Verfahren zur Herstellung neuer N,N'-Malonyl-5,6-Dihydrobenzo(c)cinnoline Gegenstand der Erfindung ist die Herstellung von N,N'-Malonyl-5,6-Dihydro-benzo(c)cinnolinen der Formel

und ihrer tautomeren Form, wobei R einen mit äther sauerstoffhaltigem Radikal substituierten Alkylrest mit insgesamt höchstens 10 Kohlenstoffatomen be deutet.
Acylderivate der obigen Formel, bei welchen R den 3-Methoxybutyl-, den Tetrahydrofurfuryl- oder den Furfurylrest darstellt, werden besonders bevor zugt.
Die neuen Acyl- bzw. Malonylderivate des 5,6- Dihydro-benzo(c)cinnolins haben wertvolle pharma kologische Eigenschaften oder sind als Zwischenpro dukte zur Herstellung von Heilmitteln sehr geeignet. Viele Vertreter haben eine ausgeprägte analgetische und antiphlogistische Wirkung.
Bemerkenswert ist, dass diese Malonylderivate saure Eigenschaften besitzen und mit Basen wie Ka- liumhydroxyd, Lithiumhydroxyd, Bariumhydroxyd, Trimethylammoniumhydroxyd, Triäthylammonium- hydroxyd und a,cv-Bis-(trimethylammonium)-hexan- dihydroxyd Salze bilden, von denen insbesondere die Alkalisalze in der Regel ohne Basenüberschuss in Wasser klar löslich sind;
solche wässerige Lösungen sind nur schwach alkalisch und hervorragend geeig net zur intramusculären oder subcutanen Injektion. Viele der in Wasser gut löslichen Salze der neuen Benzo(c)cinnolinderivate sind gute Lösungsvermittler und können zur Herstellung verhältnismässig konzen trierter wässeriger Lösungen von in Wasser wenig löslichen Heilmitteln, wie z. B. Aminopyrin, verwen det werden. Die Salze und die freien Säuren können auch in fester Form, z. B. nach überführung in Ta bletten, Dragees oder in Gelatinekapseln, oral verab reicht werden.

Da die 5,6-Dihydro-benzo(c)cinnoline bekanntlich sehr leicht auch von schwachen Oxyda tionsmitteln wie Luft in die entsprechenden Benzo(c)- cinnoline übergeführt werden, ist es zweckmässig, die Umsetzung unter Ausschluss von Luftsauerstoff, z. B. in einer Stickstoff- oder Wasserstoffatmosphäre, durchzuführen.
Nach einer andern Ausführungsform des Verfah rens wird das 5,6-Dihydro-benzo(c)cinnolin oder ein N-Acyl- oder ein N,N'-Diacylderivat davon, wie N- Acetyl-5,6-dihydro-benzo(c)cinnolin, oder N,N'-Di- acetyl-5,6-dihydro-benzo(c)cinnolin, ebenfalls unter Ausschluss von Sauerstoff, mit einem monosubsti tuierten Malonester in Gegenwart von Alkalialkoho- lat bei höheren Temperaturen kondensiert unter Ab spaltung von Äthanol.
Wegen der Unbeständigkeit der 5,6-Dihydro- benzo(c)-cinnoline ist es vorteilhaft, diese frisch her zustellen und direkt, im gleichen Reaktionsgefäss, mit den Malonylierungsmitteln umzusetzen. Beispiels weise kann man Benzo(c)cinnolin in Gegenwart eines substituierten Malonesters und von mehr als 1 Mol Natriumalkoholat, in Äthanol gelöst, bei Raumtem peratur mit einem Katalysator wie Platin oder Pal ladium mit trockenem Wasserstoff zu 5,6-Dihydro- benzo(c)
cinnolin hydrieren und dann direkt, ohne vorherige Isolierung, durch Erhitzen zum substituier ten N,N'-Malonyl-5,6-dihydro-benzo(c)-cinnolin kon densieren. Es ist somit möglich, in einem einzigen Reaktionsgefäss auf einfache Art Benzo(c)-cinnoline direkt in substituierte N,N'-Malonyl-5,6-dihydro- benzo(c)cinnoline überzuführen.
