Verfahren zur Herstellung von 17-Hydroxysteroiden <B>C</B> 1-enstand des Hauptpatentes Nr. 3'- ( -'e <B>21700</B> ist ein Verfahren zur Herstellung von 17- Hydroxyverbindungen der Pregnan- oder Allopregnanreihe, welches dadurch gekenn zeichnet.
ist, dass man 20-Ketimine der Pre- gnan- oder Allopregnanreihe mit acylierenden Mitteln umsetzt, die Reaktionsprodukte mit organischen Persäuren oxydiert und die Oxy dationsprodukte mit hydrolysierenden Mitteln behandelt.
Es wurde nun gefunden, dass man 17- Hydroxy verbindungen der Pregnenreihe ge winnen kann, indem man 20-Ketimine der Pregnenreihe, die mindestens.
eine, gegebenen- falls aeylierte, Hydroxygruppe aufweisen, mit aeylierenden Mitteln zu Acyloxy-enaeylaminen umsetzt, diese partiell zu Hydroxy-enacyl- aininen verseift, deren 1-Tydroxygruppen zu Carbonylgruppen oxydiert, die Reaktionspro dukte mit organischen Persäuren zu Epoxy- den oxydiert, und diese mit hydrolysierenden Mitteln behandelt.
Je nach den Reaktionsbedingungen können hierbei 1 oder 2 Asylreste an (las Stickstoff atom treten. In letzterem Falle ist einer der beiden Aeylreste leicht verseifbar. Die En- amin-Doppelbindung kann zwischen 17 und 20 sowie zwischen \20 und 21 liegen, jedoch ist die Lage zwischen 17 und 20 bevorzugt. Die Durchführung der Reaktion wird nicht durch weitere Substituenten oder Doppelbin dungen im Steringerüst beeinflusst.
Die freie Hydroxy- und etwa. vorhandene Aminogrup- pen werden verständlicherweise ebenfalls acy- liert, etwa vorhandene Carbonylgr.ippen even tuell in Enolacylate übergeführt. Die vorliegenden Enaminacylate verhalten sich chemisch wesentlich anders als die 417,20- Enolacetate, wie sie Gallagher beschrieben hat.
Während 20-Enolaeetate auch unter sehr milden Bedingungen unter Rückbildung der 20-Ketogruppe hydrolysiert werden, gelingt es bei den Enaminaeylaten, wenn sie 2.
Acyl- gruppen enthalten, leicht, sie zu Monoacyl- enaminen zu verseifen sowie aus eventuell im Kern des Sterinskelettes vorhandenen Acyl- oxygruppen die Asylgruppe abzuspalten, ohne dass dadurch die Enamin-Struktur aufgeho ben würde.
Weiterhin unterscheiden sich Enolacetate und Enaminacylate auch in ihrem Verhal ten bei der Halogenierung. Während ungesät tigte Enolacetate nicht partiell zu halogenie- ren sind, gelingt es beispielsweise, eine d5 ständige Doppelbindung der Enamindiacylate mit Halogen abzusättigen, ohne dass die En amin-Doppelbindimg in Reaktion tritt.
Die Anlagerung von Sauerstoff an die 17,20-ständige Doppelbindung der Enacyl- amine mittels Persäuren kann in üblicher Weise geschehen. Etwa vorhandene, gegen Persäuren empfindliche Doppelbindungen im Kern können hierbei vorübergehend durch Halogene geschützt werden. Bei 44-3-Ketonen erübrigt sich dieser Schutz. Die entstehen den Epoxyde werden mit hydrofysierenden Mitteln behandelt, wobei formal unter Bil dung von .Säureamiden 1'7-a-Hydroxypregnan- derivate entstehen.
Die neuartige Aufspal tung der Epoxyde ist für den Fachmann ausserordentlich überraschend, zumal es be kannt ist, dass Sterine, die acylierte Stick stoffatome enthalten, ausserordentlich schwer hydrolytisch zu spalten sind.
