Verfahren zur Herstellung von Antibiotika Die Erfindung bezieht sich auf ein Ver fahren zur Herstellung von wertvollen anti biotischen Stoffen, nämlich von Oxytetraeyclin und Vengicid durch aerobe Kultivierung eines .#ees-Stamm,es, das dadurch gekenn- -'treptom, zeichnet ist,
dass Streptomyces vendargensis nov. spec. verwendet und aus dem Gärungs medium das Oxytetraeyclin und das Vengicid isoliert werden.
Es wurde nämlich gefunden, dass eine neue, bisher unbeschriebene Art von Mikro- organismen, denen die Namen Streptom.yces vendargensis gegeben wurden, mehrere Anti biotika produziert, von denen das eine dem bekannten Oxytetraeyelin entspricht, wäh rend ein weiteres; für welches die Bezeich nung Vengicid vorgeschlagen wird, bisher nicht bekannt war.
Dieser neue Organismus Streptomy ces vendargensis ist gemäss Bergey5 Manual of Determinative Baeteriology (1918) als ein Actinomyeet klassifiziert worden.

Es ist bekannt, dass dieser 1VTikroorganis- mus auch ein Fungizid' abscheidet [Antibiotics @: Chemotherapy 1 (1951), 289-290].
Im Zuge einer ausgedehnten Forschung auf dem Gebiet der Antibiotika wurde nun ein neuer Actinomycet gezüehtet aus einer Erde aus Südfrankreich, derssen morpholo gische und biochemische Eigenschaften erheb lich von Streptomy ces rimosus abweichen. Die ser neue Actinomy cet wird Streptomyces ven- d.argensis bezeichnet.
Die Tabelle 1 zeigt die Eigenschaften des Streptomyces vendargensis, gezogen auf ver schiedenen Kulturböden, mit denjenigen des Streptomyces rimosus verglichen. In dieser Tabelle wird das vegetative iHycel die Kolonie genannt, während das sporogene Mycel mit Aeromycel bezeichnet. wird.

Die Tabelle zeigt, dass die Kolonien des Sti,eptoniyees rimostis auf den meisten Nä.hr- niechen dunkler sind als die des Streptomy ces vericlai-geiisis.
Ein kennzeichnender Unterschied ist der (iei-iteli. Während Streptomyee;s rimostis einen iiiehr oder weniger starken Geruch auf den mei sten Nährmedien hat, verbreitet Streptoinyces vcndargensis keinen oder fast keinen Geruch.
Wenn man die Aeromycele der beiden lritiiioni@=ceten vergleicht, erscheint das von Streptom.#-ces rimosus fast immer in geschlos senen Spiralen, während das von Streptomy ces vendargensis keine Spiralen auf irgendeinem Nährmedium bildet. Streptomyees venclargensis bildet Kolonien von dein sogenannten tuft tvpe (cf. S.
A. @V aksman, The Actinomycetes 1950).
V engieid weicht, sowohl in seinen chemi- sehen als ;seinen bioeheniischen Eigenschaften von dem vom Streptomyces rimosu;s gebildeten fun#,-iziden Stoff ab. Vengieid ist ein weisser, kristalliner Stoff mit amplioterem Charakter.
Seine Wasserlöslielikeit bei Zimmertempera tur ist. gering, und zwar ungefähr 0,4 mg/em3. Es ist in geringem Masse löslich in organiselien Lästtn\;smitteln, wie Aceton, Methanol, Buta- riol und Amylalkohol, fast unlöslich in Di- ii.tliyläther. Das plj der gesättigten Lösung in Wasser ist ungefähr 6,3.
Der Schmelzpunkt ist 241,5-243 C und- die spezifische Drehung des reinen Stoffes ist [a] D = 51,6 in 0,1n- Salzsäure. Das Molekulargewicht des Stoffes ist ungefähr 600.