Auch die Alkalisalze der 5,6-Dihydro-benzo(c)- cinnoline, welche bekanntlich aus den Benzo(c)cinno- linen und den entsprechenden Alkalimetallen bei Raumtemperatur durch Schütteln oder Verrühren in Gegenwart von geeigneten Lösungsmitteln wie Äther entstehen, sind zur Umsetzung mit den monosubsti tuierten Malonylrest abgebenden Verbindungen ver wendbar.
Für das Verfahren sind auch substituierte Malon- säuren als den substituierten Malonylrest abgebende Verbindungen verwendbar; so erhält man beispiels weise aus N-Acetyl-5,6-dihydro-benzo(c)cinnolin und substituierten Malonsäuren durch Erhitzen mit sau ren Kondensationsmitteln wie Phosphortrichlorid substituierte N,N'-Malonyl-5,6-dihydro-benzo(c)cin- noline.
Die Reaktanten werden vorzugsweise so gewählt, dass bei der Umsetzung die eingangs besonders her vorgehobenen Verbindungen entstehen. Die als Aus gangsstoffe wegen ihrer Beständigkeit gut geeigneten N-Acyl- und N,N'-Diacyl-5,6-dihydro-benzo(c)cinno- line können aus den entsprechenden 5,6-Dihydro- benzo(c)cinnolinen mit Acylierungsmitteln wie Acet- anhydrid, Keten, Benzoylchlorid oder Buttersäure chlorid erhalten werden.
Im übrigen sind die Aus gangsstoffe bekannt oder lassen sich nach an sich bekannten Methoden gewinnen.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Tem peraturen sind in Celsiusgraden angegeben.
<I>Beispiel 1</I> 0,92 Gewichtsteile Natrium werden in absolutem Äthanol gelöst. Darauf gibt man 5 Gewichtsteile 3-Methoxybutyl-malonsäure-äthylester und 4,5 Ge wichtsteile N-Acetyl-5,6-dihydro-benzo(c)cinnolin zu und kristallisiert den Alkohol aus der gelben Lösung in einer trockenen Stickstoffatmosphäre langsam ab. Der kristalline Rückstand wird während 2 Stunden bei einem Vakuum von 10-15 mm Hg auf 130 bis 135 erhitzt und nach dem Erkalten in 100 Volum- teilen Wasser aufgenommen.
Die klare, braune, wäs serige Lösung wird viermal mit 50 Volumteilen Chloroform ausgeschüttelt, und alle Chloroform schichten werden mit den gleichen 100 Volumteilen 2-n Sodalösung gewaschen.
Die vereinigten wässerigen Schichten werden mit Essigsäure angesäuert, wobei eine kristalline, hell gelbe Fällung in einer Ausbeute von 5,6 Gewichts teilen entsteht. Zur Reinigung kann man aus Ätha- nol, Essigester-Benzin 1: 1 oder aus Benzol-Benzin 1 : 2 umkristallisieren, wobei man das N,N'-(3-Meth- oxybutyl-malonyl)-5,6-dihydro-benzo(c)cinnolin der Formel auf Seite 1, wobei

in Form von blassgelben Nadeln vom Schmelzpunkt 105 erhält.
Der in diesem Beispiel als Zwischenprodukt ver wendete 3-Methoxybutyl-malonsäure-äthylester kann auf folgende Art erhalten werden: 104 Gewichtsteile 3-Methoxybutanol (im Handel erhältlich) werden mit 450 Volumteilen Benzol ver rührt und bei 20-30 gleichzeitig innert anderthalb Stunden mit 180 Volumteilen Benzolsulfochlorid und 72 Gewichtsteilen pulverisiertem Natriumhydroxyd versetzt.
