Verfahren zur Herstellung von substituierten Morpholinen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von substituierten :@rorpholinen.
Es sind bereits Verfahren zur Herstellung von Morpholinderivaten bekannt, wobei Di- äthanolamine mit wässrigen Säuren, z. B. mit 70 1/oiger Schwefelsäure, unter Erhitzen auf Temperaturen von 140-180 C behandelt wur- den, um den Morpholinringschluss zu erzielen.
Diese Verfahren sind für eine grosstech nische Anwendung ungeeignet, da sie Vorrich tungen erfordern, die aus besonders wider standsfähigem Material bestehen und ein Er hitzen unter erhöhtem Druck erlauben müssen. Ferner werden dabei erhebliche Wärmemen- gen verbraucht.
Schliesslich hat sich gezeigt, dass beim Arbeiten in: dem genannten Temperaturbereich empfindliche Substituenten in. den als Aus gangsstoffe verwendeten. Diäthanolaminen durch die Einwirkung der im Reaktion gemisch vorhandenen wässrgen Säure hydro- lvsiert werden können und dass die gewünschte Ringschlussreaktion biss zu einem gewissen Grade rückläufig ist.
Es ist daher ein Ziel der Erfindung, diese Nachteile auszuschalten und ein Verfahren zu ;schaffen, bei dem die gewünschten Morpholinderivate in einfacher und technisch durchführbarer Weise herge stellt werden können und in hoher Reinheit und guten Ausbeuten anfallen.
Es wurde nun gefunden, dass dies Ziel erreicht werden kann, wenn bei der Herstel lung von substituierten Morpliolinen aus sub- stituierten Diäthanolaminen besonders milde Reaktionsbedingungen für den Ringschluss angewandt wenden. Das erfindungsgemässe Verfahren erlaubt nun,
den Ringschluss-zu den Morpholinderiva- ten unter besonders milden Reaktionsbedin gungen, z. B. ohne zusätzliche Erwärmung oder nur unter leichter Erwärmung, durchzu führen. Im: Schweizer Patent Nr. 279777 ist ein Verfahren beschrieben, bei we'l'chem der Ringschluss schon beim Stehenlassen des ge lösten substituierten Diäthanolamins eintritt.
Ein derartig leichter Ringschlüss ist aber auf ganz bestimmte Einzelfälle beschränkt, wäh rend im allgemeinen energischere Bedingitn- gen notwendig sind.

Man kann den Ringsehluss, z. B. unter Ver- meidung von Erwärmung, mit konzentrierter Schwefelsäure bewirken. Bei Verwendung kon- zentrierter Schwefelsäure und des substituier- ten Diäthanolamins in Form der .freien Base als Ausgangsmaterial ist es wegen der Neu- tralisationswärme zweckmässig,
unter guter Kühlung zu arbeiten. Wenn man von einem Salz des substituierten Diäthanolamins aus geht, kann man dieses .ohne besondere Küh lung in, die konzentrierte Schwefelsäure ein tragen.
Nach einigen Stunden Stehenlassen hat sich das gewünschte Morpholinderivat gebil:.- det und man kann in: üblicher Weise aufarbei ten, z. B. indem man auf Eis giesst, alkalisch

macht, ausäthert und das Morpholin durch Kristallisation oder Destillation reinigt.
Auch mit verdünnten 'Säuren lässt sich die Umsetzung durchführen. Bei manchen Aus- gangsstoffen ist :es ausreichend, wenn bei Zim mertemperatur gearbeitet wird. Bei ,andern ist ein gelindes Erwärmen auf dem Wasserbad mit einer wüssrigen oder alkoholischen Säure erforderlich.
