Verfahren zur Herstellung von Derivaten des 4-Hydroxy-cumarins Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von 3-substituierten 4-Hydroxy-cumarinen und ihren 1-Thia-analogen, welche zum Teil bereits bekannt sind und sogar therapeutische Anwendung als Antikoagulantia gefunden haben, zum Teil aber bisher unbekannt waren und mittels anderer Verfahren kaum oder nur mit schlechter Ausbeute zugänglich sein dürften.
Das 4-Hydroxy-cumarin zeigt in seiner Fähigkeit zur Anlagerung an a, ss-ungesättigte Ketone zwar ein den Verbindungen mit reaktionsfähiger Methylen- gruppe ähnliches Verhalten, indessen ist es unmöglich, 4-Hydroxy-cumarine analog den Acetessigsäure- und Malonsäureestern durch Umsetzung ihrer Alkalimetallsalze mit Monohalogenverbindungen direkt oder unter Umlagerung zunächst entstandener Enol- äther in 3-Stellung zu substituieren.
Überraschenderweise wurde nun gefunden, dass 4-Hydroxy-cumarin, substituierte 4-Hydroxy-cumarine und ihre 1-Thia-analogen der Formel

Als Halogenwasserstoff kann Chlorwasserstoff mit gutem Erfolg verwendet werden und als anorganisches Säurehalogenid insbesondere Phos- phoroxychlorid, ferner zum Beispiel auch Thionyl- chlorid.
Die Reaktion kann bei Temperaturen von etwa 50 bis 150Q, meist zwischen 100 und 150 , erfolgen. Es können Lösungsmittel verwendet werden, die gegenüber Halogenwasserstoff inert sind, ein genügendes Lösungsvermögen für 4-Hydroxy-cumarine besitzen, und schliesslich einen geeigneten Siedebereich besitzen, sofern die Reaktion unter Atmosphären-

druck vorgenommen werden soll. Als geeignetes Lösungsmittel hat sich zum Beispiel Tetrachlor- äthan, das obigen Bedingungen entspricht und sich mit Wasserdampf leicht entfernen lässt, erwiesen, Als weiteres Lösungsmittel kommt zum Beispiel Chlorbenzol in Betracht.
Es ist wahrscheinlich, dass bei der Umsetzung der Benzylalkohole mit den 4-Hydroxy-cumarinen stets die Halogenwasserstoffsäureester der ersteren als Zwischenprodukte auftreten und es sich deshalb bei den beiden oben definierten Verfahrensmodifikationen um dieselbe Kondensationsreaktion handelt. Indessen ist hervorzuheben, dass als Ausgangsstoffe mit gutem Erfolg auch solche Substitutionsprodukte des Benzyl- alkohols verwendet werden können, deren Halogenwasserstoffsäureester in isoliertem Zustand unbeständig oder infolge Neigung zu plötzlicher Polymerisa- tion sogar gefährlich zu handhaben sind.
Die Möglichkeit der Verwendung dieser substituierten Benzylalkohole als Ausgangsstoffe bedeutet somit eine beträchtliche Erweiterung der Auswahl an solchen Stoffen. Anderseits sind in gewissen Fällen Halogenwasserstoffsäureester zum Beispiel durch Chlormethylierung aromatischer Ringe oder durch Halogenierung von Seitenketten leichter zugänglich als die entsprechenden Alkohole.