Nach dem Verfahren gemäss der vorlie genden Erfindung gelingt es also, 17-Hydroxy- v erbindungen der Pregnenreihe in glatter Reaktion auch im technischen Massstab dar zustellen.
Die als Ausgangsmaterial für das vorlie gende Verfahren verwendeten 20-Ketiminver- hindungen sind leicht nach den Verfahren der deutschen Patentschriften Nrn. 693351 und 713193 bzw. Schweizer Patent. Nr. 306'507 herstel'lbar.
Als in Frage kommende Verbindungen seien beispielsweise genannt: d5-Pregnen-ol- (3)-on-(20)-ketimin, 3-Formoxy-pregnen-(5)- on-(20)-ketimin, 3,11-Dihydroxy pregnen-(5)- on-(20) -ketimin, 3@-Aeetoxy-11-oxo-pregneti- (5)-on-(20)-ketimin, ferner 7- bzw.
1'2-Hydr- oxyverbindungen, wie sie beispielsweise durch Curtiussehen Abbau der fallensäuren und Überführung der erhaltenen Amine in Keti- mine naeh den Verfahren der oben genannten deutschen Patentschriften erhalten werden.
Es ist nicht erforderlich, für die. Her stellung der acylierten Enamine von reinen Ketiminen auszugehen. Es genügt, wenn man nach dem Verfahren der oben genannten Pa tentschriften 20-Steroidamine in Chloramine überführt und aus diesen durch Abspaltung von Salzsäure mittels Natriumalkoholat die Ketimine herstellt.
Man kann dabei aus der Reaktionsmischung das Lösungsmittel im Va- kuum abdestillieren und den Rüekstand mit den unten aufgeführten Acylierungsmitteln direkt umsetzen. Es ist aber anderseits auch möglich, durch Einleiten von Chlorwasserstoff unter Kühlung das Chlorhydrat des Ketimins zu gewinnen und aus diesem durch Behan deln mit Alkalialkoholat das Ketimin., in Frei heit zu setzen.
Als acylierende Mittel, wie sie gemäss dem Verfahren der vorliegenden Erfindung in Betracht kommen, seien beispielsweise ge nannt : organische Carbonsäureanhydride, wie Benzoesäureanhydrid, Essigsäureanhydrid, Propionsäureanhydrid u. ä.; insbesondere-sind die Anhydride der niederen aliphatisehen Car- bonsäuren für die Reaktion -eeignet. An Stelle der Säureanhy dride kann man auch die Säurechloride verwenden.
Für die Einfüh rung der Acetylgmuppe eignen sich ferner Isopropenylaeetat sowie Keten.
Als organische Persäuren kommen zum Beispiel in Betracht: Aromatische Persäuren, wie Perbenzoesäure, Perphthalsäure, oder ge sättigte aliphatische Persäuren, wie Per ameisensäure, Peressigsäure oder Perbernstein- säure u. ä.
Es ist nicht erforderlich, die freien Persäuren zu verwenden, sie können vielmehr auch während der Reaktion intermediär aus den vorhandenen organischen Säuren durch Zugabe von Peroxyden, wie beispielsweise Na- triumperoxy d oder Bariumperoxyd, gebildet werden. Auch eine -Mischung aus Wasserstoff- superoxyd und Eisessig kann verwendet wer den.
Als h@-drolysierende Mittel werden vor zugsweise verdünnte Alkalilau-en, wie Natron lauge oder Kalilauge, verwendet.
Die Verfahrenserzeugnisse stellen wertvolle Zwischenprodukte zur Gewinnung von in der ärztlichen Praxis eingeführten, wertvollen Nebennierenrindenhormonen dar.
Beispiel js,i:ezb-pregiindic ri-ol-(3)-ocetylaiiiiii- (,20)-nceta_t 1,06 g J5Pregnen-ol-(3)-oii- (20) -ketimin werden in 20 em3 Py ridin gelöst. und nach dem Versetzen mit 7,5 em3 Essigsäureanhy- drid 16 Stunden bei Zimmertemperatur sich selbst überlassen.