Die jeweiligen Reaktionsbedin gungen können leicht durch einfache Vorver- suche ermittelt werden. Als verdünnte Säuren, welche bei dem erfindungsgemässen Verfahren verwendet werden, können, seien zum Beispiel erwähnt: Schwefelsäure, Bromwasseratoffsäure und Salzsäure. Als besonders geeignet haben sich folgende Konzentrationen erwiesen:
0,1.- bis 30 o/age Schwefelsäure und 0,1-20 /o-ige Salzsäure und Bromwasserstoffsäure.
Zu dien bevorzugt verwendeten Ausgangs stoffen gehören u. a. folgende substituierte Diäthanolamine: l Phenyl-a-methyl-ss,ss'-dioxy- diäthyllamin, ss-Phenyl-a-äthyl-ss,ss''-dioxy-di- äthylamin, ss!-(p-Oxyphenyl)-ss,ss'-dioxy-diäthyl- amin, ss:-(m-Oxyphenyl)-ss,ss'-dioxy-diäthylamin sowie die entsprechenden Methoxyphenylver- bindungen.
Die als Ausgangsmaterial verwendeten sub stituierten Diäthanollamine können nach irgend einem geeigneten Verfahren hergestellt werden. Zweckmässig werden diese substituierten Di- äthanoIamine auf folgende Weise hergestellt Man setzt Benzyläthanolamin mit einem ge eigneten halogenierten Keton um und unter- wirft das Reaktionsprodukt der <RTI
ID="0002.0134"> katalytischen Hydrierung, wobei .der Benzylrest abgespalten und die Ketogruppe gleichzeitig zur CHOH- Cruppe reduziert wird.. Diese Reaktionen kön nen durch folgendes Schema veranschaulieht werden Die erfindungsgemäss hergestellten Mor- pho!line sind wertvolle Pharmazeutica bzw. Zwischenprodukte für die Herstellung von Pharmazeutika.
Die folgenden Beispiele sollen die Erfin dung näher erläutern.
Beispiel <I>1</I> 2-Phenyl morphol'in 10 g ss - Phenyl- ss,ss' - dioxy- diäthyltamir_- hydrothlorid werden in 15 cm3 konzentrierter Schwefelsäure gelöst und über Nascht bei Zim- inertemperätur siehengelassen. Dann wird auf Eis gegossen,
mit Natronlauge alkalisch ge macht und; die Base in Äther aufgenommen. :Kaeh dem Trocknen wird der Ätherrückstand fraktioniert destilliert. Das 2-Phenyl-morpbo- lin geht bei 145 C (14 mm) über. Ausbeute 83 %. Das Chlorhydrat wird mit alkoholischer Salzsäure und Aceton erhalten. F. = 151 C.
Beispiel <I>2</I> 2-Pltenyl-3 methyl-morpholin Nach dem gleichen Verfahren. wie in Bei spiel 1 erhält man ausgehend von ,- Phenyl-a- m:ethyldioxy -diäthylamin-hydrochlorid, das 2-Phenyl 3-methyl-morpholin in 92 o/oiger Ausbeute. Die Base ist eine Flüssigkeit, Kpl2 = 138 C.
Das Chlorhydrat kristallisiert aus alkoholischer Salzsäure und Aceton. F. =1820 C.
Beispiel <I>3</I> 2,3-Diphenyll-morpholin 10 g a,ss-Diphenyl-ss,ss'-:dzoxy-diäthylamin- hydrochlorid werden in 15 cms konzentrierter Sehwefelsäure gelöst und über Nacht bei Zim mertemperatur stehengelassen. Die dickflüssige Lösung wird auf Eis gegossen und mit kon zentriertem Ammoniak alkalisch gemacht, wobei sich .das 2,
3-Diphenyl-morpholin kristal lin abscheidet, F = 91 C. Die weitere Reini- gung-erfolgt über das Chlorhydrat, das beim Versetzen mit alkoholischer Salzsäure und Aceton kristallisiert. F. = 265 C, Ausbeute 671/o.
