Verfahren zur Herstellung von 4-Oxo-19-nor-A-homosteroid-dienen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von neuen 4-Oxo-19-nor-A-homo-steroid-dienen, insbesondere 4-Oxo-19-nor-A-homo-androstadienen, in erster Linie solchen der allgemeinen Formel

worin R eine freie oder ketalisierte Oxogruppe, eine freie, veresterte oder verätherte Hydroxylgruppe zusammen mit einem Wasserstoffatom oder einem niederen aliphatischen Kohlenwasserstoff darstellt.
Die neuen Verbindungen sind wertvolle Zwischenprodukte zur Herstellung der bekannten, hochwirksamen 4-Oxo-19-nor-A-homo-steroid-1(10),2,4a-triene. Sie besitzen aber auch selbst wertvolle pharmakologische Eigenschaften.
Besonders wichtig sind die Verbindungen obiger Formel, worin R eine Ketogruppe oder eine freie oder mit einer niederen aliphatischen Carbonsäure veresterte Hydroxylgruppe zusammen mit einem Wasserstoffatom oder einem niederen Alkyl-, wie Methyl- oder Äthyl-, einem niederen Alkenyl-, wie Vinyl-, Allyl- oder Methallyl-, oder einem niederen Alkinyl-, wie Äthinyl- oder Propinylrest darstellt.
Die neuen Verbindungen werden erhalten, wenn man ein A1,4-3-Oxo-steroid-dien, das in 10-Stellung eine mit einer starken anorganischen oder organischen Säure veresterte Hydroxymethylgruppe trägt, mit einem Alkalimetall in einem aprotischen Lösungsmittel und in Ge- genwart eines polycyclischen aromatischen Kohlenwasserstoffs umsetzt, erhaltene 4-Oxo-19-nor-A-homo-ste- roid-2,5(10)-diene zu den entsprechenden 1,5(10)-Dienen isomerisiert.
Eine mit einer starken anorganischen oder organischen Säure veresterte Hydroxylgruppe ist insbesondere eine solche die mit Halogenwasserstoffsäure, wie Chlorwasserstoff oder Bromwasserstoffsäure oder mit einer Sulfonsäure, wie Methan- oder p-Toluolsulfonsäure verestert ist.
Die Umsetzung mit dem Alkalimetall, wie Natrium, Kalium, insbesondere aber Lithium, wird vorzugsweise bei tiefer Temperatur, z. B. bei 1000 bis +150, in erster Linie bei ungeführt 400 bis - 750 durchgeführt.
Als polycyclische aromatische Kohlenwasserstoffe seien z. B. genannt: Naphthalin, die Methyl-naphthaline, die Dimethyl-naphthaline, Phenanthren, Terphenyl, Anthracen, Acenaphthen, Stilben, Fluoranthen und insbesondere Diphenyl. Aprotische Lösungsmittel sind solche die keine Protonen abgeben können, insbesondere offenkettige oder cyclische Äther, gegebenenfalls im Gemisch untereinander oder zusammen mit andern aprotischen Lösungsmitteln. Es kommen insbesondere in Betracht: niederaliphatische Äther, wie Dimethyläther, Diäthyl äther, Dipropyläther, Dibutyl- oder -isobutyläther, Äthylenglykoldiäthyläther, z. B. Äthylenglykoldimethyl- oder -diäthyläther, Propylenglykoläther, wie Propylenglykoldimethyl- oder -diäthyläther, Acetale, wie Acetaldehyddimethyl- oder -diäthylacetal, Dioxan, Tetrahydrofuran, Tetrahydropyran, Methyltetrahydrofurfuryläther und ähnliche Verbindungen.
Besonders gut bewährt hat sich Tetrahydrofuran. Ferner können auch etwa aliphatische oder aromatische inerte Kohlenwasserstoffe, wie Hexan, Benzol, Toluol oder hydroaromatische Kohlenwasserstoffe, wie Tetralin oder Decalin, verwendet werden.
Die genannte Umsetzung wird vorzugsweise so durchgeführt, dass man das Alkalimetall in einem aprotischen Lösungsmittel auf einen polycyclischen aromatischen Kohlenwasserstoff, vorteilhaft unter Stickstoff, bei Raumtemperatur einwirken lässt, wobei sich das Metall löst.
Man kühlt dann die Lösung auf die innerhalb des oben angegebenen Intervalls liegenden Temperatur ab, fügt das Steroid zu und lässt in der Kälte reagieren.
