melted at 135-136.