<I>Beispiel 4</I> 2, 6-D@iphenyl-morpholin 10 g ss,ss'-Diphenyl ss,ss'-doxy-d'iäthylamin- hy drochlorid lässt man in 15 ems konzentrier- ter Schwefelsäure gelöst über Nacht bei Zim- mertemperatur stehen. Nach dem Giessen ;auf Eis wird mit Ammoniak alkalisch gemacht.
Die Base fällt als zäher klebrige Masse aus, die in Chloroform aufgenommen wird. Der Chloro- formrückstand gibt, mit Äther angerieben, die kristalline Base vom F. = 108 C. Dias<B>Chlor-</B> hydrat wird mit alkoholischer Salzsäure und Aceton erhalten. F: = .etwa 200 C (Zers.). Ausbeute 65 0/0.
Beispiel <I>5</I> 2-Phenyl;-3-äthyl-morpholin 1-Phenyl 1-oxo 2-brom-butan wird mit Ben- zyläthanolamin umgesetzt und das erhaltene Aminoketon in:
Methanol mit Palladiumkohle hydriert. Das entstandene 1-Phenyl-l-oxy-2- äthanolamino-buttan wurde als Hydrochlorid vom F. = 147 C (Zerr.) erhalten. Die Base schmilzt nach dem Umkristallisieren aus Ben zol bei 870 C, das in , Alkohol leicht lösliche, durch Zusatz von. Äther und Oxalsäure erhal tene Bioxalat bei 900 C.
1 g der Base wurde in 1,5 cm3 konzentrier ter Schwefelsäure gelöst, wobei die Tempera tur infolge der Neutralisationswärme auf etwa 40 C anstieg. (Bei grösseren Ansätzen russ gekühlt werden).
Nach dem Stehen über Nacht wurde auf Eis gegossen, alkalisch gemacht, aus- geäthert und die getrocknete Ätherlösung auf ein kleines Volumen eingeengt und mit alkoho lischer Salzsäure angesäuert. Das kristallisierte Hydrochlorid des 2-Phenyl 3-äthyl-morpholins schmilzt bei 181 C. Ausbeute 79 0/0.
<I>Beispiel 6</I> 2-(p-Tolyl)-morpholin 67 g, p-Methyl-acetophenon in 500 cin3 Benzol werden langsam mit 25 cm3 Brom ver setzt und das Benzol. nach kurzem Stehen<B>ab-</B> destilliert.
Der Rückstand wird mit 150 g Benzyläthanolamin in 150 em3 Benzol ver setzt, wobei man dafür Sorge trägt, dass die Temperatur 550 C nicht übersteigt. Es kristas liniert Benzyläthanolamin-hydrobromid aus, das am nächsten: Tag abgesaugt und mit Benzol gewaschen wird.
Die Benzollösung. wird 2- biss 3mal mit Wasser ausgeschüttelt, über Natrium sulfat getrocknet -Lind das Benzol abdestilliert

Beispiel <I>10</I> 2,6-Diphenyl-3,5-dimethyl-morpholin 2 Mol 1-Phenyl-l-oxo-2-broin-prop@an liefern bei der Umsetzling mit. 1 Mol Benzylamin N- Benzyl-ss,ss'-diphenyl-ss,ss'-dioxo- a,aY-dimethyl- diäthylamin, das bei der katalytischen Reduk tion ss,ss'-Diphenyl-ss,ss'-dioxy-a,a'-dimethyl-di- äthyl.amin, ergibt.
1 g dieses Produktes wird in 1,5 cm3 kon- zentrierter Schwefelsäure gedöst und nach dem Stehen über Nacht bei Zimmertemperatur in üblicher Weise aufgearbeitet. Main erhält das 2,6 - Diphenyl - 3,5 - dimethyl - morpholin vom F. =147 C.

Processes for the preparation of morpholine derivatives are already known, with diethanolamines with aqueous acids, e.g. B. with 70 1 / o sulfuric acid, with heating to temperatures of 140-180 C, in order to achieve the morpholine ring closure.