Die Isomerisierung gegebenenfalls erhaltener 4-Oxo -1 9-nor-A-homo-steroid-2,5(10)-diene zu den entsprechenden 1,5(10)-Dienen erfolgt in üblicher Weise, zum Beispiel durch Reagierenlassen mit sauren oder basischen Mitteln, oder durch Adsorption an Kieselgel.
In den erhaltenen Verfahrensprodukten können, wenn ei wünscht, vorhandene veresterte Hydroxygruppen oder ketalisierte Oxogruppen nach an sich bekannten Verfahren z. B. hydrolytisch, gespalten werden.
Die erfindungsgemäss zu verwendenden Ausgangsstoffe sind bekannt oder können in an sich bekannter Weise erhalten werden, z. B. durch Veresterung der 19 -Hydroxy-3-oxo-A'4-diene oder durch Dehydrierung mit Dichlordicyan-benzochinon von 1 9-reaktionsfähig veresterten 3-Oxo-A4-enen.
Wie oben erwähnt lassen sich die neuen Endprodukte leicht in die bekannten, hochwirksamen 4-Oxo-l9-nor- -A-homo-steroid- 1(1 0),2,4triene überführen, zum Beispiel indem man die erhaltenen 4-Oxo-steroid-l ,5(10)- -diene in Gegenwart eines basischen Mittels, wie eines tertiären Amins, insbesondere einem cyclischen tertiären Amin z. B. Pyridin, Piperidin, Collidin oder Chinolin, bromiert und, wenn erwünscht, in erhaltenen Verbindungen verätherte veresterte Hydroxylgruppen und/oder ketalisierte Oxogruppen in Freiheit setzt.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen man einen Ausgangsstoff unter den Reaktionsbedingungen bildet oder in Form eines Salzes oder andere Derivate verwendet.
Die folgenden Beispiele erläutert die Erfindung ohne sie jedoch in irgendeiner Hinsicht einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
Eine Mischung von 150 ml absolutem Tetrahydrofuran, 450 mg Lithium und 9 g Diphenyl wird während 2 Stunden bei Zimmertemperatur im Stickstoffstrom gerührt. Darauf kühlt man die tiefbaue Lösung auf 700 ab und versetzt mit 6 g 3-Oxo-17-äthylendioxy-19-mesyl oxy-N androstadien unter Nachspülen mit wenig Tetrahydrofuran. Nach 30minutigem Rühren bei - 65 bis 700 wird mit 100 ml gesättigter Ammoniumchloridlösung versetzt und dreimal mit Benzol extrahiert. Der Rückstand der mit kochsalzhaltigem Wasser gewaschen, getrockneten und im Vakuum eingedampften organischen Lösungen wird in Toluollösung an 300 g Florisil adsortiert unter Nachwaschen mit 1,2 1 Toluol.
Zur Verschiebung der Doppelbindung von der 2- in die 1-Stellung belässt man das Rohprodukt während 2 Stunden auf der Säule. Mit Toluol-Essigester-(95:5)-Gemisch wird darauf das 4 -Oxo- 17 -äthylendioxy-A-homo-Al 5'l0) - 19-norandrostadien eluiert, von dem man nach Kristallisation aus Äther-Petroläther-Gemisch 2,04 g vom F. 115-116 erhält. Aus der Mutterlauge werden weitere 0,12 g derselben Verbindung gewonnen.
Aus den späteren mit Toluol-Essigester-(95:5)-Gemisch eluierten Fraktionen erhält man durch Kristallisation aus Äther-Petroläther-Gemisch 240 mg 3-Oxo-17 -äthylendioxy-5,19-cyclo-01-androsten. das nach erneutem Umlösen bei 1455-147 schmilzt. Mit reinem Essigester wird noch 200 mg Ausgangsmaterial eluiert.
Das als Ausgangsmaterial benötigte 3-Oxo-17-äthy lendioxy-l 9-mesyloxy-1,4-andrnstadien wird wie folgt hergestellt:
Eine Mischung von 56 g 3,17-Dioxo-19-mesyloxy-A4- -androsten, 42 g Dichlordicyan-benzochinon und 840 ml Dioxan wird während 18 Stunden im Stickstoffstrom unter Rückfluss gekocht. Nach Abkühlen wird abgenutscht und mit Methylenchlorid nachgewaschen. Den Rückstand des im Vakuum eingedampften Filtrates filtriert man in Methylenchloridlösung durch 420 g Aluminiumoxid (Aktivität II) unter Nachwaschen mit 4,2 1 Toluol-Essigester-(1: I)-Gemisch. Nach Eindampfen im Vakuum wird aus einem Methylenchlorid-Äther-Gemisch umkristallisiert. Dabei erhält man 42 g 3,17-Dioxo-l9 -mesyloxy-Al 4-androstadien vom F. 203,5-204,5 (Zersetzung).