Die vermutliche empirische Formel isst C"4IL"O,Nlo. Die chemische Ana- lvse, des reinen Stoffes ergab: C 47,05 0/0, 1t 4,85 Oh, N 23,25 oh und 0 24,85 % (berech- riet). In dem I?ltraviolettabsorptions,apektrtim @vtirden Maxima bei 233,

Mit einer Suspension in Nujol (1la.rkenproduh-t, Paraffinöl) bzw. in Hexa- ehlorbutadienwurden die naehfolgenclen kenn- zeiehnenden Llbsorptioiisbande im infraroten Absorptionsspektimm gemessen (in reziproken Zentimetern) :

Die biologisch aktiven Stoffe können von dem Kulturfiltrat. des Streptomyces vendar- gensis mittels Papierchromatographie getrennt werden. Wenn das Chromato-graphieren bei 26 C auf Eaton-Dikeman-Papier Nr.
H 613 mit Hilfe eines Elutionsmittels der folgenden Zusammensetzung ausgeführt wurde: 600 cm3 wassergesättigtes Butanol, 12 g para-Toluols l- fonsäur e und 4,8 eins Piperidin, ist der RF-Wert des Vengicids 0,45,
während unter den gleichen Bedingungen der fungicide Stoff aus Strepto- myces rimosus einen RF-Wert von 0,70 hat.
Nach der papierchromat.ographischen Tren nung bei einer Temperatur von 20 C auf demselben Papier, aber mit einem andern Ehitionsmittel, wurden RF-Werte von 0,45 bzw. 0,31 gefunden. In diesem Fall war das Elu- tionsmittel die oberste Schicht eines Gemisches von 4 Teilen Butanol, einem Teil Äthanol und 5 Teilen Wasser.
Durch diese Papierchromatographie wurde auch gefunden, dass Streptomyces vendargen- sis eine Anzahl anderer Antibiotika produ ziert, und. zwar einen Komplex von zwei oder drei Stoffen mit einem RF-Wert von 0,04 bis 0,08 (26 C, Lösungsmittel: wassergesättigtes Butanol, para-Toluolsulfonsäure und Piperi- din), die z. B. sehr aktiv gegen Bacillus sub- tilis (Stamm Marburg) sind.
Des weiteren zeigte es sich, dass das Kulturfiltrat des Strept.omyces vendargensis zwei Antibiotika enthält, die nur geringe Stabilität in einem sauren Medium zeigen und die einen RF-Wert von 0,05--0,13 haben (bei 20 C, Lösungsmit tel: oberste Schicht aus Butanol, Äthanol und Wasser in dem Verhältnis 4:1:5): diese Antibiotika haben einen verzögernden Effekt auf das Wachstum von u. a. Saecharomyces. Diese Stoffe werden in den Kulturfiltraten des Streptomy ces rimosus nicht gefunden.

Streptomyces vendargensis wird vorzugs weise als Tauchkultur in einem flüssigen Nähr medium unter sterilen Verhältnissen in ge schlossenen Gefässen, die mit Rührorganen versehen sind, in die zwecks Belüftung steri ler Sauerstoff oder Luft während der Züch tung zugeführt werden kann, gezüchtet.
Die Zeit., während der .die Züchtung durch geführt wird, die Temperatur und die übrigen Bedingungen, die zur Erhaltung guter Aus beuten an Antibiotika zu beachten sind, wer den im nachstehenden näher beschrieben.
Das Nährmedüun sollte einen Kohlenstoff lieferanten und einen organischen oder an- organischen Stickstofflieferanten enthalten; die Anwesenheit von Mineralsalzen, wie Phos phaten, Kalium- und Natriumsalzen, und Spu ren verschiedener Metalle ist erwünscht. Öfters aber sind die Ausgangsstoffe, die in der Praxis gebraucht werden, schon genügend mit diesen Mineralsalzen verunreinigt, so, dass es sich erübrigt, sie speziell hinzuzufügen.
Als Kohlenstofflieferanten können sowohl lösliche als unlösliche Kohlenhvdrate, wie Glukose, Saceha.rose, Laktose oder Stärke, ge braucht werden. Zuckeralkohole, wie Glycerin, sind ebenfalls geeignet..