Als Ausgangsstoffe der allgemeinen Formel 1I kommen beispielsweise in Betracht: Benzylalkohol, a-Phenyl-äthanol, -n-propanol, n-butanol, Benzhydrol, Salicylalkohol, 3,5-Dichlorsalicylalkohol, 4-Methoxy-benzylalkohol, 2-Hydroxy-3,5-dimethyl-benzylalkohol, 2-Hydroxy-3-chlor-5-methyl-benzylalkohol, 2-Methoxy-3-chlor-5-methyl-benzylalkohol, a-(2-Hydroxy-3-methyl-5-chlor-pheny1)-äthanol, a-(2-Methoxy-3-methyl-5-chlor-phenyl)- n-propanol, 2,6-Bis-hydroxymethyl-4-chlor-phenol, 4,6-Bis-hydroxymethyl-o-kresol,
2,6-Bis-hydroxymethyl-p-kresol, 2,6-Bis-hydroxymethyl-4-tert.butylphenol, 3,3'-Bis-hydroxymethyl-4,4'-dihydroxy-5,5'- dimethyl-diphenylmethan, Benzylchlorid, Benzylbromid, 2- und 4-Chlor-benzylchlorid, p-Brom-benzylbromid und Benzhydrylchlorid. , Neben 4-Hydroxy-cumarin können beispielsweise 4-Hydroxy-6-methyl-cumarin, 4-Hydroxy-8-methyl- cumarin, 4-Hydroxy-6,7-dimethyl-curnarin, 4-Hy- droxy-6-chlor-cumarin, 4-Hydroxy-6,7-dichlor-cuma- rin,
4-Hydroxy-6-chlor-8-methyl-cumarin, 4 Hydroxy- 6-brom-cumarin und 4-Hydroxy-l-thia-cumarin verwendet werden. In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Centigraden angegeben.
Beispiel 1 17 Teile 2-Hydroxy-3-chlor-5-methyl-benzylalko- hol und 20,3 Teile 4-Hydroxy-cumarin werden in 120 Volumteilen Tetrachloräthan gelöst bzw. suspendiert und in diese Suspension wird während 40 Minuten bei 50 Chlorwasserstoff eingeleitet. Man erhitzt dann auf 135 bis zum Aufhören der Chlorwasserstoffentwicklung. Nach dem Abkühlen saugt man ab und kristallisiert das 3-(2'-Hydroxy-3'-chlor- 5'-methyl-benzyl)-4-hydroxy-cumarin aus Tetra- chloräthan um; Fp. 253 bis 254 .
Durch Kochen mit Essigsäureanhydrid erhält man das Diacetat, das nach Kristallisation aus Äthylacetat bei 159 schmilzt.
In analoger Weise können zum Beispiel auch 3-(a,3'-Dimethyl-2'-hydroxy-5'-chlor-benzyl)- 4-hydroxy-cumarin, Fp. 239,5 (aus Tetrachlor- äthan); 3-(2'-Hydroxy-3',5'-dichlor-benzyl)-4-hydroxy- cumarin, Fp. 252 bis 253 (aus Tetrachloräthan); Diacetat: Fp. 167 (aus Methanol); und 3-(2'-Hydroxy-3'-methyl-5'-chlor-benzyl)-4-hydroxy- cumarin, Fp. 269 (aus Pyridin) hergestellt werden.
Beispiel 2 3,78 Teile 2,6-Bis-hydroxymethyl-4-chlor-phenol und 3,56 Teile 4-Hydroxy-cumarin werden in 120 Volumteilen Tetrachloräthan suspendiert und während einer halben Stunde wird bei 50 Chlorwasserstoff eingeleitet. Man erhitzt während 15 Minuten zum Sieden und hält dann während zwei Stunden bei 140 . Nach dem Abkühlen saugt man ab und wäscht mit Äthanol. Man kristallisiert aus Phenylacetat aus und erhält 2,6-Bis-(4'-hydroxy- cumarinyl-(3')-methyl)-4-chlorphenol vom Fp. 296 .
In analoger Weise können zum Beispiel auch 2,4-Bis-(4'-hydroxy-cumarinyl-(3')-methyl)-6-methyl- phenol, Triacetat (durch Kochen mit Acetan- hydrid) Fp. 226 bis 227 (aus Tetrachloräthan- Äthanol); 2,6-Bis-(4'-hydroxy-cumarinyl-(3')-methyl-4-tert. butyl-phenol, Fp. 252 bis 253 (aus Phenylacetat);
3,3'-Bis-(4"-hydroxy-cumarinyl-(3")-methyl)-4,4'- dihydroxy-5, 5'-dimethyl-diphenylmethan, Fp. 264 bis 265 (aus Nitrobenzol, Phenylacetat . oder Tetrachloräthan-Äthanol), Tetraacetat, Fp. 221 bis 223 (aus Toluol); und 2,6-Bis-(4'-Hydroxy-cumarinyl-(3')-methyl)- 4-methyl-phenol, Fp. 264 (aus Tetrachloräthan) erhalten werden.