Eine Lösung von 42,12 g des oben erhaltenen Diens und 910 mg p-Toluolsulfonsäure in 2,1 1 Benzol und 23 ml Äthylenglykol wird während 4 Stunden unter gutem Rühren und unter Verwendung eines Wasserabscheiders gekocht.
Nach Abkühlen und Zugabe von 63 ml Pyridin wird mit 420 ml gesättigter Natriumhydrogencarbonatlösung und mit Wasser gewaschen. Die wässrigen Lösungen extrahiert man noch einmal mit Toluol, worauf die organischen Lösungen getrocknet und im Vakuum eingedampft werden. Durch Kristallisation des Rückstandes aus Methylenchlorid erhält man 37,85 g 3-Oxo-17-äthylendioxy - 1 9-mesyloxy-¯l'-ansIrostadien vom F. 169-1700 (Zer- setzung).
Beispiel 2
1,5 g des in Beispiel 1 erhaltenen 4-Oxo-17-äthvlen- dioxy-A-homo-Al 3'l0)-l9-norandrostadiens erwärmt man im Stickstoffstrom mit einer Mischung von 20 ml Eisessig und 2 ml Wasser während 20 Minuten auf 600.
Darauf wird im Vakuum eingedampft und in Toluollösung durch 75 g Florisil unter Nachwaschen mit 2 1 eines Toluol-Essigester-(9:1 )-Gemisches filtriert. Nach Umlösen des Rückstandes des im Vakuum eingedampften Filtrates aus einem Äther-Petroläther-Gemisch werden 990 mg 4,17-Dioxo-A-homo-A )-l9-norandrosta- dien vom F. 108-111 erhalten.
Beispiel 3
Zu einer Lösung von 1,8 g Diphenyl im 30 ml Tetrahydrofuran gibt man 90 mg Lithium und lässt zwei Stunden im Stickstoffstrom bei Zimmertemperatur rühren.
Darauf wird die tiefblaue Lösung auf - 700 abgekühlt und mit 1 g 3-Oxo-17 -methyl-173-hydroxy-19-chlor- 4-androstadien versetzt unter Nachspülen mit wenig Tetrahydrofuran. Nach 45minutigem Rühren bei 65 bis - 700 versetzt man mit 20 ml gesättigter Ammonium chlorid-Lösung und extrahiert mehrmals mit Toluol. Der Rückstand der mit verdünnter Kochsalzlösung gewaschenen, getrockneten und im Vakuum eingedampften organischen Lösungen wird in Toluollösung an 60 g Fluorisil adsorbiert unter Nachwaschen mit insgesamt 240 ml Toluol. Nach 17 Stunden wird das 4-Oxo-17z-methyl-17{ss- hydroxy-A-homo-A"5 rl ) -1 9-norandrostadien mit Toluol Essigester-(97:3)-Gemisch eluiert.
Man erhält nach Kristallisation aus einem Äther-Petroläther-Gemisch 292 mg, die nach erneutem Umlösen bei 120-122 schmelzen.
Das als Ausgangsmaterial verwendete 3-Oxo-17z-me- thyl - 17,3 - hydroxy- 19 - chlor-A1,4-androstadien wird wie folgt hergestellt:
Zu einer Mischung von 19 g 3-Äthoxy-17-oxo-19 -chlor-033-androstadien und 200 ml Tetrahydrofuran gibt man unter Rühren und Eiskühlung 64 ml einer 3-m. Lösung von Methylmagnesiumchlorid in Tetrahydrofuran.
Nach Ilstündigem Rühren bei Zimmertemperatur wird unter Rühren auf 500 ml gesättigte Ammoniumchlorid Lösung und Eis geleert und mehrmals mit Toluol extrahiert. Die organischen Lösungen werden mit gesättigter Ammoniumchlorid-Lösung gewaschen, getrocknet und im Vakuum eingedampft. Da der so erhaltene Rückstand nach IR-Spektrum noch Ausgangsmaterial enthält, setzt man ihn erneut in der oben beschriebenen Weise mit 50 ml 3-m. Methylmagnesiumchlorid in Tetrahydrofuran um. Das auf diese Weise gewonnene rohe 3-Athoxy-17x- -methyl- 171B-hydroxy- 19-chlor-a35- androstadien wird ohne weitere Reinigung für die nächste Stufe verwendet.
Durch Kristallisation aus Alkohol kann ein reines Produkt vom F. 138-141,5 erhalten werden.