Die Reaktionsmisehung wird dann in Eiswasser eingerührt und nach einigem Stehen der ausgefallene Niederschlag abgesaugt, bei 700 getrocknet und aus Cyclo- hexan umkristallisiert. Ausbeute 750 mg. Schmelzpunkt 149 bis 153 (auf der Kofler- schen Schmelzbank).
Zur Reinigung kann die Substanz auch im Hoehvakuum bei 2.10 bis 220 /10-3 mm destilliert werden. Das Ultrarotabsorptions- spektrum zeigt. Banden bei 2,94 It und bei 3,05 11, die der NH=COC'H3-Gruppe zuzuord nen sind.
h) 45,17(20)-pregnadien,-ol-(3)-diacetylamin- (20)-acetat (1) Das zur Trockne eingedampfte Reak tionsgemisch, wie es bei der Einwirkung einer Lösung von 2;57 g Natrium in 195 em3 abso lutem Alkohol auf 4,69 g 45-Pregnen-ol-(3)- ehloramin-(20)-acetat erhalten wird, wird in 68 em3 Essigsäureanhydrid gelöst,
zum Sieden erhitzt und der gebildete Essigester und die Essigsäure vollständig abdestilliert. Anschlie ssend wird eine Stunde unter Rückfluss zum Sieden erhitzt. Die weitere Aufarbeitung des Reaktionsgemisches geschieht wie unter a) be schrieben. Das Rohprodukt wird aus wenig Cycloliexan und dann aus Methanol umkri stallisiert.. Ausbeute 3,2 g. Schmelzpunkt 179 bis 1820 (auf der Koflersehen Schmelzbank).
(2<B>) '2,5</B> g 45,17(20)-Pregnadien-ol-(3')-acetyl- amin-(20)- acetat werden in einer Mischung voll 25 eins Essigsättreanhydrid, 25 cm3 Iso- propenylacetat und 1,25 g p-Toluolsulfonsäure gelöst und 3 Stunden unter Rüekfluss zum Sieden erhitzt. Dann wird das abgekühlte Re aktionsgemisch in Eiswasser eingegossen, 3 Stunden gerührt und mit Äther extrahiert.
Naeli dem Waschen mit Bicarbonatlösung und Wasser wird die Ätherlösung über Natrium sulfat getrocknet. und im Vakuum zur Troekne cingedanipft. Das so erhaltene Rohprodukt wird an saurem Aluminiumoxyd chromatogra- phiseh gereinigt.
Die Elutionen mit Benzol petroläther (1:3) ergeben 850 mg d5,17(20)- Pregnadieu o'1-(3)-diacetylamin- (20) -acetat, das aus Methanol umkristallisiert werden kann. Schmelzpunkt 1,51 bis 153 (auf der Koflerschen Schmelzbank).
Der Schmelzpunktunterschied der nach b) (1) und b) (2;) erhaltenen Verbindungen be ruht offenbar auf einer cis-trans-Isomerie. <I>c)</I> 5,6-Dibromr417#20-pregnen-ol-(3)-diacetyl- amin-(20)-ctcetcat .1.12 mg 45,17(20).:Pregnadien-ol-(3)-diacetyl- amin-(20)-acetat. und 140 mg wasserfreies Na triumacetat werden in 10 cm3 Eisessig gelöst und .dazu unter Rühren eine Lösung von 160 mg Brom in 2:;
5 cm3 Eisessig zugetropft. Nach einigen Minuten wird die Reaktions mischung in 100 cm3 Wasser eingerührt, der entstandene Niederschlag abgenutscht, mit Wasser gut gewaschen, in kaltem Aceton ge löst und die Acetonlösung vorsichtig mit Was ser versetzt. Das auskristallisierende Dibro- mid wird abgesaugt. Ausbeute 400 mg. Schmelzpunkt 180 bis 196 (unter Zersetzung auf der Koflerschen Schmelzbank).