Beispiel 3 In ein Gemisch von 18,4 Teilen Benzhydrol und 20 Teilen Calciumchlorid in 100 Volumteilen Tetra-

chloräthan leitet man bei Zimmertemperatur trockenen Chlorwasserstoff bis zur Sättigung ein.
Dann giesst man vom Bodensatz ab, wäscht mit 50 Volum- teilen Tetrachloräthan nach und versetzt die vereinigten Lösungen mit 18 Teilen 4-Hydroxy-l-thia- cumarin. Die erhaltene Suspension wird so lange im Ölbad auf 130 bis 140 erwärmt, bis kein Chlorwasserstoff mehr entweicht, dann durch Wasserdampfdestillation vom Tetrachloräthan befreit, der Destillationsrückstand in Natronlauge unter Erwärmen gelöst und die geklärte Lösung angesäuert. Das ausgefallene 3-Benzhydryl-4-hydroxy-l-thiacumarin wird abfiltriert, nach Kristallisation aus Dioxan-Was- ser schmilzt es bei 235 bis 237 .
In analoger Weise erhält man das 3-(a-Äthyl-benzyl)-4-hydroxy-l-thia-cumarin, Fp. 145 bis 146 (aus Äthanol-Wasser). Beispiel 4 13,6 Teile a-Phenyl-propanol und 20 Teile 4-Hy- droxy-cumarin werden in 250 Volumteilen Tetrachloräthan bei 50 während einer Stunde mit trockenem Chlorwasserstoff behandelt, dann anschliessend bis zum Aufhören der Chlorwasserstoffentwicklung (etwa 11/, Stunden) auf 135 bis 140 im Ölbad erwärmt. Man kühlt ab, saugt das unveränderte 4-Hy- droxy-cumarin ab und wäscht mit wenig Tetrachlor- äthan nach.
Durch Eindampfen der vereinigten Lösungen im Vakuum zur Trockne und Kristallisieren aus Methylcyclohexan erhält man das 3-(a-Äthyl- benzyl)-4-hydroxy-cumarin vom Fp. 176 bis 178 . Beispiel 5 3,5 Teile a-(2-Hydroxy-3-methyl-5-chlor-phenyl)- n-propanol werden in 35 Volumteilen Tetrachloräthän gelöst, 3 Teile feingepulvertes Calciumchlorid zugegeben und bei 30 wird während 15 Minuten Chlorwasserstoff eingeleitet.
Dann filtriert man vom Calciumchlorid ab, wäscht dieses mit 15 Volumteilen Tetrachloräthan nach und versetzt das Filtrat mit 4,2 Teilen 4-Hydroxy-cumarin. Das erhaltene Gemisch wird 40 Minuten im Ölbad auf 130 bis 140 erhitzt. Nach dem Abkühlen saugt man ab, wäscht mit Ligroin und Alkohol und kristallisiert schliesslich aus Tetrachloräthan um, wobei 3-(a-Äthyl-2'- hydroxy-3'-methyl-5'-chlorbenzyl)-4-hydroxy-cumarin vom Fp. 223 bis 224 erhalten wird.
Beispiel 6 4,3 Teile a-(2-Methoxy-3-methyl-5-chlor-phenyl)- propanol werden in 15 Volumteilen Tetrachloräthan mit 4 Teilen feingepulvertem Calciumchlorid versetzt und 30 Minuten bei 60 bis 70 mit Chlorwasserstoff behandelt. Dann wird vom Calciumchlorid abgetrennt, 4,8 Teile 4-Hydroxy-cumarin zugegeben, eine Stunde auf 140 bis 150 erwärmt und eine Stunde am Rückfluss gekocht. Das Lösungsmittel wird mit Wasserdampf abdestilliert und der Destilla- tionsrückstand mit Natronlauge extrahiert.