Die Menge des Kohlen stofflieferanten in dem Medium kann sehr verschieden sein und ist abhängig von der Art des verwendeten Kohlehydrats und der sonstigen Zusammensetzung des Mediums; ge- wöhnlieh beträgt sie ungefähr zwischen 0,5 bis 5 1/a, des Gewichtes des Mediums.
AlsStickstofflieferantenkönnen eine grosse Anzahl Stoffe verwendet werden. Erwähnt seien hydrolysiertes oder niehthy.drolysierte:s Kasein, Maisquellfliissiglzeit (corn steep liquor), Pepton, Fleischextrakt, Fisehextralrt, Fisch mehl, Sojabohnenmehl, Erdnussmehl, Nitrate.
Es hängt von der Ergiebigkeit und dersonHstigen Zusammensetzung des Mittels und öfters auch von variierenden ökonomischen Faktoren ab, welcher Stickstofflieferant. im Einzelfalle am zweckmässigsten ist.
Kleine Mengen sticlLstoffhaltiger Grund stoffe, wie Hefeextrakt, distiller's solubles usw., können eine bedeutende Steigerung der Ausbeute herbeiführen.
Dde Fermentat.ionsdauer hängt von der Zusammensetzung des Nährmediums ab. G e- wöhnlieh schwankt sie zwischen 72 und 96 Stunden; die Fermentation kann erwünschten- fall -i aueli längere Zeit fortgesetzt werden, wenn die gesteigerte Ausbeute an Antibiotika die höheren Kosten einer längeren Fermen tation rechtfertigt.
Die Temperatur .der Fermentation kann zwisehen 20 und 3J C liegen. Bevorzugte Tem peraturen liegen zwisehen 26=?8 C.
Für ein optimales Wachstum des Organis mus und eine optimale Ausbeute an Anti biotika sollte das PH während der Fermenta tion innerhalb ziemlich enger Grenzen gehal ten werden, z. B. zwischen 5,0 und 8,0. Nach der Sterilisation wird das Nährmedium am besten auf ein pi; zwischen 6 und 7 einge stellt. Die Fermentation wird vorzugsweise bei einem pH zwischen. 6,5 und 7,3 ausgeführt, da die Erfahrung gezeigt hat, dass die höchste Ausbeute bei :diesem PH erzielt wird.
Das pii kann während der Fermentation konstant gehalten werden, indem man Alkalihydroxyd unter sterilen Bedingungen in regelmässigen Zeitabständen hinzufügt, gewöhnlich aber wird festes Calciumearbonat als eine Art Puffer in Mengen von 0,2-1,0 Cewielitspro- zent des Nährmediums gebraucht.
Die optimale Menge der dem Nährmedium w *ilii,encl der Fermentation zuzuführenden Luft hängt von der Form des Gefässes, der (,'eseliwindi;ii:eit und der Form der Rühr- organe ab. Gewöhnlieh liegt diese Menge zwi- sehen 0,5 und 4 Litern pro Liter Nährmedium pro Minute.
Als Impfstoff wird vormlrsweise eine 48 bis 72 Stunden alte Vorkultur des Strepto- myees vendargensi.s verwendet.
Um gute Ausbeuten zu erhalten und Schwankungen der Resultate vorzubeugen, wird die Impfung vorzugsweise mit 3-6 Volumprozent Kultur, bezogen auf das Nä.hr- medium, durehgefübrt. Bei Verwendung gro sser Fermentationsgefässe kann man auch eine grössere Menge der Kultur, z.
B. 1011/o, in dem Hauptfermenta.tionsgefäss für die Einleitung einer neuen Kultur aufbewahren, anstatt die genannte Vorkultur als Impfstoff zu verwen den. Der Fermentationsprozess kann auch konti nuierlich durchgeführt werden.
h'ür die Isolierung der Antibiotika aus dem Kulturmedium können verschiedene üb- liehe Methoden aufgewandt werden. Vorzugs weise wird die Kulturflüssigkeit, wenn die Fermentation beendet. ist, und bevor man mit der eigentlichen Isolierung beginnt, auf ein pH von 2,0-2,5, vorzugsweise auf ein ptI = 2,2, angesäuert.