Zur Hydrolyse der Enoläthergruppierung wird das Rohprodukt mit 200 ml 90-proz. Essigsäure während 24 Stunden im Stickstoffstrom gerührt. Darauf giesst man auf 3 1 gesättigte Natriumhydrogencarbonat-Lösung und extrahiert mehrmals mit Methylenchlorid. Die organischen Lösungen werden mit verdünnter Kochsalzlösung gewaschen, getrocknet und im Vakuum eingedampft.
Durch Chromatographie des Rückstandes an 950 g Florisil und Kristallisation der mit Toluol-Essigester-(95:5) und (9:1)-Gemisch eluierten Fraktionen aus Äther erhält man 12,75 g 3-Oxo-17,v-methyl-17,B-hydroxy-19-chlor-A4- -androsten vom F. 106-1090.
12,75 g der oben erhaltenen 3-Oxoverbindung, 10 g Dichlor-dicyan-benzochinon und 190 ml Dioxan werden während 20 Stunden im Stickstoffstrom unter Rückfluss gekocht. Nach Abkühlen, Abnutschen und Nachwaschen mit Toluol und Methylenchlorid wird der Rückstand des im Vakuum eingedampften Filtrates an 390 g Aluminiumoxyd (Aktivität II) chromatographiert. Aus den mit Toluol-Essigester-(99:1) und (49:1)-Gemisch eluierten Fraktionen erhält man durch Kristallisation aus einem Methylenchlorid-Äther-Gemisch 10 g 3-Oxo-17,a+-methyl- -17,B-hydroxy-l9-chlor-AI4-androstadien vom F. 193 bis 1940.
Beispiel 4
Zu einer Lösung von 656 mg 4-Oxo-17-äthylendioxy -A-horno-A1 st10)-l9-nor-androstadien in 60 ml Pyridin gibt man unter Rühren 7,40 ml einer 1,16-n. Lösung von Brom in Tetrachlorkohlenstoff und lässt während 6 Stunden bei Zimmertemperatur stehen. Dann wird auf 110 ml n. Natronlauge geleert und dreimal mit Äther extrahiert.
Die ätherischen Lösungen werden mehrmals mit n. Natronlauge und Wasser gewaschen, getrocknet und unter Zugabe von Methylenchlorid im Vakuum eingedampft.
Bei der präparativen Dünnschichtchromatographie des Rückstandes an Kieselgel im System Toluol-Aceton-(4: 1) (8 Platten à 1 m) erhält man zwei Hauptzonen, die abgekrazt und mit Toluol-Methanol-(4: 1)-Gemisch eluiert werden. Die Eluate werden mit Wasser gewaschen, getrocknet und im Vakuum eingedampft. Aus der obern Zone gewinnt man nach Kristallisation aus Äther 200 mg 4-Oxo- 17-äthylendioxy - A-homo - ()) ,2,4a¯ 9-norandrostatrien, das nach weiterem Umlösen aus einem Methylenchlorid-Äther-Gemisch bei 176,5 bis 1780 schmilzt.
Die untere Zone liefert nach Kristallisation aus Äther 160 mg einer Verbindung gleicher Konstitution, wie die der oberen Zone. Sie ist jedoch im Ring A noch durch ein Bromatom substituiert. F. 116-122" unter Zersetzung.
Beispiel 5
Zu einer Lösung von 1,8 g Diphenyl in 30 ml Tetrahydrofuran gibt man unter Rühren im Stickstoffstrom 90 mg Lithium. Nach 21/2stündigem Rühren wird die tiefblaue Lösung auf 700 abgekühlt und innert 10 Minuten mit einer Lösung von 1 g 3-Oxo-17sc-äthinyl-17,B- -hydroxy- 1 9-chlor-1,4-androstadien in 10 ml Tetrahydrofuran unter Nachspülen mit 4 ml Tetrahydrofuran versetzt. Die Reaktionslösung lässt man während 45 Minuten bei - 75 bis - 700 rühren und versetzt dann mit 20 ml gesättigter Ammoniumchlorid-Lösung. Dann wird mehrmals mit Toluol extrahiert, mit gesättigter Ammoniumchlorid-Lösung gewaschen, getrocknet und im Vakuum eingedampft.
Der Rückstand wird in Toluollösung an 50 Florisil adsorbiert unter Nachwaschen mit insgesamt 240 ml Toluol. 20 Stunden später wird das 4-Oxo -17x-äthinyl-17f-hydroxy-A-homo A (10) 19-nor-andro- stadien mit einem Toluol-Essigester-(19:1)-Gemisch eluiert. Man erhält einen kristallien schwach gelbgefärbten Rückstand, der nach Umlösen aus einem Methylenchlorid-Äther-Gemisch bei 193-195,50 schmilzt.