<I>d)</I> 45,17(2o) <I>-</I> pregnadielc-ol-(3)-acetylamizt,(20) (1) Eine Lösung von 280 mg d5.17(20)- Pregnadien ol-(3i)-acetylamin-('20) - acetat in 10 em3 Methanol wird mit einer Lösung von 130 mg Kaliumearbonat in '2,2 em3 Wasser vermischt und 105 Minuten unter Rückfluss zum Sieden erhitzt. Die abgekühlte Lösung wird in Wasser gegossen, die ausgefallenen Flocken abgesaugt und getrocknet. Sie wer den zur Reinigung aus viel Aceton umkri stallisiert. Ausbeute 190 mg.
Schmelzpunkt 240 bis 2600 (unter Zersetzung auf der Kofler- schen Schmelzbank).
(2) Eine Lösung von 500, mg 45,17(20)- Pregnadien-ol'-(3)-dia.cetylamin-(20,)-acetat in 18 em3 Methanol wird mit einer Lösung von 400 mg Ka.liumbicarbonat in 4,5 eins Wasser versetzt und 90 Minuten unter Rückfluss zum Sieden erhitzt. Die weitere Aufarbeitung ge schieht analog wie unter d) (1) beschrieben. Ausbeute 240 mg, 'Schmelzpunkt 250 bis 2700 (unter Zersetzung auf der Koflerschen Schmelzbank).
e) ,J@,r@rzo@_pre3.raactiem-osa-(3)-acet@la.@@ti7z-(20) 100 mg d5.i7(zo)-Pregnadien-ol-(3) -acetyl- amin - (20) werden in 90 cm3 heissem Toluol gelöst, mit. 23 em3 C'yclohexanon versetzt und aus der Mischung etwa 12 cm3 abdestilliert. Nach dem Zugiessen einer Lösung von 8,
00 mg AluminiumisoPro- pylat in 5 em3 Toluol wird 130 Minuten un ter Rüekfluss zum Sieden erhitzt. Nach Zu gabe von 0,3 cm-3 Eisessig in 3 cm-' Toluol wird das Reaktionsgemisch 2. Stunden der Wasserdampfdestillation unterworfen. Schliess lich wird mit. Äther extrahiert, die Äther schicht mit Wasser gewaschen, über Natrium sulfat getrocknet und im Vakuum zur Trockne eingedampft.
Das Rohprodukt wird in wenig Aceton gelöst und, durch vorsichtiges Zugeben von Petroläther zum Kristallisieren gebracht. Ausbeute 300 mg. Schmelzpunkt 173 bis 180 (auf der Koflerschen Schmelzbank) (cis- trans-Isomerengemisch). [a] D = +99 .10 (in Äthanol).
<I>f)</I> 17,20-0xido-d4-pregnen-on-(3)-aceftyZ- amin,-(20) 260 mg 44,i7(zo)-pregnadien-on-(3)-acetyl- amin-(20) werden in einer Mischung von 20 cm3 Benzol und 6 c0- Toluol gelöst, auf 0 abgekühlt und mit. 113 mg Benzoper- säure, gelöst in 0,81 cm3 Benzol, versetzt.
Nach 100 Minuten Stehen bei 0 wird mit Äther verdünnt, die Ätherlösung mit 5prozentiger Natronlauge und Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuiun bei maximal 250 zur Trockne eingedampft. Das Rohprodukt wird mit wenig Aeeton ver rieben, wobei Kristallisation eintritt. Ausbeute 121 mg. Schmelzpunkt 14-6 bis 149 unter Zer setzung (Kupferblock).
g) 17-(a)-Oxy-progesteron (1) 100 mg 17,20-Oxido-44-pregnen-on- (3)-a.cetyIamin-(20) werden in 12 em3 Me thanol suspendiert, mit 3 cm3 0,3n-Natron- lauge versetzt und in Stickstoffatmosphäre eine Stunde unter R.ückfluss zum Sieden er hitzt, wobei bereits nach einigen Minuten alles in Lösung geht. Dann wird mit. Essig säure neutralisiert. und der Alkohol im Va kuum eingedampft.