Daraufhin wird die Flüssigkeit am besten filtriert oder zentrifugiert zur Ent, ferni.ng des ibIycels. Wenn die Flüssigkeit nicht angesäuert wird, gehen 20-250/9 des Oxytetraeyclins verloren.
Nach Entfernung des Mycels und aller andern festen Bestandteile aus der Fermen tationsflüssigkeit können die Antibiotika an aktivierte Kohle bei einem pH von ungefähr 7 adsorbiert und dann mit Hilfe eines Alkohols, der mit. Wasser teilweise mischbar ist, wie Butanol, bei einem PH von ungefähr 1,5 eluiert werden. Die Edution wird vo.r7iigs- weise ausgeführt. mit einem Gemisch von Butanol, Wasser und Phenol.
Nach Konzen tration des Eluats durch Verdampfung der Lösungsmittel. im Vakuum wird die Flüssig keit zweckmässig mit einem Alkohol, der mit Wasser nicht misehbar oder teilweise misch bar ist, wie Butanol oder Amylalkohol, bei einem pH von ungefähr 9, extrahiert. Von der auf diese Weise erhaltenen Lösung der Anti biotika in z. B. Butanol kann durch Extrak tion mit Wasser bei einem pH von ungefähr 2 eine wässrige Lösung der Antibiotika er halten werden.
Aus dieser Lösung kann das Oxytetra- eyelin, gewünschtenfalls nach Konzentration, in bekannter Weise (vergleiche belgische Pa tentschrift Nr.506950) mit Hilfe von Ma- gnesiumehlorid und einem quaternären Am monsalz, wie 1-lexadeeyltrimethylammoniiim- chlorid, -bei einem pH von ungefähr 9, gefällt werden.
Das Präzipitat kann leicht zu reinem Oxy- tetraey clin oder dessen salzsaurem Salz auf gearbeitet werden. Obgleich Vengicid, eben- sowie Oxytetracyclin, ein Stoff mit einem amphoter en Charakter ist, wird es durch eine quaternäre Ammoniumbase nicht gefällt.
Das Vengicid' kann aus dem Filtrat des Präzipitats des Oxytet.racyclins, z. B. durch Extrahieren des Filtrats mit Butanol bei einem PH von ungefähr 9, erhalten werden; die auf diese Weise erhaltene butanolische Lösung des Vengicids kann wiederum mit Wasser bei einem pH von ungefähr \? extrahiert werden. Vengicid wird gegebenenfalls nach Konzen trierung des Extraktes in kristallinischer Form durch Einstellen der wässrigen Lösung auf ein pH zwischen 4,0 und 5,0 erhalten.
Das mycelfreie Kulturfiltrat kann auch gegebenenfalls nach Konzentration durch Ein engung - direkt mit einem Alkohol der Buta- nol- und Amylalkoholgruppe bei einem PH von ungefähr 9 extrahiert werden. Die auf diese Weise erhaltene Lösung der Antibiotika. kann dann mit Wasser bei einem pH von un gefähr 2 nochmals extrahiert werden.
Gemäss einem bevorzugten Isolierungsver- fahren wird ein Ionenaustauseher mit schwach saurem Charakter, insbesondere Duolite S 30 (Markenprodukt, ein synthetisches Harz auf Phenolbasis) verwendet..
Es war nicht zu erwarten, dass ein amplio- terer Stoff, wie Oxytetraey clin, .an einen sol chen schwach sauren Ionenaustauscher adsor- biert werden könnte, da normalerweise bei Verwendung von Ionenaustauschern für die Adsorption von amphoteren Stoffen solche mit entweder stark sauren oder stark bas i- sehen Eigenschaften für notwendig gehalten werden.