Das als Ausgansmaterial benötigte 3-Oxo-17z-äthinyl- - 1 7-hydroxy- 1 9-chlor-1,4-androstadien wird wie folgt hergestellt:
In 1,6 1 absolutes Tetrahydrofuran wird während 30 Minuten Acetyien eingeleitet. Darauf lässt man unter weiterem Einleiten von Acetylen und Rühren innert 25 Minuten 2 1 einer 0,36 m Lösung von Methylenmagne- siumchlorid in Tetrahydrofuran einfliessen. Nach einer weiteren halben Stunde wird der Acetylenstrom abgestellt und durch einen schwachen Stickstoffstrom ersetzt.
Zur so bereiteten Lösung von Äthinylmagnesiumchlorid gibt man 20 g 3-Äthoxy-17-oxo-19-chlor-#3,5-androstadien unter Nachspülen mit 40 ml Tetrahydrofuran. Zwei Stunden später versetzt man unter Kühlung mit 300 ml gesättigter Ammoniumchlorid-Lösung. Die Aufarbeitung erfolgt durch Extrahieren mit Toluol und Waschen mit gesättigter Ammoniumchlorid-Lösung. Zur Spaltung des Enoläthers wird der Rückstand mit 200 ml 90SOiger Essigsäure während 15 Minuten im Stickstoffstrom auf 1000 erwärmt. Den Rückstand der im Vakuum eingedampften Reaktionslösung chromatographiert man an 1 kg Florisil.
Aus den mit Toluol-Essigester-(49: 1)-Ge- misch eluierten Fraktionen gewinnt man durch Kristallisation aus Methylenchlorid-Äther-Petroläther 13,72 g 3 -Oxo- 17z-äthinyl-17z-hydroxy-19-chlor-A4-androsten vom F. 161,5 - 1630.
Eine Mischung von 12 g des oben erhaltenen Äthinylcarbinols, 12 g Dichlor-dicyan-benzochinon und 240 ml Dioxan werden während 22 Stunden im Stickstoffstrom unter Rückfluss gekocht. Dann wird abgekühlt, abgenutscht und mit Toluol und Methylenchlorid nachgewaschen. Den Rückstand des im Vakuum eingedampften Filtrates chromatographiert man an 360 g Aluminiumoxid (Aktivität II). Durch Kristallisation der mit Toluol Essigester-(9: 1)-Gemisch eluierten Fraktionen aus einem Methylenchlorid-Äther-Petroläther-Gemisch erhält man 10,12 g 3-Oxo-17sc-äthinyl-17;,3-hydroxy-19-chlor-Az4-an- drostadien vom F. 230-2330.
PATENTANSPRUCH I
Verfahren zur Herstellung von 4-Oxo-19-nor-A-homosteroid-dienen, dadurch gekennzeichnet, dass man ein /\1,4-3-Oxo-steroid-dien, das in 10-Stellung eine mit einer starken organischen oder anorganischen Säure veresterte Hydroxymethylgruppe trägt mit einem Alkalimetall in einem aprotischen Lösungsmittel und in Gegenwart eines polycyclischen aromatischen Kohlenwasserstoffs umsetzt und erhaltene 4-Oxo- 1 9-nor-A-homo-steroid-2,5(10)-diene zu den entsprechenden 1,5(10)-Dienen isomerisiert.
UNTERANSPRÜCHE

The invention relates to a process for the preparation of new 4-oxo-19-nor-A-homosteroids, in particular 4-oxo-19-nor-A-homo-androstadienes, primarily those of the general formula

wherein R represents a free or ketalized oxo group, a free, esterified or etherified hydroxyl group together with a hydrogen atom or a lower aliphatic hydrocarbon.
The new compounds are valuable intermediates for the preparation of the known, highly effective 4-oxo-19-nor-A-homo-steroid-1 (10), 2,4a-trienes. But they also have valuable pharmacological properties themselves.
Particularly important are the compounds of the above formula in which R is a keto group or a free hydroxyl group or a hydroxyl group esterified with a lower aliphatic carboxylic acid together with a hydrogen atom or a lower alkyl, such as methyl or ethyl, a lower alkenyl, such as vinyl or allyl - Or methallyl, or a lower alkynyl, such as ethynyl or propynyl radical.
The new compounds are obtained when an A1,4-3-oxo-steroid diene, which carries a hydroxymethyl group esterified with a strong inorganic or organic acid in the 10-position, with an alkali metal in an aprotic solvent and in the presence converts a polycyclic aromatic hydrocarbon, the 4-oxo-19-nor-A-homo-steroid-2,5 (10) -dienes obtained isomerized to the corresponding 1,5 (10) -dienes.