Nach weiterem Rühren während einigen Stunden wird mit 450 Volumteilen Wasser in einem Scheidetrichter gut vermischt, die wässerige Schicht abgelassen und in einem zweiten Scheidetrichter mit 100 Volumteilen Benzol gewaschen. Die vereinigten Benzolschichten werden mit 50 Volumteilen 34pro- zentigem Ammoniak während einigen Stunden ge schüttelt, darauf noch während 10 Minuten mit 100 Volumteilen 2-n Natronlauge.
Die Benzolschicht wird von der wässerigen Schicht getrennt und mit 100 Vo- lumteilen 2-n Natronlauge und mit 100 Volumteilen Wasser gewaschen. Aus der Benzollösung erhält man nach dem Eindampfen und Trocknen im Vakuum 201 Gewichtsteile Benzolsulfonsäure-3-methoxy-bu- tylester in Form eines hellgelben Öls.
171 Gewichts teile davon werden innerhalb 3 Stunden unter Rüh ren in eine unter Rückfluss kochende Lösung aus 350 Volumteilen absolutem Äthanol, 16,1 Gewichts teilen Natrium und 224 Gewichtsteilen Malonester laufen gelassen. Bevor aller Benzolsulfonsäureester zugegeben ist, beginnt die Ausscheidung von Na trium-benzolsulfonat. Nachdem man noch einige Stunden am Rückfluss gekocht hat, wird die Haupt menge Äthanol unter vermindertem Druck verdampft und mit 250 Volumteilen Wasser versetzt. Es wird nun mit Äther extrahiert, die Ätherschichten ergeben nach dem Waschen mit Wasser, Trocknen über Na triumsulfat und Eindampfen einen öligen Rückstand.
Unter Verwendung einer Fraktionierkolonne erhält man beim Destillieren im Vakuum nach dem Abtren nen des überschüssigen Malonesters eine einheitliche, farblose Fraktion vom Kp 12 13 8-l39 in einer Aus beute von 134 Gewichtsteilen, welche aus reinem 3-Methoxybutylmalonsäure-äthylester besteht.
<I>Beispiel 2</I> In eine Lösung von 0,92 Gewichtsteilen Natrium in 75 Volumteilen absolutem Äthanol werden 5,1 Gewichtsteile Tetrahydrofurfuryl - malonsäureäthyl- ester mit 4,5 Gewichtsteilen N-Acetyl-5,6-dihydro- benzo(c)cinnolin gegeben. In einer Atmosphäre von absolutem Stickstoff wird durch Erhitzen in einem Ölbad das Äthanol innerhalb 3 Stunden abdestilliert. Der kristalline, grüngelbe Rückstand wird darauf während 1 Stunde in einem Vakuum von 10 bis 15 mm Hg auf 135-140 erhitzt.
Nach dem Abkühlen löst man in 80 Volumteilen Wasser und schüttelt mit 50 Volumteilen, dann zwei mal mit 25 Volumteilen Chloroform aus. Alle Chloro- formschichten werden mit 30 Volumteilen 1-n Na tronlauge gewaschen. Die vereinigten alkalischen, wässerigen Schichten werden mit 1,2 Gewichtsteilen Aktivkohle geschüttelt und dann unter Zusatz von Diatomeenerde filtriert. Aus dem Filtrat erhält man beim Ansäuern mit Essigsäure unter Aussenkühlung mit Eiswasser eine ölige, hellgelbe Ausscheidung.
Diese wird in Chloroform aufgenommen, mit Was ser gewaschen und durch Eindampfen des Chloro forms isoliert. Zur Trocknung wird der Rückstand zweimal mit wenig Benzol versetzt und im Vakuum eingedampft. Beim Stehenlassen kristallisiert der Rückstand spontan. Beim Umkristallisieren aus 40 Volumteilen Äthanol erhält man 5,15 Gewichtsteile blassgelbe Kristalle vom Schmelzpunkt 115 , welche auf Grund der C-, H- und N-Bestimmung aus rei nem N,N'-(Tetrahydrofurfuryl-malonyl)-5,6-dihydro- benzo(c)cinnolin der Formel auf Seite 1 bestehen, worin R den Rest

Eine äusserst geringe Menge einer bräunlichen Verunreinigung kann entfernt werden, wenn man eine benzolische Lösung der neuen Substanz durch eine Säule aus Aluminiumoxyd (nach Brockmann) filtriert.