Die ausgeschiedenen Kri stalle werden abgesaugt, mit Wasser gewa schen und aus wenig Aceton umkristallisiert. Ausbeute 61 mg. Schmelzpunkt 205 bis 209 (Kupferblock) (unkorr.); Misehschmelzpunkt mit authentischem 17-(a)-Oxy-progesteron 205 bis 209 im Kupferblock (unkorr.).
(2) 270 mg 17;\?0-Oxido-44-pregnen-on-(3)- acetylamin-(20) werden in 31 em3 Methanol suspendiert, mit, 16,8 ein- 0,3n-Natronlaiige versetzt und in Stiekstoffatmosphäre eine Stunde unter Rückfluss zum Sieden erhitzt. Die weitere Aufarbeitung geschieht, wie sie vorstehend unter (1) beschrieben ist. :Ulan erhält 212 mg 17-(a)=Oxy-progesteron vom Schmelzpunkt 205 bis '09 (Kupferblock) (unkorr.) .
Das als Ausgangsmaterial benötigte 45- Pregnen-ol-(3)-on-(\_'ü)-ketimin kann in fol gender Weise gewonnen werden: In einer Lösung von 0,9 g Natrium in 68 cm3 absolutem Alkohol werden 1,6'5 g 45-Pregnen-ol-(3)-chloramin,(20) -acetat sus pendiert und 40 Minuten unter Rückfluss zum Sieden erhitzt.
In das abgekühlte Reaktions gemisch wird trockenes Chlorwasserstoffgas bis zur kongosauren Reaktion eingeleitet und dann anschliessend Nat.riumä.thylatlösung bis zur alkalischen Reaktion zugesetzt.
Nach dem Einengen im Vakuum in Stiekstoffatmosphäre wird der feste Rüekstand mit 100 em3 Benzol ausgekocht, die benzolisehe Lösung heiss ab- genutseht und im Vakuum unter Stickstoff abermals zur Trockne eingeengt. Ausbeute 1.16 g d5 - Pregnen - o1 - (3) - on-(20)-ketimin. Schmelzpunkt. 178 (Koflerbank).
Geht man nicht vom reinen 20-Ketimin, sondern von '20--Aminen aus, so kann man das unter a) beschriebene Enacet-yl'amin auch wie folgt, ohne Isolierung des Ketimins, her stellen In eine Lösung von 2;57 g Natrium in 195 em3 absolutem Alkohol werden 4,69 g 45-Pregnen-ol-(3)-ehloramin-(20)-acetat einge tragen und das Reaktionsgemiseh -10 Minuten unter Rückfluss zum Sieden erhitzt.
Hierauf wird der überschüssige Alkohol im Vakuum abgedampft.. Zum Rückstand werden vorsich tig und unter Kühlung 68 em3 Essigsäure- anhydridgegeben, Nach dem Abklingen der Reaktion wird die Reaktionslösung eine Stunde unter Rückfluss zum :Sieden erhitzt.
Naeli dem Abkühlen wird vom auskristallisier ten Na.triunlacetat und Natriumchlorid scharf abgenutseht. Das Filtrat wird im Vakuum zur Trockne eingeengt, Die Lösung des Filter r,üekstandes in 350 em3 Wasser wird in die Lösung des obigen Verda.mpfungsrückstandes in 30 e1113 Alkohol eingerührt und der ausge flockte Niederschlag nach einigem Stehen ab- genutseht, mit Wasser gewaschen und im Vakuum bei 70 getrocknet.
Das gepulverte Rohprodukt wird aus heissem Cyelohexan um- kristallisiert. Ausbeute '2 ,S g. Schmelzpunkt 150 bis 152 .