Für -die Isolierung der Antibiotika mit. Hilfe von Duolite S 30 wird am besten wie folgt vorgegangen: Dass vom Mycel befreite Kulturfiltrat wird bei einem pH von ungefähr 2,5 durch eine mit dem erwähnten Austau- scher gefüllte Säule geführt. Durch Einstel lung der Höhe der Säule und der Menge des durchzuführenden Kulturfiltrats. kann die Adsorption so geleitet.
werden, dass praktisch nur das Oxytetraeyelin adsorbiert wird, wäh rend Glas Vengicid gelöst bleibt. Letzteres kann dann entweder durch Adsorption an aktiver Kohle oder durch direktes Extrahieren,<B>7.B.</B> mit Butanol, erhalten werden. Das adsorbierte Oxytetracy elin kann mit Alkalihydroxyd elu- iert und dann in bekannter Weise zu reinem Oxytetraeyelin aufgearbeitet werden.

<I>Beispiel 6</I> 10 Liter der nach Beispiel 3 erhaltenen Flüssigkeit werden mit konzentrierter Salz säure unter Rühren auf ein pH = 2,2 einge stellt. Nachdem man eine halbe Stunde ge rührt hat, wird sie mit einer Sharpless-Zentri- fuge zentrifugiert. In dieser Weise wird eine opalisierende Flüssigkeit erhalten, die, nach dem sie einige Zeit gestanden hat, durch einen oder mehrere mit Filtrapid (Markenpro dukt, ein Filtrierhilfsmittel) versehenen Fil ter filtriert wird, bis sie klar ist.
Das klare Filtrat wird mit Hilfe von Na triumhyd.roxy,d auf ein pH = 7,3 eingestellt und während 15 Minuten mit 100 g aktivierter Kohle gerührt. Das Adsorbat wird daraufhin durch Filtrieren abgetrennt, mit Wasser ge- waschen und an der Luft getrocknet.
Die lufttrockene Kohle, an.die die Antibiotika a-dsorbiert sind, wird darauf fünfmal mit 1 Liter wassergesättigten Butanols, das 10 g Phenol enthält., bei einem pH =1,5 eluiert. Nach jeder Elution wird .die aktive Kohle wieder in dem Eluiermittel suspendiert. und mit Salzsäure wieder auf das gewünschte pl, gebracht.
Das Eluat wird dann mit N atriumhydroxyd auf ein pH = 2,5 eingestellt und in einem Rüekflussverdampfer im Vakuum (bei unge fähr 35 C) auf ein Volumen von ungefähr 1 Liter konzentriert..
Mikrobiologische Gehaltsbestimmungen zei gen, .dass die Flüssigkeit 3,8 g Oxytetraeyelin (80% der ursprünglichen Menge) und 2,56 g Vengicid (95% der ursprünglichen Menge) enthält.
Das konzentrierte Ehtat wird mit. Natrium hydroxyd auf ein pH = 9,5 eingestellt und viermal mit 0,75 Litern Butanol extrahiert. Der butanolisehe Extrakt wird mit Salzsäure auf ein pl, = 2,0 eingestellt und fünfmal mit 0,75 Litern Wasser extrahiert.
Die auf diese Weise erhaltene wässrige Lö sung enthält 2,85 g 0xytetracyelin und 2,31 g Vengicid. Die Lösung wird mit Natrium hydroxyd auf ein PH = 2,5 eingestellt und in einem Rückflussverdampfer auf iuzgefähr 1400 c m3 konzentriert, d. h.
auf eine Konzen tration von ungefähr 2 mg Oxytetracyelin pro ems. Dieser konzentrierten Lösung werden der Reihe nach 112 em3 einer 0,1 molaren Lösung von Magnesiumehlorid, 5,
6 ein-' einer 50%igen Lösung von Zitronensäure und 14 g einer 33%igen Lösung von I-Iexadecyltrime- thylammoniuinehlori:d' zugefügt. Daraufhin wird die Lösung mit Natriumhydroxyd auf ein pl, = 9,0 eingestellt.
