Process for the preparation of compounds with vitamin C and F effects
It was shown a long time ago that it is possible in principle to convert ascorbic acid into oil- and fat-soluble compounds if, for example, its sodium salt is reacted with the acid chloride of a higher fatty acid. Although the compounds of the type mentioned that have become known up to now represent antiscorbutically effective bodies, they have not achieved any practical importance as medicaments. This is evidently because the need to have ascorbic acid available in fat-soluble form is quite low, unless other advantages or special properties are of particular interest at the same time.
In addition, the ascorbyl monoesters of saturated aliphatic monocarboxylic acids with 12 to 18 carbon atoms per molecule seem to have been tried as antioxidants without any notable successes of practical importance being known.
In contrast, it has now been found that compounds with vitamin C and F action of very outstanding biological interest can be prepared by converting l-ascorbic acid with a derivative of a vitamin F-active, at least diunsaturated fatty acid which provides the acid residue esterified. Salts thereof can also be used in place of ascorbic acid. The acid chlorides of the fatty acids mentioned are particularly suitable for esterification. This leads to compounds that show a double vitamin function in animal experiments, namely both the specific vitamin C effect and, at the same time, the likewise specific vitamin F effect. In addition to the double vitamin function of these bodies, a pronounced desensitizing, anti-inflammatory and bacteriostatic effect could be determined.
The biologically active isomers of linoleic acid, linolenic acid and arachidonic acid are primarily suitable as vitamin F-active, at least doubly unsaturated fatty acids.
Beispiel
example
2 parts of finely powdered Na ascorbate are suspended in 6 parts of dried pentane or benzene. With stirring or vibration, 3 parts of a mixture of linoleic and linolenic acid chloride are slowly added, and intensive mixing is continued for several hours with heating to about 80-90 ° C. Then several pieces of ice are added and one 100 cm is slowly added Mix well, destroy the emulsion formed by adding a little isopropyl alcohol and wait for the phases to separate. Now the pentane or benzene solution is separated off, washed several times with water and finally recovered as anhydrous as possible, then over sodium sulfate sicc . dried.
Finally, asbestos and aluminum oxide are added as filter aids, suction filtered and the solvent is completely removed by vacuum distillation. The remaining mixture of the monoesters of linoleic acid and linolenic acid with ascorbic acid presents itself as an easily mobile oil with a slightly yellowish color and a pleasant but somewhat bitter smell and taste.