h) 45,17 (20) -pregnadiene, -ol- (3) -diacetylamine- (20) -acetate (1) The reac tion mixture evaporated to dryness, as it is when exposed to a solution of 2; 57 g of sodium in 195 em3 absolute alcohol on 4.69 g of 45-pregnen-ol- (3) -chloramine- (20) acetate is obtained, is dissolved in 68 em3 acetic anhydride,
heated to boiling and the ethyl acetate formed and the acetic acid completely distilled off. The mixture is then refluxed for one hour. The further work-up of the reaction mixture takes place as described under a) be. The crude product is umkri crystallized from a little cycloliexane and then from methanol .. Yield 3.2 g. Melting point 179 to 1820 (on the Koflersehen melting bench).
(2 <B>) '2.5 </B> g 45.17 (20) -Pregnadien-ol- (3') -acetyl-amine- (20) -acetate are added in a mixture full of 25 units of acetic anhydride, 25 cm3 of isopropenyl acetate and 1.25 g of p-toluenesulfonic acid dissolved and heated to boiling for 3 hours with reflux. Then the cooled reaction mixture is poured into ice water, stirred for 3 hours and extracted with ether.
After washing with bicarbonate solution and water, the ether solution is dried over sodium sulfate. and dried in a vacuum. The crude product obtained in this way is purified by chromatography on acidic aluminum oxide.
Elutions with benzene petroleum ether (1: 3) give 850 mg d5,17 (20) - Pregnadieu o'1- (3) -diacetylamine- (20) -acetate, which can be recrystallized from methanol. Melting point 1.51 to 153 (on the Kofler melting bench).
The melting point difference of the compounds obtained according to b) (1) and b) (2;) is apparently based on a cis-trans isomerism. <I> c) </I> 5,6-Dibromor417 # 20-pregnen-ol- (3) -diacetyl- amine- (20) -ctcetcat. 1.12 mg 45.17 (20) .: Pregnadien-ol- ( 3) diacetyl amine (20) acetate. and 140 mg of anhydrous sodium acetate are dissolved in 10 cm3 of glacial acetic acid and, while stirring, a solution of 160 mg of bromine in 2 :;
5 cm3 glacial acetic acid was added dropwise. After a few minutes, the reaction mixture is stirred into 100 cm3 of water, the resulting precipitate is filtered off with suction, washed well with water, dissolved in cold acetone and water is carefully added to the acetone solution. The dibromide which crystallizes out is suctioned off. Yield 400 mg. Melting point 180 to 196 (with decomposition on the Kofler melting bench).
<I> d) </I> 45.17 (2o) <I> - </I> pregnadielc-ol- (3) -acetylamizt, (20) (1) a solution of 280 mg d5.17 (20) - Pregnadien ol- (3i) -acetylamine - ('20) - acetate in 10 em3 methanol is mixed with a solution of 130 mg potassium carbonate in 2.2 em3 water and refluxed for 105 minutes. The cooled solution is poured into water, the flakes which have precipitated out are suctioned off and dried. You who the umkri crystallized for cleaning from a lot of acetone. Yield 190 mg.
Melting point 240 to 2600 (with decomposition on the Kofler melting bench).
(2) A solution of 500 mg 45.17 (20) - Pregnadien-ol '- (3) -dia.cetylamine- (20) - acetate in 18 em3 methanol is mixed with a solution of 400 mg potassium bicarbonate in 4.5 liters of water were added and the mixture was refluxed for 90 minutes. Further work-up is carried out analogously to that described under d) (1). Yield 240 mg, melting point 250 to 2700 (with decomposition on the Kofler melting bench).

00 mg of aluminum isopropylate in 5 cubic meters of toluene is heated to the boil for 130 minutes under reflux. After adding 0.3 cm -3 of glacial acetic acid in 3 cm- 'toluene, the reaction mixture is subjected to steam distillation for 2 hours. Finally, with. Ether extracted, the ether layer washed with water, dried over sodium sulfate and evaporated to dryness in vacuo.
The crude product is dissolved in a little acetone and made to crystallize by carefully adding petroleum ether. Yield 300 mg. Melting point 173 to 180 (on the Kofler melting bench) (cis-trans isomer mixture). [a] D = +99.10 (in ethanol).