Die flockige Fällung, die ungefähr 98% des in .der Flüssigkeit enthaltenen Oxyt.etra- cyclins enthält, wird filtriert und im Vakuum bei 40-15 C getrocknet.
Diese Fällung wird nun fünfmal mit, un gefähr 13 cm3 Methanol extrahiert.. Diesem methanolisehen Extrakt werden 3,5 em3 kon zentrierte Salzsäure (spez. Gewicht 1,1.9) zu- gefügt; das Chlorhydrat von Oxytetraeyclin kristallisiert dann spontan aus. Ausbeute: 2,2 g.
Das Filtrat der Fällung des Oxytetra- eyelins mit der quaternären Ammoniumbase wird fünfmal mit. 0,75 Litern Butanol bei einem p1, = 9,5 extraliiert.. Der hutanolisehe Extrakt wird mit Salzsäure auf ein pH <I>= 2</I> eingestellt und dreimal mit. 1 Liter Wasser extrahiert.
Die wässrige Lösung wird mit Na- triLUnhydroxy d auf ein p1, = 2,5 eingestellt und im einem Rüeldlussdampfer im Vakuum konzentriert auf-ungefähr 1.00 em3. Es wird dann portionsweise mit Natriumhy droxy d auf ein pl, = 4,5 eingestellt.
Das Vengieid kristallisiert als gelbe Kri stallmasse aus. Ausbeute 1,9 g.
Durch Lösen in verdünnter Salzsäure, Ent färben mit aktiver Kohle und wiederholtem Fällen bei PH = 4,5 kann der Stoff gereinigt werden. Die weissen Kristalle schmelzen bei 241,5-243 C. Diese Kristalle sind monoklin. Die Abmessungen der Kristallelemente sind laut Messungen mit. Rönt-en,stralilen: a=6,6A; Ti=19,1A; c=5,2A;
ss=7.04 . Beispiel <I>i</I> 10 Liter der nach Beispiel 3 erhaltenen Flüssigkeit, deren Fermentation vollendet ist, werden mit konzentrierter Salzsäure unter Rühren auf ein p1, = 2,2 eingestellt. Nachdem man eine halbe Stunde gerührt hat, wird die Flüssigkeit mit. Hilfe einer @hariploss-Zentri- fuge zentrifugiert. Die opalisierende Flüssig keit wird mit einem Filtrierhilfsmittel einmal oder mehrere Male filtriert.
Es wird schliess lieh ein klares Filtrat erhalten.
Nachdem es auf pl, ='-),5 eingestellt wor den ist, wird dieses Filtrat mit einer Ge_ sehwindigkeit von ungefähr 1 Liter pro Stunde durch eine Säule von Duolite S 30 geführt.
Die Säule ist. 75 ein lang, hat. einen Durchmesser von ungefähr 4,6 ein und ist mit Natriumliydroxyd regeneriert worden, wäh rend es vor jedem Gebraueli mit ungefähr 1250 ein-' eines 0,5-molaren Azetatpuffers mit pl, = 4,5 und daraufhin mit -100 em3 Wasser gewaschen wird.
Nachdem das Kulturfiltrat durch die Säule geführt. worden- ist, wird es wiederum mit -100 cni3 Wasser gewaschen. Das Filtrat ent hält nun ungefähr 901/o Vengicid, während (las Oxytetracyclin quantitativ adsorbiert wor- ci en ist.
Die Ausscheidung des Vengicids findet nach Beispiel 6 statt, d. h. es wird an aktiver Kohle adsorbiert, mit einem Wasser-Butanol- Phenol-Cemiseh eluiert, das Eluat konzen triert und mit Butanol extrahiert, die Lösung in Butanol mit Wasser extrahiert und die wässrige Lösung bei pH 2,5 konzentriert.