It is therefore an aim of the invention to eliminate these disadvantages and to create a process in which the desired morpholine derivatives can be produced in a simple and technically feasible manner and are obtained in high purity and good yields.
It has now been found that this goal can be achieved if particularly mild reaction conditions are used for the ring closure in the production of substituted morphiolins from substituted diethanolamines. The inventive method now allows
the ring closure to the morpholine derivatives under particularly mild reaction conditions, z. B. without additional heating or only with slight heating to perform durchzu. In: Swiss Patent No. 279777, a process is described in which the ring closure occurs when the dissolved substituted diethanolamine is left to stand.
Such an easy ring lock is limited to very specific individual cases, while more energetic conditions are generally necessary.
It has now surprisingly been found that substituted diet handlamines of the general formula

The reaction can also be carried out with dilute acids. With some starting materials: it is sufficient to work at room temperature. Others require gentle warming on a water bath with an aqueous or alcoholic acid.
The respective reaction conditions can easily be determined by simple preliminary tests. As dilute acids which can be used in the process according to the invention, there may be mentioned, for example: sulfuric acid, hydrobromic acid and hydrochloric acid. The following concentrations have proven to be particularly suitable:
0.1 to 30% sulfuric acid and 0.1-20% hydrochloric acid and hydrobromic acid.
The preferred starting materials used include u. a. the following substituted diethanolamines: l phenyl-a-methyl-ss, ss'-dioxy-diethyllamine, ss-phenyl-a-ethyl-ss, ss '' - dioxy-diethylamine, ss! - (p-oxyphenyl) -ss , ss'-dioxy-diethylamine, ss :-( m-oxyphenyl) -ss, ss'-dioxy-diethylamine and the corresponding methoxyphenyl compounds.
The substituted diethanolamines used as the starting material can be prepared by any suitable method. These substituted diethanolamines are expediently prepared in the following manner: Benzylethanolamine is reacted with a suitable halogenated ketone and the reaction product is subjected to <RTI
ID = "0002.0134"> catalytic hydrogenation, where the benzyl radical is split off and the keto group is reduced to the CHOH group at the same time. These reactions can be illustrated by the following scheme. The morpholines produced according to the invention are valuable pharmaceuticals or intermediates for the manufacture of pharmaceuticals.
The following examples are intended to explain the invention in more detail.
Example <I> 1 </I> 2-Phenyl morphol'in 10 g ss - phenyl ss, ss' - dioxydiethyltamir_hydrothlorid are dissolved in 15 cm3 of concentrated sulfuric acid and allowed to sit overnight at room temperature. Then it is poured onto ice
made alkaline with caustic soda and; the base taken up in ether. : After drying, the ether residue is fractionally distilled. The 2-phenyl-morpholine passes over at 145 C (14 mm). Yield 83%. The chlorine hydrate is obtained with alcoholic hydrochloric acid and acetone. F. = 151 C.
Example <I> 2 </I> 2-Pltenyl-3 methyl-morpholine Using the same procedure. As in Example 1, starting from - phenyl-a-m: ethyldioxy-diethylamine hydrochloride, 2-phenyl-3-methyl-morpholine is obtained in 92% yield. The base is a liquid, Kpl2 = 138 C.
The chlorine hydrate crystallizes from alcoholic hydrochloric acid and acetone. F. = 1820 C.
Example <I> 3 </I> 2,3-Diphenyll-morpholine 10 g of a, ss-diphenyl-ss, ss' -: dzoxy-diethylamine hydrochloride are dissolved in 15 cms of concentrated sulfuric acid and left to stand overnight at room temperature. The viscous solution is poured onto ice and made alkaline with concentrated ammonia, whereby the 2,
3-Diphenyl-morpholine precipitates crystalline, F = 91 C. Further purification takes place via the chlorohydrate, which crystallizes when mixed with alcoholic hydrochloric acid and acetone. M.p. = 265 C, yield 671 / o.