As polycyclic aromatic hydrocarbons such. B. ethylene glycol dimethyl or diethyl ether, propylene glycol ether such as propylene glycol dimethyl or diethyl ether, acetals such as acetaldehyde dimethyl or diethylacetal, dioxane, tetrahydrofuran, tetrahydropyran, methyltetrahydrofurfuryl ether and similar compounds.
Tetrahydrofuran has proven particularly effective. Furthermore, for example, aliphatic or aromatic inert hydrocarbons, such as hexane, benzene, toluene, or hydroaromatic hydrocarbons, such as tetralin or decalin, can be used.
The reaction mentioned is preferably carried out in such a way that the alkali metal is allowed to act in an aprotic solvent on a polycyclic aromatic hydrocarbon, advantageously under nitrogen, at room temperature, the metal dissolving.

The isomerization of any 4-oxo-9-nor-A-homo-steroid-2,5 (10) -dienes to the corresponding 1,5 (10) -dienes takes place in a customary manner, for example by reacting with acidic or basic agents, or by adsorption on silica gel.
In the process products obtained can, if desired, esterified hydroxyl groups or ketalized oxo groups present according to known methods, for. B. hydrolytically split.
The starting materials to be used according to the invention are known or can be obtained in a manner known per se, e.g. B. by esterification of 19-hydroxy-3-oxo-A'4-dienes or by dehydrogenation with dichlorodicyano-benzoquinone of 19-reactively esterified 3-oxo-A4-enes.
As mentioned above, the new end products can easily be converted into the known, highly effective 4-oxo-19-nor- -A-homo-steroid- 1 (1 0), 2,4trienes, for example by converting the 4-oxo steroid-l, 5 (10) - -dienes in the presence of a basic agent such as a tertiary amine, especially a cyclic tertiary amine z. B. pyridine, piperidine, collidine or quinoline, brominated and, if desired, etherified esterified hydroxyl groups and / or ketalized oxo groups in the compounds obtained sets free.
The invention also relates to those embodiments of the process in which a starting material is formed under the reaction conditions or used in the form of a salt or other derivatives.
The following examples illustrate the invention without, however, restricting it in any respect. The temperatures are given in degrees Celsius.
Example I.
A mixture of 150 ml of absolute tetrahydrofuran, 450 mg of lithium and 9 g of diphenyl is stirred for 2 hours at room temperature in a stream of nitrogen. The deep solution is then cooled to 700 and 6 g of 3-oxo-17-ethylenedioxy-19-mesyl oxy-N androstadiene are added while rinsing with a little tetrahydrofuran. After stirring for 30 minutes at -65 to 700, 100 ml of saturated ammonium chloride solution are added and the mixture is extracted three times with benzene. The residue of the organic solutions washed with saline, dried and evaporated in vacuo is adsorted in toluene solution on 300 g of Florisil, washing with 1.2 l of toluene.
To shift the double bond from the 2 to the 1 position, the crude product is left on the column for 2 hours. The 4-oxo-17-ethylenedioxy-A-homo-Al 5'10) -19-norandrostadiene is then eluted with toluene-ethyl acetate (95: 5) mixture, from which, after crystallization from an ether-petroleum ether mixture 2 .04 g received from F. 115-116. A further 0.12 g of the same compound is obtained from the mother liquor.
240 mg of 3-oxo-17-ethylenedioxy-5,19-cyclo-01-androstene are obtained from the fractions later eluted with toluene-ethyl acetate (95: 5) mixture by crystallization from an ether-petroleum ether mixture. which melts after renewed dissolution at 1455-147. A further 200 mg of starting material is eluted with pure ethyl acetate.
The 3-oxo-17-äthy lendioxy-l 9-mesyloxy-1,4-andrnstadien required as starting material is prepared as follows:
A mixture of 56 g of 3,17-dioxo-19-mesyloxy-A4- -androsten, 42 g of dichlorodicyan-benzoquinone and 840 ml of dioxane is refluxed for 18 hours in a stream of nitrogen. After cooling, it is suction filtered and washed with methylene chloride. The residue of the filtrate evaporated in vacuo is filtered in methylene chloride solution through 420 g of aluminum oxide (activity II), washing with 4.2 l of toluene-ethyl acetate (1: I) mixture. After evaporation in vacuo, it is recrystallized from a methylene chloride-ether mixture. 42 g of 3,17-dioxo-19-mesyloxy-Al 4-androstadiene with a melting point of 203.5-204.5 (decomposition) are obtained.