<I> f) </I> 17.20-0xido-d4-pregnen-on- (3) -aceftyZ- amine, - (20) 260 mg 44, i7 (zo) -pregnadien-on- (3) - acetyl amine (20) are dissolved in a mixture of 20 cm3 of benzene and 6 co-toluene, cooled to 0 and with. 113 mg benzopic acid, dissolved in 0.81 cm3 benzene, added.
After 100 minutes of standing at 0, it is diluted with ether, the ethereal solution is washed with 5% sodium hydroxide solution and water, dried over sodium sulfate and evaporated to dryness in a vacuum at a maximum of 250. The crude product is rubbed with a little acetone, with crystallization occurring. Yield 121 mg. Melting point 14-6 to 149 with decomposition (copper block).
g) 17- (a) -Oxy-progesterone (1) 100 mg of 17,20-oxido-44-pregnen-on- (3) -a.cetyIamin- (20) are suspended in 12 em3 of methanol, with 3 cm3 0.3N sodium hydroxide solution is added and the mixture is heated to boiling under R reflux for one hour in a nitrogen atmosphere, and after a few minutes everything dissolves. Then with. Acetic acid neutralized. and the alcohol evaporated in a vacuum.
The precipitated crystals are filtered off with suction, washed with water and recrystallized from a little acetone. Yield 61 mg. Melting point 205 to 209 (copper block) (uncorr.); Melting point with authentic 17- (a) -oxy-progesterone 205 to 209 in the copper block (uncorr.).
(2) 270 mg of 17; 0-oxido-44-pregnen-on- (3) -acetylamine- (20) are suspended in 31 em3 of methanol, 16.8.8 0.3N sodium hydroxide solution is added and the solution is dissolved in A nitrogen atmosphere was heated to boiling under reflux for one hour. The further work-up takes place as described above under (1). : Ulan receives 212 mg of 17- (a) = oxy-progesterone from melting point 205 to '09 (copper block) (uncorr.).
The 45-pregnen-ol- (3) -one - (\ _ 'ü) -ketimine required as starting material can be obtained in the following manner: In a solution of 0.9 g of sodium in 68 cm3 of absolute alcohol, 1.6 '5 g of 45-pregnen-ol- (3) -chloramine, (20) -acetate suspended and heated to boiling under reflux for 40 minutes.
Dry hydrogen chloride gas is introduced into the cooled reaction mixture until the Congo-acid reaction occurs, and then sodium ethoxide solution is added until the reaction is alkaline.
After concentrating in vacuo in a nitrogen atmosphere, the solid residue is boiled out with 100 cubic meters of benzene, the benzene solution is sifted off while hot and again concentrated to dryness in vacuo under nitrogen. Yield 1.16 g of d5 - pregnen - o1 - (3) - one- (20) -ketimine. Melting point. 178 (Koflerbank).
If one does not start with pure 20-ketimine, but with '20 -amines, the enacet-ylamine described under a) can also be prepared as follows, without isolating the ketimine: In a solution of 2.57 g Sodium in 195 cubic meters of absolute alcohol are 4.69 g of 45-pregnen-ol- (3) -ehloramin- (20) acetate and the reaction mixture is heated to boiling under reflux for -10 minutes.
The excess alcohol is then evaporated off in vacuo. 68 cm3 acetic anhydride are added to the residue carefully and with cooling. After the reaction has subsided, the reaction solution is heated to boiling under reflux for one hour.
After cooling, the crystallized Na.triunlacetat and sodium chloride are sharply removed. The filtrate is concentrated to dryness in vacuo. The solution of the filter residue in 350 cubic meters of water is stirred into the solution of the above evaporation residue in 30 percent of alcohol and the flocculated precipitate is sifted off after standing for a while, washed with water and dried in vacuo at 70.
The powdered crude product is recrystallized from hot cyelohexane. Yield '2, S g. Melting point 150 to 152.