Dar aufhin wird das Vengicid bei pH = 4,5 gefällt durch Zugabe von Natriumhydroxyd zur wäss- rigen Lösung; das Produkt wird durch Um kristallisation gereinigt. Die Ausbeute ist 2,0 g.
Je nach der ursprünglich anwesenden Menge Vengicid in dem Kulturfiltrat und der andern Bestandteile kann sich eine oder meh rere der obigen Stufen erübrigen.
Das Oxy tetracyclin wird aus der Duolite S 30 -Säule mit Hilfe von 2000 em3 einer 0,2511 wässrigen Natriumhydroxydlösung elu- iert; :die Lösung wird mit einer Geschwindig keit von ungefähr 1000 cm?, pro Stunde durch geführt. Dann wird dieses Eluat im Vakuum in einem Rückflussverdampfer bei pH = 2,0 auf 1000 em3 eingeengt.
Die Lösung enthält gemäss der mikrobiolo gischen Prüfung 4,27 g Oxytetracyclin (d. h. 90% der ursprünglichen Menge). Die Lösung wird mit Butanol bei pH == 9,5 extrahiert.
Der auf diese Weise erhaltene Butanolextrakt. wird im Vakuum auf ungefähr 3000 em3 konzen triert. -Die Lösung enthält dann 3,2 g Oxy- tetraeyclin. Diese Menge wird mit Hilfe der quaternären Ammoniumbase quantitativ ge- fiillt, und weiter, wie in Beispiel 6 beschrie- ben, gereinigt. Es werden 2,
6 g Chlorhydrats von Oxytetraeyelin erhal- teli.
Beispiel <I>8</I> 1.2 Liter der gemäss Beispiel 3 erhaltenen Flüssigkeit. werden nach der Fermentation mit konzentrierter Salzsäure unter Rühren auf ein pH = 2,2 eingestellt und, nachdem man eine halbe Stunde gerührt hat,, wird zen trifugiert. Die opalisierende Flüssigkeit wird im Vakuum auf 1200 cm- konzentriert und daraufhin mit einem Filtrierhilfsmittel fil triert.
Das klare Filtrat wird mit Natrium hydroxyd auf ein PH = 9,0 eingestellt und viermal mit. 900 ein?- Butanol extrahiert. Die Butanollösung wird mit Salzsäure auf ein pH = 2,0 eingestellt und fünfmal mit 1100 em3 Wasser extrahiert.
Die wässrige Lösung ent hält-, wie eine mikrobiologische Analyse zeigt, 4,50 g Oxytetracyelin (800%o der ursprünglich anwesenden Menge) und 3,01 g Vengieid (9311/o der ursprünglich anwesenden Menge).
Nach Konzentration im Vakuiun auf unge fähr 1900 em3 wird das Oxytetracyelin, nach dem in Beispiel 6 beschriebenen Verfahren, mit einer quaternären Ammoniumbase und einem Erdalkalisalz gefällt. Das Vengicid wird aus dem Filtrat, wie in Beispiel 6 be schrieben, erhalten. Die Ausbeute an Oxy- tetracyclin ist. 3,2 g und die an Vengicid 2,2 g.
<I>Beispiel 9</I> 10 Liter der nach Beispiel 3 erhaltenen Flüssigkeit werden mit Schwefelsäure unter Rühren auf ein PH = 2,2 eingestellt und, nach dem man eine halbe Stunde gerührt, hat, zentrifugiert. Die opalisierende Flüssigkeit wird im Vakuum auf 1000 eins konzentriert und daraufhin mit einem F'iltrierhilfsmittel filtriert. Das klare Filtrat wird dann mit Natriumhydroxyd auf ein pH = 9,0 eingestellt und viermal mit 0,75 Liter Butanol extrahiert.
Die Butanollösung wird viermal mit 0,75 Liter Wasser bei einem pH = 2,0 extrahiert. Auf diese Weise werden ungefähr 3,0 Liter einer, wässrigen Lösung der Antibiotika erhalten, die gemäss einer mikrobiologischen Analyse 3,8 g Oxytetracyclin und 2,4 g V engicid enthalten.