Die beim Neutralisieren mit Salzsäure anfallenden Kristalle werden zweimal mit Wasser ausgekocht und aus Methanol oder Äthanol umkristallisiert. Das erhaltene 3-(a-Äthyl-2'-methoxy-3'-methyl-5'-chlor-ben- zyl)-4-hydroxy-cumarin schmilzt bei 178 bis 179 . In analoger Weise erhält man beispielsweise das 3-(4'-Methoxy-benzyl)-4-hydroxy-cumarin, Fp. 185 bis 186 (aus Methanol).
Beispiel 7 1,9 Teile 2-Methoxy-3-chlor -5-methyl-benzyl- alkohol und 2,4 Teile 4-Hydroxy-cumarin werden in 30 Volumteilen Tetrachloräthan aufgeschlämmt, mit einem Teil Phosphoroxychlorid versetzt und zunächst auf 100 , dann eine Stunde auf l30 und schliesslich noch eine halbe Stunde zum Sieden erhitzt. Anschliessend wird das Lösungsmittel mit Wasserdampf abdestilliert, der Rückstand in 2-n Natronlauge gelöst, die Lösung filtriert und angesäuert.
Man kristallisiert aus verdünntem Äthanol und Methanol und erhält das 3-(2'-Methoxy-3'-chlor-5'-methyl-benzyl)- 4-hydroxy-cumarin, Fp. 166,5 .
Beispiel 8 1,5 Teile 2-Hydroxy-3,5-dimethyl-benzylalkohol und 1,6 Teile 4-Hydroxy-cumarin werden in 2 Teilen Phosphoroxychlorid gelöst, wobei Erwärmung eintritt; man erwärmt hierauf noch 10 Minuten auf 50 und versetzt dann mit Wasser. Das ausgefallene Reaktionsprodukt wird aus Alkohol und dann aus Tetrachloräthan kristallisiert. Das 3-(2'-Hydroxy- 3',5'-dimethyl-benzyl)-4-hydroxy-cumarin schmilzt bei 252,5 bis 2530.
Beispiel 9 16 Teile 4-Hydroxy-cumarin und 20 Teile Benz- hydrylchlorid werden in 100 Volumteilen Tetrachloräthan etwa 15 Stunden unter Rückfluss gekocht. Nach dem Abkühlen saugt man das Reaktionsprodukt ab und kristallisiert aus verdünntem Methanol. Das erhaltene 3-Benzhydryl-4-hydroxy-cumarin schmilzt bei 183 bis 184 .
Dieselbe Verbindung kann man beispielsweise auch nach den in den Beispielen 6 oder 7 beschriebenen Verfahren herstellen. Beispiel 10 16 Teile 4-Hydroxy-cumarin und 13 Teile Benzyl- chlorid werden in 100 Volumteilen Tetrachloräthan 24 Stunden unter Rückfluss gekocht. Man destilliert das Lösungsmittel mit Wasserdampf ab, löst den Rückstand in verdünnter Natronlauge, filtriert und säuert an. Nach Kristallisation aus verdünntem Alkohol schmilzt das 3-Benzyl-4-hydroxy-cumarin bei 205 bis 207 .

Example 4 13.6 parts of a-phenylpropanol and 20 parts of 4-hydroxy-coumarin are treated in 250 parts by volume of tetrachloroethane at 50 for one hour with dry hydrogen chloride, then until the evolution of hydrogen chloride ceases (about 11 /, hours) heated to 135 to 140 in an oil bath. It is cooled, the unchanged 4-hydroxy-coumarin is filtered off with suction and washed with a little tetrachloroethane.