Die neue Verbindung löst sich mit der äquiva lenten- Menge Diäthylamino-äthanol leicht in Was ser mit zitronengelber Farbe. Solche wässerige Lö sungen sind nur sehr schwach alkalisch (pH < 8).
Wenn man bei der oben beschriebenen Kon densation anstelle von 4,5 Gewichtsteilen N-Acetyl- 5,6 - dihydro - benzo(c)cinnolin 5,3 Gewichtsteile N,N'-Diacetyl-5,6-dihydro-benzo(c)cinnolin verwen det, so erhält man 4,85 Gewichtsteile aus Athanol umkristallisiertes N,N'-(Tetrahydrofurfuryl-malonyl)- 5,6 - dihydro - benzo(c)cinnolin vom Schmelzpunkt 115 .
N,N'-Diacetyl-5,6-dihydro-benzo(c)cinnolin kann auf folgende Art hergestellt werden: 40 Gewichtsteile 2,2'-Dinitro-diphenyl, 950 Vo- lumteile Äthanol, 30 Volumteile Wasser, 12 Ge wichtsteile Kaliumhydroxyd und 0,2 Gewichtsteile Platinoxyd werden in einem Rührautoklaven bei etwa 50 Atmosphären Wasserstoffdruck hydriert. Die Reaktion verläuft exotherm, und die Tempera tur wird durch Aussenkühlung auf 20-30 gehal ten. Nach ungefähr einer Stunde ist die Wasserstoff aufnahme beendet.
Man rührt noch eine bis zwei Stunden bei etwa 30 Atmosphären Wasserstoffdruck, lässt den überdruck ab und oxydiert das in guter Ausbeute entstandene 5,6-Dihydro-benzo(c)cinnolin durch Durchleiten von Luft in der Wärme zu Benzo- (c)cinnolin. Das Äthanol wird darauf verdampft und der Rückstand mit 500 Volumteilen 2-n Salz säure behandelt.
Das als Hydrochlorid in Lösung gegangene Benzo(c)cinnolin wird nach Abfiltrieren von geringen Mengen Verunreinigungen mit Ammo niak in kristalliner Form gefällt und genutscht. Nach Lösen in 300 Volumteilen siedendem Methanol, Zu gabe von 2 Gewichtsteilen Aktivkohle und Filtrieren der heissen Lösung kristallisieren mindestens 20 Ge wichtsteile Benzo(c)cinnolin in reiner Form bei Ab kühlen aus.
1,8 Gewichtsteile davon werden in 10 Volum- teilen Eisessig und 4 Volumteilen Acetanhydrid mit 0,005 Gewichtsteilen Platinoxyd bei<B>20 </B> bei Atmo sphärendruck hydriert. Die Wasserstoffaufnahme kommt nach etwa 3/4 Stunden zum Stillstand. Man lässt über Nacht stehen, erhitzt eine halbe Stunde auf 100 und versetzt nach dem Abkühlen mit 100 Volumteilen Wasser.
Durch Extraktion mit Chloro form erhält man 2,34 Gewichtsteile eines öligen Rückstandes, welcher, mit 25 Volumteilen Methanol versetzt und bei -17" stehengelassen, 0,42 Ge wichtsteile kristallines N,N'-Diacetyl-5,6-dihydro- benzo(c)cinnolin ausscheidet.
Die Mutterlauge ent hält viel Monoacetylverbindung, welche nach dem Trocknen beim Erhitzen mit 6 Volumteilen Acet- anhydrid während 4 Stunden auf 1000 in die Diace- tylverbindung übergeführt wird, welche durch Um kristallisieren aus Methanol in Form von farblosen Prismen vom Schmelzpunkt 169-170 erhalten wird.