A solution of 42.12 g of the diene obtained above and 910 mg of p-toluenesulfonic acid in 2.1 l of benzene and 23 ml of ethylene glycol is boiled for 4 hours with thorough stirring and using a water separator.
After cooling and adding 63 ml of pyridine, the mixture is washed with 420 ml of saturated sodium hydrogen carbonate solution and with water. The aqueous solutions are extracted once more with toluene, whereupon the organic solutions are dried and evaporated in vacuo. Crystallization of the residue from methylene chloride gives 37.85 g of 3-oxo-17-ethylenedioxy-19-mesyloxy-¯l'-anirostadiene with a melting point of 169-1700 (decomposition).
Example 2
1.5 g of the 4-oxo-17-äthvlen- dioxy-A-homo-Al 3'10) -19-norandrostadiene obtained in Example 1 are heated in a stream of nitrogen with a mixture of 20 ml of glacial acetic acid and 2 ml of water for 20 minutes to 600.
It is then evaporated in vacuo and filtered in toluene solution through 75 g of Florisil, washing with 2 l of a toluene-ethyl acetate (9: 1) mixture. After redissolving the residue of the filtrate evaporated in vacuo from an ether-petroleum ether mixture, 990 mg of 4,17-dioxo-A-homo-A) -19-norandrosta- diene of F. 108-111 are obtained.
Example 3
90 mg of lithium are added to a solution of 1.8 g of diphenyl in 30 ml of tetrahydrofuran and the mixture is stirred for two hours in a stream of nitrogen at room temperature.
The deep blue solution is then cooled to -700 and 1 g of 3-oxo-17-methyl-173-hydroxy-19-chloro-4-androstadiene is added while rinsing with a little tetrahydrofuran. After stirring at 65 to -700 for 45 minutes, 20 ml of saturated ammonium chloride solution are added and the mixture is extracted several times with toluene. The residue of the organic solutions washed with dilute sodium chloride solution, dried and evaporated in vacuo is adsorbed in toluene solution on 60 g of Fluorisil, washing with a total of 240 ml of toluene. After 17 hours the 4-oxo-17z-methyl-17 {ss- hydroxy-A-homo-A "5 ml) -1 9-norandrostadiene is eluted with toluene / ethyl acetate (97: 3) mixture.
After crystallization from an ether-petroleum ether mixture, 292 mg are obtained which, after being redissolved, melt at 120-122.
The 3-oxo-17z-methyl-17.3-hydroxy-19-chloro-A1,4-androstadiene used as starting material is produced as follows:
To a mixture of 19 g of 3-ethoxy-17-oxo-19-chloro-033-androstadiene and 200 ml of tetrahydrofuran are added 64 ml of a 3-m while stirring and cooling with ice. Solution of methyl magnesium chloride in tetrahydrofuran.
After stirring for 11 hours at room temperature, the mixture is poured onto 500 ml of saturated ammonium chloride solution and ice with stirring and extracted several times with toluene. The organic solutions are washed with saturated ammonium chloride solution, dried and evaporated in vacuo. Since the residue obtained in this way still contains starting material according to the IR spectrum, it is added again in the manner described above with 50 ml of 3-m. Methylmagnesium chloride in tetrahydrofuran. The crude 3-ethoxy-17x-methyl-171B-hydroxy-19-chloro-a35-androstadiene obtained in this way is used for the next stage without further purification.
A pure product with a melting point of 138-141.5 can be obtained by crystallization from alcohol.
To hydrolyze the enol ether group, the crude product with 200 ml of 90 percent. Acetic acid stirred in a stream of nitrogen for 24 hours. It is then poured onto 3 l of saturated sodium hydrogen carbonate solution and extracted several times with methylene chloride. The organic solutions are washed with dilute sodium chloride solution, dried and evaporated in vacuo.
Chromatography of the residue on 950 g of Florisil and crystallization of the fractions eluted from ether with a toluene / ethyl acetate (95: 5) and (9: 1) mixture gives 12.75 g of 3-oxo-17, v-methyl-17 , B-hydroxy-19-chloro-A4- -androsten from F. 106-1090.
12.75 g of the 3-oxo compound obtained above, 10 g of dichloro-dicyan-benzoquinone and 190 ml of dioxane are refluxed for 20 hours in a stream of nitrogen. After cooling, suction filtration and washing with toluene and methylene chloride, the residue of the filtrate evaporated in vacuo is chromatographed on 390 g of aluminum oxide (activity II). From the fractions eluted with toluene-ethyl acetate (99: 1) and (49: 1) mixture, 10 g of 3-oxo-17, a + -methyl- -17, B- are obtained by crystallization from a methylene chloride-ether mixture hydroxy-19-chloro-AI4-androstadien from F. 193 to 1940.