Dieser Extrakt wird im Vakuum konzen triert auf ein Volumen von ungefähr 1,0 Liter. Die Lösung wird in ungefähr 6 Stunden durch 4 Säulen von Duolite S 30 geführt. Die ersten drei Säulen enthalten je 30 g Duolite , während,die vierte Säule 20 g Duolite ent hält. Die 30 g Adsorptionsmittel enthaltenden Säulen sind 42 cm lang, die andere Säule ist 28 cm lang.
Das Verhältnis zwischen dem Durchmesser und :der Länge ist ungefähr 1.:15. Das Duolit in den Säulen ist mit Natriumhydroxyd regeneriert worden und, dar aufhin mit- 0,1n-Salzsäure gewaschen, bis das pH des durehfliessenden Wasehwassers 2,0 oder niedriger war.
Die Analyse der Säulen zeigt, dass das Oxytetracyelin und das Vengi- cid wie folgt über .die Säulen verteilt sind:

Die Eluierung der Säule wird mit. 0,25n- Natriumhydroxyd durchgeführt. Die vierte Säule wird nicht eluiert., sondern für die fol gende Adsorption behalten. Für jede Säule braucht man 300 em3 Natriumhydroxyd. Die gesamten Eluate werden mit konzentrierter Salzsäure auf ein pH = 2,0 eingestellt und im Vakuum auf 175 cms konzentriert.
Diese Lösung enthält 20 mg 0xytetracyclin pro em3. Sie wird neutralisiert, was eine Fällung des Oxytetracyclins zur Folge hat. Nach Filtrie ren, Waschen mit. Wasser und Trocknen wiegt :dieses Präzipitat <B>3,7</B> g. Es enthält 3,4 g Oxy- tetracyclin. Das Präzipitat wird zur weiteren Reinigung in 27 em3 Methanol suspendiert. 4,5 cm' konzentrierte Salzsäure (spez. Gewicht.
1.,9) werden der Suspension zugesetzt. Das Oxytetracyclin wird in das Chlorhydrat über geführt, während die Verunreinigungen sieh lösen. Das Salzsäuresalz .des Oxytetracyelins wird mit methanoliseher Salzsäure (0,1n) fil triert und schliesslich mit. Äther gewasehen. Die Ausbeute an Oxy tetracy elin ist<B>3,12</B> g.

Streptomyces vendargensis is preferably grown as an immersion culture in a liquid nutrient medium under sterile conditions in closed vessels which are provided with stirrers into which sterile oxygen or air can be fed during the cultivation for the purpose of ventilation.
The time during which the cultivation is carried out, the temperature and the other conditions that must be observed to maintain good yields of antibiotics are described in more detail below.
The culture medium should contain a carbon supplier and an organic or inorganic nitrogen supplier; the presence of mineral salts such as phosphates, potassium and sodium salts, and traces of various metals is desirable. Often, however, the starting materials that are used in practice are already sufficiently contaminated with these mineral salts, so that there is no need to add them specifically.
Both soluble and insoluble carbohydrates such as glucose, Saceha rose, lactose or starch can be used as carbon suppliers. Sugar alcohols such as glycerin are also suitable.
The amount of carbon suppliers in the medium can be very different and depends on the type of carbohydrate used and the other composition of the medium; Usually it is between 0.5 and 5 1 / a, the weight of the medium.

The temperature of the fermentation can be between 20 and 3 ° C. Preferred temperatures are between 26 =? 8 C.
For optimal growth of the organism and an optimal yield of anti biotics, the PH should be kept within fairly narrow limits during fermentation, e.g. B. between 5.0 and 8.0. The liquid is then best filtered or centrifuged to remove the ibIycel. If the fluid is not acidified, 20-250 / 9 of the oxytetraeycline is lost.
After removing the mycelium and all other solid constituents from the fermentation fluid, the antibiotics can be adsorbed on activated charcoal at a pH of about 7 and then with the help of an alcohol that is mixed with.