<I>Beispiel 3</I> 4,8 Gewichtsteile Furfuryl-malonsäure-diäthyl- ester und 4,5 Gewichtsteile N-Acetyl-5,6-dihydro- benzo(c)cinnolin werden in einer Lösung von 0,92 Gewichtsteilen Natrium in 75 Volumteilen Äthanol kondensiert und aufgearbeitet, wie in Beispiel 2 be schrieben. Beim Ansäuern der vereinigten alkalischen Schichten mit Essigsäure entsteht eine kristalline Fällung, welche nach dem Abfiltrieren und Aus waschen mit Wasser beim Umkristallisieren aus Äthanol 4,2 Gewichtsteile hellgelbe Kristalle vom Schmelzpunkt 1470 liefern.
Die neue Substanz kann auch aus Essigester-Benzin umkristallisiert werden und entspricht in der Konstitution dem N-N'-(Fur- furyl-malonyl)-5,6-dihydro-benzo(c)cinnolin der For mel auf Seite 1, worin R den Rest

bedeutet. <I>Beispiel 4</I> 3,60 Gewichtsteile Benzo(c)cinnolin, 4,90 Ge wichtsteile Tetrahydrofurfuryl - malonsäurediäthyl- ester, 0,018 Gewichtsteile Platinoxyd und eine Lö sung von 0,92 Gewichtsteilen Natriummetall in 75 Volumteilen Äthanol werden in ein Druckgefäss mit Rührvorrichtung gebracht.
Durch Evakuierung wird die Luft entfernt, dann sofort unter 17 Atmosphären Wasserstoffdruck (wasserfrei) gesetzt. Unter gutem Rühren ist die Wasserstoffaufnahme bei Raumtem peratur zum 5,6-Dihydro-benzo(c)cinnolin innert 1,5 Stunden beendet. Der Wasserstoffdruck wird auf 3 Atmosphären reduziert, ohne Rühren 2 Stunden lang bei 130-140 , dann noch 2 Stunden bei 150 bis 160 erhitzt. Man lässt über Nacht erkalten.
Die hellgelbe alkoholische Lösung wird unter partiellem Vakuum zur Trockne eingedampft, der kristalline, hellgelbe Rückstand in 100 Volumteilen Wasser aufgenommen und wie im Beispiel 2 beschrie ben aufgearbeitet. Man erhält so 3,65 Gewichtsteile reines N,N'- (Tetrahydrofurfuryl - malonyl)-5,6 - dihydro - benzo(c)- cinnolin vom Schmelzpunkt 115 , welches mit der in Beispiel 2 beschriebenen Verbindung identisch ist.

The reactants are preferably chosen so that the compounds particularly emphasized at the beginning are formed during the reaction. The N-acyl- and N, N'-diacyl-5,6-dihydro-benzo (c) cinnolines, which are well suited as starting materials because of their stability, can be obtained from the corresponding 5,6-dihydro-benzo (c) cinnolines with Acylating agents such as acetic anhydride, ketene, benzoyl chloride or butyric acid chloride can be obtained.

<I> Example 1 </I> 0.92 parts by weight of sodium are dissolved in absolute ethanol. Then 5 parts by weight of 3-methoxybutyl malonic acid ethyl ester and 4.5 parts by weight of N-acetyl-5,6-dihydro-benzo (c) cinnoline are added and the alcohol is slowly crystallized from the yellow solution in a dry nitrogen atmosphere. The crystalline residue is heated to 130 to 135 under a vacuum of 10-15 mm Hg for 2 hours and, after cooling, is taken up in 100 parts by volume of water.
The clear, brown, aqueous solution is shaken out four times with 50 parts by volume of chloroform, and all chloroform layers are washed with the same 100 parts by volume of 2N soda solution.
The combined aqueous layers are acidified with acetic acid, a crystalline, light yellow precipitate being formed in a yield of 5.6 parts by weight.