By evaporating the combined solutions to dryness in vacuo and crystallizing from methylcyclohexane, 3- (a-ethylbenzyl) -4-hydroxy-coumarin of melting point 176 to 178 is obtained. Example 5 3.5 parts of a- (2-hydroxy-3-methyl-5-chlorophenyl) -n-propanol are dissolved in 35 parts by volume of tetrachloroethane, 3 parts of finely powdered calcium chloride are added and hydrogen chloride is passed in for 15 minutes.
The calcium chloride is then filtered off, this is washed with 15 parts by volume of tetrachloroethane and the filtrate is treated with 4.2 parts of 4-hydroxycoumarin. The resulting mixture is heated to 130 to 140 minutes in an oil bath. After cooling, it is filtered off with suction, washed with ligroin and alcohol and finally recrystallized from tetrachloroethane, with 3- (a-ethyl-2'-hydroxy-3'-methyl-5'-chlorobenzyl) -4-hydroxy-coumarin having a melting point of 223-224 is obtained.
Example 6 4 parts of finely powdered calcium chloride are added to 4.3 parts of a- (2-methoxy-3-methyl-5-chlorophenyl) propanol in 15 parts by volume of tetrachloroethane and the mixture is treated with hydrogen chloride for 30 minutes at 60 to 70. The calcium chloride is then separated off, 4.8 parts of 4-hydroxycoumarin are added, the mixture is heated to 140 to 150 for one hour and refluxed for one hour. The solvent is distilled off with steam and the distillation residue is extracted with sodium hydroxide solution.
The crystals obtained when neutralizing with hydrochloric acid are boiled twice with water and recrystallized from methanol or ethanol. The 3- (a-ethyl-2'-methoxy-3'-methyl-5'-chlorobenzyl) -4-hydroxy-coumarin obtained melts at 178 to 179. In an analogous manner, for example, 3- (4'-methoxy-benzyl) -4-hydroxy-coumarin, melting point 185 to 186 (from methanol) is obtained.
Example 7 1.9 parts of 2-methoxy-3-chloro -5-methyl-benzyl alcohol and 2.4 parts of 4-hydroxy-coumarin are slurried in 30 parts by volume of tetrachloroethane, one part of phosphorus oxychloride is added and initially to 100, then one Heated to the boil for an hour at 1.30 and then for half an hour. The solvent is then distilled off with steam, the residue is dissolved in 2N sodium hydroxide solution, and the solution is filtered and acidified.
It is crystallized from dilute ethanol and methanol, and 3- (2'-methoxy-3'-chloro-5'-methyl-benzyl) -4-hydroxy-coumarin is obtained, melting point 166.5.
EXAMPLE 8 1.5 parts of 2-hydroxy-3,5-dimethyl-benzyl alcohol and 1.6 parts of 4-hydroxy-coumarin are dissolved in 2 parts of phosphorus oxychloride, with heating occurring; the mixture is then heated to 50 for 10 minutes and then treated with water. The precipitated reaction product is crystallized from alcohol and then from tetrachloroethane. The 3- (2'-hydroxy-3 ', 5'-dimethyl-benzyl) -4-hydroxy-coumarin melts at 252.5 to 2530.
EXAMPLE 9 16 parts of 4-hydroxycoumarin and 20 parts of benzhydryl chloride are refluxed for about 15 hours in 100 parts by volume of tetrachloroethane. After cooling, the reaction product is filtered off with suction and crystallized from dilute methanol. The 3-benzhydryl-4-hydroxy-coumarin obtained melts at 183 to 184.
The same compound can also be produced, for example, by the processes described in Examples 6 or 7. Example 10 16 parts of 4-hydroxycoumarin and 13 parts of benzyl chloride are refluxed in 100 parts by volume of tetrachloroethane for 24 hours. The solvent is distilled off with steam, the residue is dissolved in dilute sodium hydroxide solution, filtered and acidified. After crystallization from dilute alcohol, the 3-benzyl-4-hydroxy-coumarin melts at 205 to 207.