Example 4
To a solution of 656 mg of 4-oxo-17-ethylenedioxy -A-horno-A1 st10) -19-nor-androstadiene in 60 ml of pyridine, 7.40 ml of a 1.16-n is added with stirring. Solution of bromine in carbon tetrachloride and left to stand for 6 hours at room temperature. The mixture is then poured into 110 ml of sodium hydroxide solution and extracted three times with ether.
The ethereal solutions are washed several times with sodium hydroxide solution and water, dried and evaporated in vacuo with the addition of methylene chloride.
Preparative thin-layer chromatography of the residue on silica gel in the toluene-acetone (4: 1) system (8 plates of 1 m each) gives two main zones which are scratched off and eluted with a toluene-methanol (4: 1) mixture. The eluates are washed with water, dried and evaporated in vacuo. After crystallization from ether, 200 mg of 4-oxo-17-ethylenedioxy - A-homo - ()), 2,4ā 9-norandrostatriene are obtained from the upper zone, which after further dissolving from a methylene chloride-ether mixture at 176, 5 to 1780 melts.
After crystallization from ether, the lower zone yields 160 mg of a compound of the same constitution as that of the upper zone. However, it is still substituted by a bromine atom in ring A. F. 116-122 "with decomposition.
Example 5
90 mg of lithium are added to a solution of 1.8 g of diphenyl in 30 ml of tetrahydrofuran with stirring in a stream of nitrogen. After stirring for 21/2 hours, the deep blue solution is cooled to 700 and within 10 minutes with a solution of 1 g of 3-oxo-17sc-ethinyl-17, B- -hydroxy-19-chloro-1,4-androstadiene in 10 ml Tetrahydrofuran is mixed with 4 ml of tetrahydrofuran while rinsing. The reaction solution is allowed to stir for 45 minutes at -75 to -700 and then mixed with 20 ml of saturated ammonium chloride solution. It is then extracted several times with toluene, washed with saturated ammonium chloride solution, dried and evaporated in vacuo.
The residue is adsorbed on 50% Florisil in toluene solution, washing with a total of 240 ml of toluene. 20 hours later the 4-oxo-17x-ethinyl-17f-hydroxy-A-homo A (10) 19-nor-androstadiene is eluted with a toluene / ethyl acetate (19: 1) mixture. A crystalline, pale yellow-colored residue is obtained which, after dissolving from a methylene chloride-ether mixture, melts at 193-195.50.
The 3-oxo-17z-äthinyl- - 1 7-hydroxy- 1 9-chloro-1,4-androstadiene required as the starting material is produced as follows:
Acetyien is passed in 1.6 l of absolute tetrahydrofuran over 30 minutes. Then, while further passing in acetylene and stirring, 2 l of a 0.36 M solution of methylene magnesium chloride in tetrahydrofuran are allowed to flow in over the course of 25 minutes. After a further half an hour, the stream of acetylene is switched off and replaced by a weak stream of nitrogen.
20 g of 3-ethoxy-17-oxo-19-chloro- # 3,5-androstadiene are added to the solution of ethynyl magnesium chloride prepared in this way, while rinsing with 40 ml of tetrahydrofuran. Two hours later, 300 ml of saturated ammonium chloride solution are added while cooling. Working up is carried out by extracting with toluene and washing with saturated ammonium chloride solution. To cleave the enol ether, the residue is heated to 1000 with 200 ml of 90% acetic acid for 15 minutes in a stream of nitrogen. The residue of the reaction solution evaporated in vacuo is chromatographed on 1 kg of Florisil.
13.72 g of 3-oxo-17z-äthinyl-17z-hydroxy-19-chloro-A4- are obtained from the fractions eluted with toluene-ethyl acetate (49: 1) mixture by crystallization from methylene chloride-ether-petroleum ether androsten from F. 161.5 - 1630.
A mixture of 12 g of the ethynyl carbinol obtained above, 12 g of dichloro-dicyano-benzoquinone and 240 ml of dioxane are refluxed for 22 hours in a stream of nitrogen. It is then cooled, suction filtered and washed with toluene and methylene chloride. The residue of the filtrate evaporated in vacuo is chromatographed on 360 g of aluminum oxide (activity II). By crystallization of the fractions eluted with toluene / ethyl acetate (9: 1) mixture from a methylene chloride-ether-petroleum ether mixture, 10.12 g of 3-oxo-17sc-ethinyl-17;, 3-hydroxy-19-chloro- Az4-androstadien from F. 230-2330.
PATENT CLAIM I
